Analysis of Differentially Expressed Genes in Kiwifruit Actinidia chinensis var. 'Hongyang' (참다래 '홍양' 품종의 차등발현유전자 분석)

  • We used suppression subtractive hybridization (SSH) combined with mirror orientation selection (MOS) method to screen differentially expressed genes from red-fleshed kiwifruit 'Hongyang'. As a result, the 288 clones were obtained by subcloning PCR product and 192 clones that showed positive clones on colony PCR analysis were selected. All the positive clones were sequenced. After comparisons with the NCBI/Genbank database using the BLAST search revealed that 30 clones showed sequence similarity to genes from other organisms; 10 clones showed significant sequence similarity to known genes. Among these clones, 3 clones (AcF21, AcF42 and AcF106) had sequence homology to 1-aminicyclopropane-carboxylic acid (ACC)-oxidase (ACO) that known to be related to fruit ripening. The expression patterns of differentially expressed genes were further investigated to validate the SSH data by reverse transcription PCR (RT-PCR) and quantitative real-time PCR (qReal-time PCR) analysis. All the data from qReal-time PCR analysis coincide with the results obtained from RT-PCR analysis. Three clones were expressed at higher levels in 'Hongyang' than 'Hayward'. AcF21 was highly expressed in the other genes at 120 days after full bloom (DAFB) and 160 DAFB of 'Hongyang'.

Correlation Analysis between Productivity of Forage Sorghum × Sudangrass Hybrids [Sorghum bicolor (L.) Moench] and Climatic Factors in Central Northern Region of South Korea (중북부지역에서 수수×수단그라스 교잡종의 생산성과 기후요인과의 상관분석)

  • Sorghum×sudangrass hybrid (Sorghum bicolor (L.) Moench, SSH) is one of the most important summer forage crop and it is widely used for silage in Korea. Agriculture is highly dependent on the climate condition and experiencing significant loss of productivity due to climate change. This study was conducted to investigate the correlation analysis between productivity of forage SSH and climatic factors in Central Northern region of South Korea for 3 years (2017 to 2019). Plant height and dry matter yield of SSH were significantly higher in Gyeonggi-do than Ganwon-do. The productivity of SSH is more closely related with temperature than other climatic factors. Maximum temperature and Growing degree days in May and June showed a positive correlation. However, correlation between production of SSH and precipitation was not clear in this study, but rainy days showed a negative correlation (0.42). In conclusion, temperature is most important climatic factor to the maintenance of plant yield.

Validation of Satellite Altimeter-Observed Sea Surface Height Using Measurements from the Ieodo Ocean Research Station (이어도 해양과학기지 관측 자료를 활용한 인공위성 고도계 해수면고도 검증)

  • Satellite altimeters have continuously observed sea surface height (SSH) in the global ocean for the past 30 years, providing clear evidence of the rise in global mean sea level based on observational data. Accurate altimeter-observed SSH is essential to study the spatial and temporal variability of SSH in regional seas. In this study, we used measurements from the Ieodo Ocean Research Station (IORS) and validate SSHs observed by satellite altimeters (Envisat, Jason-1, Jason-2, SARAL, Jason-3, and Sentinel-3A/B). Bias and root mean square error of SSH for each satellite ranged from 1.58 to 4.69 cm and 6.33 to 9.67 cm, respectively. As the matchup distance between satellite ground tracks and the IORS increased, the error of satellite SSHs significantly amplified. In order to validate the correction of the tide and atmospheric effect of the satellite data, the tide was estimated using harmonic analysis, and inverse barometer effect was calculated using atmospheric pressure data at the IORS. To achieve accurate tidal corrections for satellite SSH data in the seas around the Korean Peninsula, it was confirmed that improving the accuracy of tide data used in satellites is necessary.

Effects of 17 β -estradiol, bisphenol A and genistein on the expression of the glutathione peroxidase gene of Philasterides dicentrarchii (Ciliophora: Scuticociliata)

  • A subtracted cDNA library of a marine scuticociliate, Philasterides dicentrarchii, in response to 17β-estradiol exposure was constructed using suppression subtractive hybridization (SSH). As a result of SSH, 275 clones were isolated, and among them, only glutathione peroxidase (GPX) gene was isolated as an antioxidative enzyme responding to 17β-estradiol. The semi-quantitative reverse-transcriptase polymerase chain reaction (RT-PCR) analysis revealed that the transcription of GPX gene of P. dicentrarchii was clearly increased by exposure to 17β-estradiol. The GPX transcription was also clearly increased by exposure to xenoestrogens such as bisphenol A (BPA) and genistein.

  • 인터넷환경에서 네트워크구축이 보편화된 요즈음, 보안문제가 이슈로 대두되고 있다. 대형 네트워크상에서는 벌써 수년 전부터 방화벽이 보안장비의 중추를 담당하며 사용되고 있으나, 중소규모의 네트워크 환경을 유지관리하기 위한 보호대책은 미비한 편이다. 특히 라우터 전후에서 기능을 하는 방화벽 외에 대형 네트워크 안에서 중소규모의 네트워크를 운영하고자 할 때, 새로운 방화벽 서버를 사용할 수 있다면, 작은 규모의 네트워크에 적합한 패킷필터링이 가능하여, 외부의 침입으로부터 좀 더 안전한 네트워크의 구성이 가능해진다. 본 논문에서는 상대적으로 비용이 저렴하고 setup이 간단한 Linux 방화벽을 통해 외부망에서 접근할 수 없는 사설(private) IP주소를 사용하는 사설망(Private Network)안에 서버를 구성했을 때, Secure Shell에서 지원하는 Port forwarding 기능을 사용하여, 사설망안에 위치한 웹서버, 메일서버등에 접근할 수 있는 방법을 제시하고자 한다.

Isolation of Cold Acclimation-related Genes in Wheat Chromosome Substitution Line 5D

  • To identify low temperature-induced genes of wheat chromosome substitution line 5D, suppression subtractive hybridization (SSH) was performed with mRNAs from leaf samples that treated with low temperature ($4^{\circ}C$). A cDNA library was constructed using mRNA isolated from wheat chromosome substitution line 5D leaves treated with low temperature ($4^{\circ}C$). The nucleotide and deduced amino acid sequences of the putative gene products were compared. wfr-9 and wfr-32 showed identity over 90% related to vernalization gene. Other two genes, wfr-77 and wfr-83 which is related to freezing-resistant gene have also identity over 90%. This result suggest that those genes may be transcribed into antifreeze proteins which are accumulated within leaf apoplasts, when wheat chromosome substitution line 5D is acclimated during low temperature treatment.

Differentially Expressed Genes Related to Cold-resistance in Barley (Hordeum vulgare L. cv. Nagaoka)

  • To investigate genes related to vernalization and cold- resistance in barley (Hordeum vulgare L. cv. Nagaoka), differentially expressed genes were identified from cold-resistant barley leaves with suppression subtractive hybridization (SSH) and Northern blot analyses. The nucleotide and the deduced amino acid sequences of the putative gene products were compared. The bvrn-7 showed high homology(84%) with gene related to vernalization, and the bvrn-3, bvrn-12, bvrn-28, bvrn-29 and bvrn-36 related to cold-resistant genes had high identity of 88~98% with low temperature-induced genes. The results indicate that the 6 genes were closely related to vernalization and cold-resistance during low temperature treatment.

  • File Transfer Protocol (FTP) is a protocol for transferring files between computers, FTPS with TLS, and SFTP based on similar SSH. The keynote to apply one-time password (OTP) has been diffused to resolve identity and password theft cases. In this paper, it has been confirmed that both data transmission and identity authentication using OTP at TLS level are possible without significant change in user client.

  • 4 차 산업혁명의 시대를 맞아 사물인터넷 및 5G 를 활용한 수많은 사물들이 인터넷을 기반으로 연결되고 있다. 또한 이러한 사물들을 관제 및 유지 보수하기 위해서 장비들에 보안 관제 시스템을 구축하고 모니터링을 하기 위한 많은 비용과 관리의 어려움을 겪고 있다. 만약, 장비들이 스스로 능동적인 방어를 하게 된다면 유지관리의 가장 큰 문제가 해결될 것이다. 이러한 능동적인 보안을 통해 보호대상 시스템의 다양한 특징들을 시간의 변화에 따라 역동적으로 변경하는 MTD(Moving Target Defense)기법들이 개발되고 있다. 본 논문에서는 네트워크 기반의 NMTD(Network-based MTD)를 이용하여 호스트 서버에 IP 와 PORT 로 접속하는 SSH 에 적용하여 능동적으로 보호하고, OTP 를 활용하여 사용자 식별을 통해 SSH 에 대한 내부자 접속에 대한 보안을 강화하는 시스템을 설계 및 구현하였다.

Gene Expression Profiling in the Pituitary Gland of Laying Period and Ceased Period Huoyan Geese

  • Huoyan goose is a Chinese local breed famous for its higher laying performance, but the problems of variety degeneration have emerged recently, especially a decrease in the number of eggs laid. In order to better understand the molecular mechanism that underlies egg laying in Huoyan geese, gene profiles in the pituitary gland of Huoyan geese taken during the laying period and ceased period were investigated using the suppression subtractive hybridization (SSH) method. Total RNA was extracted from pituitary glands of ceased period and laying period geese. The cDNA in the pituitary glands of ceased geese was subtracted from the cDNA in the pituitary glands of laying geese (forward subtraction); the reverse subtraction was also performed. After sequencing and annotation, a total of 30 and 24 up and down-regulated genes were obtained from the forward and reverse SSH libraries, respectively. These genes mostly related to biosynthetic process, cellular nitrogen compound metabolic process, transport, cell differentiation, cellular protein modification process, signal transduction, small molecule metabolic process. Furthermore, eleven genes were selected for further analyses by quantitative real-time PCR (qRT-PCR). The qRT-PCR results for the most part were consistent with the SSH results. Among these genes, Synaptotagmin-1 (SYT1) and Stathmin-2 (STMN2) were substantially over-expressed in laying period compared to ceased period. These results could serve as an important reference for elucidating the molecular mechanism of higher laying performance in Huoyan geese.
