Total Phenolic Contents and Biological Activities of Korean Seaweed Extracts

  • Crude extracts of thirty seaweeds collected in Korea were obtained using 50% ethanol, and total phenolic contents and antioxidant activities were compared. Two brown algae, Ecklonia cava (E. cava) and Sargassum siliquastrum (S. siliquastrum), showing high antioxidant activity based on 1,1-diphenyl-2-picrylhydrazyl (DPPH) radical scavenging activity and superoxide anion radical scavenging activity were further investigated for their inhibitory effects on tyrosinase activity. The E. cava extract had the highest total phenolic content among the seaweeds extracts. Total phenolic contents were strongly correlated with antioxidant activity in the thirty seaweed extracts ($R^2\;=\;0.9169$). The E. cava and S. siliquastrum extracts exhibited higher inhibition to tyrosinase activity than butylated hydroxytoluene (BHT) and epigallocatechin gallate (EGCG).

Antiviral effect of fucoxanthin obtained from Sargassum siliquastrum (Fucales, Phaeophyceae) against severe acute respiratory syndrome coronavirus 2

  • Human coronavirus diseases, particularly severe acute respiratory syndrome coronavirus 2, still remain a persistent public health issue, and many recent studies are focusing on the quest for new leads against coronaviruses. To contribute to this growing pool of knowledge and explore the available marine natural products against coronaviruses, this study investigated the antiviral effects of fucoxanthin isolated from Sargassum siliquastrum-a brown alga found on Jeju Island, South Korea. The antiviral effects of fucoxanthin were confirmed in severe acute respiratory syndrome coronavirus 2-infected Vero cells, and its structural characteristics were verified in silico using molecular docking and molecular dynamic simulations and in vitro colorimetric method. Fucoxanthin inhibited the infection in a concentration-dependent manner, without showing cytotoxicity. Molecular docking simulations revealed that fucoxanthin binds to the angiotensinconverting enzyme 2-spike protein (binding energy -318.306 kcal mol-1) and main protease (binding energy -205.118 kcal mol-1). Moreover, molecular dynamic simulations showed that fucoxanthin remains docked to angiotensin-converting enzyme 2-spike protein for 20 ns, whereas it breaks away from main protease after 3 ns. Also, the in silico prediction of the fucoxanthin was verified through the in vitro colorimetric method by inhibiting the binding between angiotensinconverting enzyme 2 and spike protein in a concentration-dependent manner. These results indicate that fucoxanthin exhibits antiviral effects against severe acute respiratory syndrome coronavirus 2 by blocking the entry of the virus. Therefore, fucoxanthin from S. siliquastrum can be a potential candidate for treating coronavirus infection.

Hepatoprotective Effect of Extracts and Phenolic Compound from Marine Algae in Bromobenzene-treated Rats (해조류 추출물과 페놀성화합물의 in vitro 및 in vivo 간보호활성)

  • The methanol extracts of some marine algae were tested for investigating the effects on the formation of lipid peroxide and the activities of free radical generating enzyme in vitro in bromobenzene-treated rat. The extracts of Enteromorpha compressa, Capsosiphon fulvescens, Gelidium amansii, Hizikia fusiformis, Sargassum siliquastrum and Sargassum thunbergii which decreased the formation of lipid peroxide, inhibited the activity of xanthine and aldehyde oxidases by adding of each extracts. Phloroglucinol isolated from Ecklonia stolonifera reduced bromobenzene-induced hepatic lipid peroxidation. This compound administered daily over one week before intoxication with bromobenzene did not affect the activities of aminopyrine N-demethylase, aniline hydroxylase and glu tathione S-transferase. Epoxide hydrolase activity was decreased by bromobenzene, which was restored by pretreatment of phloroglucinol, The results suggest that the bromobenzene-induced hepatic lipid peroxidation by phloroglucinol is reduced by enhancing the activity of epoxide hydrolase.

  • Eight species of Sargassum subgenus Bactrophycus section Halochloa and Repentia were collected from the coasts of Korea: Sargassum coreanum J. Agardh, Sargassum sagamianum Yendo, Sargassum micracanthum (Kützing) Endlicher, Sargassum macrocarpum C. Agardh, Sargassum autumnale Yoshida, Sargassum siliquastrum (Mertens ex Turner) C. Agardh, Sargassum serratifolium (C. Agardh) C. Agardh, Sargassum yezoense (Yamada) Yoshida et Konno. These species were investigated on morphological variations and geographical distributions in Korea. The valuable taxonomic characters between similar species of these section were discussed with special reference to types of holdfasts, shape of main branch, leaves and vesicles.

Community Structure of Benthic Marine Algae at Daejin and Jukbyeon on the Mid-East Coast of Korea (동해 중부 대진과 죽변 연안의 저서 해조류 군집구조)

  • The species composition and variation of benthic marine algae at Daejin and Jukbyeon on the mid-east coast of Korea were investigated seasonally from August 2006 to April 2007. As a result, the total of 85 species, including 8 chlorophytes, 26 phaeophytes, 50 rhodophytes and 1 sea grass were identified. 52 species were found at Daejin and 74 species at Jukbyeon. Dominant species in importance value were Corallina pilulifera, Chondrus ocellatus, melobesioidean algae, Sargassum horneri, Sargassum yezoense and Phyllospadix japonica at Daejin, and Corallina pilulifera, melobesioidean algae, Phyllospadix japonica, Acrosorium polyneurum, Sargassum siliquastrum and Hizikia fusiformis at Jukbyeon, respectively. The vertical distribution of algae were characterized by Corallina pilulifera, Hizikia fusiformis, Sargassum spp. (S. siliquastrum, S. yezoense) and Symphyocladia latiuscula at intertidal zone, Sargassum spp. (S. horneri, S. serratifolium, S. yezoense), melobesioidean algae, Phyllospadix japonica and Corallina pilulifera at 1 m depth, melobesioidean algae, Phyllospadix japonica and Sargassum spp. (S. horneri, S. yezoense) at 5 m depth and melobesioidean algae, Phyllospadix japonica and Sargassum spp. (S. horneri, S. micracanthum, S. yezoense) at 10 m depth.

Investigation of Antimicrobial Activity of Brown Algae Extracts and the Thermal and pH Effects on Their Activity

  • The antimicrobial activity of water and ethanol extracts from 30 species of algae was measured using the agar diffusion method and minimum inhibitory concentration (MIC) test. In agar diffusion method, the 95% ethanol extracts from 12 of the algae showed growth inhibition against the tested microorganisms. In particular, Ishige okamurai, Ecklonia stolonifera, Sargassum siliquastrum, Sargassum thunbergii, Colpomenia bullosa, and Ecklonia cava had strong antibacterial activities against Gram-positive bacteria at 4 mg/mL. In the results of the MIC test, S. siliquastrum showed the most antimicrobial activity, where its MIC values ranged from 0.005 to 0.0075% against Listeria monocytogenes, Clostridium perfringens, and Basillus subtilis. In the thermal stability test, for the ethanol extracts of I. okamurai, E. cava, S. siliquastrum, S. thunbergii, and C. bullosa, the extracts proved to maintain high antimicrobial activities when they were treated at $121^{\circ}C$ for 15 min. In the pH stability test, the antimicrobial activity of the S. siliquastrum ethanol extract was stable from pH 2 to 10, whereas the activity of the other species ethanol extracts were weakened under pH 10 against several microbes.

Effect of Extracts from Sargassum siliquastrum on Shelf-life and Quality of Bread (꽈배기 모자반 추출물 첨가에 의한 빵의 저장성 및 품질 증진 효과)

  • The purpose of this study was to examine the qualities of breads added with 0.01%, 0.1% and 1% of Sargassum siliquastrum extracts (SSE). The result of total microbial count showed that breads with SSE were reduced with increasing storing time, especially bread with 1% SSE was reduced about 2 log cycle as compared to that of control. The moisture content of the bread added SSE and non-added bread were not significantly different during the early storage period, but after 9 days of storage, the moisture content of control was 13% decreased while that of the SSE added bread was decreased by only $5{\sim}6%$. In the color, lightness and yellowness of bread diminished with increasing amounts of SSE in bread while conversely, redness increased. In sensory evaluation, bread containing 0.01% and 0.1% SSE were preferred than the control over total preference, while bread containing 1% SSE showed the lowest preference. These results suggest that the addition of 0.1% SSE in bread had a good effect on improving the preservation and development of quality.

Inhibition Activities of Sea Weeds on Prolyl Endopeptidase, Tyrosinase and Coagulation (해조류 메탄올 추출물의 Prolyl Endopeptidase, Tyrosinase 저해 및 항응고 활성 스크리닝)

  • About forty sea weeds were screened for their inhibitory effects against prolyl endopeptidase, tyrosinase and thrombus coagulation. Out of them, methanolic extract of Ecklonia cava, Sargassum patens, Sargassum hemiphyllum, Sargassum thunbergii, Sargassum singgildianum, Hizikia fusiformis, and Ishige okamurae inhibited more than 90% of prolyl endopeptidase activity at 40 ppm. Sargassum siliquastrum and Ecklonia cava exhibited 51% and 76% of inhibitory activity against tyrosinase at 40 ppm, respectively. In APTT assay system, Sargassum singgildianum, Pterocladia capilacea and Hizikia fusiformis delayed coagulation of thrombus about two times (210, 211, and 198% over control at ca 367 ppm, respectively) and in TT assay, Lomentaria catenata, Laurencia okamurae, and Hizikia fusiformis did most effectively (216,197, and 251% at ca 367 ppm, respectively).

Effects of Gamma Irradiation on Antioxidant Properties and Physical Characteristics of Sargassum siliquastrum Water Extract (꽈배기 모자반 물 추출물의 항산화능과 물리적 특성에 대한 감마선 조사의 영향)

  • In this study, the antioxidant properties and physical characteristics of irradiated Sargassum siliquastrum water extract were evaluated. Samples were irradiated with $Co^{60}{\gamma}$-ray at doses ranging from 3 to 20 kGy. They were then analyzed to investigate antioxidant properties, including total phenolic compound content and DPPH (1,1-diphenyl-2-picrylhydrazyl) radical scavenging activity. In addition, physical properties such as viscosity and color were evaluated. The results demonstrated that total phenolic compound content and DPPH radical scavenging activity were significantly improved (p<0.05) by irradiation. In terms of physical properties, viscosity and color were reduced in the irradiated extracts as compared to non-irradiated samples. In conclusion, gamma irradiation improved the antioxidant properties of Sargassum siliquastrum water extract, improving its original weak point as a natural antioxidant when applied in the food industry.
