The study was aimed at evaluating the antioxidant and antimicrobial activities of methanol extract of Caesalpinia bonducella leaves (MECB) (Family: Caesalpiniaceae). The effect of MECB on antioxidant activity, reducing power, free radical scavenging (DPPH radical, nitric oxide radical, superoxide anion radical, hydroxyl radical and hydrogen peroxide radical scavenging), total phenolic content and antimicrobial activities were studied. The antioxidant activity of MECB increased in a dose dependent manner. About 50, 100, 250 and 500 g of MECB showed 53.4, 61.2, 69.1 and 76.2 % inhibition respectively on peroxidation of linoleic acid emulsion. Like antioxidant activity, the effect of MECB on reducing power increased in a dose dependent manner. The free radical scavenging activity of MECB was determined by DPPH radical scavenging method. The potency of this activity was increased with increased amount of extract. MECB was found to inhibit the nitric oxide radicals generated from sodium nitroprusside $(IC_{50}\;=\;102.8\;g/ml)$ whereas the $IC_{50}$ value of curcumin was 20.4 g/ml. Moreover, the MECB was found to scavenge the superoxide generated by photoreduction of Riboflavin. MECB was also found to inhibit the hydroxyl radical generated by Fenton reaction, where the $IC_{50}$ value is 104.17 g/ml compared with catechin 5 g/ml, which indicates the antioxidant activity of MECB. The MECB capable of scavenging hydrogen peroxide in a concentration-dependent manner. The amounts of total phenolic compounds were also determined. Antimicrobial activities of MECB were carried out using disc diffusion methods with five Gram positive, four Gram negative and four fungal species. The results obtained in the present study indicate that MECB leaves are potential source of natural antioxidant and antimicrobial agents.
To discover the sources with antioxidant and tyrosinase inhibitory activities in Korean traditional medicines, 10 extract of medicinal plants were screened for their potential to scavenge stable 1,1-diphenyl-2-picryhydrazyl (DPPH) free radical, inhibit hydroxyl radical (${\cdot}OH$), total phenolic content, and inhibition of tyrosinase. The potency of DPPH radical scavenging activity was shown in the extract of Ulmus davidiana var. japonica Nakai that has a greater effect with $IC_{50}$ values of $6.49{\pm}5.43{\mu}g/mL$, than BHA ($IC_{50}=20.99{\pm}0.74{\mu}g/mL$), L-ascorbic acid $(IC_{50}=20.59{\pm}1.06{\mu}g/mL),\;and\;{\alpha}-tocopherol\;(IC_{50}=25.55{\pm}0.26{\mu}g/mL)$ as a positive control. The ${\cdot}OH$ scavenging activities were observed in the plants tested. Acanthopanax senticosus, Cirsium setiders, U. davidiana exhibited scavenging activity of more than 60% at $500{\mu}g/mL$. The scavenging activity(%) of BHA and a-tocopherol were 64.32 and 55.87% at $500{\mu}g/mL$, respectively. The total phenolic content was determined, in order to assess its effect on the extract's antioxidant activity. The total phenoic content of $33.37{\pm}0.52mg/g$ was conformed by methanolic extract of U. davidiana. The U. davidiana and Morus bombycis exhibited tyrosinase inhibitory activity with a $34.28{\pm}1.32\;and\;75.57{\pm}1.10%$, respectively. In particular, M. bombycis has stronger tyrosinase inhibitory activity than albutin with $36.48{\pm}3.56%$ as a positive control. This work showed that the inhibitory abilities of Korean medicinal plants, such as U. davidiana and M. bombycis, on DPPH free radical, inhibit hydroxyl radical (${\cdot}OH$), and inhibition of tyrosinase and total phenolic content, can be useful in the prevention and treatment of free radical-relate disease. Investigations into further isolation of inhibitory principles of U. davidiana and M. bombycis are now in progress.
Since reactive oxygen species (ROS) and hydroxyl radicals ($^-OH$) play an important role in the pathogenesis of many human degenerative diseases, much attention has focused on the development of safe and effective antioxidants. Preliminary experiments have revealed that the methanol (MeOH) extract of the stem of Prunus davidiana exerts inhibitory/scavenging activities on 1, 1-diphenyl-2-picrylhydrazyl (DPPH) radicals, total ROS and peroxynitrites ($ONOO^-$). In the present study, the antioxidant activities of this MeOH extract and the organic solvent-soluble fractions, dichloromethane (CH$_2$Cl$_2$), ethyl acetate (EtOAc), and n-butanol (n-BuOH), and the water layer of P. davidiana stem were evaluated for the potential to inhibit $^-OH$ and total ROS generation in kidney homogenates using 2',7'-dichlorodihydrofluorescein diacetate (DCHF-DA), and for the potential to scavenge authentic $ONOO^-$. We also evaluated the inhibitory activity of seven flavonoids isolated from P. davidiana stem, kaempferol, kaempferol 7-Ο-$\beta$-D-glucoside, (+)-catechin, dihydrokaempferol, hesperetin 5-Ο-$\beta$-D-glucoside, naringenin and its 7-Ο-$\beta$-D-glucoside, on the total ROS, $^-OH$ and $ONOO^-$ systems. For the further elucidation of the structure-inhibitory activity relationship of flavonoids on total ROS and 'OH generation, we measured the antioxidant activity of sixteen flavonoids available, including three active flavonoids isolated from P. davidiana, on the total ROS and 'OH systems. We found that the inhibitory activity on total ROS generation increases in strength with more numerous hydroxyl groups on their structures. Also, the presence of an ortho-hydroxyl group, whether on the Aring or S-ring, and a 3-hydroxyl group on the C-ring increased the inhibitory activity on both total ROS and $^-OH$ generation.
Vitamin E is a fat-soluble vitamin that exists in eight different forms, which are $\alpha$, $\beta$, $\gamma$, and $\delta$ tocopherol, and $\alpha$, $\beta$, $\gamma$, and $\delta$ tocotrienol. Tocopherols and tocotrienols are important antioxidant in foods, feeds and their raw materials, where they scavenge lipid radicals. Each form has its own biological activity, the measure of potency or functional use in the body. Antioxidants such as vitamin E act to protect cells against the effects of free radicals, which are potentially damaging by-products of the body's metabolism. Free radicals can cause cell dam-age that may contribute to the development of cardiovascular disease and cancer, The content of tocopherols and tocotrienols vary depending on the environmental condition such as growing regions, temperature, crops and varieties. This report deals with chemical and physical properties and extends to their nutritional and health effect on the tocopherols and tocotrienols in cereal grains.
Schisandra chinensis have been traditionally used in Asia for the treatment of dyspnea, cough, mouth dryness, spontaneous diaphoresis, nocturnal diaphoresis, nocturnal emission, dysentery, insomnia and amnesia. The purpose of this study is to evaluate the protective effects of Schisandra chinensis on oxidative DNA damage and lipid peroxidation induced by ROS in non cellular and cellular system. DPPH radical, hydroxyl radical and hydrogen peroxide scavenging assay were used to measure the antioxidant activities. Phi X-174RF I plasmid DNA cleavage assay and intracellular DNA migration assay were used to evaluate the protective effect on oxidative DNA damage. MTT assay and lipid peroxidation assay were used for evaluating the protective effect on oxidative cell damage. It was found to scavenge DPPH radical, hydrogen peroxide and hydroxyl radical and it inhibited oxidative DNA damage, lipid peroxidation and cell death induced by hydroxyl radical. These data indicate that Schisandra chinensis possesses a spectrum of antioxidant and DNA-protective properties
The antioxidant activity of the extract of Erigeron annuus was assessed by means of two different in vitro tests: bleaching of the stable 1,1-diphenyl-2-picrylhydrazyl radical (DPPH test) and the scavenging of authentic peroxynitrite in company with peroxynitrite generation from 3-morpholinosydnonimine (SIN-1). In both tests, the 85% aq. MeOH and n-BuOH soluble fractions of the crude extract showed a significant scavenging effect on peroxynitrite and DPPH radical in comparison to L-ascorbic acid. And bioassay-guided fractionation of the n-BuOH soluble fraction led to the isolation of three compounds: Apigenin (1), quercetin-3-O-glucoside (2), and caffeic acid (3). The structures of the isolated compounds were elucidated on the basis of their spectroscopic data and their antioxidant activities were measured by determining their capacity to scavenge peroxynitrite and the DPPH radical.
The antioxidative and hepatoprotective potentials of two anthraquinones, alaternin (2-hydroxy-emodin) and emodin, to scavenge and/or inhibit hydroxyl radicals generated by the Fenton reaction and to protect tacrine-induced cytotoxicity in human liver derived HepG2 cells were evaluated, respectively. The inhibitory activity on hydroxyl radical generated in a cell-free chemical system (FeSO$_4$/$H_2O$$_2$) was investigated by a fluorescence spectrophotometer using a highly fluorescent probe, 2$^1$,7$^1$-dichlorofluorescein. The hydroxyl radical scavenging activity was determined by electron spin resonance spectroscopy using 5,5-dimethy-1-pyrroline-N-oxide as hydroxyl radicals trapping agents. Tacrine-induced HepG2 cell toxicity was determined by a 3-[4,5-dimethylthiazole-2yl]-2,5-diphenyltertrazolium bromide assay. Although the scavenging activity of alaternin on hydroxyl radical was similar to that of emodin in dose-dependent pat-terns, the inhibitory activity exhibited by the former on hydroxyl radical generation was stron-ger than that of the latter, with $IC_{50}$/ values of 3.05$\pm$0.26 $\mu$M and 13.29$\pm$3.20 $\mu$M, respectively. In addition, the two anthraquinones, alaternin and emodin showed their hepatoprotective activ-ities on tacrine-induced cytotoxicity, and the EC$_{50}$ values were 4.02 11M and 2.37 $\mu$M, respec-tively. Silymarin, an antihepatotoxic agent used as a positive control exhibited the EC$_{50}$ value of 2.00 $\mu$M. These results demonstrated that both alaternin and emodin had the simultaneous antioxidant and hepatoprotective activities.ies.
The purpose of this study was to evaluate the biological activities of extracts of ramie (Boehmeria nivea (L.) Gaud.), hereafter referred to as Bn. Bn extracts from various collecting area were extracted with methanol. Two extracts from our study, Bn-13 and -82, showed significant antioxidant properties, likely due to their ability to scavenge free radicals. In addition, Bn extracts showed stronger anti-bacterial activity against Escherichia coli (Bn-40), Stapylococcus aureus (Bn-33), and Helicobacter pylori (Bn-05). In addition, this study was conducted to evaluate the anti-inflammatory effects of Bn extracts in lipopolyssacharide (LPS)- and interferon-${\gamma}$ (IFN-${\gamma}$)-stimulated RAW 264.7 macrophages cells. Bn-37 significantly inhibited the production LPS/IFN-${\gamma}$-induced nitric oxide. The most noteworthy anti-cancer effect was found in Bn-23. Bn-08 showed inhibition of aldose reductase. This study provides basic information for the development of functional foods.
Crude extracts of Limonium tetragonum and their solvent-partitioned fractions were evaluated for their potential to scavenge authentic $ONOO^-$, and $ONOO^-$ derived from 3-morpholinosydnonimine (SIN-1). Four flavonol glycosides (1-4) were isolated by activity-guided separation. Their chemical structures were elucidated by extensive 2 D NMR experiments and by comparison with published spectral data. These compounds were also estimated for their peroxynitrite scavenging effects. The scavenging ratios of compounds 1-4 on authentic $ONOO^-$ were 56, 37, 56, and 54%, respectively, at a concentration of 1 ${\mu}M$. On the other hand, the inbihition ratios of compounds 1-4 against $ONOO^-$ generation from SIN-1 were 59, 39, 44, and 54% at the same concentration, respectively.
Isoflavones 4'-O-methylalpinumisoflavone (1) and 5, 7, 4'-trihydroxy-6,8-diprenylisoflavone (2) were isolated from stem bark of Cudrania tricuspidata (Carr.) Bureau. These compounds were the first reported from this plant. Antioxidant activities 1 and 2 were evaluated by measuring their ability to scavenge DPPH and ABTS radicals. Diprenylisoflavone 2 showed strong scavenging activity against ABTS ($IC_{50}\;=\;16.3\;{\mu}M$), three-fold higher compared to genistein, which do not possess prenylated group which indicates strong scavenging activity against ABTS radical of isoflavone 2 was due to prenyl group.
