The Effects of Pregnancy-Related Water Extract of 3 Types Herbal Medicines and Ethanol Extract of Moutan Radicis Cortex on the Endometrial and Placental Cells (임신 관련 3종 한약 처방 물 추출물과 목단피 에탄올 추출물이 자궁내막과 태반세포에 미치는 영향)

  • Objectives: This study was performed to investigate the effects of pregnancy-related four herbal medicines (Samul-tang, Onkyung-tang, Chokyungjongok-tang and Moutan Radicis Cortex) on the endometrial and placental cells. Methods: In this study, we examined viability and decidualization of telomerase immortalized human endometrial stromal cell lines (T-HESCs) and viability and invasion ability of human first trimester trophoblast cell lines Sw.71 by four herbal medicines (Samul-tang, Onkyung-tang, Chokyungjongok-tang and Moutan Radicis Cortex) Results: In the study, we showed that Samul-tang, Onkyung-tang, Chokyungjongok-tang increased decidualization marker prolactin (PRL) and insulin-like growth factor-binding protein 1 (IGFBP1) in T-HESCs. Moutan Radicis Cortex decreased the mRNA level of PRL and IGFBP1, and the protein level of PRL and IGFBP1 had no significant effect. Moreover, four herbal medicines reduced invasion ability of Sw.71 cells. Conclusions: These results suggest that Samul-tang, Onkyung-tang, and Chokyungjongok-tang have beneficial effects on successful embryo implantation and pregnancy maintenance by increasing decidualization markers such as PRL and IGFBP1. Moutan Radicis Cortex reduces the mRNA levels of PRL and IGFBP1, which may adversely affect pregnancy. Further investigations are needed to elucidate the significance of the decreased invasive ability of Sw.71 cells induced by four herbal medications.

The Microbiological Quality Estimation of Samul Chol-Pyon during the Storage (사물절편의 미생물학적 품질안전성)

  • This study was investigated to prove the relation between the ingredient of Samul Chol-Pyon and its microorganism pollution level during the storage. As a result, the pollution degree in total aerobic bacteria, yeast, mold, and colitis germs of rice power turned out to have a lower one than the oriental medicine materials do. In case of preserved write Chol-Pyon, the total aerobic bacteria pollution level was 8.8 $\times$ 10$^3$CFU/g the highest degree among other ones in their among other ones in their early pollution levels and in the oriental medicine materials, the pollution level was degreased as its annex increased. Moreover, yeast propagated fast in its first day of storage, but mold grew somewhat slowly than yeast and total aerobic bacteria did. In every case, the range of colitis germs growth was between 10$^2$-10$^3$CFU/g and it was similar to the each one of total aerobic bacteria, yeast, and molds. On its third day of storage, the pollution level of mold showed 10$^4$-10$\^$5/CFU/g.

Effect of Preservation Conditions on the Stability of Samul-tang Decoctions (사물탕 전탕액의 보관 조건에 따른 안정성 분석)

  • Consumer interest in the stability of medicinal herb extracts during storage has increased. Although the advent of new technologies has improved preservation conditions, increasing the storage time, there are few studies on the preservation of herb extracts. The purpose of this study was to perform microscopic observations of Samul-tang decoctions under various preservation conditions. The storage temperature (a high temperature, room temperature, with or without light, refrigeration, or cryopreservation) and storage time (0, 15, 30, 90, and 180 days) were given to each condition Macroscopic morphology, pH, UV absorption, HPLC, and bacteriological studies were performed to determine microscopic changes in Samul-tang decoctions. The biological activity (tyrosinase inhibition) of the Samul-tang decoctions was also examined. There were no major changes in the indicated observation items when the extracts were stored in each condition. However, at higher storage temperatures and longer storage times, microscopic changes increased, although no bacteria were detected. Furthermore, the higher the storage temperature was and the longer the storage time was, the bigger the change was, despite of minor microscopic changes. Therefore, to maintain the stability of herbal extracts during storage, it is recommended to keep the Samul-tang decoction in the preservation condition of refrigeration and cryopreservation or without light rather than high temperature and room temperature as possible.

Effect of Hyulbuchukeo-tang and its Component Groups on Alloxan Induced Hepatoxicity in Rats (혈부축어탕(血府逐瘀湯)과 그 구성약물군(構成藥物群)이 Alloxan 유도 당뇨(糖尿) 백서(白鼠)의 간독성에 미치는 영향)

  • This study was performed to observe the effect of hyulbuchukeo-tang and its component groups on recovery of hepatoxicity in alloxan treated rats. The experimental group was divided into 3 groups: hyulbuchukeo-tang(HCT) and its components groups, sayeok-san(H-sa) and dohong-samul-tang (H-do). The results were obtained as follows: 1. The level of Glucose was considerably reduced by HCT, H-sa, H-do with high significancy. 2. In the change of GOT activity, HCT and H-do were decreased with high significancy, H-do was decreased with some significancy. 3. In the change of GPT activity, HCT was decreased with high significancy, H-sa and H-do were decreased with some significancy. 4. In the change of ${\gamma}-GPT$ activity, HCT was decreased with high significancy, H-do was decreased with some significancy. 5. In the change of ALP activity. only HCT was decreased with some significancy. 6. In the change of LDH activity, HCT was decreased with high significancy, H-sa and H-do were decreased with some significancy. 7. In the change of bilirubin contents. only HCT was decreased with some significancy. As result, hyulbuchukeo-tang(HCT) has significant effects on recovery of hepatoxicity, and its components groups. sayeok-san(H-sa) and dohong-samul-tang(H-do) have some effects. It suggests that sayeok-san(H-sa) and dohong-samul-tang(H-do) have more effects on recovery of hepatoxicity in case of the combination of the two (that is hyulbuchukeo-tang) than in case of the separation.

Case of a Hemorrhagic Cystitis Patient Taking Antiplatelets Treated with Jeoryeong-tang-hap-Samul-tang (항혈소판제제를 복용 중인 출혈성 방광염 환자의 저령탕합사물탕 치험 1례)

  • Hemorrhagic cystitis refers to massive inflammation and the diffuse vesical bleeding of the bladder. In patients with hematuria complaining of dysuria and pain, it is necessary to differentiate various causes, including cystitis, nephritis, and prostatitis. After the diagnosis of hemorrhagic cystitis, antibiotics usually treat and prevent further urinary tract infections. In the present case, a 78-year-old female patient with hemorrhagic cystitis presenting with hematuria underwent Korean medical treatment with Jeoryeong-tang-hap-Samul-tang for 29 days. The effect of the treatment was assessed with the hematuria grading scale (HGS) per week, urinalysis per two weeks, and the NRS (numeric rating scale) of dysuria per day. After treatment, both HGS and NRS scores decreased, and protein, blood, and red blood cells (RBCs) in urinalysis improved. This case report suggests that Jeoryeong-tang-hap-Samul-tang might be an effective option for hemorrhagic cystitis patients who continuously take antiplatelets.

Single Oral Dose Toxicity Evaluation of Samul-tang, a Traditional Herbal Formula, in Crl:CD (SD) Rats

  • Background: Samul-tang (Si-Wu-Tang, SMT) is a traditional herbal formula, which has been widely used to treat various diseases such as menstrual irregularity, bleeding and leucorrhea. Although many studies have investigated the pharmacological properties of SMT, its toxicity information has not yet been fully elucidated. Methods: Five Sprague Dawley (SD) rats of each sex were given a single dose (5000 mg/kg) of SMT by gavage; control rats received the vehicle only. After the single administration, mortality, clinical signs, body weight changes and gross findings were monitored for 15 days in accordance with Good Laboratory Practice (GLP) principles. Results: In a single oral dose toxicity study, there was no adverse effect on mortality, clinical sign, body weight change or gross finding in any treatment group. Conclusions: The results indicate that SMT did not induce toxic effects at a dose level up to 5000 mg/kg in rats and its median lethal dose ($LD_{50}$) was considered to be over 5000 mg/kg/day body weight for both genders.

Experimental Study of Samul-tang Extract and Bambusae caulis in liquamen on the Regional Cerebral Blood Flow and Mean Arterial Blood Pressure in Normal Rats (사물탕합죽력(四物湯合竹瀝)이 국소 뇌혈류량과 평균 혈압에 미치는 실험적 연구)

  • This study was designed to investigate the effects of Samul-tang Extract and Bambusae caulis in liquamen(SEB) on the changes in regional cerebral blood flow (rCBF) and mean arterial blood pressure (MABP) in normal rats. The results were as follows ; SBE significantly increased rCBF in a dose-dependent manner, and increased MABP in a dose-dependent manner. This result suggests that SBE significantly increased rCBF by dilating pial arterial diameter. Increase of SBE-induced rCBF was significantly inhibited by pretreatment with indomethacin (1 mg/kg, i.p.), an inhibitor of cyclooxygenase and increase of SBE-induced rCBF was inhibited by pretreatment with methylene blue ($10\;{\mu}g/kg$, i.p.), an inhibitor of guanylate cyclase. Increase of SBE-induced MABP was significantly increased by pretreatment with methylene blue but increase of SBE-induced MABP was decreased by pretreatment with indomethacin. These results suggested that the action of SBE was mediated by cyclooxygenase.

Immuno stimulatory activities of Samul-tang, Sagunja-tang, Pamul-tang and Sipjeondaebo-tang in vitro (사물탕, 사군자탕, 팔물탕, 십전대보탕의 약리 활성 비교 연구)

  • Objectives : Samul-tang(SM). Sagunja-tang(SG). Pamul-tang(PM) and Sipjeondaebo-tang(SJ) was used many diseases such as sterility. menstrual disorder. general prostration. recruitment in Korea. We investigated the immune stimulatory activities of SG. SM. PM and SJ in in vitro. Methods: For comparision for effective of SM. SG. PM and SJ. this study examined anti-inflammation(NO. PGE2 assay). anti-oxidation(DPPH assay) and immune response in in vitro assay. For immune response activities. this study used NO synthesis on RAW 264.7 cells, splenocyte proliferation and cytokine assay(IL-2, IL-4) in splenocyte. Results: The results showed that SG. SM. PM and SJ were no significant effect anti-oxidation and anti-inflammatory effects. For immune response. they showed the splenocyte proliferation and macrophage proliferation. We confirmed that they synthesised NO a dose-dependent manner significantly and secreted the IL-4. Conclusions : These results suggested the SG, SM, PM and SJ had immune stimulatory activity. A efficacy of immune response in them had produced similar results.

Regulatory Effects of Samul-tang on Axonal Recovery after Spinal Cord Injury in Rats

  • In oriental medicine, Samul-tang (SMT) has been used for the treatment of cardiovascular diseases and neuronal disorders. Here, possible effects of SMT on axonal regeneration after the spinal cord injury were examined. SMT treatment induced increases in regeneration-related proteins GAP-43, cell division cycle 2 (Cdc2) and phospho-Erk1/2 in the peripheral sciatic nerves after crush injury. Increased levels of Cdc2 and phospho-Erk1/2 were observe mostly in the gray matter area and some in the dorsomedial white matter. These increases correlated with increased cell numbers in affected areas. Moreover, axons of corticospinal tract (CST) showed increased sprouting in the injured spinal cord when administrated with SMT compared with saline-treated control. Thus, the present data indicate that SMT may be useful for identifying active components and for therapeutic application toward the treatment of spinal cord disorders after injury.

Analysis of Studies on Samul-tang for Fundamental Establishment of Evidence Based Medicine (EBM 기반구축을 위한 사물탕 연구 문헌 분석)

  • To establish the fundament for EBM of Traditional Korean Medicine, the papers on Samul-tang which was frequently used in medical institutions of Traditional Korean Medicine were analyzed through researching domestic and international literatures. The papers were classified by the registration of domestic or international journals, by the year of publishment, by experimental methods, by laboratory animals used in biological experiment and by the kinds of studies on biological efficacy. Of total 67 papers on Samul-tang, 58 volumes were registered in domestic journals and 9 volumes were in international journals of which 8 volumes were in SCI journals. Since 1978, publishments of papers have continuously increased. The papers on instrumental analyses were 6, biological studies were 58 volumes, clinical studies were 3. Instrumental analyses were preceeded with standard compounds(gallic acid, albiflorin, paeoniflorin, benzoic acid, ferulic acid, 5-HMF). And biological studies showed improvement of cardiovascular function and circulation, antianemia, brain protection, immunoregulation, antistress, radioprotection, antifatigue, antiinflammation and antiallergy, antioxidative effect. Through clinical studies, antifatigue, improvement of insomnia and osteoporosis were reported. Samul-tang could be used to tonify and activate blood. And further study on clinical field need to be conducted in accordance with biological study.
