Prevalence and Toxin Characteristics of Bacillus cereus Isolated from Drinking Cups in Spring (약수터 음용도구의 Bacillus cereus 분포 및 독소 특성)

  • The purpose of this study was to investigate the microbiological contamination of water and drinking cups in springs and to estimate the toxin gene, enterotoxin production ability and antibiotic susceptibility of foodborne pathogens. Ten spring water and 34 drinking cups were tested. The average number of total aerobic bacteria and coliform bacteria in spring water were 1.8 log CFU/mL and 1.2 log CFU/mL, and in drinking cups were $4.7log\;CFU/100cm^2$ and $1.7log\;CFU/100cm^2$. Salmonella spp., Staphylococcus aureus, E. coli O157:H7, Listeria monocytogenes, Vibrio parahaemolyticus, Yersinia enterocolitica were not isolated from all of samples but Bacillus cereus was detected in 5 (14.7%) of 34 drinking cups. The nheA and entFM genes were major enterotoxin genes in B. cereus isolated from drinking cups. All of B. cereus tested in this study produce non-heamolytic enterotoxin but only 2 isolates possessed heamolysin BL enterotoxin producing ability. B. cereus was resistant to ${\beta}-lactam$ antibiotics. These results revealed that the sanitary conditions of drinking cups in spring should be improved promptly. The substitution carrying a personal drinking cup for the public drinking cups equipped in springs is suggested to prevent food-borne illness.

Investigation of Microbial Contamination Levels between GAP and non-GAP Certified Farms of Lettuce and Cucumber (GAP 인증·미인증 상추 및 오이 농가의 미생물학적 오염도 평가)

  • The aims of this study were to investigate microbial contamination levels and to survey sanitation management between Good Agricultural Practices (GAP) and non-GAP farms of lettuce and cucumber. The samples (lettuce, cucumber, soil, agricultural water, gloves, and packing plastic bag) were tested to analyze sanitary indicator bacteria (total aerobic bacteria, coliforms and Escherichia coli) and major pathogenic bacteria (Staphylococcus aureus, Bacillus cereus). In the lettuce farms, the contamination levels of total aerobic bacteria and coliforms in GAP farms were little lower than non-GAP farms or similar. Staphylococcus aureus and Bacillus cereus in soil and agricultural water of GAP farms were detected at higher levels than non-GAP farms in soil and agricultural water. In the case of cucumber farms, levels of total bacteria and Bacillus cereus in soil and total bacteria and coliform in gloves of GAP farms were higher than those of non-GAP farms, and other bacteria contamination levels in collected samples were similar. These results indicate that agricultural products produced from GAP farm still exhibited potential microbial risks. According to the field survey, a sanitation management in GAP farms was insufficient. These results could be useful as basic data to suggestion of plan for preventing microbial contamination and to improvement of GAP certification.

Evaluation on HACCP prerequisite-program performance within general hospital foodservice operations (종합병원 급식소의 HACCP 선행요건 관리 수행도 평가)

  • Purpose: This study was conducted to examine prerequisite-program performance of the hospital foodservice operation and develop measures for improvement of prerequisite-program performance. Methods: Data were collected through surveys administered to 168 hospital dieticians in the Seoul, Incheon, Gyeonggi-do, Daegu, and North Gyeongsang areas. Out of total questionnaires, 65 questionnaires were usable and the response rate was 38.7%. Statistical analyses were performed using the SPSS program (ver 20.0) for ${\chi}^2-test$ and one-way ANOVA. Results: According to the result of the prerequisite-program evaluation, the percentage of 'appropriate', 'needs to be improved', and 'inappropriate' was 44.6%, 47.7%, and 7.7%, respectively. The score for the 'inappropriate' group was significantly lower than that of the 'appropriate' group or 'needs to be improved' group on the food safety management (p < 0.001), preparation facility management (p < 0.001), water management (p < 0.001), and storage transportation management (p < 0.05) parts. Holding rate of foodservice facility and equipments in the 'appropriate' group were significantly higher than in the others on convection oven (p < 0.01), air conditioner (p < 0.01), three-compartment sink (p < 0.01), hot-holding equipment (p < 0.01), cold-holding equipment (p < 0.05), exclusive thawing refrigerator (p < 0.05), and sterilizer for sanitary shoes (p < 0.05) items. Conclusion: To improve the quality of hospital foodservice, foodservice managers and HACCP specialists should develop and implement a prerequisite-program and a HACCP plan considering the characteristics of the hospital foodservice operation.

Microbiological Evaluation of Chilled Freshes Raw-fish Manufacturers before and after HACCP System Establishment (싱싱회류 생산업체의 HACCP 시스템 구축 전 후의 미생물학적 평가)

  • Raw-fish food contains a lot of moisture and is a high-protein food. It is a first-stage processed food taking a lot of manual work. Therefore, it is classified as a PHF food, very liable to cause a bacterial food-poisoning. But its manufacturers are usually small-sized and a systematic sanitation management is difficult to expect. But the manufacturer participating in this study produces chilled fresh raw-fish food. Fish are sliced into two fillets, which are packaged under vacuum, kept and distributed in refrigerators, and sold within a day. It is a newly-developed kind of raw-fish food, and a more improved kind of raw-fish food making possible a systematic sanitation management. The HACCP (Hazard Analysis and Critical Control Point) is a systematic and continuous process-control method which is very efficient for controling food sanitation and reducing the expenses. A new HACCP model has been developed to be applied to a large-sized chilled fresh raw-fish food manufacturer. To ascertain its efficiency, the baterial examination was done to its workplace and products. The significance test was done on its data by "SPSS 12.0 for Window" and "Mann-Whitney U Test". The numbers of bacteria on its final products were significantly different in flatfish and porgy. The number of bacteria tended to decrease in each time-differential sampling (P<.00l). The final food products showed no food-poisoning bacteria in all the time-differential tests and in all the samplings, which proves that the CCP of the HACCP system is under control. After the SSOP program was applied, no pathogenic bacteria were found in the work-place, and the kinds and numbers of bacteria decreased. The numbers of general bacteria and colon bacilli also showed a significant difference from those before the SSOP program in the filleting board (P<.05), in the skinning board (P<.0l), in the neck-removing knife (P<.05), and in the filleting knife (P<.01). The working equipments, periodically disinfected, also showed a significant difference in sanitary conditions (in the dehydrator, P<.05). The number of bacteria found on the food-touching surface was within the standard (below 500/l00 cm$^2$) After the SSOP program was applied, the general bacteria and colon bacilli were not found. The quality of water used in the food processing was also within the standard. The numbers of bacteria falling from the air in the work-place were negligible in all the samplings (<30CFU/l000ι). The staphylococci and fungi were not found.

SSOP Program Development for HACCP Application in Fresh Raw Fish Manufacturing (싱싱회류 생산업체의 HACCP 적용을 지원하기 위한 SSOP Program 개발과 성과)

  • This study aims at developing an easily-applicable Sanitation Standard Operating Procedure(SSOP) program for Fresh raw-fish manufacturing. It compares the sanitation inspection results of SSOP-program-applied procedure and non-applied procedure. The samplings for bacteriological examination were collected from the touching surface of food and from the personal workers. The sanitation status was recorded in the check-up list. The results were statistically analyzed to ascertain the efficiency of the SSOP program. The present official KFDA check-up list for sanitation inspection is composed of 51 items. Each item is judged to be fit or unfit to the standards. When the unfit items are more than 6, the work-place is judged to be insanitary. But in this study, the results of sanitation inspection are scaled in points in the 70% allotment, and the results of bacteriological examination are scaled in points in 30%, totaling 100%. Below 60 scores are judged to be insanitary and administratively punished; 61-75 scores are judged to be average; 76-85 good; above 86 excellent. The sanitation inspection scores by the improved check-up list went up by two grades from 71.8 (before SSOP program) to 88.6 (after SSOP program). The results of bacteriological examination also improved from 57.5 to 98.2. The total scores of both results converted in the allotment of 70% and 30% also showed an improvement by two grades from 66.4 (before SSOP program) to 93.4 (after SSOP program). The above results show that the manufacturers participated in this study are following the 8 essential sanitary procedures which are needed to anchor the HACCP system.

The Effects of Expectancy-Disconfirmation of Attraction Affecting Visitor's Satisfaction in Natural Recreation Forests - Focused on Recreational Resources and Facilities - (자연휴양림(自然休養林) 매력물(魅力物)의 기대불일치(期待不一致)가 이용만족도(利用滿足度)에 미치는 영향(影響) - 자연휴양림의 휴양자원과 시설을 중심으로 -)

  • The purpose of this study was to identify the effects of expectancy-disconfirmation (ED) of attraction affecting visitor's satisfaction in natural recreation forest (NRF). After reviewing the literature about recreational resources (RRs), facilities, ED, and visitor's satisfaction in NRF, this study analogized 11 attractions of NRF and constructed the conceptual framework. This study had obtained data through a questionnaire, which surveyed 415 visitors at 3 NRFs in 204 summer season. This study found that 1) The expectancy-performance of visitor's interesting resources is the highest among RRs, 2) There was mean difference between expectancy and performance of attraction at one percent significant level, 3) Safety facility had the highest ED value and plant resources had the lowest ED value, 4) The higher ED value of attractions, the lower value of visitor's satisfaction, 5) in multivariate analysis, such variables as physical resource, social culture resource, lodging facility, foundation facility, safety facility, sanitary facility, and education facility have been turned out be statistically significant at one percent level, 6) The relative contribution of the ED of foundation facility, lodging facility, and education facility on visitor's satisfaction have ben determined to have respectively 2.50, 1.48, and 1.88 times more important than that of physical resources.

Impact of Pollution Sources on the Bacteriological Water Quality in the Yongnam-Gwangdo Shellfish Growing Area of Western Jinhae Bay, Korea (진해만 서부 용남·광도해역의 세균학적 수질에 미치는 육상 오염원의 영향)

  • To evaluate the bacteriological water quality in Yongnam-Gwangdo, located in western Jinhae Bay, seawater samples were analyzed using sanitary indicator bacteria at 57 sampling stations. According to survey results from January 2007 to December 2009, the range of the geometric mean and the estimated 90th percentile for coliforms and fecal coliforms in the samples were <1.8-16.5 and 1.8-246.8 MPN/100 mL and <1.8-7.1 and 1.8-74.8 MPN/100 mL, respectively. The samples, including those taken from stations located in Wonmunman, Gwangdo, and Dangdong, showed high levels of microbial contamination caused by the climate and weather patterns in the marine environment. The bacteriological water quality in the area met Korean criteria for a designated shellfish growing area for export and National Shellfish Sanitation Program criteria for an approved shellfish growing area, except at station #49. A total of 24 direct pollution sources were discharged into the shellfish growing area. The radius of impact was calculated for each pollution source to assess the effect on the shellfish growing area. The calculated radius of impact for most of the pollution sources was below 300 m. However, the radius of impact for the combined pollution sources in Kyeonnaeryang was 93-1973 m. There were significant differences between the calculated closed sea area and actual monitoring results. The closed sea area values calculated from the fecal coliform load in drainage water tended to be higher than the actual monitoring results. Tidal currents and environmental factors such as salinity, water temperature, sunlight, and microbiological factors affect the survival of fecal indicator bacteria in seawater.

  • Vibrio mimicus is a closely related species with V. cholerae, and has been reported to be associated with gastrointestinal infections. Although extraintestinal infections of these vibrios have also been reported in Japan and Southeast Asia. But little research papers on V. mimicus was reported in Korea. Therefore, we tried to isolate V. mimicus from the environmental sea water from April to July in Pusan, Korea. Among the isolated strains, we selected the strongest hemolytic strain and then named V. mimicus SM-9. In this paper, we checked the antibiotic susceptibility and psychrotrophic characteristics of the isolated strain. Hemolytic activity of the hemolysin produced by the isolated strain was also measured. V. mimicus was not detected from the sea water samples in April and May, but its detection rate was relatively high in June and July in Pusan, Korea. The bacteriological characteristics of V. mimicus SM-9 were Gram-negative rods, motile, oxidase positive, Voges-Proskauer negative and sucrose negative. In 23 kinds of antibiotics susceptibility test, V. mimicus SM-9 showed susceptibility to the most of antibiotics submitted while it was resistive against lincomycin, oxacillin, rifampin and vancomycin. Hemolytic activity of the hemolysin produced by V. mimicus SM-9 was highest in stationary phase of the growth curve in BHI broth at 37$^{\circ}C$ and its activity was reached 18 HU per $m\ell$ of culture supernatant. For checking the psychrotrophic property of V. mimicus SM-9, the decreasing rate of the strain in phosphate buffer solution and yellowtail flesh homogenate was examined during the storage at 4, 0, -4 and -2$0^{\circ}C$. The decreasing rates of the selected strain stored in phosphate buffer solution were greater than those in fish homogenate. Decreasing rates of V. mimicus SM-9 stored in phosphate buffer solution were not significantly different by the storage temperatures. The viable cell counts of the strain were decreased as 5 log cycles after 120 hours at all the tested temperatures. While decreasing numbers of the strain in fish homogenates were 2*4 log cycles after 120 hours. The decreasing pattern of the strain numbers were very slow after 200 hours at all the stored temperatures.

Evaluation of the Level of microbial Contamination in the Processing Company of Nuroong-ji (누룽지 생산시설에 대한 미생물학적 오염도 평가)

  • This study was conducted to evaluate the microbial contamination levels in the processing company of Nuroong-ji. Microbial contamination levels were examined for sanitary indication bacteria such as aerobic plate count, coliforms and fungi, and pathogenic bacteria such as Escherchia coli O157:H7, Salmonella spp., Staphylococcus aureus, and Listeria monocytogenes. Contamination levels were detected differently according to handling materials and purposing work-space. The equipments and raw materials were not seriously contaminated but there were necessary to attend the cross-contamination. A high contamination level was detected at the process where the interference of the employees was relatively higher than the other process. Standardization of the roasting process (l20~$170^{\circ}C$, about 10 min) could be necessary to control the microbial organism effectively on Nuroong-ji manufacturing process. At small/medium size foodstuff manufacturers, it is the most important to improve the recognition level of individual hygiene but also expand a hygiene facility.

Evaluation of the Influence of Inland Pollution Sources on Shellfish Growing Areas after Rainfall Events in Geoje Bay, Korea (강우에 따른 거제만해역 육상오염원의 영향평가)

  • The influences of inland pollution sources because of rainfall events on the bacteriological water quality in Geoje Bay, a major shellfish production area in Korea, were investigated. The sanitary status of sea water and shellfish after rainfall events was also evaluated. The flow rates of 13 streams around Geoje Bay showed 6 to 7-fold increases after 15 to 21 mm of rainfall. Peak pollution was observed in the Naegan Stream, the Gandeok Stream and the Seojeong Stream. The calculated impact area of inland pollution sources was 3.1 $km^2$ immediately after 15 mm of rainfall and expanded to 3.5 $km^2$ after 24 hours. These calculations of impacted area matched results from fecal coliform analyses with sea water. The distance between the major pollution source in the bay (the Gandeok Stream) and the station with the worst bacteriological water quality immediately after 15 mm of rainfall, which was below the Korean standard, was 0.8 km in a straight line; this distance increased to 2.0 km after a period of 24 hours. The area impacted by inland pollution sources after a 15 mm rainfall event was wider than after a 21 mm rainfall. Although the flow rate from inland pollution sources was higher, the concentration of fecal coliform in the discharged water was lower after higher rainfall events. These observations corresponded with the results of fecal coliform analyses with sea water samples. According to the evaluation of the influences of inland pollution sources and fecal coliform analyses on sea water and shellfish samples in Geoje Bay, pollutants from inland sources did not reach the boundary line of the shellfish growing area after rainfall events of 15 or 22 mm. The bacteriological water quality of the shellfish growing area in Geoje Bay met the Korean standard and US NSSP requirements for approved shellfish growing areas.
