This study was performed to determine the antimutagenic and cytotoxic effect of Aster scaber root ethanol extract on Salmonella typhymurium TA98, TA100 and cancer cell lines using Ames test and cytotoxicity assay, respectively. Cancer cell lines include chronic myelogenous leukemia(K562), human gastric carcinoma(KATOIII), human hepatocellular carcinoma(Hep3B) and human breast adenocarcinoma(MCF-7). Futher fractionations with hexane, chloroform, ethyl acetate, butanol and water from ethanol extract of Aster scaber root were performed to obtain effective fraction. Ethanol extract and ethyl acetate fraction showed 79% and 82% inhibitory effect on the mutagenesis induced by N-methyl-N'-nitro-N-nitrosoguanidine(MNNG) against TA100, while 48% and 60% inhibition was observed on the mutagenesis induced by 4-nitroquinoline-l-oxide(4NQO) against TA98. In the meanwhile, ethyl acetate fraction showed 78% and 85% inhibitory effect on the mutagenesis induced by benzo(${\alpha}$)pyrene[B(${\alpha}$)P] against TA98 and TA100, respectively, while 83% inhibition was observed on the mutagenesis induced by 3-amino-l,4-dimethyl-5H-pyrido(4,3-b) indole(Trp-P-1) against TA98. Ethyl acetate fraction (0.125 mg/ml) showed the strongest cytotoxic effect against K562, KATOIII, Hep3B and MCF-7 at the same concentration compared to those of other fractions. Ethanol extract and water fraction showed the least inhibitory effect.

The effects of dietary copper (Cu) supplementation in broiler chickens challenged with a single injection of Salmonella typhymurium lipopolysaccharide (LPS) on the antioxidant capacity and plasma levels of ceruloplasmin (Cp) were evaluated. The broiler chickens were provided with a basal diet or diets supplemented with 8 and 50 mg/kg Cu from 1d of age. At 25d of age, 48 chickens with similar body weight were selected from each diet. Half of the chickens in each dietary treatment were injected intraperitoneally (i.p.) with LPS (1 mg/kg body weight). The other half was injected with saline, serving as the control. Body weight gain and feed consumption were significantly suppressed by LPS challenge during the first 12-h after injection, regardless of dietary Cu levels. Feed efficiency was reduced by LPS injection during the 72-h experimental period. Dietary Cu levels had no significant effect (p>0.05) on the plasma ceruloplasmin concentrations in chickens injected with saline. In contrast, high dietary level of Cu elevated plasma Cp levels in chickens with LPS challenge. Short-term LPS challenge had no significant effect on the antioxidant ability of broilers, as indicated by superoxide dismutase, ferric reducing/antioxidant power and the thiobarbituric acid reacting substances in the plasma. The result suggests that high dietary Cu level (as much as 50 mg/kg supplementation) is favorable for coping with short-term LPS challenge through upregulating plasma Cp levels.
The antimicrobial activities of extracts of Prunus mume by sugar against food poisoning bacteria, and the effects of heat and pH treatment on these antimicrobial activities, were investigated. The level of total solids, pH, and acidity of P. mume extracts were 55.08% (w/w), pH 3.1, and 1.52%, respectively. P. mume extracts showed the strongest antimicrobial activity against Vibrio parahaemolyticus, among the bacteria tested. P. mume extracts significantly inhibited the growth of V. parahaemolyticus, Bacillus cereus, and Staphylococcus aureus at levels of 1-2% (w/v) of extracts in media. The antimicrobial activities of P. mume extracts were neither affected by heating at $65-125^{\circ}C$ for 30 minutes, nor by neutralization of extract to pH 7.0.

Effect of dietary brown seaweed(Undaria pinnatifida) levels on the anti-oxidant enzyme system was evaluated in blood of broiler chicks activated innate immune response. Day-old broiler chicks were fed diets containing 0.0(basal), 1.0, 2.0 and 4.0 % of brown seaweed for 4 weeks. The innate immune response was activated by injecting Salmonella typhymurium lipopolysaccharide(LPS) i.p. at 8, 10 and 12 day of age. The activation of innate immune response enhanced(p< 0.01) and the brown seaweed 1.0 and 2.0 % diets reduced(P< 0.05) the superoxide dismutase(SOD) activity in erythrocyte cytosol significantly. The activation of innate immune response elevated significantly the levels of peroxide and the activity of peroxidase in plasma, while the levels of dietary brown seaweed resulted in a significant linear increase in peroxidase activity in plasma of normal bird. Experience of the innate immune response in 4 week-old chicks reduced linearly the plasma level of peroxide with the level of brown seaweed and enhanced the plasma peroxidase activity. Also, the plasma of normal birds fed the brown seaweed showed higher level of peroxide and lower activity of peroxidase. The results indicated that dietary brown seaweed affected SOD and peroxidase activities in blood of broiler chicks during activation of innate immune response.

Three experiment were designed to determine the effects of dietary(n-3) fatty acids on the growth-suppressive effects of the inflammatory response and anti-oxidant enzyme activity and balances of nitrogen and energy. Day-old chicks were fed diets containing 1.0 % soybean oil in experiment 1, or diets containing 1.0 % of fish oil in experiment 2, and then on 7-d of age chicks were fed diets containing 1.0 % of either soybean oil or fish oil. In experiment 3, day-old chicks were fed on diets containing 1.0 % of either soybean oil or fish oil. In chicks of half pens within each dietary treatments of each experiments, the acute phase response were induced by injecting with Salmonella typhymurium lipopolysaccaride(LPS) i.p. 3 times at 8, 10 and 12-d of age. During acute phase response, chicks fed dietary fish oil from 1-d of age enhanced energy balances and did not give negative effect on nitrogen balance and uric acid excretion, but elevated MnSOD and Cu/ZnSOD activity In liver and erythrocyte cytosols compared with those fed dietary soybean oil. The results indicated that enhanced daily gain and metabolizable energy value of diet in birds fed fish oil from one-d of age seems due to change on the antioxidants systems during acute phase response.
This research was performed to investigate the immnomodulative effects of ploysaccharides extracted from the fruiting body of Agarcus blazei cultivated with the media which are fermented with sugar cane bagasse containing Pueraria thunbergiana in open-air storage. In MTT test, methanol extracts from the fruiting body of A. blazei cultivated with P. thunbergiana media showed in colon carcinoma line(HT29) by 1.5∼3.5 fold and human heptoma cell line (HepG2) by 1.3 ∼2.4 fold antitumor activites compared to two types media (rice straw plus sugar cane bagasse, rice straw only) often used in the fauns. To clarify the antimutagenic principles, three extracts, Ab-l, Ab-2 and Ab-3, were separated by the solvent fractionations such as hot water, cold & hot sodium hydroxide respectively, and their antimutagenic effects was determined against N-methyl-N'-nitro-N-cnitrso-guanidine(MNNG) using Salmonella typhymurium. There was no significant differencies of inhibition levels among the used media, but Ab-3 tractions still showed a high antimutagenicity in the Ames test regardless of cultivating areas or media. To prove the cell immunofunction, nitric oxide (NO) produced from Raw 264.7 matrophage cultured with three fractions (Ab-l, Ab-2, Ab-3) was measured, and showed generally increase about 45 ∼58 percent compared to another two media (rice straw plus sugar cane bagasse, rice straw only), in the fraction of hot alklai extracts of the fruiting body cultivated with P. thunbergiana, which means that the media selection could be very important factors for improving medicinal effects in agaricus blazei fruiting body.
In this study, the effects of dietary krill meal levels on cellular immunity in LPS-injected broiler chicks was evaluated. One day-old male broiler chicks(Ross) were fed on the experimental basal meal(0.0% krill meal), or diets containing 0.5%, 1.0% and 2.0% krill for 3 weeks, and the acute phase response was activated by intraperitoneally injection of Salmonella typhimurium lipopolysaccharide(LPS) 3 times at 9, 11, and 13 days of age. 1. Acute phase response induced a significant reduction in(p<0.05) daily weight gain and feed intake, and increases in liver and spleen weight. However, it was not affected by dietary krill meal levels. 2. The krill meal diets reduced TNF-α activity as compared to the basal diet after 24 hours (acute phase response) and 1 week(recovery from the acute phase response) following LPS injection (p<0.05). The acute phase response induced a significant increase(p<0.05) in TNF-α activity relative to the control in chicks fed on a basal diet, but this was also unaffected by dietary krill meal levels. 3. Acute phase response-mediated ovotransferrin levels(relative to what was measured in the control bird) were increased in birds fed on the basal, 1.0% and 2.0% krill diets, and were reduced in birds fed on the 0.5 % krill diet. 4. In LPS-injected chicks, 1.0% and 2.0% krill meal diets induced a significant reduction in(p<0.05) the Con A-induced proliferation of PBMC and splenocytes relative to what was observed in the chicks fed on a 0.5% krill diet, whereas the splenocytes proliferated in a linear fashion with the krill levels in the diets of the control birds. The results showed that the dietary levels of krill meal reduced TNF-α activity in the blood and also influenced blood ovotransferrin levels and the proliferation of PBMC and splenocytes, and krill meal is considered to be associated with both innate and cellular immunity in broiler chicks.
To select new useful domestic plants with antimicrobial activities, thirty five samples of berry plant were distributed from Plant Diversity Research Center in Korea Research Institute of Bioscience and Biotechnology. Their extracts with methanol were tested against Escherichia coli, Salmonella typhimurium, Staphylococcus aureus, Listeria monocytogenes by paper disc method. The methanol extracts from Viburnum dilatatum, Viburnum erosum, Aralia continentalis, Camellia japonica, Acer trumcatam, Arasaema takesimense and Rhysocarpus intermedius were effective against S. aureus and the results were as follows: Viburnum erosum 6.5 mm, Viburnum dilatatum and Aralia continentalis 7.0 mm, Rhysocarpus intermedius 8.0 mm, Acer trumcatum 9.0 mm, Camellia japonica and Arisaema takesimense 9.5 mm. And only the methanol extract from Camellia japonica was effective against L. monocytogenes, 7.0 mm. Antimicrobial activities of E. coli and S. typhymurium were not detected. The minimum inhibitory concentration(MIC) of berry of Camellia japonica was examined as 1,250 $\mu\textrm{g}$/mL against S. aureus and as 1,250∼2,500 $\mu\textrm{g}$/mL against L. monocytogenes.

Influences of dietary brown seaweed(BSW) on the nutrient metabolism, anti-oxidant enzyme activity and cell-mediated immune response were studied in broiler chicks activated acute phase response. 72 Hatched male broiler chicks(Ross) were divided into 12 pens, 6 heads per pen, and fed the BSW 0.0% (Basal) or 2.0% diet, respectively, and injected with the Salmonella typhimurium lipopoly saccharide(LPS) for activation of the acute phase response three times at 8, 10 and 12 d of age. During 4 wks of experimental feeding, growth performance of broiler chicks was not affected by dietary BSW and the acute phase response. Compared with control birds, the acute phase response did not affect the daily weight gain in birds fed BSW 2.0% diet, decreased nitrogen balance(NB) or metabolizable energy(ME) utilization per metabolic body size(kg0.75), and enhanced activities of peroxidase or extracellular SOD(EcSOD), tumor necrosis factor-alpha and ovotransferrin in plasma and MnSOD and CuZnSOD in erythrocyte cytosol. Compared to BSW 0.0% diet, 2.0% diet enhanced protein retention(NB) per kg0.75 regardless the acute phase response, did not affect uric acid nitrogen excretion(UAN) per kg0.75 in birds during the acute phase response, decreased(p<0.05) the UAN excretion per kg0.75 in control birds. And BSW 2.0% diet also decreased(p<0.05) plasma peroxide level and erythrocyte peroxidase or MnSOD activity but increased plasma peroxidase and EcSOD activity and interleukin-1 activity secreted from LPS-stimulated PBMC in 4 week broiler chicks.
Cirsium japonicum var. ussuriense were extracted with methanol and then fractionated with nhexane, EtOAc and BuOH to get active fractions. And their antioxidant and antimicrobial activities in each fraction were determined. Ethyl acetate and butanol fraction of Cirsium japonicum var. ussuriense showed strong antioxidant activities, but hexane fraction did not show any activities. But in the antimicrobial test, Ethyl acetate fraction showed strong antimicrobial activities except to Aspergillus awamori, Asperigillus niger. Especially, Ethyl acetate fraction showed the strongest activities against Bacillus subtilis. And aqueous fraction showed the strongest activities against Cladosporium herbarum, Hypocrea nigricans. This study was performed to determine the antimutagenic and cytotoxic effect of Cirsium japonicum var. ussuriense methanol extract on Salmonella typhimurium TA98, TA100 and cancer cell lines using Ames test and cytotoxicity assay, respectively. Cancer cell lines include human lung carcinoma(A549), human breast adenocarcinoma(MCF-7) and human hepatocellular carcinoma (Hep3B). Futher fractionations with hexane, ethyl acetate, butanol and water from methanol extract of Cirsium japonicum var. ussuriense were performed to obtain effective fraction, methanol extract showed 60.14% inhibition effect on the mutagenesis induced by MNNG against TA100, while 77% and 72.5% inhibition was observed on the mutagenesis induced by 4NQO against TA98 and TA100, respectively. and methanol extract showed 82.25% and 73.7% inhibitory effect on the mutagenesis induced by Trp-P-1 against TA98 and TA100, respectively. methanol extract showed the strongest effect against A549, MCF-7 and Hep3B at the same concentration compared to those of other fration.
