The effect of water extract composed of panax ginseng radix rubra, acanthopanacis cortex, and cordyceps (PAC) on diabetic animal models were investigated in two different diabetic animal models. FAC water extract significantly reduced the plasma glucose levels on day 30 as compared with the diabetic control group in $KKA^Y$ obese, hyperglycemic and hyperglycemic and hyperinsulinemic mice, and also reduced the plasma glucose levels as well as total cholesterol in multiple low dose (MLD) strep tozotocin-induced diabetic SD rats. PAC water extract also showed an inhibitory effect on reduction of body weight and on development of MLD STZ-induced diabetic state. Elevated kidney hypertrophy, which is a characteristic feature shown in early stage of diabetic nephropathy and calculated as the ratio of kidney mass (g) relative to the body weight (g), was also markedly improved in PAC water extract- treated group as compared to the diabetic control group. Taken together, these data suggest that PAC water extract may have a potential as a antidiabetic agent in type 2 diabetes mellitus.

This study was designed to investigate whether Sasa borealis leaves extracts (SLE) may inhibit yeast ${\alpha}-glucosidase$ and ${\alpha}-amylase$ activities and postprandial hyperglycemia in STZ-induced diabetic mice. Freeze-dried SLE was extracted with 70% methanol and followed by a sequential fractionation with dicholoromethan, ethylacetate, butanol, and water. Both ethylacetate and butanol fractions showed high inhibitory activities against the ${\alpha}-glucosidase$ and ${\alpha}-amylase$ enzymes. The $IC_{50}$ of ethylacetate and butanol fractions against ${\alpha}-glucosidase$ were 0.54 and 0.63 mg/mL, respectively, indicating a greater inhibition effect than acarbose (0.68 mg/mL) (p<0.05). Likewise, the two fractions exhibited a smaller $IC_{50}$ against ${\alpha}-amylase$, compared with acarbose (p<0.05). However, the yield of ethylacetate fraction of SLE was relatively small. Postprandial blood glucose testing of normal mice and STZ-induced diabetic mice by starch soln. loading (2 g/kg B.W.) showed that postprandial blood glucose level at 30, 60, and 120 min were markedly decreased by single oral administration of SLE butanol fraction (200 mg/kg B.W.) in both normal (p<0.0l) and diabetic mice (p<0.0l). Furthermore, the incremental area under the curve (AUC) was significantly lowered via SLE administration (5,745 versus 12,435 $mg{\cdot}mim/dL$) in the diabetic mice (p<0.0l). The incremental AUC in normal mice corroborated the hypoglycemic effect of SLE (p<0.0l) found in the diabetic mice. These results suggest that SLE may delay carbohydrate digestion and thus glucose absorption. In addition, SLE may have the potential to prevent and treat diabetes via its ability on lowering postprandial hyperglycemia.

The purpose of this study was to assess the effects of Cordyceps militaris extract supplementation on the improvement of blood glucose, lipid compositions and hepatic functional enzyme activities in the serum of streptozotocin(STZ, 50 mg/kg BW, IP injection)-induced diabetic rats fed the experimental diets for 5 weeks. The concentrations of blood glucose and lipid compositions(total cholesterol, ${\beta}$-lipoprotein, free cholesterol, cholesteryl ester, triglyceride and phospholipid) and atherosclerotic index in serum were significantly lower in the DM+CM group than in the DM group, whereas the ratio of HDL-cholesterol concentration to total cholesterol in the DM+CM group were higher than in the DM group. The activities of hepatic functional enzymes(aminotransferase, LDH and ALP) in serum were lower in the Cordyceps militaris extract administration group(group DM+CM) than in the hyperglycemic DM group. The results shown above suggested that Cordyceps militaris extract supplementation effectively improves blood glucose and atherosclerosis in the serum of diabetic rats.
Background: Traditional plant drugs, are less toxic and free from side effects compared to general synthetic drugs. They have been used for the treatment of diabetes and associated renal damage. In this study, we evaluated effect of Hachimi-jio-gan against diabetic renal damage in a rat model of type 1 diabetic nephropathy induced by subtotal nephrectomy plus streptozotocin (STZ) injection, and in Otsuka Long-Evans Tokushima Fatty (OLETF) rats and db/db mice as a model of human type 2 diabetes, and its associated complications. To explore the active components of Hachimi-jio-gan, the antidiabetic effect of corni fructus, a consituent of Hachimi-jio-gan, and 7-O-galloyl-${{\small}D}$-sedoheptulose, a phenolic compound isolated from corni fructus, were investigated. Methods and Results: We conducted an extensive literature search, and all required data were collected and systematically organized. The findings were reviewed and categorized based on relevance to the topic. A summary of all the therapeutic effects were reported as figures and tables. Conclusions: Hachimi-jio-gan serves as a potential therapeutic agent to against the development of type 1 and type 2 diabetic nephropathy. From the results of characterization active components of corni fructus, 7-O-galloyl-${\small}D$-sedoheptulose is considered to play an important role in preventing and/or delaying the onset of diabetic renal damage. 7-O-Galloyl-${\small}D$-sedoheptulose is expected to serve as a novel therapeutic agent against the development of diabetic nephropathy.
First, mice were dosed with 50mg/kg of streptozotocin(STZ) twice every 24 hours to cause high blood-sugar. Then, after three days, mice were injected with 100mg/kg of STZ again. Two different dosages of Saengjihwangeumja-gami were given to the experiment groups: SA group, 15mg/kg/day, and SB group, 90mg/kg/day, in order to determine the effects of Saengjihwangeumja-gami, which has been known to be good for DM(Diabetes Mellitus). By observing weight and blood-sugar level changes, blood tolerance, the numerical value of BUN(Blood Urea Nitrogen) and creatinine in blood, and through light-electronicmicroscopic and immunohistologic investigations of pancreas and kidneys, the following results were obtained: 1. The experiment groups showed a high suppressive effect of weight-loss. 2. The experiment groups' blood-sugar and blood tolerance showed an effective lowering of blood-sugar levels. 3. The experiment groups did not show any noticeable change in the numerical value of BUN and creatinine in blood compared with that of the control groups. 4. The experiment groups showed a higher Insulin positive reaction of pancreatic islets ${/beta}-cell$ than the control groups. 5. The experiment groups showed a higher immuno-reaction against IGF- II than the control groups. 6. Observation of apoptosis of the pancreatic islets showed that the cells of experiment groups were less injured compared with those of the control groups, and fewer apoptag-positive reaction cells were seen in experiment groups than in the control groups. 7. Uunder electron-microscopy, the insulin-containing granules in pancreatic islets ${/beta}-cells$ had increased more in the experiment groups than in the control groups. 8. Under light microscopy, the injury on the inner & outer membrane of the glomerulus and epithelial cells of capillaries and cells among vessels were fewer in the experiment groups than in the control groups. 9. More apoptag-positive reaction cells in the kidney were seen in the control groups than in the experiment groups. 10. PAS-positive reaction substances had increased more in the substrate among the vessels of a glomerulus belonging to the control group than those of the experiment group. 11. Uunder electron-microscopy, the nucleonic membrane, nucleoplasm and mitochondria of proximal and distal renal tubular were more injured in the control groups than in the experiment groups. In conclusion, strong evidence for the efficacy of Saengjihwangeumja-gami in lowering blood-sugar, and in recovery and generation of pancreatic tissues injured by DM was observed. Results suggest Saengjihwangeumja-gami is an effective treatment for DM. Further study of the principles of blood-sugar dropping effects of Saengjihwangeumja-gami are needed, as well as further study of recovery and regeneration of pancreatic tissues injured by DM.
The effects of Allium vegetables on blood glucose levels and lipid metabolism in streptozotocin (S12) induced diabetic rats were investigated. Diabetes mellitus was induced by S1'2 injection (45 mg/kg 5.w.) into the tail vein. Sprague-Dawley rats weighing $220\;{\pm}\;10\;g$ were randomly assigned to 7 groups: normal, S1'2-control and five Allium groups (Allium cepa, Allium fistulosum, Allium sativum, Allium tuberosum and Allium victorialisL Normal and S12control groups were fed an AIN-93 diet and five Allium groups were fed a modified diet containing. 10% Allium powder each for 4 weeks. Body weight, diet intake, food efficiency ratio (FER) and organ weights- were monitored. Activities of aspartate aminotransferase (AST) & alanine aminotransferase (ALT) were observed: Plasma lev~ls of glucose, free fatty acid, triglyceride and HDL-cholesterol were analyzed. Levels of glycogen, cholesterol and triglyceride in liver were determined. Levels of malondialdehyde (MDA) in liver, lung, kidney, and pancreas were assayed. The hepatic contents of chromium (Cr) , iron (Fe), zinc (Zn) and manganese (Mu) were measured. The Allium sativum group had weight gain and suppressed a hypertrophy of the kidney significantly. The activity of ALT was significantly lowered in the diabetic groups except Allium sativum group compared to STZ-control group. The Allium sativum and Allium tuberosum groups showed the hypoglycemic effects at 4 weeks. There were no significant differences between the control and all the other diabetic groups in the plasma levels of cholesterol, HDL-cholesterol, triglycerides and free fatty acids. Most of the Allium groups except Allium fistulosum were observed significantly lowered level of MDA in the lung compared to STZ-control group. The diabetic rats fed the Allium cepa and Allium sativum have shown significantly lowered hepatic Zn contents. The results suggested that the intake of the Allium vegetables may be effective in the antihyperglycemia by lowering blood glucose levels.
The effects of acetic acid fermented juice prepared with submerged culture media of Antrodia camphorata mycelium (AJA: pH 3.2, acidity 2.0, Brix degree 3.2) on blood glucose levels and serum lipid profiles of rats in which diabetes was induced with streptozotocin (STZ) were investigated. Rats were divided into normal controls (NC), diabetic controls (DM), and groups receiving diluted (1:1, with water) AJA (A1) and undiluted AJA (A2). The volume of liquid given to both A1 and A2 animals was 0.5 mL/100 g body weight. In the A1 and A2 groups, compared with the DM group, polyphagia, liver enlargement, and weight loss caused by diabetes were considerably alleviated, but did not attain the levels of NC group rats. In the A1 and A2 groups, the levels of blood glucose were 17.1% and 28.2% lower than in the DM group. There was no significant difference in the levels of fructosamine between the DM and A1 group, but the A2 group had a level 16.3% lower than the DM group. In the A1 and A2 groups, compared with the DM group, serum triglyceride levels decreased by 44.1-48.0%, serum total cholesterol by 24.0-31.1%, and serum LDL-cholesterol by 25.2-51.1%. The level of HDL-cholesterol in A2 animals rose by 45.9% compared to NC rats. The results show that AJA may be a useful beverage for diabetes patients, offering both antihyperglycemic activity and improvement in levels of serum lipids.
To elucidate the effect of water and methanol extracts of Cordyceps militaris, Paecilomyces japonicus and their mycelin on diabetes and organs in STZ-induced diabetic rats, weight of organs (liver, kidney, spleen, thymus), plasma level of blood glucose, total protein, triglyceride, free fatty acid, hepatic total protein, triglyceride and glycogen were determined as compared with those of negative control group. The blood glucose level of CM-1 and CM-M group showed significantly reduced, and all groups except CM-2 increased in body weight. CM-1 decreased the liver weight, and PJ-2 decreased the kidney weight. In all groups except PJ-2, plasma total protein level was increased, and the triglyceride, and CM-3 and CM-H decreased the free fatty acid was decreased in CM-3, PJ-1 and PJ-2 treated groups. In hepatic tissue, total protein was significantly increased in CM-H and CM-M treated group, and in all groups except CM-2, the triglyceride were significantly decreased and glycogen was increased. In conclusion, CM-1 and CM-M that possess potential antidiabetic activity increased glycogen and lowered serum glucose level, thus they might improve metabolic disorder originated from diabetes by increasing serum protein and reducing excess triglyceride in serum and liver tissue.

The purpose of this study was to investigate the effects of regular treadmill running on GLUT4 protein of skeletal muscle in STZ-diabetic rats. I used 19 male Sprague-Dawley rats weighing 140 to 160 grams. Rats were randomly assigned into normal, diabetes(DM) and DE(DE) groups. The exercise was loaded with treadmill running for 5 days per week during 4 weeks. All experimental procedures were carried out following overnight fasting 48 hours after last exercise. Gain(gm) in body weight in DM rats(822.4) was lowered compared to normal rats(1092.8), and decreased by exercise. Plasma glucose concentration(mg/dl) in DM rats was 1433.1 which is higher than that of normal group of 1036.4. The concentration of DE group was lower than that of DM rats. Plasma insulin concentration(${\mu}U/ml$) of DM and DE rats was significantly lowerd compared to normal rats. There was no difference of plasma concentrations of FFA and HDL cholesterol among noraml, DM and DE groups. Plasma triglyceride concentration(mg/dl) was significantly highered in DM group compared to those of DM group, the concentration of DE group was lower. Glycogen concentration(mg/gm wet weight) of the plantaris muscle in DM and DE groups was significantly reduced compared to normal group. Glucose transporter 4(GLUT4) protein of soleus was analyzed by Western blot. In DM group, the GLUT4 protein level was markdly decreased compared to normal group, but the level was recovered to the level of normal group by 4 weeks treadmill running. In conclusion, the insulin resistance induced by STZ administration was partially improved by 4 weeks physical training in rats.

In this study, we evaluated the antioxidant and antidiabetic effects of buckwheat. The diabetic animal models were divided into four groups: normal mice group (NOR), streptozotocin-induced diabetic mice group (STZ), group treated with seeds of common or tartary buckwheat (SCB or STB), and the group treated with whole plants of common or tartary buckwheat (PCB or PTB). Rutin content was 44-48 times higher in STB or PTB than in SCB. Oral glucose tolerance and insulin resistance were significantly reduced by treatment with STB, PCB, and PTB. Treatment with PTB also decreased the serum glucose level significantly and the serum insulin levels slightly compared with the STZ group. These results suggest that rutin content and antioxidant activity are closely related to the antidiabetic effect of the treatment. Our results demonstrate that the seeds of tartary buckwheat and whole plants of either common or tartary buckwheat have antidiabetic effects-attenuating blood glucose in an animal model of type II diabetes.
