Parasites of Freshwater Fishes in Cheju-do (제주도산(濟州道産) 담수어류(淡水魚類)에 기생(寄生)하는 기생충(寄生蟲)에 관한 연구(硏究))

  • Parasites of freshwater fishes in Cheju-do were studied from May 1989 to April 1990, and incidence of infection in 16 fish species was reported. Protozoan parasites (Trichodina sp., Chilodonella sp., Ichthyophthirius sp., Vorticella sp., Myxidium sp., Myxobolus sp., Henneguya sp:, Ichthyobodo sp., and Trychophrya sp.), water mold (Saproregnia sp.), two monogenes (Dactylogyrus sp. and Gyrodactylogyrus sp.), Trematods, Cestods, Nematods, Acanthocephalas, parasitic copepods(Lernaea sp. and Pseudergasilus sp.) and a Hirudinea were recognized as freshwater fish parasites in Cheju-do. Trichodina sp. showed the highest infection rate (18.3%). Fifty seven individual fishes out of 311 were infected by this parasite. Nematods showed the second highest infection rate (13.5%). Dactylogyrus sp., Acanthocephalas, and Trematods showed the third (4.8%), fourth (4.2%), and fifth (2.6%) infection rate respectively. Of the 16 fish species Cryptocentrus filifer (Gobiidae) showed the highest infection rate. Nineteen fish out of 28 have Trichodina sp., and 14 fish out of 28 have Nematods. Those infection rates were 67.9% and 50.0% respectively. No parasites were collected from the fishes of Gwangryung vally, Dosoon-chun, Gangjeong-chun, and Hyodon-chun.

Current status of pathogen infection in cultured eel Anguilla japonica between 2000 and 2010 (2000~2010년 우리나라 양식산 뱀장어, Anguilla japonica의 병원체 감염현황)

  • Disease survey was conducted to investigate the cause of high mortality in 23 farms of eel, Anguilla japonica, during the period from 2000 to 2010. Seven kinds of fish pathogens were confirmed in the tested fish, which included: Pseudodactylogyrus sp. (infection rate: 65.5%, 19/29 samples), aquabirnavirus (ABV, 62.5%, 15/24 samples), Edwardsiella sp. (44%, 11/25 samples), anguillid herpesvirus 1 (AngHV1, 16.7%, 2/12 samples), Heterosporis sp. (10.3%, 3/29 samples), Vibrio sp. (8%, 2/25 samples), Trichodina sp. (3.4%, 1/29 samples). The rate of single infection was 44.8% (13/29 samples), while 62.1% (18/29 samples) showed mixed type of infection with 2 to 5 different pathogens. The most predominant mixed infection were Pseudodactylogyrus sp.-Edwardsiella sp. (10.3%, 3/29 samples), Pseudodactylogyrus sp.-ABV (10.3%, 3/29 samples), Edwardsiella sp.-ABV (6.9%, 2/29 samples). Infection by the above infectious parasites, bacteria and virus was confirmed in tested eels of juvenile to adult sizes that were cultured at $25{\sim}30^{\circ}C$ in almost the examined farms.

Characterization of Bacteria Isolated from Pine Wood Nematodes in Korea (국내 소나무재선충에서 분리한 세균의 특성)

  • A survey of bacterial species associated with Korean isolates of pine wood nematode (PWN) was performed. A total of 110 bacterial isolates were obtained from the PWN isolates that were previously isolated from Pinus densiflora and P. koraiensis. Among the bacterial isolates, Cedecea neteri was most frequent (64 isolates) followed by Ewingella americana (21 isolates), Pseudomonas sp. (15 isolates), Flavobacterium sp. (8 isolates) and Rahnella aquatilis (2 isolates). Both E. americana and Pseudomonas sp. which are assumed to be closely associated with PWN were examined for their phytotoxicity to P. thunbergii seedlings. Ethyl acetate extracts of Psuedomonas sp. (Ba2 strain) cultures were found to induce wilting and mortality in the tested seedlings. The three bacterial species, Pseudomonas sp. (Ba2 strain), E. ameircana (Ba4 strain) and C. neteri (Ba10 strain) were examined in vitro for their sensitivity to 21 kinds of antibiotics. All of the strains were highly susceptible to carbenicillin, doxcycline and tetracycline.

Identification and Antioxidant Activity using Electron Spin Resonance Spectrometry of Antioxidant Producing Marine Actinomycetes Streptomyces sp. ACT-18 (항산화물질을 생산하는 해양방선균 Streptomyces Sp. ACT-18의 동정 및 Electron Spin Resonance Spectrometry를 이용한 항산화활성)

  • For the research of the natural marine antioxidant, an antioxidant-producing marine actinomycetes was isolated from sea water in Jeju coastal area. The strain was identified based on 16S rDNA sequencing, the morphology by a method of scanning electron microscopy, physiological and biochemical characteristics and cellular fatty acid analysis. The isolated strain ACT-18 was gram positive, aerobic, non-motile spores. Substrate mycelia are dark green and yellow gray aerial mycelia. The cell size of the strain was $0.5{\sim}1.0\;{\mu}m$. 16S rDNA sequence analysis showed that were Gram-positive bacteria grouped on Streptomyces sp. Results of cellular fatty acid analysis showed that major cellular fatty acids were $C_{15:0}$ anteiso (39.33%), $C_{16:1}$ cis 9 (11.96%), $C_{16:0}$ (13.08%) and $C_{17:0}$ anteiso (10.99%). The antioxidant activity of methanol extract from Streptomyce sp. ACT-18 was evaluated by measuring 1,1-diphenyl- 2-picrylhydrazyl (DPPH), hydroxyl, and alkyl radical scavenging activity using an electron spin resonance (ESR) spectrometer. DPPH radical scavenging activity of SBME (Streptomyces Broth Methanol Extract) A-18 was 46% at 0.1 mg/mL. Hydroxyl radical scavenging activity of SBME A-18 was 63% at 0.1 mg/mL. Alkyl radical scavenging activity of SBME A-18 was 39% at 0.1 mg/mL.

Biodegradation of diesel oil and n-alkanes (C18, C20, and C22) by a novel strain Acinetobacter sp. K-6 in unsaturated soil

  • A large residual fraction of aliphatic components of diesel prevails in soil, which has adverse effects on the environment. This study identified the most bio-recalcitrant aliphatic residual fraction of diesel through total petroleum-hydrocarbon fractional analysis. For this, the strain Acinetobacter sp. K-6 was isolated, identified, and characterized and investigated its ability to degrade diesel and n-alkanes (C18, C20, and C22). The removal efficiency was analysed after treatment with bacteria and nutrients in various soil microcosms. The fractional analysis of diesel degradation after treatment with the bacterial strains identified C18-C22 hydrocarbons as the most bio-recalcitrant aliphatic fraction of diesel oil. Acinetobacter sp. K-6 degraded 59.2% of diesel oil and 56.4% of C18-C22 hydrocarbons in the contaminated soil. The degradation efficiency was further improved using a combinatorial approach of biostimulation and bioaugmentation, which resulted in 76.7% and 73.7% higher degradation of diesel oil and C18-C22 hydrocarbons, respectively. The findings of this study suggest that the removal of mid-length, non-volatile hydrocarbons is affected by the population of bio-degraders and the nutrients used in the process of remediation. A combinatorial approach, including biostimulation and bioaugmentation, could be used to effectively remove large quantities of aliphatic hydrocarbons persisting for a longer period in the soil.

Monophyly of the Family Desmoscolecidae (Nematoda, Demoscolecida) and Its Phylogenetic Position Inferred from 18S rDNA Sequences

  • To infer the monophyletic origin and phylogenetic relationships of the order Desmoscolecida, a unique and puzzling group of mainly free-living marine nematodes, we newly determined nearly complete 18S rDNA sequences for six marine desmoscolecid nematodes belonging to four genera (Desmoscolex, Greeffiella, Tricoma and Paratricoma). Based on the present data and those of 72 nematode species previously reported, the first molecular phylogenetic analysis focusing on Desmoscolecida was done by using neighbor joining (NJ), maximum parsimony (MP), maximum likelihood (ML) and Bayesian inference (BI) methods. All four resultant trees consistently and strongly supported that the family Desmoscolecidae forms a monophyletic group with very high node confidence values. The monophyletic clade of desmocolecid nematodes was placed as a sister group of the clade including some members of Monhysterida and Araeolaimida, Cyartonema elegans (Cyartonematidae) and Terschellingia Iongicaudata (Linhomoeidae) in all the analyses. However, the present phylogenetic trees do not show any direct attraction between the families Desmoscolecidae and Cyartonematidae. Within the monophyletic clade of the family Desmoscolecidae in all of the present phylogenetic trees, there were consistently observed two distinct subgroups which correspond to the subfamilies Desmoscolecinae [Greeffiella sp. + Desmoscolex sp.] and Tricominae [Paratricoma sp. + Tricoma sp].

Isolation of Plasmids from the Moderately Halophilic Bacteria (Moderate 호염성 세균의 Plasmid 유전자 분리)

  • Moderately halophilic bacteria were collected from solar salt with Larsen medium containing $10\%$ NaCl. A total of 56 strains were isolated and tested for the presence of plasmid DNA by agarose gel electrophoresis. Twelve isolates ($21\%$) carried at least one kind of plasmid. Six different isolates among them were selected to study the molecular weight of plasmids and the morphological and physiological characters. Vibrio sp. 14, Alcaligenes sp. 63, Pseudomonas sp. 11, Flavobacterium sp. 38, Bacillus sp. 16, and Alcaligenes sp. 52 carried at least one plasmid of about 7.2 kbp, 6.4 kbp, 6.85 kbp, 8.5 kbp, 8.75 kbp, and 6.8 kbp respectively.

Effect of Seaweeds added in preparation of Tofu (두부제조시 해조류 첨가 효과)

  • This study was performed to investigate some quality characteristics of 새려 prepared from soybean milk and various seaweed(Undaria pinnatifida, Laminaria japonica, Porpyra tenera, Enteromorpha sp., Codium sp.) pulps in the ration of 9:1(v:v) with 20% MgCl2. The yields of tofu containing Undaria pinnatifida, Laminaria japonica, or Enteromorpha sp. increased but porphyra tenera, Codium sp. decreased in comparison with tofu prepared from whole soybean milk. The protein content of tofu containing Undaria pinnatifida, Laminaria japonica, Porphyra tenera, or Codium sp. increased but Enteromprpha sp. decreased in comparison with the tofu prepared from whole soybean milk. The content of Ca in Undaria pinnatifida, Porphyra tenera added tofu was higher than that of the tofu prepared from whole soybean milk or other seaweeds added tofu. In sensory evaluation the texture, color, taste of tofu were favored with the addition of sea mustard(Undaria pinnatifida) pulp than that of the tofu prepared from whole soybean milk or tofu prepared other seaweed. Tofu prepared was possible with adding 0.5~1.5% sea mustard to soybean milk but the feasible added amount level was 1% of sea mustard. The yields, protein Ca, and K content of tofu were increased by the more adding amount of sea mustard tan tat of the tofu prepared from whole soybean milk. The hardness values of 1% sea mustard added tofu were decreased than that of the tofu prepared from whole soybean milk ; on the other hand, elasticity, cohensiveness, gumminess and brittleness of tofu with sea mustard increased. The L and a values of tofu were lower and b values were higher with the addition of 1% sea mustard. The content of histidine, tyrosine, leusine, and phenylalanine were decreased but the other amino acid were increased in tofu prepared from 1% sea mustard pulp added to soybean milk. The saturated fatty acid and monoene fatty acid content of tofu were increased and C18:2, C18:3(${\gamma}$), C18:3, C20:5 of polyene fatty acid were slightly decreased in tofu prepared from 1% sea mustard pulp added to soybean milk.

Assay of ${\beta}$-Glucosidase Activity of Bifidobacteria and the Hydrolysis of Isoflavone Glycosides by Bifidobacterium sp. Int-57 in Soymilk Fermentation

  • The isoflavone glycosides are hydrolyzed by ${\beta}$-glucosidase from gut microbes to the bioactive aglycones. However, the specific bacteria from the human intestinal tract that are involved in the metabolism of these compounds are not known. This study was undertaken to develop a fermented soymilk which converts isoflavones to the more bioactive aglycones form using a Bifidobacterium strain. The ${\beta}$-glucosidase activity of 15 Bifidobacterium strains were measured during cell growth. Among them, Bifidobacterium sp. Int-57 was selected for this study, because it has the highest ${\beta}$-glucosidase activity. Growth, acid development, ${\beta}$-glucosidase activity, and the hydrolysis of daidzin and genistin were investigated in four soymilks inoculated with Bifidobacterium sp. Int-57. After 12 h of fermentation, the counts of viable Bifidobacterium sp. Int-57 in all the soymilks reached a level of more than $10^8$ cfu/ml, which was then maintained. The pH of soymilks started to decrease rapidly after 6 h of fermentation and leveled off after 18 h. The titratable acidity of BL# 1 soymilk, BL#2 soymilk, and JP#l soymilk increased from 0.18 to 1.21, 1.15, and $1.08\%$ over the fermentation period, respectively. After 24 h of fermentation, the $\beta$-glucosidase activity in BL#1 soymilk, BL#2 soymilk, JP#l soymilk, and JP#2 soymilk increased to 59.528, 40.643, 70.844, and 56.962 mU/ml, respectively. The isoflavone glycosides, daidzin and genistin, in soymilks were hydrolyzed completely in the relatively short fermentation time of 18 h. These results show that Bifidobacterium sp. Int-57 can be used as a potential starter culture for developing fermented soymilk which has completely hydrolyzed isoflavone glycosides.
