Drug Resistance Profiles of Salmonella sp. and Shigella sp. Isolated from Diarrheal Patients in Pusan, Korea (부산지역의 설사환자로부터 분리한 Salmonella sp. 및 Shigella sp.의 약제내성 유형에 관한 연구)

  • A total of 34 Salmonella sp. an d25 Shigella sp. were isolated from 311 patients with diarrhea. The isolation rates of Salmonella sp. ad Shigella sp. were 10.9% and 8%, respectively. The serogroups of 34 Salmonella sp. were in order of group D(50%), group B(38.25), group E(8.8%) and group C 92, 9%0. the serogroups of 25 Shigella sp. were group D(96%) and group B(4%). Seasonal distribution of isolated Salmonella sp. and Shigella sp. were shown the most high at July, 17.65% and 64%, respectively. Age group distribution of isolated Salmonella sp. and Shigella sp. were shown the most high at twenties and thirties (23.5%), and teens(76%), respectively. The Salmonella isolates were resistant in order of prevalence use of streptomycin(SM) (100%), erythromycin (EM) and movobiocin (NB)(90.6%), penicillin G(PG) (65.6%) and cephalexin (CPX)(46.9%). the isolates of Shigella sp. were resistant in order of prevalence use of EM (95.8%), NB(91.7%), SM(87.5%). Eighteen kinds of resistant patterns of Salmonella ioslates were detected. The multiple resistance patterns of Shigella isolates were mostly SM, EM, NB type (79.2%). The minimum inhibitory concentration of Salmonella sp. and Shigella sp. and Shigella sp. isolated from patients with diarrhea were tabulated.

Production and Characterization of Egg Yolk Antibodies (IgY) against Flagella Antigen of Salmonella sp. (살모넬라 편모 항원에 대한 난황항체(IgY)의 생산 및 특성)

  • Egg yolk antibodies(IgY) from laying hens immunized with antigens from Salmonella choleraesuis, Salmonella typhimurium and Salmonella dublin were produced. The Antigenic proteins isolated from those flagella of Salmonella sp., determined by SDS-PAGE, were pure and had a molecular mass of approximately 53.4, 51 and 54.6 kDa, respectively. The IgY titers were found at two weeks after first immunization and increased gradually to maximum of 330,000 300,000 and 440,000 respectively. According to the results of specificity test by ELISA, the IgY raised against Salmonella sp. were found highly specific activity levels. Concentration of Salmonella sp. incubated with anti-Salmonella sp. IgY were drastically reduced to the levels of 2.8∼4.0 log CFU/ml. The contents of IgY in an egg yolk was approximately 31∼33 mg/ml.

Effects of Bacillus subtilis on Growth Performance and Resistance to Salmonella Infection in Broiler Chickens

  • The experiment was undertaken to see the effects of Bacillus sp. on the growth performance and disease resistance to Salmonella sp. infections. The use of probiotic microbes in poultry is commonly practiced. In this study, Bacillus subtilis was tested using a total of 120 chicks of age of 1 day after hatching. The growth traits examined were body weight gain and feed conversion rate. And also, the Salmonella resistance of Bacillus subtilis was tested after the chicks were orally administered with Salmonella pullorum by gavage force injections. The result showed that Bacillus subtilis yielded a high feed efficiency, consequently increased growth rate. For the effect of Bacillus subtilis on Salmonella infection, Bacillus subtilis significantly improved the resistance to Salmonella pullorum infection. Various clinical symptoms of Salmonella infection were highly decreased by addition of Bacillus sp.

Characterization of Bacteriophages against Salmonella Gallinarum (Salmonella Gallinarum 박테리오파지의 특성)

  • Bacteriophages are viruses that exclusively infect bacterial cells, and lytic bacteriophages can be used as a safe alternative to antibiotics for the prevention and treatment of animal diseases. In this study, we attempted to isolate and characterize bacteriophages for Salmonella enterica serovar Gallinarum (Salmonella Gallinarum), the causative agent of fowl typhoid in chickens. Ten bacteriophages were isolated from samples of sewage from seven poultry slaughterhouses. One of these isolate, designated as $SG{\Phi}-YS$ SP and classified in the family Myoviridae, produced plaques with seven Salmonella Gallinarum strains. However, no plaques were produced with any of the Salmonella enterica serovar Enteritidis strains tested, suggesting that this bacteriophage is Salmonella Gallinarum specific. To assess the lytic ability of $SG{\Phi}-YS$ SP against Salmonella Gallinarum, bacterial growth rates following inoculation of the bacteriophage were compared with the control. The $SG{\Phi}-YS$ SP treatment, with a multiplicity of infection of 10, reduced the growth of Salmonella Gallinarum by 2.21 log cfu/mL at 6 h, and 2.13 log cfu/mL at 9 h, suggesting that this bacteriophage isolate could be used for the prevention or treatment of Salmonella Gallinarum infection in chickens.

Antibacterial Activity by Lactobacillus bulgaricus SP5 against Pathogenic Bacteria (병원성 미생물에 대한 Lactobacillus bulgaricus SP5의 항균활성)

  • This study was carried out to get basic resources for the industrial use of Lactobacillus bulgaricus SP5. The antibacterial activity of the supernatant obtained from Lactobacillus bulgaricus SP5 was tested against the pathogenic bacteria such as Escherichia coli KCCM 11234, Salmonella enteritidis KCCM 3313, Salmonella enteritidis KCCM 12021, and Salmonella typhimurium KCCM 40253. The supernatant of L. bulgaricus SP5 showed antibacterial activity against tested pathogenic bacteria. The antibacterial activity was examined after adjusting pH and heat treatment of supernatant. Heat treatment of supernatant had antibacterial activity against pathogenic bacteria at all temperature. However, pH changes showed no antibacterial activity. Antibacterial activity of the supernatant was confirmed to be due to organic acids (lactic, acetic, phosphoric, succinic, pyroglutamic, citric, malic, and formic acid).

Antimicrobial drug susceptibility and plasmid profiles of Salmonella species isolated from poultry (가금에서 분리한 Salmonella속 균의 항균물질에 대한 감수성 및 plasmid profile)

  • In this study, we aim to find the presence of virulence-related plasmid in Salmonella isolates from poultry, and the difference between S pullorum and S gallinarum on the plasmid profile and antibiotics resistance. We used seventeen isolates of Salmonella spp that were isolated from poultry. Thirteen isolates, S typhimurium(ST), S pullorum(SP) and S gallinarum(SG), contained virulence-related plasmids. These are 95Kd plasmid in ST and 85Kd plasmid in SP and SG. Three(1/4 of ST, 1/1 of SE, and 1/9 of SP) isolates have no detectable plasmids. The isolates of ST have relatively variable plasmid profile but the isolates of S pullorum except No 12(additional 3.0Kb plasmid) have common 85K6, 8.1Kb, 4.0Kb and 2.3Kb plasmid and two of three isolates of S gallinarum have common 85Kb, 4.0Kb and 2.3Kb plasmid but the rest has only 85Kb plasmid. Interestingly, all of the isolates of SP have 8.1Kb plasmid, and same size of plasmid is also found in one of ST isolates. All of the isolates have the resistance to penicillin, chloramphenicol, rifampicin, streptomycin, sulfamethazine and some isolates show the resistance to ampicillin and tetracycline. There is no relatedness between plasmid profile and antibiotics resistance and no differences between SP and SG in antibiotics resistance. Therefore further differentiation of each isolates by restriction enzyme assay and, if possible, charaterization of each plasmid, especially, 8.1Kb plasmid in SP and ST, may be necessary.

  • Salmonella enterica serovars Gallinarum(SG, causative agent of fowl typhoid) and S Pullorum(SP, causative agent of pullorum disease) are very important bacterial pathogens in poultry industry. They share some common antigenic properties though the characteristics of outbreaks are quite different. To discriminate between SG and SP, we developed two rapid diagnostic methods, allele-specific real-time PCR and 3'-tailed PCR over 2 single nucleotide polymorphisms ($237^{th}\;and\;598^{th}$). In both methods, $237^{th}$ allele was found to be a good target for differential diagnosis, while $598^{th}$ allele produced some non-specific reactions.

  • The specificity and sensitivity of oligonucleotide primers were examined for the rapid detection of Salmonella in meat samples. The oligonucleotide primers used in this study were designed with the modification of mdh and invA sequence in the chromosome of Salmonella Typhimurium. Through the subsequent analysis of the specificity and sensitivity of the primers, two types of oligonucleotide primers, SLM1 and SLT4 were selected for the detection of Salmonella genus specific and S. Typhimurium species specific, respectively. The lowest detection limit of each primer was represented as 1 cell per reaction when reacted with a prepared DNA solution. The detection efficiency of the two primers was analysed with beef and pork samples intentionally contaminated with a mixture of Salmonella culture, and three preparation methods -, namely direct reaction after extraction, enrichment after extraction, and DNA extraction after enrichment for PCR reaction, - were also compared. No differences were found in the results according to meat sources and preparation methods.

Salmonella sp. 의 신속한 동정을 위한 증진배양의 개선에 관한 연구

  • The development of an enrichment method for the rapid and effective identification of Salmonella spp. in sewage or food was studied. As a growth factor for Salmonella, 10 mM cyclic adenosine monophosphate (cAMP) in trypticase soy broth with 0.6% yeast extract (TSBYE) increased cell number five-folds and 0.6% yeast extract in selenite broth increased cell number ten-folds of control. Bile salts in selenite broth was tested for the selection of S. enteritidis in a mixture with Staphylococcus aureus, Pseudomonas aeruginosa, Lactobacillus plantarum and Escherichia coli. The latter four strains were effectively inhibited at 0.1% bile salt. A two-step culture method was used to enrich Salmonella spp.; a primary-enrichment and secondary- enrichment culture. At a primary-enrichment step, selenite broth with 0.6% yeast extract and 10 mM cAMP was used, and at a secondary-enrichment step, 0.1% bile salt was additionally used. Culture times of a primary- enrichment and a secondary-enrichment step were 8 hr and 6 hr, respectively. In this procedure, cell number increased from 10$^{0.3}$ to 10$^{8.5}$ with inhibition of other strains within 14 hr. In the case of an initial cell concentrarion as low as 10$^{-2}$ cfu/ml, a cell number increased to 10$^{7}$ cfu/ml by using a 10 hr primary-enrichment and 6 hr secondary-enrichment procedure.

Serological Distribution and Antibiotic Susceptibility of Salmonella sp. Isolated from Pusan Area in 2000 (2000년도 부산지역에서 분리된 살모넬라속균의 혈청학적 분포 및 항균제 감수성)

  • A total of 79 Salmonella spp. were isolated from Pusan area in 2000. The serotypes of 79 Salmonella isolates were classified as 42 strains of S. typhi(53.1%), 24 strains of S. enteritidis(30.4%), 9 strains of S. montevideo(11.4%), 2 strains of S. typhimurium(2.5%), 1 strain of S. infantis(1.3%) and 1 strain of S. indiana(1.3%) strains(16.5%) of Salmonella sp. were isolated at May July, respectively. The isolates of S. typhi were sensitive to most sntibiotics except streptomycin. All isolates of S. typhi were especially sensitive to tobramycin, gentamicin, colistin, kanamycin, samikacin, sulfamethozazole/ trimethoprim, cefriaxone, ceftazdime, cifrofloxacin, cefoxitin and cefotaxime. Isolates of S. enteritidis wer presented higher resistance than isolates of S. typhi. Twenty-four strains of S. enteritidis were sensitive to kanamycin, amikacin cifrofloxacin, cefoxitin and cefotaxime, however 13 strains(54.2%) of S. enteritidis were resistant to carbenicillin, ampicillin and ticarcillin. Nine strains of S. montevideo were sensitive to most antibiotics except carbenicillin and streptomycin. Each 1 stain of S. indiana and S. infantis was sensitive to most antibiotics used in this study except streptomycin. Three kinds of resistant pattern (CB, SM, TE, AM, TC). In the case of S. enteritidis isolates, 9 kinds resistant pattern were detected. Most frequent resistant pattern of S. enteritidis isolates was CB, AM, TC type(16.7%)
