5-Aza-2'-deoxycytidine Inhibits the Maintenance of Cancer Stem Cell in a Mouse Model of Breast Cancer (마우스 유방암 모델에서 5-Aza-2'-deoxycytidine의 암줄기세포 유지 억제 효과)

  • Aberrant DNA methylation plays an important role in the development of cancer. It has been reported recently that DNA hypermethylation is involved in the maintenance of cancer stem cells. The present study was designed to test the hypothesis that the demethylating agent, 5-aza-2'-deoxycytidine (AZA), can inhibit the potential for maintenance of cancer stem cells. To validate this hypothesis, we used 4T1 syngeneic mouse models of breast cancer. The AZA pre-treated 4T1 cells showed a dramatic inhibition of tumorsphere formation, compared to their counterparts in vitro. In addition, the AZA treatment significantly suppressed the expression of stem regulator genes, such as oct-4, nanog and sox2, compared to counterparts in vivo. Therefore, selective inhibition of DNA methylation may be useful for stem-specific cancer therapy.

An Analysis of Changes in Air Pollutant Emissions Due to the Introduction of the Special Act on the Improvement of Air Quality in Port Areas -Focusing on Incheon Port (항만지역등 대기질 개선에 관한 특별법 도입에 따른 대기오염물질 배출량 변화 분석 -인천항을 중심으로 )

  • This study aims to analyze the effect of reducing air pollutant emissions of the ship fuel oil sulfur content regulation policy, which has been gradually introduced for three years. In addition, the emission reduction effect of VSR and AMP was also analyzed. The analysis was conducted on NOx, CO, VOC, SOx, TSP, PM10, and PM2.5, which are provided by EEA, and the spatial scope of the analysis was conducted on Incheon Port, which is located in the metropolitan area of Korea and has a large ripple effect on air pollution. Three scenarios were constructed for analysis. Scenario 1: If there is no policy, Scenario 2: If only fuel oil sulfur content regulation was implemented, Scenario 3: The analysis was conducted by reflecting fuel oil sulfur content regulation, VSR, and AMP. As a result of the analysis, in the case of scenario 1, 4,801 tons, 4,932 tons, and 5,144 tons of air pollutants were emitted during the three-year period. In Scenario 2, 4,219 tons, 4,152 tons, and 3,989 tons were discharged, and in Scenario 3, 4,198 tons, 4,138 tons, and 3,973 tons were discharged. The findings of this study are anticipated to be applied as fundamental research in port air environment management and Incheon Metropolitan City air management.

Evaluation of Biological Activity of Veronica incana Extracts (Veronica incana 추출물의 생물학적 활성 평가)

  • Objectives : The aim of this study is to evaluate the potential biological activity of Veronica incana extracts (VIE) through in vitro, ex vivo, and in vivo experiments. Methods : In vitro, we conducted analyses on the total polyphenol (TP) and total flavonoid (TF) levels, alongside DPPHand ABTS radical scavenging activities. Ex vivo evaluations on adipose tissue measured glycerol release as a marker of lipolysis. In LPS-induced RAW 264.7 cells, we quantified nitric oxide (NO) production. Following H2O2 induction in U2OS cells, we performed mitochondrial assays such as MitoSox and MitoTracker. Moreover, Bodipy assays were conducted in 3T3-L1 cells. In vivo, we performed anti-osteoarthritis effect of VIE against monosodium iodoacetate (MIA)-induced osteoarthritis in rats. Results : The results presented encompass a myriad of models, from cell culture to animal experiments as well as ex vivo studies. VIE demonstrated high TP and TF contents, potent DPPH and ABTS scavenging activities, and regulated glycerol release. Moreover, the inhibition of NO production in LPS-induced inflammation was notably confirmed and the reduction of lipid droplets was distinctly shown. Furthermore, in H2O2-induced U2OS cells, MitoSox was effectively reduced while MitoTracker noticeably increased. In vivo assays confirmed a significant increase in hindpaw weight distribution (HWD) decreased by MIA after VIE treatment. Additionally, VIE inhibited serum inflammatory cytokines (TNF-𝛼, IL-6, and IL-1𝛽) and MDA levels in joint tissue. Conclusion : In conclusion, Veronica incana exhibited various pharmacological effects including antioxidant, anti-obesity, and anti-inflammatory properties.

Production of Biodiesel from Fleshing Scrap Using Immobilized Lipase-catalyst (Lipase-catalyst를 이용한 프레싱 스크랩의 바이오디젤 제조에 관한 연구)

  • This study was carried out to investigate the reaction of lipase-catalyst transesterification using animal fat recovered from fleshing scrap generated during leather making process. Transesterification reaction between fat and primary or secondary alcohol was carried out under the condition of immobilized enzyme catalyst. The conversion rate was the highest when 1.5 mole of methanol was injected by 4 times. As for lipase, Candida antarctica showed the highest conversion rate of 82.2% among the 4 different lipases. It was found that water contained in the fat causes lower conversion rate. The condition of 1.2wt. % of water in the fat decreased the conversion rate by 40%. It was considered that the resulted reactant, fatty acid ester could be used as raw material for biodiesel with the characteristics of not generating SOx and diminishing smoke.

An Study on Estimating Cargo Handling Equipment Emission in the Port of Incheon (인천항 하역장비 대기오염물질 배출량 산정 연구)

  • Currently, in-port emissions are a serious problem in port cities. However, emissions, especially non-greenhouse gases, from the operation of cargo handling equipment (CHE) have received significant attention from scientific circles. This study estimates the amount of emissions from on-land port diesel-powered CHE in the Port of Incheon. With real-time activity data provided by handling equipment operating companies, this research applies an activity-based approach to capture an up-to-date and reliable diesel-powered CHE emissions inventory during 2017. As a result, 105.6 tons of carbon monoxide (CO), 243.2 tons of nitrogen oxide (NOx), 0.005 tons of sulfur oxide (Sox), 22.8 tons of particulate matter (PM), 26.0 tons of volatile organic compounds (VOCs), and 0.2 tons of ammonia (NH3) were released from the landside CHE operation. CO and NOx emissions are the two primary air pollutants from the CHE operation in the Port of Incheon, contributing 87.71% of the total amount of emissions. Cranes, forklifts, tractors, and loaders are the four major sources of pollution in the Port of Incheon, contributing 84.79% of the total in-port CHE emissions. Backward diesel-powered machines equipped in these CHE are identified as a key cause of pollution. Therefore, this estimation emphasizes the significant contribution of diesel CHE to port air pollution and suggests the following green policies should be applied: (1) replacement of old diesel powered CHE by new liquefied natural gas and electric equipment; (2) the use of NOx reduction after-treatment technologies, such as selective catalytic reduction in local ports. In addition, a systematic official national emission inventory preparation method and consecutive annual in-port CHE emission inventories are recommended to compare and evaluate the effectiveness of green policies conducted in the future.

Study of Effective Cryoprotectants on the Cryopreservation of Porcine Mesenechymal Stem Cells (돼지 중간엽 줄기세포 동결에 있어서 동결보호제에 따른 특성 연구)

  • The objective of this study was to investigate the effective cryoprotectants for the cryopreservation of porcine mesenechymal stem cells (pMSCs). In order to understand the effectiveness of various cryoprotectants on pMSCs, we studied the most commonly used cryoprotectants; dimethyl sulfoxide (DMSO), ethylene glycol (EG), DMSO and EG. pMSCs were isolated from bone marrow matrix of piglet (2 month) and characterized by alkaline phopshatase (AP) activity, colony forming, and differentiation to adipocyte. In slow cooling cryopreservation, the pMSCs were exposed to cell medium containing Dulbecco's Modified Eagle's Medium (DMEM) supplemented with 10% DMSO, 1.5M EG and 5% DMSO/0.75M EG, respectively, and freezed to $-1^{\circ}C$/min from $25^{\circ}C$ up to $-80^{\circ}C$ in a cryo-container. The proportion of viable cells and the growing rates in fresh pMSCs were significantly (P<0.05) higher than those of other groups, but did not differ between the cryopreserved groups. The expression of Sox-2 and Nanog gene was increased by extending culture time in cryopreserved groups. The expression of Bax gene in cryopreserved groups was similar with fresh pMSCs. Moreover, the gene expression of adipocyte-specific marker as well as chondrogenic/osteogenic factors in cryopreserved groups was similarly to fresh pMSCs. Taken together, our results suggested that all these cryoprotectants of 10% DMSO, 1.5M EG and 5% DMSO/0.75M EG could be used for cryopreservation of the pMSCs.

Establishment and Characterization of Multipotent Germ Line Stem Cells (MGSCs) from Neonatal Mouse Testis (신생 생쥐 고환에서 기인한 다분화능 생식줄기세포주의 확립 및 특성 분석)

  • Objective: The aim of this study was to investigate whether multipotent germline stem cells (MGSCs) can be established from neonatal mouse testis. Methods: Various cells containing MGSCs were collected from neonatal testis of ICR mice and allocated to plates for in vitro culture. After 7 days in culture, the cells were passed to a fresh culture plate and continuously cultured. From the third or fourth passage, the presumed MGSCs were cultured and maintained on mitomycin C-inactivated STO feeder cells. The MGSCs were cultured in a condition where mouse embryonic stem cells (ESCs) are cultured. Characteristics of the MGSCs were evaluated by RT-PCR, immunocytochemistry, alkaline phosphatase activity, karyotyping, and transmission electron microscopy. Results: Two MGSCs lines were established from 9 pooled sets of neonatal testicular cells. MGSCs colonies were morphologically undistinguishable from ESCs colonies and both MGSC lines as well as ESCs expressed undifferentiated stem cell markers, such as Thy-1, Oct-4, Nanog, Sox2 and alkaline phosphatase. Fine structure of undifferentiated MGSCs were similar to those of ESCs and 60% of MGSCs (12/20) had normal karyotype at passage 10. They were able to form embryoid bodies (EBs) and MGSC-derived EBs expressed marker genes of three germ layers. Conclusion: We could establish the MGSCs from neonatal mouse testis and they were differentiated to multipotent lineages of three germ layers. Molecular characteristics of MGSCs were similar to those of ESCs. Our results suggest a possibility that multipotent stem cells derived from testis, the MGSCs, could replace the ESCs in biotechnology and regenerative medicine.

Preliminary Study on Factor Technology of Selective Catalytic Reduction System in Marine Diesel Engine (선박용 디젤엔진 SCR 시스템 요소 기술에 관한 기초 연구)

  • From 2016, controls on reduction of NOx and SOx emissions from the vessels that are operated in the emission control area were tightened. The selectivity catalytic reduction system of the denitrification equipment which NOx among the above controlled materials is very effective and used commercially very much. But it has the disadvantage that CSR is activated at high temperatures. Therefore, the SCR and SCR activation instrument that can react even at low temperatures by using micro-nano bubbles so that the above problems can be minimized were developed. And the computational fluid dynamics technique was used by ANSYS-CFX package to prepare the plan that improves the SCR system's efficiency. Simulation for the viscous flow analysis of the SCR system was executed by applying the Navier-Stokes equation to it as a governing equation. For the SCR system's shape, 3D modeling was done by using CATIA V5. SCR jet nozzle's position was checked by changing it to the intervals of 1/3, 1/2, and 2/3 from the inlet of the vent pipe to compare the SCR system's efficiency. And the number of nozzles was compared and analyzed by simulating 4, 6, and 8 holes to check an effect of the number on the SCR system's efficiency. The simulation result has found that the closer nozzles are to the inlet of the vent pipe and the more nozzles are, the more efficiency is improved.

  • Pseudomonas aeruginosa is an opportunistic human pathogen, causing a wide variety of infections including cystic fibrosis, microbial keratitis, and burn wound infections. The cell-to-cell signaling mechanism known as quorum sensing (QS) plays a key role in these infections and the QS systems of P. aeruginosa have been most intensively studied. While many literatures that introduce the QS systems of P. aeruginosa have mostly focused on two major acyl-homo serine lactone (acyl-HSL) QS signals, N-3-oxododecanoyl homoserine lactone (3OC12) and N-butanoyl homoserine lactone (C4), several new signal molecules have been discovered and suggested for their significant roles in signaling and virulence of P. aeruginosa. One of them is PQS (Pseudomonas quinolone signal; 2-heptyl-3-hydroxy-4-quinolone), which is now considered as a well-characterized major signal meolecule of P. aeruginosa. In addition, recent researches have also suggested some more putative signal molecules of P. aeruginosa, which are diketopiperazines (DKPs) and pyocyanin. DKPs are cyclic dipeptides and structurally diverse depending on what amino acids are involved in composition. Some DKPs from the culture supernatant of P. aeruginosa are suggested as new diffusible signal molecules, based on their ability to activate Vibrio fischeri LuxR biosensors that are previously considered specific for acyl-HSLs. Pyocyanin (1-hydroxy-5-methyl-phenazine), one of phenazine derivatives produced by P. aeruginosa is a characteristic blue-green pigment and redox-active compound. This has been recently suggested as a terminal signaling factor to upregulate some QS-controlled genes during stationary phase under the mediation of a transcription factor, SoxR. Here, details about these newly emerging signaling molecules of P. aeruginosa are discussed.

Helicobacter pylori Eradication Reduces the Metachronous Recurrence of Gastric Neoplasms by Attenuating the Precancerous Process

  • Purpose: The importance of Helicobacter pylori eradication after endoscopic resection (ER) of gastric neoplasms remains controversial. In this study, we clarified the importance of H. pylori eradication for metachronous lesions after ER. Materials and Methods: This study included 3,882 patients with gastric neoplasms who underwent ER. We included patients infected with H. pylori who received eradication therapy. Among them, 34 patients with metachronous lesions after ER and 102 age- and sex-matched patients (nonmetachronous group) were enrolled. Background mucosal pathologies such as atrophy and intestinal metaplasia (IM) were evaluated endoscopically. The expression levels of CDX1, CDX2, Sonic hedgehog (SHH), and SOX2 were evaluated based on H. pylori eradication and the development of metachronous lesions. Results: The eradication failure rate was higher in the metachronous group than in the nonmetachronous group (P=0.036). Open-type atrophy (P=0.003) and moderate-to-severe IM (P=0.001) occurred more frequently in the metachronous group. In patients with an initial diagnosis of dysplasia, the eradication failure rate was higher in the metachronous group than in the nonmetachronous group (P=0.002). In addition, open-type atrophy was more frequent in the metachronous group (P=0.047). In patients with an initial diagnosis of carcinoma, moderate-to-severe IM occurred more frequently in the metachronous group (P=0.003); however, the eradication failure rate was not significantly different between the two groups. SHH and SOX2 expression was increased, and CDX2 expression was decreased in the nonmetachronous group after eradication (P<0.05). Conclusions: Open-type atrophy, moderate-to-severe IM, and H. pylori eradication failure were significantly associated with metachronous lesions. However, eradication failure was significantly associated with dysplasia, but not carcinoma, in the metachronous group. Thus, H. pylori eradication may play an important role in preventing metachronous lesions after ER for precancerous lesions before carcinomatous transformation.
