Effects of Some Metabolic Inhibitors on Phototactic Movement in Cyanobacterium Synechosystis sp. PCC 6803 PTX (람세균 Synechocystis sp. PCC 6803 PTX의 주광성 운동에 미치는 몇가지 대사 억제제의 효과)

  • For understanding physiological nature of phototaxis in Synechocystis sp. PCC 6803 PTX(S. 6803 PTX), we examined the effects of some metabolic inhibitors and cation ionophore on the phototactic movement. In the presence of DCMU, which blocks the photosynthetic electron transport just after photosystem II acceptor, there was no inhibitory effect on the phototaxis up to $100\;\mu\textrm{M}$. Instead, the respiratory electron chain inhibitor such as sodium azide dramatically impaired the phototaxis in S. 6803 PTX. These observations indicate that the phototaxis is linked not to photo-phosphorylation, but to respiratory phosphorylation. When the cells were treated with un couplers such as CCCP or DNP, which dissipate the electrochemical gradient of proton($\Delta\mu_{H}+$) across the cytoplasmic membrane, these chemicals did not affect phototaxis. In contrast, when cells were treated with DCCD or NBD which deprive cells of A TP but leave $\Delta\mu_{H}+$ intact across the membrane, the phototactic movement was severly reduced. These results imply that ATP production, not proton motive force, is involved in the phototactic movement in this organism as a driving motive force. The application of specific calcium ionophore A23187 strongly impaired positive phototaxis. Calcium fluxes should be engaged in the sensory trans-duction of phototactic orientation. Finally, when ethionine was supplimented to culture media, the photomovement of this organism was inhibited. This implies that methylation/demethylation mechanism controls the process of phototaxis in S. 6803 PTX like chemotaxis in E. coli and Salmonella typhimurium.murium.

Evaluation of Antimicrobial Activity of Farnesoic Acid Derivatives

  • The biological activities of farnesoic acid derivatives against pathogenic fungi and bacteria were investigated. Farnesoic acid and its derivatives showed growth inhibitory activities against various bacteria. Among the compounds tested, geranylgeranoic acid (3) had potent antibacterial activity against Salmonella typhimurium, Proteus vulgaris, and Bacillus subtilis with minimum inhibitory concentration (MIC) in the range of $6.25-12.5{\mu}g/ml$. On the other hand, amide derivatives of farnesoic acid showed some antifungal activities. In particular, 3,7,11-trimethyl-dodeca-2,6,10-trienoic acid amide (5a) had a potent antifungal activity against Aspergillus niger, Candida albicans, and Trichophyton sp. with MIC in the range of $6.25-12.5{\mu}g/ml$.

  • Of 80 cases of Salmonellosis reported in a provincial area in 1992, 90% were related to outbreaks, which occurred from Apr. to Nov. And the serotypes include the following: typhimurium, hadar, and enteritidis etc. Among these outbreaks the authors carried out the epidemiological investigation for 'I-eup'(highest attack region) outbreak. The results obtained were as follows: 1. The S. typhimurium isolated from stools of 7 cases among 29 cases. 2. The common-source of exposure was boiled pork and/or skewered slices of seasoned meat. 3. The incidence rate was 65.1%(29 cases). 4. The incidence showed significant difference between males(32.1%) and females(80.0%). 5. The incidence in under 20 years of age and 40 years of age were much higher than other age groups but there was no significant difference among much age groups. 6. The mean and the median incubation period were 18.5 and 17.0 hours respectively. 7. The secondary attack was not appeared.

The Effects of the Fermentation Broth of Fruits and Vegetables on Antimicrobial Activity and Egg Quality with its Supplementation in Feed (과채발효액의 항균 및 탈취력 효과와 사료 첨가시 계란 품질에 미치는 영향)

  • This study was conducted to investigate the effect of the fermentation broth of fruits and vegetables and various kinds of lactobacillus containing in the broth on anti-microbial activity, anti-acidity, its organic acid concentrations and deorderization. In addition, the quality of eggs laid from the hens supplemented with the fermentation broth was determined. The fermentation broth contained over 8 kinds of lactobacillus and other microbes. Lactobacillus (L.) hilgardii, L. reuteri, L. nagelii, L. plantarum, and Zygosaccharomyces bisporus were survived especially in pH 2.0 among them. The concentrations of acetic acid and lactic acid in the broth were higher than minimum inhibitory concentration values (MIC) to Escherichia (E.) coli and Salmonella sp. Overall the broth contained sufficient organic acid to inhibit the growth of E. coli and Salmonella sp. In addition, L. nagelii and L. plantarum had the greatest anti-microbial activity against E. coli, Salmonella (S.) typhimurium, Staphylococcus aureus and S. gallinarium among lactobacillus in the broth. The broth had greater anti-microbial activity than individual lactobacillus. The fermentation broth had deodorization capacity of ammonia and hydrogen sulfate and the 50% of their concentrations were removed after 30 min treatment. Egg production rates, egg weight, and feed efficiency were increased in eggs laid from hens that supplemented with 0.1 % fermented broth compared to the control. The supplementation also increased the thickness and strength of egg shell to reduce broken eggs and decreased cholesterol levels in egg yolk. In conclusion, the fermentation broth of fruits and vegetables can be a natural alternative supplementation of feed in laying hens.

Biochemical and Molecular Identification of Antibacterial Lactic Acid Bacteria Isolated from Kimchi (김치에서 항균활성 유산균의 분리 및 동정)

  • Total 480 lactic acid-producing bacteria were isolated from five kinds of kimchi, and their antibacterial activity was tested against Salmonella enterica serovar Typhimurium, Bacillus subtilis, and Pseudomonas aeruginosa using an agar diffusion assay. Among them, 340 isolates showed a halo on MRS agar against one or more indicator strains, which were identified using multiplex PCR, an API 50CHL kit, and a 16S rDNA sequence analysis. As a result, 169 Lactobacillus plantarum, 20 Lactobacillus fermentum, two Lactobacillus paracasei ssp. paracasei, two Lactobacillus sp., and 15 Pediococcus sp. were identified. This may be the first report on the isolation of antibacterial Lactobacillus fermentum from kimchi.

Changes in Quality Characteristics of Cheongkukjang added with Deodeok (더덕이 첨가된 청국장의 품질특성 변화)

  • In order to demonstrate the usefulness of Deodeok. the functions of Deodeok and the qualities of Cheongkukjang were investigated. DPPH (1.1-diphenyl-2-picryl-hydrazyl) radical scavenging activity of Deodeok extracted with 70% ethanol was higher than that of water extract. The direct antimutagenic effect of ethanol extract of Deodeok was examined by Ames test using Salmonella typimurium TA 98. The inhibition rates on ethanol extract at concentrations of 200, 1.000, 2.000, 3.000 and 4.000 ${\mu}g$/plate were 5.75, 31.38, 34.75, 53.50 and 83.75%, respectively. The inhibition rates of ethanol extract was higher over 2 times than that of water extract. The qualities including physicochemical and sensory properties of Deodeok Cheongkukjang were investigated over the following range of Deodeok levels; 5, 10, 15 and 20% (w/w). The strain used in Cheongkukjang manufacture was Bacillus sp. B-3 with the highest enzyme activities such as amylase and protease. During fermentation at $40^{\circ}C$ for 6 days. amino-type nitrogen content of Cheongkukjang containing 10% (w/w) Deodeok was more than others. When Deodeok content exceeded 15% (w/w), higher contents resulted in lesser amino-type nitrogen production. The results showed that Deodeok had influenced the growth of Bacillus sp. B-3. The L-value of Deodeok Cheongkukjang was decreased according to increasing the Deodeok contents. Sensory evaluation showed that Deodeok Cheongkukjang containing 10% (w/w) Deodeok was superior to other tested.

Standardization of Kinds of Ingredient in Chinese Cabbage Kimchi (배추김치의 재료 종류 표준화)

  • Kinds of ingredient of Chinese cabbage kimchi were standardized by the sensory evaluation, chemical properties, and functional properties of antimutagenic effect and inhibitory effect on the growth of AGS human gastric adenocarcinoma cells. The kinds of ingredient in control kimchi from the previous study, but Gueun salt instead of Chunil salt, exhibited better overall acceptability and less moldy smell and moldy flavor than any other kinds of ingredient added chinese cabbage kimchi in the taste. The kimchi showed chemical properties of properly fermented kimchi, pH 4.3 and acidity 0.72% and also contained 1.6 g% reducing sugar and $2.2{\times}10^8\;CFU/mL$ Leuconostoc sp. The juice of standardized kimchi with the above kinds of ingredient showed not only high antimutagenicity (74%) against aflatoxin $B_1$ in Salmonella typhimurium TA100 but also strong inhibitory effect (60%) on the growth of AGS human gastric adenocarcinoma cells in SRB assay. From the taste, chemical and functional properties, the standardized kinds of ingredient were Youngyang taeyangcho red pepper powder, anchovy juice, Gueun salt, Garak sin 1 ho Chinese cabbage.

Effect of Supplementary Actinomycetes (Nocardia sp. CS682) Ferment on the Performance, Blood Parameters, Immunoglobulin and Small Intestinal Microflora Contents in Broilers (방선균(Nocardia sp. CS682) 발효물의 급여가 육계의 생산성, 혈액성상, 면역글로불린 및 소장 내 미생물 함량에 미치는 영향)

  • This study was conducted to investigate the effects of dietary supplementation of CS682, a fermentation product of Actinomycetae Nocardia sp. CS682, and DCS682$^{(R)}$, a commercial product, on the performance, blood parameters, small intestinal microflora, and immunoglobulin contents in broilers. In Exp. 1, a total of 240 ROSS$^{(R)}$ broiler chickens of 1d old were assigned to six dietary treatments: Control, Antibiotics (6 ppm avilamycin), CS682-0.25 (CS682 0.25%), CS682-0.50, CS682-0.75 and CS682-1.00. There were significant (p<0.05) differences among treatments in feed conversion. The CS682-0.25 treatment was significantly (p<0.05) lower than Antibiotics and other CS682 treatments in 0~2 wk feed conversion. The CS682 treatments influenced MCV (mean corpuscular volume) in blood. The cfu of Escherichia coli in small intestinal content was lowest in Antibiotics treatment followed by CS682 treatments and Control. In Exp. 2, a total of 1,000 ROSS$^{(R)}$ broiler chickens of 1 d old were assigned to five dietary treatments: Control, Antibiotics (6 ppm avilamycin), DCS682-0.05 (DCS682$^{(R)}$ 0.05%), DCS682-0.10 and DCS682-0.20. There were significant differences (p<0.05) among treatments in mortality. The DCS682-0.20 treatment was lower than DCS682-0.10 in 0~3 wk and lower than Control in 0~5 wk mortality. Antibiotics treatment was lowest in all microbial population in small intestinal content. The cfu of E. coli and Salmonella typhimurium of DCS682 treatments were higher than Antibiotics treatment but lower than the Control. The results of present broiler experiments indicated that supplementation of 0.20~0.25% CS682 and DCS682, improve feed conversion, mortality and control harmful intestinal microbes.

The First Report to Evaluate Safety of Cyanobacterium Leptolyngbya sp. KIOST-1 for Use as a Food Ingredient: Oral Acute Toxicity and Genotoxicity Study

  • Leptolyngbya sp. KIOST-1 (LK1) is a newly isolated cyanobacterium that shows no obvious cytotoxicity and contains high protein content for both human and animal diets. However, only limited information is available on its toxic effects. The purpose of this study was to validate the safety of LK1 powder. Following Organisation for Economic Co-operation and Development (OECD) guidelines, a single-dose oral toxicity test in Sprague Dawley rats was performed. Genotoxicity was assessed using a bacterial reverse mutation test with Salmonella typhimurium (strains TA98, TA100, TA1535, and TA1537) and Escherichia coli WP2 uvrA, an in vitro mammalian chromosome aberration test using Chinese hamster lung cells, and an in vivo mammalian erythrocyte micronucleus test using Hsd:ICR (CD-1) SPF mouse bone marrow. After LK1 administration (2,500 mg/kg), there were no LK1-related body weight changes or necropsy findings. The reverse mutation test showed no increased reverse mutation upon exposure to 5,000 ㎍/plate of the LK1 powder, the maximum tested amount. The chromosome aberration test and micronucleus assay demonstrated no chromosomal abnormalities and genotoxicity, respectively, in the presence of the LK1 powder. The absence of physiological findings and genetic abnormalities suggests that LK1 powder is appropriate as a candidate biomass to be used as a safe food ingredient.

From food wastes into useful probiotics: Development of Lactic acid bacteria as useful probiotics for animal feed without antibiotics using food waste (음식물류폐기물의 자원화를 위한 항세균 활성 유산균의 개발)

  • For the conversion of food waste into a good recycling material such as animal feed without antibiotics, thirteen lactic acid bacteria, which can be used as good probiotics for animal feed, were isolated from the intestine and feces in pigs. All isolates showed strong tolerance to high salt (4% of NaCl), acid(pH4.0), and bile juice (0.8% of oxgall). The growth rate was best at $37^{\circ}C$ in all strains. Among the isolates, Lactobacillus plantarum CJY-22, L. brevis CJY-42, L. arizonensis CJY-3, and Pediococcus sp. CJY-41 showed higher and broader spectrum of antimicrobial activities against six different pathogens such as Salmonella, typhimurium. L. plantarum CJY-22 has also grown well at $25^{\circ}C$, making this strain as an appropriate candidate for the fermentation of food waste at room temperature, thus conducting the fermentation process cost-effectively.
