Effect of Plant Extracts with Superoxide Dismutase-like Activity on Survival of Fruit Flies under Oxidative Stress (Superoxide Dismutase유사활성을 지닌 식물체가 Oxidative Stress를 받고 있는 초파리의 수명에 미치는 영향)

  • Biological effect of aqueous extracts of 12 plants which showed superoxide dismutase (SOD)-like activity in vitro was evaluated using Drosophila melanogaster. Survival percentage of the flies was a criterion of effect when the flies were exported to paraquat, which generated superoxide anion radical in vivo. When flies were co-administered with paraquat and aqueous extracts of garlic, lettuce, kiwi, and nameko, they showed no defensive effect against of oxygen toxicity. If flies were exposed to 60 mM paraquat after adaptation to feed containing plant extracts with SOD-like activity for 10 days, however, survival percentage of flies fed with phytochemicals was $35{\sim}63%$ while that percentage of flies fed without phytochemical was only 11%. This result indicated that adaptation of flies to plant extracts with SOD-like activity could prevent the flies from oxidative injury. On the other hand, lettuce, kiwi, nameko, onion, persimmon, fern brake and cauliflower showed a reparative effect on an oxidative stress. Dropwort, shiitake, agaric mushroom and broccoli did not show such an effect.

Physiological Activities of Gardeniae Fructus Extracts by Microwave-Assisted Extraction as Affected by Solvents (마이크로웨이브로 추출한 용매별 치자(Gardeniae Fructus) 추출물의 생리활성)

  • Gardeniae Fructus (GF) is a fruit of Gardenia jasminoides Ellis (Rubiaceae) which has been used in traditional medicine. It contains not only geniposide but also resveratrol, a kind of stilbene as a natural antioxidant. In this study, we investigated physiological activities of GF extracts by measuring electron donating ability (EDA), nitrite scavenging ability (NSA), super oxide dismutase (SOD)-like activity and inhibitory effect of tyrosinase activity (ITA). As a solvent, it was extracted with water, 50 and 100% ethanol using microwave. At the concentration of 6 mg/mL, EDA was the highest at 50% ethanol extract but the lowest at 100% ethanol extract, $96.43{\pm}0.25%\;and\;77.06{\pm}0.22%$ respectively. NSA showed the strongest activity at 50% ethanol extract like EDA (6 mg/mL: $75.26{\pm}0.28%$). ITAs were below 20% in all samples and their activities reduced according to increasing concentration. Likewise, SOD-like activities decreased with increasing concentration of extracts in all samples. In conclusion, GF extracts showed low SOD-like activities and ITAs but EDAs and NSAs were fairly high; especially EDA of 50% ethanol extract (6 mg/mL) was high as those of 0.1 and 1% L-ascorbic acid. Therefore the results suggest that GF extracts may be useful as potential sources of natural antioxidants.

Physiological activitive of Grifola frondosa by log cultivation and bottle cultivation (원목재배와 병재배 잎새버섯(Grifola frondosa)의 생리활성)

  • This research was carried out to determine the differences of physiological activites between Grifola frondosa of log cultivation(LC) and Grifola frondosa of bottle cultivation(BC). Total flavonoids content, total phenolics content, electron donating ability(EDA), nitrite-scavenging ability(NSA), SOD-like activity and inhibitory effect of Xanthine oxidase were examined. The highest value of total flavonoid content is $5.96{\pm}0.81mg/g$ in water extract from Grifola frondosa of log cultivation at $40^{\circ}C$ (LC-W40) but, one of total phenolics compound is $44.53{\pm}0.89mg/g$ in water extract from Grifola frondosa of bottle cultivation at $40^{\circ}{\cdots}$ (BC-W40). The EDA using DPPH of BC-W40 extract showed the highest value of $97.14{\pm}0.71%$. Nitrite-scavenging ability was $62.55^{\circ}{\ae}0.36%$ in extract from Grifola frondosa of BC-W40 at pH 1.2. The value was SOD-like activity showed the highest value of $18.95^{\circ}{\ae}1.39%$ in extract from LC-W40. Xanthine oxidase inhibitory activity was the highest value of $54.31{\pm}0.40%$ in extract from Grifola frondosa BC-W40, and dependent on concentrations. These results showed that a the antioxidant effects of Grifola frondosa is excellent. However, physiological activities of Grifola frondosa were not depend on caltivation method regulary, and were different according to kind of solvents, concentraitions and physialogical factors examined such as EDA, SOD-like activity and NSA.

Polyphenol Contents and Antioxidant Activity of Extracts from Angelica dahurica Root after Different Conditions of Microwave-assisted Extraction (마이크로웨이브 추출조건에 따른 백지 추출물의 폴리페놀 함량과 항산화 작용)

  • This study investigated the antioxidant activities of extracts from Angelica dahurica roots after microwave-assisted extraction with different levels of energy (120, 240W) and extraction time (5, 10, 15 and 30 min). The highest extraction yield was 11.77 mg% in water at 240W for 30 min followed by 11.42 mg% in water at 120Wand 30 min. The highest total polyphenol contents was 32.36 mg/g in an ethanol extract, followed by 31.77mg/g in water extract at the same conditions of 240W, 30 min. The electron donating abilities both the ethanol extract obtained using 240W and 30 min and the water extract obtained employing 120W and 5min showed the highest values, 83.55% and 82.49% respectively at a concentration 1.0mg/mL. The highest superoxide dismutase (SOD)-like activity was 14.16% in ethanol extract at 120Wand 15min, followed by 13.22% in the water extract at 120W and 5 min. The best extraction yield and polyphenol content after microwave-assisted extraction were achieved with 240W and 30 min using water. The best condition for extraction of electron donating ability and SOD-like activity from A dahurica roots were 120W and 5 minutes using water.

Antioxidant Activity and Cell Differentiation Effects of Monascus purpureus Pigment on Osteoblast-like MC3T3-E1 Cells (홍국색소의 항산화 활성 및 조골세포 분화에 미치는 영향)

  • The purpose of this study was to investigate antioxidant activity and cell differentiation effects of Monascus purpureus pigment on osteoblast-like MC3T3-E1 cell. In order to examine the antioxidant activities of Monascus purpureus pigment, DPPH radical scavenging, ABTS radical scavenging and SOD-like activities were investigated. DPPH radical and ABTS radical scavenging activities of Monascus purpureus pigment were increased in a dose-dependent manner, and maximum activity were 94% and 99% at a concentration of 1,000 ㎍/ml, respectively. Additionally, SOD-like activity of Monascus purpureus pigment showed 62% at a concentration of 1,000 ㎍/ml. MC3T3-E1 cells did not show cytotoxicity in the concentration range of Monascus purpureus pigment 1~100 ㎍/ml. The ALP activity was increased by addition of Monascus purpureus pigment, and the maximum activity was 124% as compared with control. In addition, nodule formation, a late differentiation factor for bone formation, was increased by adding Monascus purpureus pigment compared to control. These results suggest that Monascus purpureus pigment is expected to be a natural source for developing functional materials to prevent bone-related diseases by osteoblast differentiation.

Study on the Development of Cosmetic Emulsion Cream for Patients with Atopic Dermatitis using Scutellaria Baicalensis (황금(黃芩)을 이용한 아토피성 피부용 한방화장품 제형화에 관한 연구)

  • Objectives : This study was conducted to determine if Scutellaria baicalensis can be used in cosmetic emulsion cream for patients with atopic dermatitis. Methods : Scutellaria baicalensis extract was obtained with the use of butylene glycol through the pressurized solvent extraction(PSE). The antioxidative activity was assessed through SOD-like activity measurement and skin irritating potential was tested using human patch test. Antimicrobial activity was measured by the clear zone formed against Staphylococcus aureus and Escherichia coli and the rheological effects on the emulsion creams were examined using oscillation test. Results : The SOD-like activity increased dose-dependently and was about 90% at 1,000ppm of Scutellaria baicalesis extract. And Scutellaria baicalensis extract did not show any potential to be irritating to the human skin, but it could not be used as an antimicrobial agent for its poor antimicrobial activity against Staphylococcus aureus. The complex modulus decreased by 1,000 pascals and the loss angle also decreased by 20% with the addition of Scutellaria baicalensis extract into the cosmetic emulsion creams, that is, the extract can confer more elastic property on the vehicle. Conclusion : From those results, Scutellaria baicalensis extract can be effectively used as an antioxidant and reinforces the elastic skincare film formed by the application of cream for patients with atopic dermatitis.

Biological Activities of Scolopendrid Pharmacopuncture (수순(水醇)추출법으로 조제된 오공 약침액의 생리활성 효과)

  • Reactive Oxygen Species(ROS) are continuously produced at a high rate as a by-product of aerobic metabolism. Since tissue damage by free radical increases with age, the reactive oxygen species(ROS) such as hydrogen peroxide($H_2O_2$), nitric oxide(NO). Several lines of evidence provided that ROS appears to cause to develop aging-related various diseases such as cancer, arthritis, cardiovascular disease. Our reserch objective was to examine the in vitro biological activity of Scolopendrid Pharmacopuncture, including the total poly-phenol content, DPPH radical scavenging, ABTS radical scavenging, Superoxide dismutase(SOD)-like activity, Nitrite scavenging ability. The total poly-phenol contents of Scolopendrid Pharmacopuncture was 35.859mg/L. Elctron donation ability on DPPH was 36.82%. The 2,2'-azinobis-3-ehtlbezothiazoline-6-sulfonic acid radical decolorization (ABTS) was 84.7%. The superoxide dismutase (SOD)-like activities of Scolopendrid Pharmacopuncture was 44.33%. The nitrite scavenging effects were pH dependent, and were highest at pH 1.5(45.2%) and lowest at pH 6.0(11.3%). We conclude that Scolopendrid Pharmacopuncture may be useful as potential sources of antioxidant.

Nitrite Scavenging and Alcohol Metabolizing Activities of Hot Water Extract from Makgeoly and Its Angiotensin Converting Enzyme Inhibitory Effect (막걸리 열수 추출물의 아질산염 소거능, 알코올 분해능 및 angiotensin converting enzyme 저해 효과)

  • In this study, we investigated the antioxidant activities, alcohol metabolizing activities, nitrite scavenging ability, angiotensin converting enzyme (ACE), and elastase inhibitory effects of hot water extract from Makgeoly (HWM). Antioxidant activities were measured by using 2,2.diphenyl.1.picryl.hydrazyl (DPPH) free radical scavenging activity and SOD (superoxide dismutase).like activity. The DPPH radical scavenging activity and SOD.like activity of HWM were remarkably increased in a dose.dependent manner and were 48.0% and 98.7% at 10 mg/ml, respectively. To determine the influence of HWM on alcohol metabolizing activity, the generating activities of reduced.nicotinamide adenine dinucleotide (NADH) by alcohol dehydrogenase (ADH) and aldehyde dehydrogenase (ALDH) were measured. The facilitating rates of ADH and ALDH activity by HWM were remarkably increased in a dose.dependent manner and were 70.2% and 64.1% at 10 mg/ml, respectively. The inhibitory activity against angiotensin converting enzyme (ACE) of HWM was increased in a dose.dependent manner and was 74.2% at 10 mg/ml. The nitrite scavenging ability of HWM showed the most remarkable effect at pH 1.2 and 2 mg/ml. These results indicated that HWM may have valuable biological properties owing to their antioxidant activities, ADH and ALDH activity, nitrite scavenging ability, and ACE inhibitory activity.

The Antioxidant Effect of Portulaca oleracea Extracts and Its Antimicrobial Activity on Helicobacter pylori (쇠비름 추출물의 항산화 효과 및 Helicobacterpylori에 대한 항균 활성)

  • This study was performed to investigate the effect of Portulaca oleracea extract on the antioxidant and antimicrobial activities against Helicobacter pylori. The each solvent extracts prepared from Portulaca oleracea were investigated by measuring total phenolic compounds, electron donating ability, superoxide dismutase-like ability and thiobarbituric acid reactive substances The herb extractor extract yielded the highest content of total phenolic compounds(72.2 mg%). The electron donating abilities(EDA) of ethyl acetate and methanol extracts showed high antioxidant activity. The superoxide dismutase (SOD)-like abilities of ethyl acetate and petroleum ether extracts also showed some activity. The antioxidant activity of thiobarbituric acid reactive substances was not significant. The petroleum ether extract of Portulaca oleracea showed the highest antimicrobial activity at 10,000 ppm concentration.

Physiological Activities of Rubus coreanus Miquel (복분자 딸기의 생리활성)

  • Physiological activities of unripened fruit, ripened fruit and leaf of Rubus coreanus Miquel were examined. Total polyphenolic compound content, electron donating ability (EDA), nitrite scavenging activity and SOD-like activity were examined using extracts of Rubus coreanus Miquel extracted with various extraction solvents such as 80% methanol, 75% acetone and water. The leaf part of Rubus coreanus Miquel included higher contents of total polyphenol compound compared with those of the other sample groups, unripened and ripened fruit. The total polyphenol compound content of leaf (100 g, dry base) extracted with 75% acetone showed the highest value of $5.06{\sim}5.87g$. As for EDA, unripened fruit showed over 90% of electron donating ability. No significant difference in EDA was found among the extracts prepared with various extraction solvents, 80% methanol, 75% acetone and water. Rubus coreanus Miquel extracts showed different nitrite scavenging abilities under different pH conditions. The nitrite scavenging abilities at pH 1.2 were in the range of $41.25{\sim}63.24%$ whereas they were $1.59{\sim}10.99%$ at pH 4.2 and $-2.84{\sim}7.94%$ at pH 6.0. The high levels of SOD-like activities were found in ripened fruit when different extracting methods were applied. Agar diffusion tests were accomplished to examine the antimicrobial activities of the extracts prepared from unripened fruit, ripened fruit and leaf with various extraction solvents. All of the extracts revealed antimicrobial activity against Bacillus cereus whereas no antimicrobial activities were observed against Escherichia coli, Salmonella typhimurium and Staphylococcus aureus.
