  • Salmonella species are the most prevalent etiologic agents of food-borne acute gastroenteritis. Direct isolation of bacteria from the contaminated food, stool and animal tissues has been used for the diagnosis of salmonellosis routinely. However, isolation of bacteria is time consuming work and not so highly sensitive. In recent years, improved methods of polymerization chain reaction(PCR) and probe hybridization technique have led to the developement of diagnostic assays which employ to detect various human and animal pathogenic bacteria. In this study, we have performed the polymerization chain reaction to detect Salmonella pullorum from tissues and stool samples of chickens with two specific primers, ST5 and ST8C. The target DNA fragment of PhoE gene was successfully amplified from liver, spleen, pancreas, heart, lung, ovary, oviduct and feces samples. The amplified DNA fragments were hybridized with Salmonella typhymurium TA3000 PhoE probe by Southern hybridization. The PCR to amplify the PhoE gene was highly rapid and sensitive method to detect Salmonella pullorum from tissues and stool samples.

  • As Salmonella enterica serovar Pullorum remains a major economic problem for the poultry industries of countries with no efficient control measures, we presented a multidrug resistance strain S06004 (isolated from a clinically sick chicken in China in 2006) for genome sequencing. The genome comparison showed that the strain contained two prophages, the ST104 and prophage-4 (Fels2) of E. coli LF82, which were not detected in the only published genomes of S. Pullorum RKS5078 and CDC1983-67. In addition, the GyrA Ser83 point mutation, drugresistant genes, and many antibiotic pump systems that are present in S06004 may be contributing to the multidrug resistance of this strain.

  • Salmonellosis is a highly contagious bacterial disease that threatens both human and poultry health. Tests that can detect Salmonella in the field are urgently required to facilitate disease control and for epidemiological investigations. Here, we combined loop-mediated isothermal amplification (LAMP) with a chromatographic lateral flow dipstick (LFD) to rapidly and accurately detect Salmonella. LAMP primers were designed to target the Salmonella invA gene. LAMP conditions were optimized by adjusting the ratio of inner to outer primers, $MgSO_4$ concentration, dNTP mix concentration, amplification temperature, and amplification time. We evaluated the specificity of our novel LAMP-LFD method using six Salmonella species and six related non-Salmonella strains. All six of the Salmonella strains, but none of the non-Salmonella strains, were amplified. LAMP-LFD was sensitive enough to detect concentrations of Salmonella enterica subsp. enterica serovar Pullorum genomic DNA as low as $89fg/{\mu}l$, which is 1,000 times more sensitive than conventional PCR. When artificially contaminated feed samples were analyzed, LAMP-LFD was also more sensitive than PCR. Finally, LAMP-LFD gave no false positives across 350 chicken anal swabs. Therefore, our novel LAMP-LFD assay was highly sensitive, specific, convenient, and fast, making it a valuable tool for the early diagnosis and monitoring of Salmonella infection in chickens.

  • The present study was conducted to investigate the preventive effect of feed supplemented with 1.0% oriental herbal medicine feed additives (OHMFA) on the colonization of Salmonella spp. and incidence of death in broiler chickens. The frequency of Salmonella spp. in feces samples treated with OHMFA (25/239; 10.5%) was significantly reduced (p<0.05) than that of control group (83/347; 23.9%). A total of 108 Salmonella spp, belonging to four different serotypes, were isolated from three broiler farms. Among the serotypes isolated, Salmonella typhimurium was the moot prevalent (60.2%), followed by S. enteritidis (20.4%), S. gallinarum (13.0%) and S. pullorum (6.4%). In incidence of death in chickens during experiment, the mortality of rate OHMFA group (3.9%) was significantly lower (p<0.01) than that of control group (5.7%). These results show that the administration of OHMFA may prevent the colonization of Salmonella in chickens.

  • Pullorum disease and fowl typhoid are septicemic avian diseases transmitting through egg by transovarian infections. A series of tests has been performing in breeding flocks of chickens and test plans for proper inspection have been modified by government veterinary institute to control of such diseases. To improve inspection plans, different test methods were compared using fowl typhoid positive samples from a poultry farm located in Jeonbuk state in 2012. Based on first inspection, 11 samples among total 200 samples were positive by rapid slide agglutination (RSA) test and 7 samples among RSA positive samples were finally diagnosed as Salmonella Gallinarum infection by ELISA, bacterial isolation, PCR, and histopathologic examination. In the second inspection, 20 samples among total 100 samples were positive by RSA test. Among RSA positive ones, 19 samples were positive by ELISA, S. Gallinarm were successfully isolated in 3 samples, and 16 samples were positive by PCR in the cecal tonsils where were not successful for bacterial isolation. Based on histopathologic examination, severe inflammation in the 13 cecal tonsils and infiltration of lymphocytes and heterophils in the 11 livers were observed. Therefore, we suggest that bacterial isolation, PCR, and histopathologic examination methods in the third inspection need to be further used in various tissues for correct diagnosis and for final eradication of pullorum disease and fowl typhoid in breeding flocks of chickens.

  • This study was carried out to identify the occurrence of transovarian transmitted diseases and antibody positive rates among Korean native breeder chickens. The infection rates with Salmonella pullorum and Salmonella gallinarum among 16-week-old, 36-week-old, and 56-week-old breeder chickens and the antibody positive rates to Egg Drop Syndrome '76, Mycoplasma gallisepticum and Mycoplasma synoviae among 16-week-old, 18-week-old, and 56-week-old breeder chickens were identified, and the antibody positive rates to seven major transovarian transmitted diseases among 1-day-old chicks were investigated. As a result, no infection with Salmonella pullorum and Salmonella gallinarum was found among the investigated subjects of all ages. Vaccinated breeder chickens showed the 100% antibody positive rate to Egg Drop Syndrome '76, and unvaccinated breeder chickens showed the 100% antibody negative rate to Mycoplasma gallisepticum and Mycoplasma synoviae, confirming that there was no infection with Mycoplasma gallisepticum and Mycoplasma synoviae. As a result of the antibody tests of the 1-day-old chicks for transovarian transmitted diseases, it was found that vaccinated chicks showed good antibody positive rates to avian encephalomyelitis, chicken infectious anemia, and avian reovirus, confirming that they had power of defense against the relevant infectious diseases, and that unvaccinated chicks showed the 100% antibody negative rates to avian leukosis, chicken reticuloendotheliosis, and Mycoplasma synoviae, confirming that there was no infection with the relevant diseases. Given that the results of this study showed that among the transovarian transmitted diseases of chickens, there was no history of infection with diseases against which vaccination was not administered and high antibody positive rates were found with diseases against which vaccination was administered, it is judged that chickens with good power of defense against diseases were bred, and it is deemed that constant monitoring and vaccination against transovarian transmitted diseases will be necessary for the control and prevention of the diseases.

  • The present study was conducted to investigate the epidemiological properties of salmonellosis of poultry farms in Kyongbuk province during the relied from November 1998 to November 1999 And antibiotic susceptibility and biochemical characteristics of 120 Salmonella cultures isolated from chicken samples were also investigated. The results obtained through this study were summarized as follows, 1. Among 667,200 chickens of 31 flocks in 17 layer farms and 9 broiler farms, 61,350 chickens of 31 flocks were died with salmonellosis. The death rate of 25 farms varied from 0.1% to 75.0%, and the average death rate was 9.2%. 2. According to etiological agents, fowl typhoid was shown the most predominant outbreak among the salmonellosis during a year, which accounted for 8R.0%(22/25) of the total case 3. The serotypes of 120 Salmonella isolates were identified as 7 strains(5.8%) of S pullorum, 10 strains(8.4%) of S typhimurium and 103 strains(85.8%) of S gallinarum. 4. Most outbreak of fowl typhoid were prevalent on the layer chicken farms(77.1%), and the summer season(45.5%) also appeared the most hazardous season during the year. 5. It seemed that the Hyline breed(70.6%) was the most susceptible among the layer chicken breeds, and followed by Isabrown(23.5%), Tetra(5.9%) in order. 6. In layers, 76.4% of fowl typhoid occurred commonly from 14 to 40 weeks including the early laying peroid, but in broiler farms, all cases was outbreak within first second weeks. 7 All the strains of S pullorum were resistant to lincomycin(Lm), penicillin(Pm), and steptomycin(Sm), but sensitive to amikacin(Ak), ampicillin(Am), cephalothin(Ce), ciprofloxacin (CiP), chloramphenicol(Cm), colistin(Co), enrofloxacin(Enr), furazolidone(Fu), gentamicin(Gm), kanamycin(Km), neomycin(Nm), polymyxin(Po), and teracycline(Tc). • This paper deals with the distribution, reservoir and mode of spread of salmonella infection on 7 pig farms in Taegu, Gyeongbuk, Gyeongnam and Chungnam and a slaughter house in Teagu during the period from May 1984 to May 1985. Isolated salmonella were examined for serotypes and biotyping of S. typhimurium. The results obtained were summarised as follows; 1. Of total 7,995 samples from 7 pig farms and a slaughter house, 319 salmonella were isolated from 234 samples (2.9%) and their serovar strains were S. derby 77, S. infantis 41, S. enteritidis 20, S. typhimurium 18, S. bredeney 16, S. london 14, S. paratyphi B 9, S. anatum 8, S. montevideo 8, S. senftenberg 7, S. thompson 6, S. pullorum 4, S. paratyphi A 1 and untypable 70. 2. The incidence rate of diarrhea of piglets, weaned pigs and fattening pigs was 9.8%, 2.3% and 0.5%, respectively whereas the rate by salmonella infection was 4.2%, 1.2% and 11.3%, respectively. 3. The isolation rate of salmonella was higher in summer and autumn. 4. The isolation rate of salmonella varied from 1.1% to 4.5% in 7 pig farms, it was higher in sewages(4.4%), weaned pigs(3.7%), boars(3.7%) and other(3.7%) included soils, manure and wild rats according to samples. Three out of 7 pig farms were contaminated heavily with various serovars of salmonella. 5. The isolation rate of salmonella from pigs slaughtered was 8.1%, it was 13.6% in rectal contents and 1.6% in mensenteric lymph nodes. 6. Eighteen strains of S. typhimurium were classified into 3 different biovars(1, 10 and 10a) by the method of Brandis and were subdivided into 6 different full biovars(1a, 1d, 1dh, 3d, 26i and 26ei) by the method described by Duguid et al. Appearance of different biovars indicated the occurrence of different exotic infection sources on the farms.
