전남지역 도축돈에서 분리된 Salmonella typhimurium의 병원성에 관한 연구 (Studies on the pathogenicity of Salmonella typhimurium isolated from slaughtered pigs in Chonnam area)

  • Non-typhoidal Salmonella serovars remain a potential threat to human health and many animals including beef cattle, broiler chickens, and pigs which possible sources of non-typhoidal salmonellosis in human. In this study, the cecal contents of slaughtered pigs were examined for Salmonella serovar prevalence. The characteristics of the isolates, including antimicrobial resistance patterns and virulence genes, were studied along with the reference strain S typhimurium ATCC 13311. Out of 640 sample, 137 Salmonella(21.4%) were isolated and their serovar were identified S typhimurium 83 strains(60.6%), S agona 10 strains(7.3%), S schwarzengrund 4 strains(2.9%), S derby 4 strains(2.9%), S ayinde 1 strains(0.7%), and untypable 35 strains(25.5%). All 83 S typhimurium strains(100%) were multi-drug resistance to at least 7 antibiotics, and 20 strains(24.1%) of 83 isolates were R-type ACSSuT. Examination of virulent gene by PCR revealed that 73 S typimurium field isolates(88%) have a invA gene and 24 strains(28.9%) have a spvC gene. Consequently, S typhimurium infection in slaughtered pigs was relatively to appear high prevalence in their herds which suggested that it should be necessary for herd health monitoring and surveillance.

  • 저자들은 평소 건강하던 9세 남아에게서 Salmonella enteritidis 위장관염에 합병된 급성 비결석성 담낭염을 임상증상, 혈액 검사 및 배양 검사, 복부 초음파 검사등을 통하여 진단 할 수 있었다. 또한 항생제 치료와 비수술적 방법인 경피경간 담낭배액술을 통하여 성공적으로 치료 하였기에 이에 보고하는 바이다.

Salmonellosis in swine: Clinical perspectives

  • Salmonella is one of the most important food-borne zoonotic pathogens, causing acute or chronic digestive diseases such as enteritis. The acute form of enteritis is common in young pigs of 2 - 4 months of age. The main symptoms include high fever ($41-42^{\circ}C$), loss of appetite, and increased mortality within 2 - 4 days of onset of the disease. It is often the cause of increasing mortality, decreasing growth rate and reducing feed efficiency of piglets. In the case of chronic enteritis in pigs, the main symptom is weight loss due to the continuing severe diarrhea. Salmonella enterica serovar Typhimurium and Salmonella enterica serovar Choleraesuis are typical pig adapted serotypes, which cause one of four major syndromes: enteric fever, enterocolitis/diarrhea, bacteremia and chronic asymptomatic carriage. These syndromes cause a huge economic burden to swine industry by reducing production. Therefore, it is necessary that swine industries should strive to decrease Salmonellosis in pigs in order to reduce economic losses. There are several measures, such as vaccination to prevent salmonellosis, that are implemented differently from country to country. For the treatment of Salmonella, ongoing antibiotic treatment is needed. However constant doses of antibiotics can be a problem because of antibiotic resistance. Therefore, the focus should be made more on prevention than treatment. In this review, we addressed the basic information about Salmonella, route of infection, clinical symptoms, and prevention of Salmonellosis.

Quantitative risk assessment of foodborne Salmonella illness by estimating cooking effect on eggs from retail markets

  • In this study, we performed a quantitative microbial risk assessment (QMRA) of Salmonella through intake of egg consumption after cooking (dry-heat, moist-heat, and raw consumption). Egg samples (n = 201) from retail markets were analyzed for the presence of Salmonella. In addition, temperature and time were investigated during egg transit, storage, and display. A predictive model was developed to characterize the kinetic behavior of Salmonella in eggs, and data on egg consumption and frequency were collected. Eventually, the data was simulated to estimate egg-related foodborne illnesses. Salmonella was not found in any of the 201 egg samples. Thus, the estimated initial contamination level was -4.0 Log CFU/g. With R2 values of 0.898 and 0.922, the constructed predictive models were adequate for describing the fate of Salmonella in eggs throughout distribution and storage. Eggs were consumed raw (1.5%, 39.2 g), dry-heated (57.5%, 43.0 g), and moist-heated (41%, 36.1 g). The probability of foodborne Salmonella illness from the consumption of cooked eggs was evaluated to be 6.8×10-10. Additionally, the probability of foodborne illness not applied cooking methods was 1.9×10-7, indicating that Salmonella can be reduced by cooking. Therefore, the risk of Salmonella infection through consumption of eggs after cooking might be low in S. Korea.

  • This paper dealt with the distribution of Salmonella (S) infection on 4 herds in Kyungju and Taegu during the period from May to October 1986. Isolated Salmonella were examined for serotypes, antimicrobial drug resistance and detection of R plasmid. The results obtained were summarised as followings: 1. Of total 4.622 samples from 4 herds, 67 Salmonella were isolated from 51 samples(1.1%), and their serovar strains were S typhimurium 6, S derby 5, S infantis 4, S bareilly 4, S dublin 3, S anatum 2, S montevideo 2 and untypable 41. 2. The isolation rate of Salmonella was higher in summer and autumn. 3. Of the 67 strains examined, 45 (67.2%) were resistant to one or more antibiotics, such as ampicillin (Am), cephalothin (Ce), chloramphenicol (Cm), rifampicin (Rf), sulfadimethoxine (Su), and tetracycline (Tc), and higher resistant to Sm (40.2%), Ce (31.3%), Am (23.9%). 4. Of the 45 resistant Salmonella strains, 44 (97.8%) harbored conjugative R plasmids and the transfer frequency of Sm (100%), Ce (95.2%), Tc (91.0%) and Su (80.0%) resistance was much higher than that of the other drug resistance. 5. The most common resistant patterns were Sm, Ce, AmCeCmSmSuTc, and AmCe. 6. In 4 herds, the incidience of drug resistance was 57.7%~100% and transfer frequency of conjugative R plasmid was 96.1%~100%.

Protective Effects of a Novel Probiotic Strain of Lactobacillus plantarum JSA22 from Traditional Fermented Soybean Food Against Infection by Salmonella enterica Serovar Typhimurium

  • Lactobacillus species have been shown to enhance intestinal epithelial barrier function, modulate host immune responses, and suppress the growth of pathogenic bacteria, yeasts, molds, and viruses. Thus, lactobacilli have been used as probiotics for treating various diseases, including intestinal disorders, and as biological preservatives in the food and agricultural industries. However, the molecular mechanisms used by lactobacilli to suppress pathogenic bacterial infections have been poorly characterized. We previously isolated Lactobacillus plantarum JSA22 from buckwheat sokseongjang, a traditional Korean fermented soybean food, which possessed high enzymatic, fibrinolytic, and broad-spectrum antimicrobial activity against foodborne pathogens. In this study, we investigated the effects of L. plantarum JSA22 on the growth of S. Typhimurium and S. Typhimurium-induced cytotoxicity by stimulating the host immune response in intestinal epithelial cells. The results showed that coincubation of S. Typhimurium and L. plantarum JSA22 with intestinal epithelial cells suppressed S. Typhimurium infection, S. Typhimurium-induced NF-κB activation, and IL-8 production, and lowered the phosphorylation of both Akt and p38. These data indicated that L. plantarum JSA22 has probiotic properties, and can inhibit S. Typhimurium infection of intestinal epithelial cells. Our findings can be used to develop therapeutic and prophylactic agents against pathogenic bacteria.

  • Bacteriologic diagnosis of enteric infection remains to be an important role of clinical laboratory because of the prevalence of the infection. Often the determination of etiologic agent and its susceptibility to antibiotics are of vital importance for a proper management of the infection. In our previous paper, an analysis of the isolation of enteric pathogens for the years 1969-73 was reported to clarify the status of those years. The present analysis was made based on the data obtained during the years 1974-78, to see if any change of the status was rendered. 1. During the 5-year period, from the cultures of 7,308 stool or rectal specimens 833 patients yielded enteric pathogens: 468 Shigella, 295 Salmonella, 30 Vibrio parahaemolyticus and 40 enteropathogenic Escherichia coli(EPEC). 2. Of the 295 Salmonella, 271 were S. typhi Isolation of 12 S.paratyphi-A, 1 Salmonella group B, 4 group C, 5 group D and 2 group E meant a definite increase of these sero-groups, S. typhi was most frequently isolated in August and in December, and from 30- to 39-year-old patients. 3. Of the 468 Shigella, 10 were subgroup A, 338 subgroup B, 3 subgroup C and 117 subgroup D. Most of the subgroup B belonged to type 1,2, or 3. The proportion of S. sonnei decreased from 31.3% in 1974 to 18.2% in 1978. In foreign patients, S. sonnei remained to be the frequntly isolated species. Shigella isolation was frequent in August and in 2- to 5-year-old patients. 4. V. parahaemolyticus was isolated from 30 and EPEC from 40 patients. 5. Ninty-nine per cent and 99.5% of the S. typhi isolates were susceptible to chloramphenicol and to ampicillin respectively. 92.8% of S sonnei were susceptible to ampicillin. S. flexneri type 2 was notable for their markedely decreased proportion being susceptible to ampicillin: 84.4% in 1974 and 25.6% in 1978.

조제분유와 이유식의 미생물학적 안전성 (Microbiological Safety of Infant Formula and Baby Food)

  • Powered infant formula and baby food contaminated with Enterobacter sakazakii were reported to cause infection among infants and to be associated with sporadic cases and outbreaks of sepsis, menigitis, cerebritis, and necrotizing enterocolitis. Salmonella contamination of infant formula has also been responsible for multiple outbreaks. Other species of Enterobacteriaceae in powdered infant formula may be causative agents, about which there has been no report. Other pathogenic bacteria have been isolated from powdered infant formula but they were not associated with outbreaks among infant. While Enterobacter sakazakii caused disease in all age groups, premature infants under 28 days old and with birth weight are most sensitive to its infection. Even if low contamination level of the bacteria in powdered infant formula and baby food may not cause infection, the possibility to multiplicate during preparation and storage of reconstituted formula may increase. The etiological factors and pathogenecity of S. sakazakii have not been elucidated. There were wide variability in phenotype and genotype between its strains. S. sakazakii has been isolated from factory facility and surroundings more frequently than Salmonella and thus factory environment should be the source for post-processing contamination of the formula with S. sakazakii. Considering current technology to manufacture power infant formula and baby food it is impossible to sterilize powdered formula but the frequency of outbreak hazard by S. sakazakii can be reduced by pasteurizing the formula base before drying and shortening storage time of the reconstituted formula.

Surveillance of Bacterial Pathogens Associated with Acute Diarrheal Disease in the Republic of Korea During One Year, 2003

  • An epidemiological survey of human enterobacterial infections was conducted to determine the prevalence of enteropathogens in the Republic of Korea during one year, 2003. We tested for infectious diseases in 26,992 stool samples obtained from people who visited clinics located in six big cities and six rural provinces. From these samples, we isolated 1,291 cases of enteritis bacterial infection (4.8%). In the urban areas, 821 cases of bacterial infection (6.4%) were identified and, in the rural areas, 479 bacterial strains (3.3%) were isolated. Seasonal patterns were seen for diarrhea associated with S. aureus, S. coli and V. parahaemolyticus, while Salmonella and Shigella infections showed slight seasonal variation. We found that S. aureus and Salmonella were more frequently isolated from children and the elderly; however, the prevalence of E. coli, V. parahaemolyticus, and Shigella were similar in different age groups. Routine monitoring of these infections is considered a worthwhile means by which to elucidate their epidemiology and modes of transmission and ultimately to control them more effectively. Continuous laboratory-based surveillance for findings of enteritis bacterial infection should be emphasized in the prevention of these infections.

Lysophosphatidylcholine Enhances Bactericidal Activity by Promoting Phagosome Maturation via the Activation of the NF-κB Pathway during Salmonella Infection in Mouse Macrophages

  • Salmonella enterica serovar Typhimurium (S. Typhimurium) is a facultative intracellular pathogen that causes salmonellosis and mortality worldwide. S. Typhimurium infects macrophages and survives within phagosomes by avoiding the phagosome-lysosome fusion system. Phagosomes sequentially acquire different Rab GTPases during maturation and eventually fuse with acidic lysosomes. Lysophosphatidylcholine (LPC) is a bioactive lipid that is associated with the generation of chemoattractants and reactive oxygen species (ROS). In our previous study, LPC controlled the intracellular growth of Mycobacterium tuberculosis by promoting phagosome maturation. In this study, to verify whether LPC enhances phagosome maturation and regulates the intracellular growth of S. Typhimurium, macrophages were infected with S. Typhimurium. LPC decreased the intracellular bacterial burden, but it did not induce cytotoxicity in S. Typhimurium-infected cells. In addition, combined administration of LPC and antibiotic significantly reduced the bacterial burden in the spleen and the liver. The ratios of the colocalization of intracellular S. Typhimurium with phagosome maturation markers, such as early endosome antigen 1 (EEA1) and lysosome-associated membrane protein 1 (LAMP-1), were significantly increased in LPC-treated cells. The expression level of cleaved cathepsin D was rapidly increased in LPC-treated cells during S. Typhimurium infection. Treatment with LPC enhanced ROS production, but it did not affect nitric oxide production in S. Typhimurium-infected cells. LPC also rapidly triggered the phosphorylation of IκBα during S. Typhimurium infection. These results suggest that LPC can improve phagosome maturation via ROS-induced activation of NF-κB pathway and thus may be developed as a therapeutic agent to control S. Typhimurium growth.
