Accurate Measurement of Isotope Amount Ratios of Lead in Bronze with Multicollector Inductively Coupled Plasma Mass Spectrometry

  • Isotope amount ratios of lead in a bronze sample have been successfully determined using multicollector inductively coupled plasma mass spectrometry (MC-ICP-MS). Matrix separation conditions were tested and optimized using ion exchange chromatography with anion-exchange resin, AG1-X8, and sequential elution of the 0.5 M HBr and 7 M $HNO_3$ to separate lead from very high contents of copper and tin in bronze matrix. Mercury was also removed efficiently in the optimized separation condition. The instrumental isotope fractionation of lead in the MC-ICP-MS measurement was corrected by the external standard sample bracketing method using an external standard, NIST SRM 981 lead common isotope ratio standard followed by correction of procedure blank to obtain reliable isotope ratios of lead. The isotope ratios, $^{206}Pb/^{204}Pb$, $^{207}Pb/^{204}Pb$, $^{208}Pb/^{204}Pb$, and $^{208}Pb/^{206}Pb$, of lead were determined as $18.0802{\pm}0.0114$, $15.5799{\pm}0.0099$, $38.0853{\pm}0.0241$, and $2.1065{\pm}0.0004$, respectively, and the determined isotope ratios showed good agreement with the reference values of an international comparison for the same sample within the stated uncertainties

Axisymmetric Swirling Flow Simulation of the Draft Tube Vortex in Francis Turbines at Partial Discharge

  • The flow in the draft tube cone of Francis turbines operated at partial discharge is a complex hydrodynamic phenomenon where an incoming steady axisymmetric swirling flow evolves into a three-dimensional unsteady flow field with precessing helical vortex (also called vortex rope) and associated pressure fluctuations. The paper addresses the following fundamental question: is it possible to compute the circumferentially averaged flow field induced by the precessing vortex rope by using an axisymmetric turbulent swirling flow model? In other words, instead of averaging the measured or computed 3D velocity and pressure fields we would like to solve directly the circumferentially averaged governing equations. As a result, one could use a 2D axi-symmetric model instead of the full 3D flow simulation, with huge savings in both computing time and resources. In order to answer this question we first compute the axisymmetric turbulent swirling flow using available solvers by introducing a stagnant region model (SRM), essentially enforcing a unidirectional circumferentially averaged meridian flow as suggested by the experimental data. Numerical results obtained with both models are compared against measured axial and circumferential velocity profiles, as well as for the vortex rope location. Although the circumferentially averaged flow field cannot capture the unsteadiness of the 3D flow, it can be reliably used for further stability analysis, as well as for assessing and optimizing various techniques to stabilize the swirling flow. In particular, the methodology presented and validated in this paper is particularly useful in optimizing the blade design in order to reduce the stagnant region extent, thus mitigating the vortex rope and expending the operating range for Francis turbines.

  • 본 연구에서는 혈액 중 저농도의 카드뮴 분석을 위하여 시료를 1% Triton X-100으로 5배 희석시킨 다음에 흑연로 원자흡수분광법을 이용하여 분석하였다. 매트릭스 개선제로서 2% $(NH_4)_2HPO_4$와 L'vov platform이 삽입된 pyrocoated 흑연로를 사용하여 비교적 높은 회화온도 ($600^{\circ}C$)에서 카드뮴의 손실을 최대한 억제시키면서, 혈액내의 방해 성분으로 인한 영향을 감소시켰다. 분석의 정확성을 평가하기 위하여 NIST 표준물질(혈청 중 Cd) 및 외부 분석 기관(일본)에서 분석한 결과를 비교 검토하였다. 이 실험에서 얻은 상대 표준편차는 1.0ng/ml 농도에서 10% 정도이며 검출한계값은 0.1ng/ml였다.

대중매체 영향에 따른 일부 대학생의 식품안전 지식 조사 (Studies on Food Safety Knowledge of College Students according to Mass-Media Impact)

  • This study was conducted to assess food safety knowledge of college students according to mass-media reports. To accomplish this, a survey was conducted to investigate the reliability, of primary mass-media sources on Bovine Spongiform Encephalopathy(BSE) as well as the preferences of college students. The most common sources of media consumed by male and female students were terrestrial television(TV)(60%), the internet(21%), and newspapers(11%) and terrestrial TV(60%), newspapers (18%), and the internet(17%), respectively. The preferences for media of major and non-major students were terrestrial TV(64%), newspapers(17%), and the internet(14%) and terrestrial TV(51%), the internet(22%), and newspapers(19%), respectively. The reliability of media as viewed by male students were as follows: terrestrial TV(53%) and the internet(40%), whereas for female students, these values were terrestrial TV(55%) and the internet(37%). The reliability of media as viewed by major and non-major students was as follows: terrestrial TV(67%) and the internet(28%), and terrestrial TV(44%) and the internet(44%), respectively. In the case of male students, the primary sources for information regarding BSE were terrestrial TV(54%) and the internet(38%), whereas for female students, the primary sources were terrestrial TV(57%) and internet (37%). In the case of major students, the primary sources for information regarding BSE were terrestrial TV(52%) and the Internet(40%), and for non-major students, the internet(47%) and terrestrial TV(39%). Both male and female students were found to have less knowledge regarding prions, SRM(Specified Risk Materials), MM(methionine-methionine) type genes, and cow above 30 months associated with BSE compared to the other factors associated with this disease. The BSE-related knowledge held by major and non-major students was revealed unknown more than the majority of responses. Based on these results, greater effort should be made to provide meaningful information to improve the food-safety knowledge of college students according to mass-media.

Metaheuristic models for the prediction of bearing capacity of pile foundation

  • The properties of soil are naturally highly variable and thus, to ensure proper safety and reliability, we need to test a large number of samples across the length and depth. In pile foundations, conducting field tests are highly expensive and the traditional empirical relations too have been proven to be poor in performance. The study proposes a state-of-art Particle Swarm Optimization (PSO) hybridized Artificial Neural Network (ANN), Extreme Learning Machine (ELM) and Adaptive Neuro Fuzzy Inference System (ANFIS); and comparative analysis of metaheuristic models (ANN-PSO, ELM-PSO, ANFIS-PSO) for prediction of bearing capacity of pile foundation trained and tested on dataset of nearly 300 dynamic pile tests from the literature. A novel ensemble model of three hybrid models is constructed to combine and enhance the predictions of the individual models effectively. The authenticity of the dataset is confirmed using descriptive statistics, correlation matrix and sensitivity analysis. Ram weight and diameter of pile are found to be most influential input parameter. The comparative analysis reveals that ANFIS-PSO is the best performing model in testing phase (R2 = 0.85, RMSE = 0.01) while ELM-PSO performs best in training phase (R2 = 0.88, RMSE = 0.08); while the ensemble provided overall best performance based on the rank score. The performance of ANN-PSO is least satisfactory compared to the other two models. The findings were confirmed using Taylor diagram, error matrix and uncertainty analysis. Based on the results ELM-PSO and ANFIS-PSO is proposed to be used for the prediction of bearing capacity of piles and ensemble learning method of joining the outputs of individual models should be encouraged. The study possesses the potential to assist geotechnical engineers in the design phase of civil engineering projects.

An In Silico Drug Repositioning Strategy to Identify Specific STAT-3 Inhibitors for Breast Cancer

  • Breast cancer continues to pose a substantial worldwide health challenge, thereby requiring the development of innovative strategies to discover new therapeutic interventions. Signal Transducer and Activator of Transcription 3 (STAT-3) has been identified as a significant factor in the development of several types of cancer, including breast cancer. This is primarily attributed to its diverse functions in promoting tumour formation and conferring resistance to therapeutic interventions. This study presents an in silico drug repositioning approach that focuses on identifying specific inhibitors of STAT-3 for the purpose of treating breast cancer. We initially examined the structural and functional attributes of STAT-3, thereby elucidating its crucial involvement in cellular signalling cascades. A comprehensive virtual screening was performed on a diverse collection of drugs that have been approved by the FDA from zinc15 database. Various computational techniques, including molecular docking, cross docking, and cDFT analysis, were utilised in order to prioritise potential candidates. This prioritisation was based on their predicted binding energies and outer molecular orbital reactivity. The findings of our study have unveiled a Dihydroergotamine and Paritaprevir that have been approved by the FDA and exhibit considerable promise as selective inhibitors of STAT-3. In conclusion, the utilisation of our in silico drug repositioning approach presents a prompt and economically efficient method for the identification of potential compounds that warrant subsequent experimental validation as selective STAT-3 inhibitors in the context of breast cancer. The present study highlights the considerable potential of employing computational strategies to expedite the drug discovery process. Moreover, it provides valuable insights into novel avenues for targeted therapeutic interventions in the context of breast cancer treatment.

Bacillus subtilis DEAD-Box RNA Helicase 유전자 결손 균주들의 저온 민감성 생장 (Cold-Sensitive Growth of Bacillus subtilis Mutants Deleted for Putative DEAD-Box RNA Helicase Genes)

  • Bacillus subtilis에 존재하는 DEAD-box RNA helicase에 대한 유전자 상동성 검색을 통해 yqfR, yfmL, ydbR, deaD 의 4종류의 유전자를 확인하였고 이들 유전자 각각의 결손 돌연변이체를 제조하였다. 이들 돌연변이체들의 특성을 알아보기 위하여 LB 배양액을 사용하여 여러 온도에서의 생장 속도를 조사하였다. LB 배양액에서 $37^{\circ}C$의 생장 결과 ydbR 결손 균주가 다소 생장이 느려지나($T_d$=53 min) 다른(yqfR, yfmL, deaD) 결손 돌연변이체들은($T_d$=30-40 min) 결손이 없는 야생형 균주 CU1065와($T_d$=32 min) 유사하였다. 반면에 $22^{\circ}C$에서의 생장은 CU1065 ($T_d$=102 min)에 비해 yqfR ($T_d$=151 min), yfmL ($T_d$=214 min), ydbR ($T_d$=343 min) 결손 균주 순으로 생장속도가 느린 저온 민감성을 보인다. deaD의 $22^{\circ}C$에서의 생장 속도는 ($T_d$=109 min) CU1065와 ($T_d$=102 min) 매우 유사하여 저온 민감성을 보이지 않았다. 그리고 이들 유전자들의 이중, 삼중, 사중의 결손 균주들을 제조하였고, 여러 온도에서 ($42^{\circ}C$, $37^{\circ}C$, $22^{\circ}C$) LB 배양액을 사용하여 생장 속도를 측정 하였다. 다중 결손은 단일 결손보다 더 심한 저온 민감성을 보이며, 이중 결손의 경우, ydbR과 yfmL의 결손이 다른 조합의 결손보다 보다 큰 저온 민감성을 나타내었다 ($T_d$=984 min). 이러한 저온 민감성은 E. coli의 csdA 혹은 srmB 결손의 결과와 유사하며 리보솜 조립과 관련이 있는 생리적 기능으로 보인다.

동토지반에서 종방향 및 횡방향 사면의 경사에 따른 트렌치 안전성 분석 (Analysis of Trench Slope Stability in Permafrost Regions According to the Vertical and Horizontal Angle of Slope)

  • 동토지역에서의 파이프라인 시공 시 계절변화와 그에 따른 지반의 강도변화가 트렌치의 안정성에 영향을 미칠 수 있다. 영국, 러시아, 국내 파이프라인 기준들을 분석하여 직경 30in. 파이프의 트렌치 형상을 도출하였다. 러시아 야쿠츠크(Yakutsk) 지역의 계절별 지반조건에서 횡방향 사면경사($0^{\circ}$, $10^{\circ}$, $20^{\circ}$, $30^{\circ}$)와 종방향 사면경사($20^{\circ}$, $30^{\circ}$, $40^{\circ}$)에 따른 트렌치의 안정성을 분석하였다. 강도감소법을 이용하여 트렌치와 사면의 안정성 해석을 수행하였다. 그 결과, 사면의 경사가 높을수록 안전율이 낮게 나왔으며 여름철 지반조건에서는 횡방향 사면경사가 $30^{\circ}$일 때 트렌치의 안정성을 확보하기 어려우며 지상으로부터 1m 아래에서 예상 파괴면을 확인하였다. 종방향 사면의 경사가 낮을 때에는 트렌치 부근의 파괴가 일어날 가능성이 높았지만 종방향 사면의 경사가 높을 때에는 트렌치 부근의 파괴 보다는 사면 전체의 파괴가 주를 이루는 것으로 분석되었다. 겨울철 지반조건에서는 지표면의 온도가 영하로 내려가서 지반 공극 내 얼음 발생으로 인하여 점착력이 발생되는 효과가 있어, 특수한 외부 하중이나 급격한 온도변화가 없을 경우에는 트렌치 사면 경사 $0{\sim}40^{\circ}$에서 안전성의 문제는 없을 것으로 판단되었다.

암환자에게 다빈도로 활용되는 한약제제 10종에 대한 요오드의 함량 분석 (Determination of Iodine Contents in Ten kinds of Frequently used Oriental Herb Medicinal Products for Cancer Patient)

  • Background and Objectives: Iodine is an essential constituent of the thyroid hormones associated with the growth and development of humans and animals as an inorganic nutrition. This element may be accumulated in human blood, tissues and body through the intake of foodstuffs, a beverage, a nutritional supplement and a medicine, among others. The aim of the research is to find out a better medicinal stuff for the thyroid cancer patient who required a low level of iodine diet. Methods: Neutron activation analysis (NAA) used for the iodine analysis is one of nuclear analytical techniques using radiation and radioisotopes and very useful as sensitive analytical technique for performing both qualitative and quantitative multi-elemental non-destructive analysis of major, minor and trace components in variety of environmental and biological materials. In this study, iodine contents in ten kinds of oriental herb medicinal products, which is frequently used to cancer patients are determined by using instrumental neutron activation analysis (INAA) at the HANARO research reactor. The samples prescribed are manufactured as powdered form for taking medicine easily. The analytical quality control is performed to assure an uncertainty of the measurement and to compensate the measured data using a biological certified reference material, NIST SRM 1572, Citrus Leaves. The measured value is $1.89{\pm}0.35mg/kg$, and the relative error is 2.88%, and relative standard deviation is 19 % due to high counting error by small counts of gamma ray spectrum. The standard deviations for other elements such as Cl, K, Mn and Na were in the range of 2 to 8%. Result: The level of iodine contents of Biki-huan, Chungryong-huan and Chungcho-huan, samples detected is less than 6 mg/kg except Hangam Plus sample (more than 210 mg/kg) and six samples were not detected. Iodine in the samples of Shoxiho-tang, Shopunghualhyl-tang, Shocungryong-tang, Banhasaxim-tang, Insampaedox-san and Myunyuk Plus were not measured, but possible level of content can be estimated from the detection limits. In addition, the concentrations of some major elements like Cl, K, Mn, Na, in the samples were determined with the detection limits. Conclusions: Most of samples showed low iodine contents of less than 6 mg/kg but it turned out that most of testing samples can be used to classify the level of iodine diet samples considering the recommended low level of iodine diet 50 ${\mu}g$/day, and a better medicinal stuff for the thyroid cancer patient can be found.

Two Stage Gasifier에서의 우드펠릿 가스화 특성 연구 (A Study on Characteristics of Wood Pellet Gasification in Two Stage Gasifier)

  • 화석연료는 사용 후 재생이 불가능하고 매장량이 한정되어 있으며, 연소 시 발생되는 각종 공해물질로 인해 환경문제를 야기하고 있다. 이러한 맥락에서 차세대 청정대체에너지로서 주목을 받고 있는 것이 바로 수소에너지이다. 현재 가장 경제성이 있는 수소제조방법으로 알려진 천연가스 Steam Reformig(SRM)은 천연가스의 매장량 한계성으로 인해 그 제조비용이 높아지고 있어, 바이오매스 및 유기성 폐기물의 가스화를 통한 수소생산방법이 자원의 재순환, 페기물 처리, 열원의 이용, 직접적인 $CO_2$ 삭감 등의 부수적인 효과가 높아 경제성 있는 수소제조법으로 많은 연구가 진행되고 있다. 이에 본 연구에서는 잠재적으로 고갈 염려가 있는 화석연료를 대체하고, 화석연료의 연소 시 발생되는 환경문제를 해결하고자 열분해로와 고온개질기로 구성된 Pilot-scale Two Stage Gasifier를 개발하고, 본 장치 내에서의 biomass의 가스화 특성을 평가하고자 한다. 열분 해로에서의 가스화 실험 결과, 열분해로의 전환율은 약 70%로 나타났으며, $H_2$, $CH_4$, CO, $CO_2$의 평균 생성량은 각각 16.7, 11.3, 37.2, 26.6 L/mim의 결과를 보였다. 고온개질기로부터의 생성가스 수율의 결과로부터, 고온개질기에 적용된 $1100^{\circ}C$의 초고온에서의 개질 반응에 의해 $CH_4$의 대부분이 환원됨을 확인할 수 있었다. 본 연구로부터 개발된 장치의 냉가스 효율은 53.2%로 비교적 높은 결과가 얻어졌으며, 수소에 대한 평균 생성량은 55.4 L/min의 결과를 보였다.
