Oxidative stress causes tissue damage and facilitates the progression of metabolic diseases, including diabetes, cardiovascular heart diseases, and obesity. Lipid accumulation and obesity-related complications have been observed in the presence of extensive oxidative stress. As part of an ongoing study to develop therapeutic supplements, Sargassum sp. were tested for their ability to scavenge free radicals and intracellular reactive oxygen species (ROS), as well as to suppress lipid accumulation. Three species, S. hemiphyllum, S. thunbergii, and Sargassum horneri, were shown to scavenge free radicals in a di(phenyl)-(2,4,6-trinitrophenyl)iminoazanium (DPPH) assay. In addition, Sargassum sp. was shown to scavenge intracellular ROS and to decrease nitric oxide (NO) production in $H_2O_2$ and lipopolysaccharide (LPS)-induced in RAW264.7 mouse macrophages, respectively. Taken together, the results suggest that Sargassum sp. possess huge potential to relieve oxidative stress and related complications, as well as lipid-induced oxidation. They indicate that S. hemiphyllum, S. thunbergii, and S. horneri are potent functional supplements that can produce beneficial health effects through antioxidant and antiobesity activities, with S. hemiphyllum being the most potent among the Sargassum sp. tested. A potential mechanism for the effect of Sargassum sp. on the suppression of lipid accumulation in differentiating 3T3-L1 mouse preadipocytes through deactivation of the peroxisome proliferator-activated receptor ${\gamma}$ (PPAR ${\gamma}$) is presented.

This study was conducted to determine the antioxidant activities of ethanol and water extracts of Sargassum hemiphyllum. Antioxidant activities were evaluated by assessing total phenolic contents, 2,2-diphenyl-2-picrylhydrazyl (DPPH) radical scavenging activity, chelating effect, reducing power, and using the rancimat method. Total phenolic contents in the ethanol and water extracts were 17.91 and 13.44 mg gallic acid equivalents/g, respectively. Ethanol extract showed higher DPPH radical scavenging activity than water extract and similar activity to that of BHT. The reducing power of ethanol and water extracts increased in a concentration-dependent manner. Particularly, ethanol extract was more effective than water extract. Water extract showed a higher chelating effect compared to ethanol extract. The antioxidant index measured by rancimat was lower than those in BHT, but the ethanol extract showed a higher value than the water extract. The ethanol extract showed higher antioxidant activity than the water extract, except for the chelating effect. These results suggest that the ethanol extract of Sargassum hemiphyllum has more potent antioxidant activity and may be used as a source of natural antioxidants.
This study was carried out to determine the inhibitory effects of solvent extracts from Sargassum hemiphyllum on growth of cancer cell lines (AGS human gastric adenocarcinoma and HT-29 human colon cancer cells) and production of lipid peroxides. Inhibitory effects of acetone with methylene chloride extract from S. hemiphyllum on the growth of AGS and HT-29 cancer cells were increased as dose dependent patterns (p<0.05). The methanol extract was more effective on inhibition of growth of AGS. The treatments of hexane, 85% aq. methanol, butanol and water fractions significantly inhibited the growth of cancer cells (p<0.05) and the inhibitory effect was stronger in HT-29. In DCFH-DA (dichlorodihydrofluorescin diacetate) assay, acetone with methylene chloride and methanol extracts showed a stronger inhibitory effect on the production of cellular lipid peroxides (p<0.05) compared with the butanol and hexane fractions. These results indicate that the consumption of S. hemiphyllum may be recommended as a potent functional food for preventing cellular oxidation and cancer.
There is growing interest in natural cosmetic ingredients as natural cosmetics become popular. As a part of effort to look for natureal cosmetic agent, seaweed Sargassums were tested for functional cosmetic agents. Effective materials were extracted from Sargassum coreanum. Sargassum hemiphyllum, and Sargassum patens by simple hot water extraction. Antioxidation, whitening, anti-wrinkle, UV absorption and anti-inflammation effects were studied for functional cosmetic agents. Sargassum extracts indicated excellent cell viability, strong anti-oxidation effect by DPPH radical scavenging activity and showed significant whitening effect from tyrosinase inhibition. However, effects of antiwrinkle, UV absorption and anti-inflammation were negligible. In conclusion, Sargassum coreanum extracts showed good possibility for anti-oxidation and whitening cosmetic agent.
As a part of our search for novel antioxidants from the seaweeds, we have investigated radical scavenging effect for their crude extracts using 1,1-diphenyl-2-picrylhydrazyl (DPPH) radical, authentic peroxynitrite, and 3-morpholinsydnonimine (SIN-1), a peroxynitrite-generating species in vitro. Thirty-four seaweeds were screened for $ONOO^-$ and DPPH radical scavenging activities. A potent inhibitory effect against peroxynitrite generated by SIN-1 at $5{\mu}g/ml$ of methanol extracts was observed in order of Ishige okamurae(95.3%), Sargassum hemiphyllum(90.2%), Symphyocladia latiuscula(89.6%), Porphyra suborbiculata(86.7%), and Gelidium amamsii(85.9%), Also, a significant scavenging effect against direct authentic peroxynitrite was revekaled for methanol extracts of Ishige okamurae(66.2%) and Sargassum hemiphyllum(55.2%) and the acetone/methylene chloride(1:1) extract of Gigatina tenella (61.0%). In our measurement for evaluating the capacity to scavenge the stable free radical of DPPH, acetone/methylene chloride(1:1) extracts of Symphyocladia latiuscula, Gloiopeltis furcata, and Sargassum thunbergii and the methanol extract of Sargassum sp. showed an inhibitory potency of 85.8%, 82.8%, 74.1%, and 64.0%, respectively.
Eight species of Sargassum subgenus Bactrophycus section Teretia were collected from the coasts of Korea: S. confusum C. Agardh, S. pallidum (Turner) C. Agardh, S. muticum (Yendo) Fensholt, S. thunbergii (Mertens ex Roth) Kuntze, S. fulvellum (Turner) C. Agardh, S. hemiphyllum (Turner) C. Agardh, S. nipponicum Yendo, and S. miyabei Yendo. These species were investigated on morphological variations and geographical distribution in Korea. The valuable taxonomic characters between similar species of this section were discussed with special reference to thickness of leaves, types of holdfasts, shape of leaves and vesicle, and length of rhizoids.
About forty sea weeds were screened for their inhibitory effects against prolyl endopeptidase, tyrosinase and thrombus coagulation. Out of them, methanolic extract of Ecklonia cava, Sargassum patens, Sargassum hemiphyllum, Sargassum thunbergii, Sargassum singgildianum, Hizikia fusiformis, and Ishige okamurae inhibited more than 90% of prolyl endopeptidase activity at 40 ppm. Sargassum siliquastrum and Ecklonia cava exhibited 51% and 76% of inhibitory activity against tyrosinase at 40 ppm, respectively. In APTT assay system, Sargassum singgildianum, Pterocladia capilacea and Hizikia fusiformis delayed coagulation of thrombus about two times (210, 211, and 198% over control at ca 367 ppm, respectively) and in TT assay, Lomentaria catenata, Laurencia okamurae, and Hizikia fusiformis did most effectively (216,197, and 251% at ca 367 ppm, respectively).

The objective of this study was to investigate the useful components and potential antioxidant activities of 30 marine algae, collected from Jeju Island in Korea. The contents of bioacitve substance of extracts from marine algae, such total polyphenol and carbohydrate, were determined. The extract of Distromium decumbens had the highest amount of total polyphenol content (21.27%), and that of Gracilaria incurvata Okamura had the highest amount of total carbohydrate content (10.18%). The antioxidant activities of extracts obtained from algae were tested through the evaluation of DPPH radical and hydroxyl radical scavenging activity. The extracts of Distromium decumbens, Sargassum hemiphyllum (Turner) C.Agardh, Sargassum serratifolium (C. Agardh) C. Agardh and Acrosorium yendoi Yamada were found to have more than 80% DPPH radical scavenging activity and that of Dictyota okamurae (Dawson) $H{\ddot{o}}tning$, Schnetter, et Prud'homme van Reine, Myagropsis myagroides (Martens ex Turner) Fensholt, Sargassum serratifolium (C. Agardh) C. Agardh and Cladophora wrightiana Harvey showed more than 50% hydroxyl radical scavenging activity. These results suggest that algae collected from Jeju Island would be good raw materials for antioxidant.
Macroalgae are important primary producers in marine ecosystem. They don't only play an important role as bioindicators but also provide economic resources for humans. Seasonal and vertical variations in seaweed species composition and biomass were examined to determine the ecological status of seaweed beds around Udo, near Jeju Island. We obtained samples at two sites in the high-intertidal to subtidal zones using the quadrat method between June 2010 and May 2011. A total of 262 species were collected, including 31 green, 61 brown, and 170 red algae. The composition of algal species revealed a decrease in species growing in cold water in comparison with the list 20 years ago. The macroalgal mean biomass (g wet wt $m^{-2}$) was 3,476 g and 2,393 g from the two sites, respectively. Ecklonia cava had the greatest biomass at both sites. The seasonal dominant species by biomass at site 1 from the low-intertidal to 1-3 m depth of the subtidal zone was mostly comprised of thick-leathery form, such as Sargassum hemiphyllum, S. coreanum, and Ecklonia cava, whereas site 2 was comprised of the turf form, such as Chondrophycus intermedius, Chondracanthus intermedius, Dictyopteris prolifera, and Gelidium elegans. The current ecological status of the seaweed community in Udo is stable based on diversity and dominance indices.

Marine algae have long been recognized as a health and beauty food, based on its anti-tumor, anti-inflammatory and anti-obesity activities. In this study, methanol extracts were prepared from 10 different phaeophyta, after which DPPH radical scavenging and cytoprotective activities of HT-22 cells against ${\beta}$-amyloid protein ($A{\beta}$), which has neurotoxic effects, were investigated. In DPPH experiments, Ecklonia cava and Ishige okamurai showed strong ROS scavenging activities, whereas eight other phaeophyta including Petalonia binghamiae (P. bin) showed weak ROS scavenging activities. To validate the cytoprotective effects of 10 different phaeophyta in $A{\beta}$-induced HT-22 cells, protein expression levels of APP, BACE1, iNOS, phosphorylated ERK1/2, phosphorylated p38 and phosphorylated JNK1/2 were determined along with MTT assay. In the MTT assay, P. bin showed the best effective cytoprotective activity at a concentrations of $25{\mu}g/mL$, whereas Sargassum confusum, Colpomenia sinuosa, Myelophycus simplex, and Sargassum hemiphyllum showed potential. Determination of protein expression levels related to $A{\beta}$-induced neurotoxicity in the five selected phaeophyta showed that P. bin inhibited BACE1 and iNOS expression in $A{\beta}$-induced HT-22 cells. These results indicate that the cytoprotective effects of P. bin are mediated by suppressing the pathways involving $A{\beta}$-induced ERK and p38 activation.
