숙성에 의한 흑밤의 영양성분 분석과 항산화 활성 (Analysis of Nutritional Composition and Antioxidant Activity of Black Chestnut by Aging)

  • Black chestnut (BC) was obtained through aging of fresh chestnut (FC) at 80℃ for 15 days. Proximate and mineral compositions along with colors of FC and BC were evaluated. With aging, moisture contents decreased by 50%, whereas sugar contents, carbohydrate contents, and calories increased. Contents of minerals (Fe, P, Ca, Na, Mg, K) were significantly higher in FC than in BC, showing an order of Mg < Ca < P < K in both FC and BC. Using a Hunter color system, it was found that lightness (L), redness (a), and yellowness (b) values of FC were higher than those of BC. Antioxidant and cytotoxic activities of hot water and ethanol (50, 80, 100%) extracts prepared from FC and BC were evaluated. Extraction yields were lower with FC than with BC. Among water and ethanol extracts, water extract showed the highest DPPH radical scavenging activity for both FC and BC. IC50 values for ABTS+ radical scavenging activities increased after aging. Cytotoxicities of FC and BC extracts were similar to each other. They were different against various cell lines (3T3, HeLa, and Sarcoma-180). These results suggest that BC could be used as a new processed food using chestnut.

The Activity of Xanthine Oxidase (Type O) in Some Partial Portions of Rat Skin

  • To evaluate the physiological significance of xanthine oxidase (XO) in rat skin, the activity of XO (type O) in skin was compared with that of small intestine or liver. Concomitantly, XO activities in some partial portions (scalp, leg, dorsal and ventral part) of skin were determined and then compared with each partial portion. XO activity of skin was lower than that of small intestine and rather higher than that of liver. Furthermore, the activity of XO in skin, after clipping of hairs and then in 5 days, was more increased than that of rat which was clipped before having been sacrificed. As for activities of free radical scavenging system (GPx, GST, SOD), skin is lower than liver and small intestine. Although it is hewn that the oxygen free radical generated by XO system lead to injurious effect on the cell, the XO activity of ventral part which is to be less exposed to xenobiotics and biological agents was the lowest among those of ventral, dorsal, leg and scalp parts in skin. In conclusion, it may be hypothesized that XO system in skin act on defence mechanism.

육체운동에 의해 유발되는 화학물질에 대한 반응성의 변화 (Alteration in Response to Chemicals Induced by Physical Exercise)

  • Acute or repeated physical exercise affects a large number of physiological parameters including hemodynamics, respiration, pH, temperature, gastrointestinal function and biotransformation, which determine the pharmacokinetics of drugs and chemicals. The rate and the amount of a chemical reaching the active site are altered by physical exercise, which results in significant changes in pharmacolosical/toxicological activity of the chemical. This aspect of physical exercise has vast implication in therapeutics and in safety evaluation, particularly for chemicals that have a low margin of safety. However there appears to be a wide inter- and intraindividual variation in the effects of physical exercise depend-ing on the duration, intensity and type of exercise, and also on the properties of each chemical. It is suggested that more studies need to be done to determine which factor(s) plays a major role in the disposition of chemicals in human/animals performing physical exercise. Certain chemicals induce severe toxicity due to metabolic conversion to reactive intermediate metabolites. it is suggested that repeated exercise may enhance the free radical scavenging system by increasing the activity of antioxidant enzymes. This area of research remain to be explored to elucidate the interaction of exercise and chemical on the antioxidant system.

유색감자 추출물의 항산화 및 항고혈압 활성 (Antioxidant and Inhibition on Angiotensin Converting Enzyme Activity of Colored Potato Extracts)

  • 괴경 내부에 적색과 보라색 안토시아닌 색소가 풍부하게 함유되어 있고, 색상의 기호도 및 건강 기능성으로 인해 소비자로부터 기호도가 증대된 유색감자의 생리적 활성을 검토하기 위해 유색감자의 대조구로 괴경 내부가 백색인 수미 품종을 사용하고, 유색감자로서 괴경 내부가 적색인 대관 1-102호, 괴경 내부가 보라색인 자심품종과 대관 1-104호로 구분하여 항산화, 활성산소 소거능 및 항고혈압 활성을 비교, 검토한 결과는 아래와 같다. 1. 유색감자 추출물의 항산화 활성 검정결과 추출물의 농도가 $10{\mu}g/mL$ 수준에서는 유색감자별 차이를 나타내지 않았으나, $50{\mu}g/mL$을 처리한 경우 시료 간 차이를 뚜렷하게 나타내었으며, 유색감자 중 괴경내부의 색상이 적색인 대관 1-102호가 34%의 높은 저해율을 나타내어 30%의 저해율을 나타낸 활성평가 대조구인 $\alpha-tocopherol$보다 높은 항산화 활성을 나타내었다. 2. 유색감자 추출물의 자유 라디칼 소거 활성에서는 괴경 내부가 보라색인 대관 1-104호가 50%의 소거능을 나타내는 농도가 $9.83{\mu}g/mL$로 가장 높은 라디칼 소거활성을 나타내었으며, 이 결과 또한 대조시약인 $\alpha-tocopherol$이나 BHT보다 높은 소거 활성을 나타내었다. 3. 유색감자 추출물의 xanthine oxidase 활성 억제효과를 검토한 결과 유색감자 간에 큰 차이를 나타내어 대관 1-102호와 1-104호의 경우 xanthine oxidase 활성을 50% 저해하는 데 필요한 농도는 약 $26{\mu}g/mL$ 수준으로 가장 높게 조사되었으며, 다음으로 수미($46{\mu}g/mL$), 자심($49{\mu}g/mL$)의 순으로 조사되었다. 4. 유색감자 추출물의 ACE 저해 활성을 검토한 결과 항고혈압 대조구 활성물질인 captopril의 수준에는 미치지 못하나 유색감자 간에는 명확한 차이를 나타내었다. 괴경내부색상이 적색인 대관 1-102호가 $117.7{\mu}g/mL$으로 가장 높은 ACE 저해 활성을 나타내었으며, 내부색상이 보라색인 대관 1-104호와 자심은 $130{\mu}g/mL$로 유사한 양상을 나타내었으며, 괴경내부 색상이 백색을 나타내는 수미품종은 $192{\mu}g/mL$으로 가장 낮은 활성을 나타내었다. 5. 이상의 결과를 종합해 볼 때 괴경 내부에 적색 혹은 보라색 안토시아닌을 다량 함유한 유색감자는 기존 백색을 가진 일반감자에 비해 항산화, 자유레디칼 소거활성 및 항고혈압 활성이 높고, 시각적 식미감을 증대시키므로 유색감자는 기능성이 증대된 식용감자로서의 이용가치가 충분한 것으로 판단된다.

가열처리에 의한 심황색소의 화학적 특성, 산화방지 활성 및 세포독성 변화 (Changes in chemical properties, antioxidant activities, and cytotoxicity of turmeric pigments by thermal process)

  • 본 연구에서는 식품의 가공이나 조리 시 빈번히 적용되는 가열처리에 의한 심황색소의 화학안정성, 산화방지활성 및 세포독성의 변화를 조사하였다. $95^{\circ}C$에서 각 시간별로 가열처리한 심황색소는 가열시간이 증가할수록 발색도가 감소하였으며, 형광도는 초기 가열 시에 증가하다 감소하는 양상을 보였다. HPLC 분석 결과 3종의 쿠쿠미노이드 중 쿠쿠민이 가열처리에 가장 민감하였으며 BMC가 가장 안정하였다. 가열 처리 후의 심황색소에 의해 ABTS 라디칼, AAPH peroxyl 라디칼 및 아질산염 소거활성이 증가하였으나, DPPH 라디칼 소거활성에는 변화가 없었다. 가열처리 시간의 증가에 따라 심황색소로 부터의 $H_2O_2$ 생성능이 증가한 반면, 정상장관계 세포 INT 407 및 대장암 세포 HCT 116를 대상으로 한 세포독성평가에서는 가열처리 후 심황색소의 활성이 유의적으로 약화되었다. 본 연구결과는 다양한 가공식품에 첨가되는 심황색소의 화학안정성 및 생리활성이 가열처리에 의해 크게 영향을 받으며, 생리활성 증진을 목적으로 첨가되는 심황색소에 이러한 가열처리 효과가 고려되어야 함을 시사한다.

산지별 유자의 생리활성 (Biological Activities of Yuza Grown in Different Areas)

  • 거제, 고성, 고흥 및 남해산 유자의 생리활성 규명에 관한 연구의 일환으로 유자를 과육과 과피로 분리하여 주요 생리활성 성분을 분석하고 과피 물추출물의 전자공여능, 아질산염소거능 및 tyrosinase 저해활성을 비교하였다. 비타민 C 함량은 과육에서 6.34${\Pm}$0.12-10.74${\Pm}$1.25 mg/100 g의 범위였으며, 과피에서는 과육에 비하여 2.17-3.57배 정도 더 높은 함량이었다. 카로티노이드 함량도 비타민 C와 동일한 경향이었으며 고성산과 남해 재래종의 과피에서 6 mg/100 g 이상으로 타 시료에 비해 유의적으로 높은 함량이었다. 총 페놀 화합물의 함량은 남해 개량종에서 가장 높아 과육과 과피에서 각각 11.11${\Pm}$0.10 mg/100 g 및 31.76${\Pm}$0.30 mg/100 g이었다. Hesperidin과 naringin의 함량도 과육보다는 과피에서 4.81-7.80, 3.07-4.62배 정도 더 높은 함량이었으며, 과피에서 hesperidin은 45.30${\Pm}$1.18-85.76${\Pm}$3.72 mg/100 g, naringin은 28.69${\Pm}$0.85-44.96${\Pm}$1.75 mg/100 g의 범위였다. 과피 물추출물의 전자공여능은 5 mg/mL 이상 첨가 시 활성은 52.78${\Pm}$1.13-66.88${\Pm}$0.53%의 범위였으며 남해산 시료에서 유의적으로 활성이 높았다. 아질산염 소거활성은 0.5 mg/mL 농도에서는 10% 미만으로 낮았으나 시료의 첨가농도가 증가할수록 활성이 높아져 10 mg/mL 농도에서는 24.95${\Pm}$1.82-29.72${\Pm}$1.84%의 범위였다. Tyrosinase 저해활성은 2.5 mg/mL의 농도에서는 남해산 시료에서만 5.06${\Pm}$1.27%의 낮은 활성이 확인되었으나, 10 mg/mL 농도에서는 32.76${\Pm}$1.43-69.21${\Pm}$1.91%로 활성이 증가하였다.

Cytoprotective Constituents of Alpinia katsumadai Seeds Against Glutamate-Induced Oxidative Injury in HT22 Cells

  • Glutamate-induced oxidative injury contributes to neuronal degeneration in many central nervous system (CNS) diseases, such as epilepsy and ischemia. Bioassay-guided fractionation of the MeOH extract of the seeds of Alpinia katsumadai Hayata (Zingiberaceae) furnished three phenolic compounds, alpinetin (1), pinocembrin (2), and (+)-catechin (3). Compounds 2 and 3 showed the potent neuroprotective effects on glutamate-induced neurotoxicity and reactive oxygen species (ROS) generation in the mouse hippocampal HT22 cells. In addition, Compounds 2 and 3 showed significant DPPH free radical scavenging effect. These results suggest that compounds 2 and 3 could be the effective candidates for the treatment of ROS-related neurological diseases.

Biological Activities and Partial Characterization of Beauveria bassiana Mycelium

  • Some biological activities of Beauveria bassiana were studied to elucidate pharmacological function of B. bassiana-infected larva of the silkworm. The mycelium consisted mainly of carbohydrate (65.8%), followed by protein (15.9%) and fat (8.3%). Glucose (68.8%), mannose (7.1%), and galactose (6.1%) were major components in carbohydrates. Ten amino acids including glutamine, threonine, valine, aspartic acid, alanine, leucine, serine, glycine, arginine, and isoleucine were found in protein as major amino acids. Various extracts were prepared from the freeze-dried mycelium of B. bassiana by systemic extraction and their biological activities were investigated. Among tested fractions, the hot-water extract (HW) contributed significantly to the anti-coagulant activity, anti-complementary activity, and stimulation of intestinal immune system. The methanol extract (ME) increased acetylcholinesterase (AChE) inhibition activity and reactive oxygen species (ROS) scavenging activity.

Antioxidant Flavonoids from the Twigs of Stewartia koreana

  • In the course of screening for antioxidant compounds by measuring the radical scavenging effect on 1,1-diphenyl- 2-picrylhydrazyl (DPPH), a total extract of the twigs of Stewartia koreana (Theaceae) was found to show potent antioxidant activity. Subsequent activity-guided fractionation of the methanolic extract led to the isolation of six phenolic compounds, ampelopsin (1), catechin (2), proanthocyanidin-A2 (3), fraxin (4), (2R, 3R)-taxifolin-3-${\beta}$-D-glucopyranoside (5), and (2S, 3S)-taxifolin-3-${\beta}$-D-glucopyranoside (6), as active principles. Their structures were elucidated by spectroscopic studies. Compounds 1-6 were isolated for the first time from this plant. Among them, three compounds 1-3 showed the significant antioxidative effects on DPPH, and riboflavin originated superoxide quenching activity. In riboflavin-nitrobluetetrazolium (NBT)-light system, compound 1 showed better superoxide quenching activity than vitamin C.

Characterization of Biocompatible Lipid-Based Vesicles Contained with Medicinal Herb Extracts

  • In order to increase the medicinal herbs efficiency of drug delivery, vesicles contained with medicinal herbs were prepared by phosphatidylcholines and surface active agent. Vesicles loaded with medicinal herbs were characterized by UV-spectroscopy, Zetasizer. The antioxidant activity of vesicles was measured by DPPH assay and ABTS radical scavenging assays. Also, an analysis was conducted to determine the effects of anti-inflammatory of vesicles contained medicinal herbs. In addition, the whitening effects of vesicles contained medicinal herbs extract were studied via tyrosinase inhibition assay. The results of vesicles were as follows. Vesicles appeared an average diameter of approximatively 164-599 nm. All studied vesicles contained with medicinal herbs showed antioxidant, anti-inflammatory and whitening effects in a dose-dependent manner. Therefore, this experiment achieves its purpose of synthesizing of vesicles. In conclusion, we recommended that the vesicles loaded with medicinal herbs have ability for anti-aging materials. Specifically, it will apply to cosmetic ingredients.
