Subacute Toxicity Study on Sagunja-tang(Sijunzi-tang) in SD Rats (SD 랫트를 이용하여 사군자탕(四君子湯)의 아급성 독성에 관한 연구)

  • Sagunja-tang(Sijunzi-tang) has been traditionally prescribed as a restorative. The present study was undertaken to determine the possible toxic effects of Sagunja-tang on SD rats. In this study, we investigated the subacute toxicity of water-extracted Sagunja-tang(Sijunzi-tang) on SD rats. Twenty rats were orally adiministered Sagunja-tang for 28 days at a dose of 0 mg/kg(control group) or 1500mg/kg(treated group), respectively. All of subjects was survived. No significant difference in abnormal clinical signs, related to hematological values, serum biochemical values, water and feed intake, coagulation time, autopsy analysis, organ weight, tissue microscopically, funduscopy, urine intake and urinalysis, was detected. Compared with the control group, we could not find any subacute toxic alteration in treated group (1500mg/kg) for 28 days. This result suggests that Sagunja-tang(Sijunzi-tang). a herbal medicine prescription, is a safe prescription to body.

The effect of Sagunja-tang on TNBS-induced Inflammatory Bowel Disease in Mouse (사군자탕(四君子湯)이 TNBS로 유발(誘發)된 생쥐의 염증성(炎症性) 장질환(腸疾患)에 미치는 영향(影響))

  • Objectives : The present study aimed to find out the effect of Sagunja-tang on the prevention and treatment of inflammatory bowel disease using mice with TNBS-induced inflammatory bowel disease. Methods : Mice with TNBS-induced inflammatory bowel disease were medicated with Sagunja-tang, and the weight changes, colon length, lipid peroxidation, and myeloperoxidase activity were observed. Levels of the inflammatory markers interleukin (IL)-$1{\beta}$ and cyclooxygenase-2 (COX-2), its transcription factor activation, phospho-NF-${\kappa}$B (pp65), in the colon by enzyme-linked immunosorbent assay and immunoblot analysis were also measured. Finally, the activation of fecal bacterial enzyme, ${\beta}$-glucuronidase and degradation activation of fecal glycosaminoglycan (GAG) and hyaluronic acid were observed. Results : We found that oral administration of Sagunja-tang inhibited TNBS-induced colon shortening and also inhibited myeloperoxidase activity in the colon of mice as well as IL-$1{\beta}$ and COX-2 expression. Sagunja-tang also inhibited TNBSinduced lipid peroxidation and pp65 activation in the colon of mice. In addition, Sagunja-tang inhibited ${\beta}$-glucuronidase activation and fecal hyaluronic acid degradation activation. Conclusions : It is supposed that Sagunja-tang has a potential therapeutic effect on inflammatory bowel disease through the inhibition of both NF-${\kappa}$B activation and lipid peroxidation, and the improvement of intestinal conditions.

Herbal Cocktail Sagunja Protects $H_2O_2$-induced H9c2 Cardiomyoblast Cell Death through the Induction of Heme Oxygenase-1

  • Sagunjatang (Sagunja), containing Radix Astragali, Radix Ginseng, Fructus Schizandrae, Radix Ophiopogonis and Radix Glycyrrhizae, has been used as a prescription for ischemic heart and brain diseases in Korean traditional medicine. This study was designed to investigate the protective mechanisms of Sagunja on $H_2O_2$-induced cytotoxicity of H9c2 cardiomyocytes. Treatment with $H_2O_2$ resulted in death of H9c2 cells, characterized by apparent apoptotic features, including the fragmentation of nucleus and increase in sub-GO/G1fraction of cell cycle. However, Sagunja markedly suppressed the apoptotic characteristics of H9c2 cells induced by $H_2O_2$ with decrease of intracellular peroxide level. In addition, Sagunja suppressed the features of mitochondrial dysfunction, including change of mitochondrial membrane potential, in $H_2O_2$- treated cells. Additionally, Sagunja induced the expression of HO-1 protein in both time-and dose-dependent manner. The role of HO-1 in ROS-scavenging activity of Sagunja is proposed.

  • Decoction of oriental medicinal preparation is prepared in various manners, and changes of chemical constituents might be occurred depending on the processing techniques. The present study was undertaken to investigate the phγsio-chemical and pharmacological equivalence between two extraction methods of Nokyong-sagunja-Tang. Samples were Prepared as follows ; Sample-I was prepared by simultaneously extracting Sagunja-Tang and velvet antler in one vessel. Sample-lI was prepared by adding velvet antler exact to the water extract of Sagunja-Tang. Both sanples showed similar results of physiochemical parameters such as pH, yield, TLC and HPLC chromatogram, and contents of ginsenoside $Rb_1$ and glycyrrhizin. Also, there were little different between two samples in pharmacological effects such as DPPH free radical scavenging effect, and inhibitory effects on xanthine oxidase, hyaluronidase, trypsin, TBA-Rs formation and hemolysis in vitro. And both samples showed no significant difference in antifatigue activities in mice. These results suggest that there might be little difference between two extraction process when velvet antler added to Sagunia-Tang.

Acute Toxicity Study on Sagunja-tang(Sijunzi-tang) in ICR Mice (ICR 마우스를 이용하여 사군자탕(四君子湯)의 급성 독성에 관한 연구)

  • Objectives : The purpose of this study was to investigate acute toxicity of orally-treated Sagunja-tang(Sijunzi-tang) in ICR mice. Methods : In this study, we investigated the acute toxicity of water-extracted Sagunja-tang(Sijunzi-tang). Twenty-five mice completed 14 days of oral Sagunja-tang(Sijunzi-tang) at respective doses of 0 (control group), 2560, 3200, 4000 and 5000 mg/kg. Results : We observed survival rates, clinical signs of male ICR mice with acute toxicity, change of body weight and autopsy. Conclusions : Compared with the control group, we could not find any toxic alteration in anytreated groups (2560, 3200, 4000 and 5000mg/kg). LD50 of Sagunja-tang(Sijunzi-tang) was over 5000 mg/kg and it is very safe for ICR mice.

Effects of Sagunjatang and Sagunja-tang plus Mylabris phalerata on Human Stomach Cancer Cells (사군자탕(四君子湯) 및 사군자탕가반묘(四君子湯加斑猫)가 위암세포에 미치는 영향)

  • The efficacy of Sagunja-tang and Sagunja-tang plus Mylabris phalerata against the human stomach cancer was examined and molecular biological fight of its actions was studied. In the efficacy test of anti-stomach cancer cells growth using the MTT assay, administration of Sagunja-tang resulted in no significant change of stomach cancer cells growth, with the control group. Administration of Sagunja-tang plus Mylabris phalerata resulted in a decrease of stomach cancer cells growth in proportion to the concentration of mylabris phalerata and time, which was significantly different from the control group(significance recognized when p<0.05). In the test using the apoptosis assay, administration of Sagunja-tang showed an increase in apoptosis of human stomach cancer cells, with no significant difference from the control group. Administrating Sagunja-tang plus Mylabris phalerata showed an increase in apoptosis of stomach cancer cells in proportion to the concentration of mylabris phalerata and time, which was significantly different from the control group(significance recognized when p<0.05). In the test using the quantitative RT-PCR to examine stomach cancer cells growth and revelation of apoptosis related genes, administrating Sagunja-tang plus Mylabris phalerata resulted in a decrease of Bcl-2, an anti-apoptosis gene, in proportiong to concentration. No significant change was examined in the revelation of CDK1, Cdc2, Cyclin D1, PCNA, c-myc, which are genes related to the stomach cancer cells growth, and Bax, Bel-XL, the genes related to apoptosis, and p53. Referring to the results above, Sagunja-tang plus Mylabris phalerata may be considered to have an anti-growth efficacy against human stomach cancer cells, and an inducement efficacy. Therefore, it can be clinically implemented in the human stomach cancer.

Analysis of Studies on Sagunja-tang(Sijunzi-tang) to Establish the Fundament for Evidence Based Medicine (EBM 기반구축을 위한 사군자탕 연구 문헌 정량 분석)

  • To establish the fundament for EBM of Traditional Korean Medicine, the papers on Sagunja-tang(Sijunzi-tang) frequently used in medical institutions of Traditional Korean Medicine were analyzed through researching domestic and international papers. The papers were classified by the registration of domestic or international journals, the year of publishment, experimental fields and the kinds of studies on biological activities. Of total 228 papers on Sagunja-tang(Sijunzi-tang), 121 volumes were selected according to creteria. 47 volumes were published in domestic journals, 71 in Chinese journal, 2 in Japanese journal, and 1 in Taiwan journal. The papers on instrumental analyses were preceeded by HPLC, MS, GC with standard compounds of herbal medicine in Sagunja-tang(Sijunzi-tang). The papers on biological activities of Sagunja-tang(Sijunzi-tang) showed improvement of gastrointestinal activity and blood circulation, immunoactivity, anti-cancer, anti-oxidant, anti-fatigue, anti-stress, pharmacokinetics, hepatic protection, radioprotection, muscular activity, hematopoiesis and nontoxical effect. Further studies including gastrointestinal disorder, immune related disease, cancer, oxidative injury and pharmacokinetic study need to be preceeded to establish the fundament for EBM of Sagunja-tang(Sijunzi-tang).

Difference of Sagunja Temperament type on Communication Ability and Interpersonal Relations Ability of University Students (대학생의 사군자 기질에 따른 의사소통 능력과 대인관계 능력 차이)

  • The purpose of this study is to investigate the difference between communication ability and interpersonal relationship ability according to the types of Sagunja Temperament. As a result of studying 350 university students in Busan, the first, Temperament type was chrysanthemum, orchid, plum, and bamboo temperament. Second, as a result of analyzing the differences in communication abilities by types of the Sagunja Temperament, there was the highest communication ability of bamboo substrate, and there were significant differences in plum, orchid, and chrysanthemum. The ability of interpersonal relationship according to the types of the Sagunja Temperament showed significant difference in 'first relationship', 'claiming other people's displeasure', 'emotional support and advice' excluding self-exposure and interpersonal conflict handling. Third, there was a significant correlation between communication ability and interpersonal relationship ability. In conclusion, this study proved that the difference of communication ability and interpersonal relationship ability difference according to the types of the Sagunja Temperament is statistically significant, and the significance of this study can be found that tried to reinterpret our tradition in various aspects by applying the Sagunja Temperament that matches the Korean emotion.

Prediction of cerebral infarction suppression mechanism of the Sagunja-Tang through network pharmacology analysis (네트워크 약리학 분석을 통한 사군자탕(四君子湯)의 뇌경색 억제 기전 예측)

  • Objectives : Sagunja-Tang is a famous prescription used in Korean medicine for the purpose of promoting vital energy, and there are few studies using Sagunja-Tang on cerebrovascular diseases yet. As previous studies confirmed that Sagunja-tang is highly likely to be used effectively for stroke, this study was intended to predict the mechanism through which Sagunja-tang would act effectively on stroke. Methods : In this study, a network pharmacology analysis method was used, and oral bioavailability (OB), drug likeness (DL), Caco-2 and BBB permeability were utilized to select compounds with potential activity. For the values of each variable used in this study, OB ≥ 30%, DL ≥ 0.18, Caco-2 ≥ 0, and BBB ≥ 0.3 were applied. Using the above variables, the relations between target genes and diseases that are presumed to be involved in the selected bioavailable compounds were constructed in a network format, and proteins thought to play a major role were identified. Results : Among the compounds included in Sagunja-Tang, 26 bioavailable compounds were selected and it was confirmed that these compounds can be effectively used in cerebrovascular diseases such as Alzheimer's disease and stroke. These compounds are considered to act on proteins related in cell death and growth. The most important mechanism of action was predicted to be apoptosis, and the protein that is thought to play the most key action in this mechanism was caspase-3. Conclusions : In our future study, Sagunja-Tang will be used in an ischemic stroke mouse model, and the mechanism of action will be explored focusing on apoptosis and cell proliferation.

Effect of Sagunja-tang on Immune Function of Mouse Immune Cells (四君子湯이 免疫機能에 미치는 影響)

  • Objectives : The extract of Sagunja-tang has been traditionally used for restorative treatment of constitutional weakness, vascular and immune disorder, and nervous disease in Oriental country. This study investigated the regulatory effects of Sagunja-tang on the expression, production, and activity of immune mediators.Methods : In this study, the extract of Sagunja-tang was prepared by extracting with distilled water at 100$^{\circ}C$ for 2.5h. The extract was freeze-dried following filtration through 0.45${{\mu}m}$ filter. The extract was dissolved in Hank's balanced salt solution (HBSS) and filtered again through 0.45${{\mu}m}$ filter before use. The level of nitrite, an oxidative product of nitric oxide(NO) was measured in the culture medium by the Griess reaction. The levels of prostaglandin E2(PGE2), Th1 cytokines (IFN-${\gamma}$, IL-2) and Th2 cytokines(IL-4, IL-5, IL-13) were measured by enzyme-linked immunosorbent assay and the protein levels of inducible nitric oxide synthase (iNOS) and cyclooxygenase-2 (COX-2) expression were determined by Western blot analysis. Also examined the effects of the extract on T-cell proliferation and cytotoxic activity of natural killer cells.Results : In this investigation, Production levels of Th2 cytokines (IL-4, IL-5, IL-13) was inhibited in a dose dependent manner by treatment with the extract. I also found that the extract increased T-cell proliferation and cytotoxic activity of natural killer cells in a dose-dependent manner.Conculsions : These results suggest that the water extract of Sagunja-tang may be useful for a therapeutic drug against a sickly constitution and immune diseases, probably by regulating the production of immune mediators.
