수산 미이용자원 중에 존재하는 효소적 산화 억제제의 검색 1. 감자 Lipoxygenase-1에 의한 효소적 산화에 대한 억제 (Screening for Potato Lipoxygenase-1 Inhibitor in Unused MArine Resources by the Polarographic Method)

  • 성인병 에방물질의 가능성을 지닌 5-lipoxygenase 활성 저해제가 8종의 수산 미이용자원의 순차 용매 추출구간들 중에 존재하는지 여부를 수산 미이용자원의 고부가 가치적 이용 방안 모색을 위해 시도해 보았다. 그 결과, 대상자료에 대한 여러 용매 추출구들의 거의 대부분의 경우에 있어서 5-lipoxygenase 효소저해능을 나타내었다. 그 중 수산동물의 경우, 불가사리의 에테르 추출구와 아세톤 추출구, 우렁쉥이 껍질의 아세톤 추출구, 오징어먹즙의 아세톤 추출구와 메탄올 추출구에서 $IC_{50}$이 각각 72.5, 65, 13.3, 13.3 및 $75\mu\textrm{g}/ml$로 높은 5-lipoxygenase 활성저해 효과를 나타내었다. 시험대상 해조류 중 구멍쇠미역의 아세톤 추출구, 모자반의 물 추출구, 에테르 추출구 및 아세톤 추출구, 산말의 에테르와 아세톤 추출구에서 $IC_{50}$이 각각 15.5, 46.5, 5.03, 1.1, 30.5 및 $35\mu\textrm{g}/ml$로서 매우 높은 저해 효과를 보였다. 이상의 활성물질에 대한 보다 더 순수한 분리 후 동정을 행하여 대량 생산만 된다면, 효소적 산화방지제로서 뿐 아니라 생체내 산화조절제로서도 이용가능할 것이므로 수산 미이용자원의 고부가 가치적 이용방안의 하나가 되리라 여겨진다.

Investigation of ${\alpha}$-Glucosidase Inhibitory Activity of Ethanolic Extracts from 19 Species of Marine Macroalgae in Korea

  • In the present work, we have collected 19 species of macroalgae (9 Phaeophta and 10 Rhodophyta) f rom all around of Korea: Dictyopteris divaricata, D. prolifera, Myelophycus cavus, Papenfussiella kuromo, Petalonia zosterifolia, Petrospongium rugosum, Rugulopteryx okamurae, Sargassum fulvellum, S. muticum, Callophyllis japonica, Gloiopeltis tenax, Gracilaria longissima, Gracilaria vermiculophylla, Grateloupia asiatica, Grateloupia lanceolata, Grateloupia sparsa, Grateloupia turuturu, Grateloupia sp, and Polyopes affinis. The macroalgal species were extracted by 70% ethanol (EtOH) for 24 h and evaluated its inhibitory effects on ${\alpha}$-glucosidase. Among ethanol extracts, Myelophycus cavus showed the most effectively inhibitory activity ($IC_{50}$, 2.17 ${\mu}g/ml$) against ${\alpha}$-glucosidase, followed by Sargassum fulvellum (<$IC_{50}$, 8.13 ${\mu}g/ml$), Dictyopteris prolifera ($IC_{50}$, 16.66 ${\mu}g/ml$), Rugulopteryx okamurae ($IC_{50}$, 50.63 ${\mu}g/ml$), and Petrospongium rugosum ($IC_{50}$, 101.62 ${\mu}g/ml$). Furthermore, MTT (3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyl tetrazolium bromide) assay showed no cytotoxicity on mouse pre-adipocytes cell line (3T3-L1). These results suggest that some edible macroalgae merit further evaluation for clinical usefulness as anti-diabetic functional foods.

해조류 추출물의 항산화 및 nitrite 소거활성 평가 (Evaluation of Antioxidant and Nitrite Scavenging Activity of Seaweed Extracts)

  • 유용생리활성의 해조류를 이용한 식품소재 개발 연구의 일환으로, 35종 해조류(갈조류 17종, 홍조류 11종 및 녹조류 7종)의 메탄올 추출물을 대상으로 in-vitro 항산화 및 nitrite 소거활성을 평가하였다. 해조류 추출물($500\;{\mu}g/ml$) 중 감태, 곰피, 대황, 넓패, 패, 모자반, 알송이모자반, 야마자모자반 및 잘피의 9종에서 60% 이상의 DSA 활성을 나타내었으며, ASA활성은 DSA활성과 높은 상관관계를 나타내었다(상관계수 0.855). 환원력 평가 결과에서는 감태, 곰피 및 대황의 3종만이 0.88 ($Abs_{700}$) 이상의 강력한 활성을 나타내었다. 한편 식품에서의 발암인자인 nitrosoamine생성억제와 관련된 NSA평가의 경우, 전체 35종의 해조류 중 19종에서 60% 이상의 NSA를 나타내었으며, 특히 기존에 알려지지 않은 모자반, 야마다모자반, 알송이모자반, 패, 넓패에서 우수한 활성을 확인하였다. 1차 선별된 9종 해조류의 DSA, ASA, NSA의 $IC_{50}$를 조사한 결과, 기존의 항산화능이 알려진 곰피, 대황, 감태 이외에도 야마다모자반 및 넓패에서 강력한 항산화 활성과 nitrite 소거활성을 확인하였다. 본 연구결과는 야마다 모자반 및 넓패를 이용하여 항혈전, 항산화, NSA 활성을 동시에 나타내는 기능성 식품소재로의 개발이 가능함을 제시하고 있다.

각종 해조류 추출물들의 인간 피부세포주 HaCaT에 대한 활력 영향 (Effects of Several Seaweed Extracts on the Viability of Human Keratinocyte HaCaT Cells)

  • The cellular viability of the human keratinocyte cell line HaCaT was compared after adding seaweed extracts to the culture medium. The viability was measured using a quick, quantitative, spectrophotometric crystal violet inclusion method. Of 36 common seaweed species tested, methanol extracts from Sargassum sagamianum and Gigartina tenella enhanced the viability of HaCaT cells by 1.6-fold, as compared to control cells, while methanol extracts from Dictyota dichotoma, Pachymeniopsis elliptica, and Enteromorpha linza decreased the viability to less than half that of controls.

참모자반 (Sargassum fulvellum) 물 추출물의 염증 억제 활성 (Anti-inflammatory Activity of the Water Extract of Sargassum fulvellum)

  • The anti-inflammatory effects of Sargassum fulvellum water extracts (SFWE) were investigated using lipopolysaccharide (LPS)-induced inflammatory response in this study. To examine the potential anti-inflammatory properties of SFWE, the NO, TNF-${\alpha}$, IL-6, IL-$1{\beta}$, and cell proliferation were measured. It was confirmed that the NO and TNF-${\alpha}$ secretion were significantly suppressed when SFWE was added to LPS-stimulated RAW 264.7 cells. Moreover, the expression of IL-6 and IL-$1{\beta}$ cytokines was suppressed by SFWE in a dose-dependent manner. Especially, IL-6 inhibition activities were over 50% at 1% of SFWE. The cytotoxicity of SFWE and the proliferation of macrophages was measured by MTT assay. As a result, there was no cytotoxicity in the macrophage proliferation treated with SFWE compared to the control. In conclusion, these results suggested that the SFWE may have significant effects on inflammatory factors and can be a potential anti-inflammatory therapeutic materials.

  • Sparassis latifolia is a fungus abundant in β-glucan and amino acids and is highly valued as a medicinal mushroom. Among amino acids, γ-aminobutyric acid (GABA) is a free amino acid and has biological effects, such as increase/decrease of hypertension, improvement of cerebral blood flow, and prevention of dementia. In this study, biological elicitors were used to increase bioactive substances as a biofortification method. Sodium alginate extracted from seaweed (Sargassum horneri, Sargassum fulvellum, Sargassum fusiforme) were used as the elicitor. The levels of β-glucan and GABA in the mycelium and fruiting body grown by adding the elicitor to the medium were investigated. Addition of sodium alginate positively affected GABA production and negatively affected the β-glucan production in these fungi. Sodium alginates extracted from S. fulvellum induced the highest increase in GABA in the mycelium and fruiting bodies. Moreover, we investigated the effects of the extracts from mycelium and fruiting bodies on dendrite development in primary cortical neurons. We found that the extract from the fruiting bodies of sodium alginate treated fungi with increased levels of GABA inhibited the dendrite outgrowth of excitatory neurons, but not inhibitory neurons.

Anti-inflammatory and antioxidant activities of Sargassum horneri extract in RAW264.7 macrophages

  • [Purpose] In this study, we investigated whether a 70% ethanolic (EtOH) extract of Sargassum horneri had antioxidant and anti-inflammatory effects in lipopolysaccharide (LPS)-stimulated macrophage-like RAW 264.7 cells. [Methods] The proximate composition, fatty acids, amino acids, and dietary fiber of S. horneri, various biologically active compounds, and antioxidant activity were analyzed. [Results] The DPPH and ABTS free radical scavenging activities, as well as the reduction power, of the S. horneri extract used here were significantly increased in a concentration-dependent manner. This indicates that S. horneri contains bioactive compounds, such as phenols and flavonoids, that have excellent antioxidant activity. The cellular viability and metabolic activity results confirmed that the extract had no discernible toxicity at concentrations up to 100 ㎍/mL. The levels of nitrites and cytokines (PGE2, TNF-α and IL-6), which mediate pro-inflammatory effect, were significantly inhibited by treatment with either 50 or 100 ㎍/mL S. horneri extract, whereas that of IL-1β was significantly inhibited by treatment with 100 ㎍/mL of the extract. Similarly, the expression of iNOS and COX-2 proteins also decreased according to 50 or 100 ㎍/mL extract concentrations. NF-κB binding to DNA was also significantly inhibited by treatment with 100 ㎍/mL of extract. [Conclusion] These results suggest that 70% EtOH extracts of S. horneri can relieve inflammation caused by disease or high intensity exercise.

Polyphenol-rich Sargassum horneri alleviates atopic dermatitis-like skin lesions in NC/Nga mice by suppressing Th2-mediated cytokine IL-13

  • Atopic dermatitis (AD) is one of major skin inflammatory diseases characterized by excessive Th2-mediated immune responses. Recent evidence provides that interlukin-13 (IL-13) plays the role of a key Th2 cytokine that drives the inflammation underlining AD. Due to adverse effects of commercially available synthetic drugs, the need for treatments based on natural products is gaining much attention. Sargassum horneri is an edible brown algae known for beneficial bioactivities including anti-inflammation. We investigated if polyphenol-rich S. horneri extracts (SHE) could suppress AD-like skin lesions in NC/Nga mice and if that involved inhibition of the infiltration of Th2-mediated cytokine IL-13. We observed markedly increased infiltration of IL-13 positive cells in AD-like skin lesions of mice but SHE treatments decreased it. Also, the dermal expression of IL-13 was sufficient to cause inflammatory responses in mice skin resembling human AD. SHE suppressed the dermal infiltration of inflammatory cells where IL-13 plays a crucial role in skin tissues and in the recruitment of inflammatory cells. Furthermore, it was confirmed that SHE reduced T cell, dendritic cell, and macrophage populations in spleen. Moreover, SHE decreased the collagen deposition in skin and ear dermis resulting in reduced fibrosis that occurs in AD due to excessive collagen. Taken together, our results reveal that SHE suppressed the infiltration of inflammatory cells into skin dermis by decreasing the infiltration of IL-13 positive cells. Therefore, SHE could be taken as a useful therapeutic agent to alleviate AD.

Growth Enhancement of the Microalga Tetraselmis suecica by an Extract of the Green Alga Monostroma nitidum

  • Cell growth of the marine microalga Tetraselmis suecica was regulated by the addition of seaweed extracts in its culture medium. Of 26 species of seaweed tested, water-soluble extracts from Monostroma nitidum and Pachymeniopsis elliptica and methanol-soluble extract from Sargassum confilsum enhanced the growth of T suecica cells. The water extract of M. nitidum was the most effective, producing up to a 2-fold increase in cell density with the addition of 1 mg/mL of extract to the culture medium. Cell size, gross biochemical composition, fatty acids, and digestion efficiency all differed marginally between cultures of T suecica grown with and without the M. nitidum extract.

여드름균(Cutibacterium acnes)에 대한 해조류 추출물의 항균효과 (In vitro Antimicrobial Activities of Edible Seaweeds Extracts Against Cutibacterium acnes)

  • The study was aimed at developing an alternative therapeutic agent against acne vulgaris, one of the most common skin diseases, to meet the continuing demand for new therapies. Acne vulgaris is often associated with the acne-causing bacteria such as Cutibacterium acnes. To investigate the safety of agents against acne vulgaris, we evaluated the potential antibacterial activities of edible seaweeds against C. acnes in Korea. Forty-one edible seaweeds, including the brown, green, and red varieties, were selected for the antibacterial test. In comparison with other seaweeds, 70% ethanolic extracts of brown seaweeds, such as Cladophora wrightiana var. minor, Eisenia bicyclis, Ecklonia cava, Ishige foliacea, Ishige okamurae, Sargassum filicinum, and Sargassum miyabei Yendo, exhibited potential antibacterial activity against C. acnes with minimum inhibitory concentrations ranging between 64 and 128 ㎍/mL. To investigate the active anti-acne agents and to enhance our understanding of the antibacterial activities against C. acnes, further solvent-fractionation experiments are warranted. The findings imply that brown seaweeds can be a potential source of natural agents against acne vulgaris.
