Free Radical Scavenging Activity of the Seed of Phaseolus calcaratus Roxburgh

  • The seed of Phaseolus calcaratus Roxburgh (PHCR) is traditionally used for anti-pyretic and antiinflammatory effects. Although these effects are believed to be related to its antioxidant potential, little information is available for the mechanisms by which PHCR seed might scavenge free radicals or otherwise act as an antioxidant. In the present study, we purified some fractions from the ethanol extract of PHCR seed and evaluated each fraction's ability to scavenge free radicals generated by cell-free systems. We also identified active compound that is putatively responsible for free radical scavenging by analyzing NMR spectra. PHCR samples exhibited a concentration-dependent radical scavenging activity against hydroxyl radicals, superoxide anions, and DPPH radicals. Of the samples tested, a methanol-eluted sub-fraction from the PHCR extract, named $FF_4$, scavenged these radicals more effectively than the other fractions. We identified catechin-7-O-$\beta$-Dglucopyranoside as the active compound responsible for free radical scavenging potential of $FF_4$.

  • 거대배아미 에탄올 추출물의 항산화활성을 지질과산화 억제활성 및 활성산소종의 소거활성을 중심으로 일반미와 대조하여 검토하였다. 연구 결과, 발아처리는 전반적으로 환원력 및 지질과산화 억제활성, superoxide radical 및 hydroxyl radical 소거활성을 상승시켰으며, 발아처리에 의한 항산화 활성의 증가현상이 화청거대배아미에서 가장 뚜렷하였다. Superoxide radical 소거작용에 있어서는 남풍거대배아미가 전체적인 소거활성은 높았으나, 발아처리에 의한 활성의 상승률은 역시 화청거대배아미가 가장 높았고, 그 작용기작은 시료에 의한 직접적인 radical 소거에 있는 것으로 나타났다. 생체 독성이 가장 큰 hydroxyl radical 소거활성을 조사한 결과, 화청거대배아미가 전체적인 소거활성 뿐 아니라, 발아처리에 의한 소거활성의 증가율도 가장 높았으며, 그 작용기작도 $Fe^{2+}$의 포족이 아닌 직접적인 라디칼 소거임이 밝혀졌다. 이와 같은 in vitro에서 관찰된 시료의 ROS 소거활성은 TPA하여 유도된 HL-60 세포의 ROS 생산을 억제하는데 유효하게 작용하였다.

  • Primary neuronal culture was studied for observing effect of ginsenoside Rgl (Rgl) on serum-free medium induced apoptosis. Results showed that Rgl at concentration of 1 umol$.$ L-1 and 10 umol$.$L-1 could inhibit apoptosis, decrease intracellular calcium concentration in cultured cortical neurons, enhance SOD activity in both aged rat cortex and cultured cortical neurons, scavenge cytotoxic oxygen free radicals, decrease NO content and NOS activity in aged rat cortex and cultured cortical neurons, increase bel-2 gene expression in rat brain. These results provided new data for elucidating the anti-aging effect of Rgi. Rgl is considered to be a useful drug for treatment of Alzheimer's disease and brain aging.

  • Chlorophytum borivilianum Baker (Antharicaceae) commonly referred as 'Safed Musli' has been widely used in the Indian traditional systems of medicine to treat various diseases like rheumatism apart from having immunomodulating property and is used as general tonic. It is also known as 'Ayurvedic viagra' for its aphordisiac properties. C. borivilianum was screened for the first time to determine its antioxidant activity, isolation of the sapogenins and standardization of the isolated sapogenin fraction using HPTLC. Potent antioxidant activity of ethanolic extract was found by their ability to scavenge DPPH (84.51%), hydroxyl radical (48.95 %), ferryl bi-pyridyl complex (84.53%) along with the inhibition of lipid peroxidation (67.17%) at $100\;{\mu}g/ml$ concentration. The ethanolic extract also exhibited significant inhibition of superoxide anion radical generated by photochemiluminescence. Thus, the potent antioxidant activity validates the innumerable therapeutic claims of the plant in the traditional system especially its use as a Rasayana drug.

Evaluation of the Antioxidant Potential of Korean Indigenous Plant Extracts by Free Radical Scavenging Activity

  • Since reactive oxygen radicals play an important role in carcinogenesis and other human diseases including neurodegenerative states, antioxidants present in natural products have received considerable attention for alleviation of these disease states. Therefore, in order io identify antioxidants in plant extracts, fifty-seven methanolic extracts derided from indigenous Korean plants were primarily assessed for potential to scavenge stable 1,1-diphenyl-2-picrylhydrazyl (DPPH) free radicals. As a result, nine plant extracts were found to exhibit the DPPH free radical scavenging activity in the criteria of $IC_{50}<40\;{\mu}g/ml$. In particular, the extracts of Melioma oldhami $(IC_{50}=0.1\;{\mu}g/ml)$, Myrica rubra $(IC_{50}=16.2\;{\mu}g/ml)$, Sympolocos paniculata $(IC_{50}=23.0\;{\mu}g/ml)$, Carpinus laxiflora $(IC_{50}=25.1\;{\mu}g/ml)$, and Cleyera japonica $(IC_{50}=26.2\;{\mu}g/ml)$ showed a potent radical scavenging activity. Further study for the identification of active compounds from these lead extracts might be warranted.

플레인 요구르트와 소이 요구르트의 미생물학적, 물리화학적 및 항산화 활성 (Microbiological, Physicochemical, and Antioxidant Properties of Plain Yogurt and Soy Yogurt)

  • 양배추 피클로부터 분리된 유산균으로 발효하여 제조한 플레인 요구르트와 소이 요구르트를 저장하는 동안 미생물학적 및 물리화학적 변화와 항산화 특성을 조사하였다. 분리 균주는 당분해능과 유전자 염기서열 분석을 통해 L. casei PC05와 L. acidophilus PC16으로 동정되었다. Lactobacillus acidophilus PC16 균주로 발효시킨 요구르트의 적정산도, 점도 및 균수는 L. casei PC05 균주에 의해 제조된 요구르트에 비해 높게 나타났으며, 특히 플레인 요구르트보다 소이 요구르트에서 세균수와 물리화학적 요인의 측정값이 유의하게 높은 것으로 확인되었다. 한편, L. acidophilus PC16 균주로 발효시킨 요구르트는 DPPH 라디칼 소거능과 철이온 킬레이팅 활성이 높았으나, L. casei PC05 균주의 요구르트는 superoxide anion 제거능과 SOD 활성이 높은 것으로 나타났다. 대부분의 항산화능은 플레인 요구르트보다 소이 요구르트에서 더 높게 나타났으며, 요구르트의 지질과산화 억제능은 항산화 활성에 기인하는 것으로 추정된다. 또한 발효직후 요구르트의 미생물학적, 물리화학적 및 항산화 활성은 $4^{\circ}C$에서 8일간 저장하는 동안 거의 일정한 수준으로 유지되었다.

Antioxidant Effect of Homogentisic Acid on Hydrogen Peroxide Induced Oxidative Stress in Human Lung Fibroblast Cells

  • Homogentisic acid was found to scavenge intracellular reactive oxygen species (ROS), and 1,1-diphenyl-2-picrylhydrazyl (DPPH) radicals and thus prevented lipid peroxidation in human fibroblast (Wl 38) cells. The radical scavenging activity of homogentisic acid was found to protect Wl 38 cells against hydrogen peroxide $(H_2O_2)$ induced oxidative stress, via the activation of extracellular signal regulated kinase (ERK) protein. Homogentisic acid increased the activity of catalase. Hence, from the present study, it is suggested that homogentisic acid protects Wl 38 cells against $H_2O_2$ damage by enhancing the intracellular antioxidative activity.

Baicalein and Baicalin from the Radix of Scutellaria baicalensis Georgi Inhibits Oxidative DNA Damage and Apoptosis via its Antioxidant Activity

  • In this study, we evaluated and compared the protective effects of two major constituents, baicalein and baicalin, against oxidative DNA and cell damages caused by hydroxyl radical. Antioxidant properties were evaluated using DPPH and hydroxyl radicals scavenging assays and $Fe^{2+}$ chelating assay. ${\varphi}X$ 174 RFI plasmid DNA and intracellular DNA migration assay were used to evaluate the protective effect against oxidative DNA damage. Also, MTT and lipid peroxidation assays were used to evaluate their protective effects against oxidative cell damage. Both baicalein and baicalin prevented intracellular DNA and cells from oxidative damage caused by hydroxyl radical via antioxidant activities. Baicalein demonstrated a stronger antioxidant activity in scavenging DPPH radicals and chelating $Fe^{2+}$ while baicalin scavenged hydroxyl radicals more efficiently. The differences in the level of baicalein and baicalin pose a different pathological pathway for each. The antioxidant activity of baicalin was due to its ability to scavenge hydroxyl radical whilst baicalein was a stronger $Fe^{2+}$ chelator. Further investigation to compare the molecular mechanisms of antitumor activities of baicalein and baicalin is vital to anticancer research.

민들레 물추출물의 항산화 및 자유라디칼 소거활성 (Antioxidative and Free Radical Scavenging Activity of Water Extract From Dandelion (Taruaxacum officinale))

  • 민들레 잎과 뿌리로부터 물추출물을 제조하여 항산화 활성 및 라디칼 소거활성을 분석하기 위하여 linoleic acid에 대한 과산화물 생성저해율, DPPH radical에 대한 소거활성, chemiluminescence를 이용한 hydroxyl radical에 대한 소거활성, EPR을 이용한 superoxide anion radical에 대한 소거활성및 hydrogen peroxide에 대한 소거활성을 측정하였다. 그 결과, 민들레 잎의 물추출물이 뿌리의 물추출물 보다 지방산에 대한 과산화물 생성 저해율이 높았고, DPPH radical, hydroxyl radical, superoxide anion radical 및 hydrogen peroxide에 대한 소거활성 역시 매우 높은 것으로 나타났다.

In Vitro Antioxidant Activity Profiles of ${\beta}$-Glucans Isolated from Yeast Saccharomyces cerevisiae and Mutant Saccharomyces cerevisiae IS2

  • To explore the possible usefulness of ${\beta}$-glucans as natural antioxidants, the antioxidant profiles of ${\beta}$-glucan, extracted from Saccharomyces cerevisiae KCTC 7911, and water soluble and insoluble mutant ${\beta}$-glucan, isolated from yeast mutant S. cerevisiae IS2, were examined by five different in vitro evaluation methods: lipid peroxidation value (POV), nitric oxide (NO), 1,1-diphenyl-2-picrylhydrazyl (DPPH) radical, reducing power, and ${\beta}$-carotene diffusion assay. The antioxidant activities of all ${\beta}$-glucans evaluated in POV test were comparable to or better than that of the known antioxidant, vitamin C. Remarkably, the ${\beta}$-glucan and water insoluble mutant ${\beta}$-glucan possessed 2.5-fold more potent activity than vitamin C at a dosage of 2 mg. Although vitamin C showed 100-fold greater activity than all ${\beta}$-glucans in NO and DPPH tests for measuring the radical scavenging capacity, all ${\beta}$-glucans revealed higher radical scavenging activity than the known radical scavenger, N-acetyl-L-cysteine (NAC), in DPPH test. The water insoluble mutant ${\beta}$-glucan had 2.6- and 5-fold greater antioxidative activity than water soluble ${\beta}$-glucan in NO and DPPH tests, respectively, showing that all ${\beta}$-glucans were able to scavenge radicals such as NO or DPPH. While all ${\beta}$-glucans revealed lower antioxidant profiles than vitamin C in both reducing power activity and ${\beta}$-carotene agar diffusion assay, the ${\beta}$-glucan and water insoluble mutant ${\beta}$-glucan did show a marginal reducing power activity as well as a considerable ${\beta}$-carotene agar diffusion activity. These results confirmed the potential usefulness of these ${\beta}$-glucans as natural antioxidants.
