다중 선형 회귀 기반 기계 학습을 이용한 인공지지체의 사각 기공 형태 진단 모델에 관한 연구 (A Study on Square Pore Shape Discrimination Model of Scaffold Using Machine Learning Based Multiple Linear Regression)

  • In this paper, we found the solution using data based machine learning regression method to check the pore shape, to solve the problem of the experiment quantity occurring when producing scaffold with the 3d printer. Through experiments, we learned secured each print condition and pore shape. We have produced the scaffold from scaffold pore shape defect prediction model using multiple linear regression method. We predicted scaffold pore shapes of unsecured print condition using the manufactured scaffold pore shape defect prediction model. We randomly selected 20 print conditions from various predicted print conditions. We print scaffold five times under same print condition. We measured the pore shape of scaffold. We compared printed average pore shape with predicted pore shape. We have confirmed the prediction model precision is 99 %.

  • A novel approach to the manufacture of biocompatible ceramic scaffold for tissue engineering using micro-stereolithography system is introduced. Micro-stereolithography is a newly proposed technology that enables to make a 3D micro structure. The 3D micro structures made by this technology can have accurate and complex shape within a few micron error. Therefore, the application based on this technology can vary greatly in nano-bio fields. Recently, tissue-engineering techniques have been regarded as alternative candidate to treat patients with serious bone defects. So many techniques to design and fabricate 3D scaffolds have been developed. But the imperfection of scaffold such as random pore size and porosity causes a limitation in developing optimum scaffold. So scaffold development with controllable pore size and fully interconnected shape have been needed for a more progress in tissue engineering. In this paper, bone scaffold was developed by applying the micro-stereolithography to the mold technology. The scaffold material used was HA(Hydroxyapatite) nano powder. HA is a type of calcium phosphate ceramic with similar characteristic to human inorganic bone component. The bone scaffold made by HA is expected, in the near future, to be an efficient therapy for bone defect.

조직공학적 섬유륜재생을 위한 PLGA 지지체 제조시 다공크기의 영향에 관한 연구 (The Effect of Pore Sizes on Poly(L-lactide-co-glycolide) Scaffolds for Annulus Fibrosus Tissue Regeneration)

  • 생분해성 고분자인 poly(L-lactide-co-glycolide) (PLGA)를 이용한 조직공학용 다공성 지지체에서의 공극률, 공극의 크기, 공극의 모양 등은 주입된 세포들이 안착하여 증식하는데 있어서 중요한 요건 중 하나이다. 본 연구에서는 섬유를 세포와 다공크기와의 관계를 파악하고자 다공형성물질인 염화나트륨을 다섯 개의 범위로 분류하여 용매캐스팅/염추출법을 이용한 다양한 다공크기를 갖는 다공성 지지체를 제조하였다. (3-(4,5-dimethylthiazole-2-yl)-2,5-diphenyltetrazolium-bromide) (MMT) 분석방법을 이용하여 제조된 지지체에 파종된 섬유륜 세포의 생존율과 증식률을 확인하였으며, in vitro 환경에서의 콜라겐 양과 DNA량을 측정하였다. In vitro 환경에의 세포간의 발생하는 여러 상호작용을 확인하기 위하여 면역결핍 쥐의 피하에 섬유륜 세포가 파종된 지지체를 이식하여 sulfated g1ycosaminoglycan(SGAG)의 합성정도와 조직학적인 평가를 수행하였다. 결론적으로 $180{\sim}250{\mu}m$ 다공크기를 갖는 지지체에서 높은 세포 생존율과 체내에서의 원할한 세포외기질의 형성을 보임으로써 여타의 지지체보다 섬유를 조직 재생에 적절할 것으로 사료된다.

압출 적층 조형법과 입자 추출법을 결합한 이중 공극 BCP/Silica 인공지지체의 제작 (Fabrication of BCP/Silica Scaffolds with Dual-Pore by Combining Fused Deposition Modeling and the Particle Leaching Method)

  • 조직 공학에서는 전통적인 인공지지체 제작 방식인 가스 발포, 염 침출, 스폰지 복제 그리고 동결주조 법 등이 이용되고 있다. 하지만 다양한 공극 형태 및 크기를 가지고 있어서 세포 상호 작용 효과 및 충분한 기계적 특성에 한계가 있다. 그러나 열 용해 적층 법은 조직공학에서 폴리머 재료를 이용하여 다양한 3차원 인공지지체를 제작할 수 있는 가장 적절한 기술이다. 따라서 본 연구에서는 PCL 몰드를 제작하고 실리카와 알긴산 나트륨 염을 포함하는 세라믹 슬러리를 제조하여 몰드에 주입시켰으며, 1일 동안 자연 건조를 시켰다. 제작된 3차원 슬러리 몰드는 PCL 몰드의 제거 및 슬러리를 경화시키기 위해 $100^{\circ}C$의 오븐에서 2시간 열처리 되었고, 열처리 후에 $1100^{\circ}C$에서 소결되었다. 제작된 인공지지체는 주사전자현미경을 통해 관찰되었고, 압축 시험을 통해 알긴산 나트륨 염의 혼합량에 따른 인공지지체의 기계적 특성은 평가되었다.

Performances and Electrical Properties of Vertically Aligned Nanorod Perovskite Solar Cell

  • Organolead halide perovskite have attracted much attention over the past three years as the third generation photovoltaic due to simple fabrication process via solution process and their great photovoltaic properties. Many structures such as mesoporous scaffold, planar heterojunction or 1-D TiO2 or ZnO nanorod array structures have been studied to enhance performances. And the photovoltaic performances and carrier transport properties were studied depending on the cell structures and shape of perovskite film. For example, the perovskite cell based on TiO2/ZnO nanorod electron transport materials showed higher electron mobility than the mesoporous structured semiconductor layer due to 1-D direct pathway for electron transport. However, the reason for enhanced performance was not fully understood whether either the shape of perovskite or the structure of TiO2/ZnO nanorod scaffold play a dominant role. In this regard, for a clear understanding of the shape/structure of perovskite layer, we applied anodized aluminum oxide material which is good candidate as the inactive scaffold that does not influence the charge transport. We fabricated vertical one dimensional (1-D) nanostructured methylammonium lead mixed halide perovskite (CH3NH3PbI3-xClx) solar cell by infiltrating perovskite in the pore of anodized aluminum oxide (AAO). AAO template, one of the common nanostructured materials with one dimensional pore and controllable pore diameters, was successfully fabricated by anodizing and widening of the thermally evaporated Al film on the compact TiO2 layer. Using AAO as a scaffold for perovskite, we obtained 1-D shaped perovskite absorber, and over 15% photo conversion efficiency was obtained. I-V measurement, photoluminescence, impedance, and time-limited current collection were performed to determine vertically arrayed 1-D perovskite solar cells shaped in comparison with planar heterojunction and mesoporous alumina structured solar cells. Our findings lead to reveal the influence of the shape of perovskite layer on photoelectrical properties.

임의 형상 제작 기법을 이용한 3차원 세포지지체 제작에 관한 연구 (A Study on the Fabrication of 3D Scaffolds Using the Solid Freeform Method)

  • With the goal of tissue regeneration for organs damaged through an accident or a disease, research on tissue engineering has been conducted to produce 3-D scaffolds that can support the cells in the attachment and growth for the cell proliferation and differentiation. A scaffold requires a suitable pore size and porosity to increase the nutrient circulation or oxygen supply for the attachment and growth of cells. The existing production methods such as solvent-casting particulate leaching, phase separation, and fiber bonding have certain disadvantages. With these methods, it is difficult to obtain a free desired shape. In addition, certain pore sizes and interconnectivities among the pores may not be guaranteed. To solve these problems, this study has fabricated a scaffold with a 3-D shaped nose using Alginate, which is a natural polymer obtained through Fused Deposition Modeling (FDM), one of the CAD/CAM-based Solid Freeform Fabrication (SFF) methods.

레이저 소결 적층 시스템을 이용한 3차원 수산화인회석 인공지지체 제작에 관한 연구 (A Study on Fabrication of 3D Hydroxyapatite Scaffolds Using a Laser Sintering Deposition System)

  • Calcium-phosphate-based bioceramics are promising biomaterials for scaffolds because they can assist in bone regeneration. In this study, a laser sintering deposition system was developed, and 3D hydroxyapatite (HA) scaffolds were fabricated. The main process conditions of the HA scaffolds were laser power, table velocity, and laser focal distance. As the laser power increased, the line width, line height, and layer thickness also increased. Further, the line width, line height, and layer thickness decreased as the table velocity increased. As the laser focal distance increased, the line width increased, but the line height and layer thickness decreased. The fabricated green scaffolds were sintered at 1050 ℃ and 1150 ℃. The sintered scaffolds had a uniform and continuous interconnected shape, with pore sizes ranging from 850 to 950 ㎛ having 53% porosity. The compressive strength of the scaffolds decreased from 0.72 MPa (1050 ℃) to 0.53 MPa (1150 ℃). The biocompatibility of the scaffolds was investigated by analyzing the adhesion of osteoblast-like MG-63 cells cultured on the surfaces of the scaffolds. The results indicate that the scaffold sintered at 1050 ℃ had good mechanical and biological properties compared to that at 1150 ℃.

혈관모사 마이크로채널이 장착된 3D 종양 세포 배양 시스템의 제작 및 검증 연구 (Fabrication and validation study of a 3D tumor cell culture system equipped with bloodvessle-mimik micro-channel)

  • Recently, three-dimensional (3D) cell culture systems, which are superior to conventional two-dimensional (2D) vascular systems that mimic the in vivo environment, are being actively studied to reproduce drug responses and cell differentiation in organisms. Conventional two-dimensional cell culture methods (scaffold-based and non-scaffold-based) have a limited cell growth rate because the culture cannot supply the culture medium as consistently as microvessels. To solve this problem, we would like to propose a 3D culture system with an environment similar to living cells by continuously supplying the culture medium to the bottom of the 3D cell support. The 3D culture system is a structure in which microvascular structures are combined under a scaffold (agar, collagen, etc.) where cells can settle and grow. First, we have manufactured molds for the formation of four types of microvessel-mimicking chips: width / height ①100 ㎛ / 100 ㎛, ②100 ㎛ / 50 ㎛, ③ 150 ㎛ / 100 ㎛, and ④ 200 ㎛ / 100 ㎛. By injection molding, four types of microfluidic chips were made with GPPS (general purpose polystyrene), and a 100㎛-thick PDMS (polydimethylsiloxane) film was attached to the top of each microfluidic chip. As a result of observing the flow of the culture medium in the microchannel, it was confirmed that when the aspect ratio (height/width) of the microchannel is 1.5 or more, the fluid flows from the inlet to the outlet without a backflow phenomenon. In addition, the culture efficiency experiments of colorectal cancer cells (SW490) were performed in a 3D culture system in which PDMS films with different pore diameters (1/25/45 ㎛) were combined on a microfluidic chip. As a result, it was found that the cell growth rate increased up to 1.3 times and the cell death rate decreased by 71% as a result of the 3D culture system having a hole membrane with a diameter of 10 ㎛ or more compared to the conventional commercial. Based on the results of this study, it is possible to expand and build various 3D cell culture systems that can maximize cell culture efficiency by cell type by adjusting the shape of the microchannel, the size of the film hole, and the flow rate of the inlet.

Chondrogenic Differentiation of Bone Marrow Stromal Cells in Transforming Growth $Factor-{\beta}_{1}$ Loaded Alginate Bead

  • We developed alginate beads loaded with transforming growth $factor-{\beta}_{1}(TGF-{\beta}_{1})$ to examine the possible application of the scaffold and cytokine carrier in tissue engineering. In this study, bone marrow stromal cells (BMSCs) and $TGF{\beta}_{1}$ were uniformly encapsulated in the alginate beads and then cultured in vitro. The cell morphology and shape of the alginate beads were observed using inverted microscope, scanning electron microscope (SEM), histological staining and RT-PCR to confirm chondrogenic differentiation. The amount of the $TGF{\beta}_{1}$ released from the $TGF-{\beta}_{1}$ loaded alginate beads was analyzed for 28 days in vitro in a phosphate buffered saline (pH 7.4) at $37^{\circ}C$. We observed the release profile of $TGF-{\beta}_{1}$ from $TGF-{\beta}_{1}$ loaded alginate beads with a sustained release pattern for 35 days. Microscopic observation showed the open cell pore structure and abundant cells with a round morphology in the alginate beads. In addition, histology and RT-PCR results revealed the evidence of chondrogenic differentiation in the beads. In conclusion, these results confirmed that $TGF-{\beta}_{1}$ loaded alginate beads provide excellent conditions for chondrogenic differentiation.
