In order to examine the antioxidant action of Loranthus parasiticus (L.) merr (LP), the study was done through measurement of parameters such as LPO, GSH, SOD, catalase, GOT, GPT and ALP. The results were obtained as follows: For the weight changes, in the left cerebrum, right cerebrum, cerebellum and testis, the group given LP showed small increase compared to the control group. But the weight changes were not significant. In the left cerebrum, the group given LP showed significant decrease compared to the control group in the content of LPO and significant increase compared to the control group in the activity of SOD. But in the right cerebrum, cerebellum the changes were not significant. For the changes of contents of SOD and catalase in the liver, the group given LP showed significant increase compared to the control group. For the changes of the activity of SOD in the kidney, the group given LP showed significant increase compared to the control group. For the changes of contents of LPO and GSH in the spleen, the group given LP were showed no significant decrease compared to the control group. And for the changes of the activities of SOD and catalase, the group given LP showed no significant increase compared to the control group. For the changes of content of LPO in the testis, the group given LP showed significant decrease compared to the control group. And for the changes of the activity of catalase, the group given LP showed significant increase compared to the control group. For the changes of GOT, GPT in the serum, the group given LP showed significant decrease compared to the control group. From above results, the antioxidant action of LP is effective. And it is expected to be necessary to the study of the mechanism in the antioxidant of LP.
To study the effects of soil moisture and texture on characteristics of growth, content of saikosaponins and activity of antioxidative enzymes in Bupleurum falcatum L., content of saikosaponins(a, c and d) and activities of superoxide dismutase(SOD) and peroxidase(POD) were investigated with two Bupleurum genotypes(Jangsoo and Samdo). Two Bupleurum genotypes were grown under different soil moisture(deficit, normal, surplus) and soil texture(sandy, sandy loam, loam) conditions. Among the tested soil conditions, dry weight accumulation rate of both cultivars could be ranked in the order surplus > normal > deficit soil for soil moisture and sandy > sandy loam > loam for soil texture. Under the surplus soil condition, growth retardation of Samdo cultivar was more severer than that of Jangsoo. Furthermore, content of saikosaponin a, d, and c also could be ranked in the order deficit > normal > surplus and sandy > sandy loam > loam for soil moisture and texture, respectively. Although both Jangsoo and Samdo cultivars grown under water deficit condition showed the highest POD and SOD activity, in general POD and SOD activity in both shoot and root was remarkably high in Jangsoo cultivar compared with Samdo. Saikosaponin content of root was positively correlated with POD and SOD. However, shoot and root length were negatively correlated with POD.
This study was performed to assess age-related changes in DNA damage and antioxidative capacity in 4, 8, 12, 16, 20, and 24 months old Sprague-Dawley male rats. The following were measured the degree of oxidative DNA damage as indicated by levels of 8-hydroxy-2'-deoxyguanosine (80HdG) in the kidney ; the peroxidized lipid concentrations in the plasma and the liver, as indicated by the levels of thiobarbituric acid reactive substances (TBARS); and the levels of antioxidant enzyme activities in the erythrocytes and the liver. Both body weight (BW) and epididymal fat pad (EFP) weight per BW increased with age until 16 months, then decreased slightly from 20 to 24 months. However, the weights of the liver, kidney and spleen per BW decreased with age. Concentrations of 8-OHdG in the kidney increased with age, only slightly front 4 to 16 months, and then markedly from 16 to 24 months. TBARS concentrations in the plasma and liver were shown to increase with age, being lowest in the 4 month-old group and highest in the 24 month-old group. Superoxide dismutase (SOD) activity in the erythrocytes increased with age Catalase activity in the erythrocytes increased from 4 to 16 months, then decreased from 20 to 24 months. Glutathione peroxidase (GSH-Px) activity in the erythrocytes showed no age-related change. Liver SOD activity decreased with age, particularly from 16 to 20 months, but catalase and GSH-Px activities in the liver showed no significant changes. These results showed that during the normal aging of SD rats, DNA damage in the kidney and TBARS concentrations in the plasma and liver increased with age, particularly after 16 months, and the imbalance of antioxidative enzyme activities in the erythrocytes accelerated with age.

Response surface methodology (RSM) was employed to optimize extraction conditions in order to find the maximal functional properties of fluid Cheonggukjang. Based on central composite design, a study plan was established with variations of microwave power, ethanol concentration, and extraction time. Regression analysis was applied to obtain a mathematical model. The maximum inhibitory of tyrosinase activity was found as 26.75% at the conditions of 30.56W microwave power, 2.40 g/mL of ratio of solvent to sample content and 10.00 min extraction time, respectively. The maximum superoxide dismutase (SOD)-like activity was 53.23% under the extraction conditions of 108.42 W, 4.38 g/mL and 7.84 min. Based on superimposition of three dimensional RSM with respect to extraction yield, inhibitory of tyrosinase activity and SOD-like activity obtained under the various extraction conditions, the optimum ranges of extraction conditions were found to be microwave power of $55{\sim}75$ W, ratio of solvent to sample content of $2{\sim}5$ g/mL and extraction time of $3.5{\sim}15$ min, respectively.
Biological effect of aqueous extracts of 12 plants which showed superoxide dismutase (SOD)-like activity in vitro was evaluated using Drosophila melanogaster. Survival percentage of the flies was a criterion of effect when the flies were exported to paraquat, which generated superoxide anion radical in vivo. When flies were co-administered with paraquat and aqueous extracts of garlic, lettuce, kiwi, and nameko, they showed no defensive effect against of oxygen toxicity. If flies were exposed to 60 mM paraquat after adaptation to feed containing plant extracts with SOD-like activity for 10 days, however, survival percentage of flies fed with phytochemicals was $35{\sim}63%$ while that percentage of flies fed without phytochemical was only 11%. This result indicated that adaptation of flies to plant extracts with SOD-like activity could prevent the flies from oxidative injury. On the other hand, lettuce, kiwi, nameko, onion, persimmon, fern brake and cauliflower showed a reparative effect on an oxidative stress. Dropwort, shiitake, agaric mushroom and broccoli did not show such an effect.

Physiological activities of pileus and stipe extracts from fresh Pleurotus eryngii were examined. Electron donating ability (EDA), superoxide dismutase (SOD)-like activity, tyrosinase inhibitory effect, angiotensin converting enzyme (ACE) inhibitory activity, total polyphenol contents and nitrite scavenging ability were examined for extracts of pileus and stipe of Pleurotus eryngii extracted with water, $50\%$ and $100\%$ ethanol. Volume of the solvent used was 50 time of sample on a weight/volume basis. EDA was $88\%$ in $50\%$ ethanol extract (pileus). Also, SOD-like activity was $62.57\%$ in water extract (pileus). Tyrosinase inhibitory effects of all samples were higher than $0.1\%$ L-ascorbic acid solution. ACE inhibitory activity of water extract of pileus was found to be the highest value of $95.14\%$. In addition, total polyphenol contents were the highest in water and $50\%$ ethanol extracts of pileus $(1427.25\;mg\%,\;1426.82\;mg\%)$. Finally, nitrite scavenging ability of $50\%$ ethanol extract of pileus at pH 1.2 showed approximately $95\%$. The results will be useful for understanding the physiological activities of Pleurotus eryngii extracts.

Gardeniae Fructus (GF) is a fruit of Gardenia jasminoides Ellis (Rubiaceae) which has been used in traditional medicine. It contains not only geniposide but also resveratrol, a kind of stilbene as a natural antioxidant. In this study, we investigated physiological activities of GF extracts by measuring electron donating ability (EDA), nitrite scavenging ability (NSA), super oxide dismutase (SOD)-like activity and inhibitory effect of tyrosinase activity (ITA). As a solvent, it was extracted with water, 50 and 100% ethanol using microwave. At the concentration of 6 mg/mL, EDA was the highest at 50% ethanol extract but the lowest at 100% ethanol extract, $96.43{\pm}0.25%\;and\;77.06{\pm}0.22%$ respectively. NSA showed the strongest activity at 50% ethanol extract like EDA (6 mg/mL: $75.26{\pm}0.28%$). ITAs were below 20% in all samples and their activities reduced according to increasing concentration. Likewise, SOD-like activities decreased with increasing concentration of extracts in all samples. In conclusion, GF extracts showed low SOD-like activities and ITAs but EDAs and NSAs were fairly high; especially EDA of 50% ethanol extract (6 mg/mL) was high as those of 0.1 and 1% L-ascorbic acid. Therefore the results suggest that GF extracts may be useful as potential sources of natural antioxidants.

The antioxidative activities and biological properties in the EtOAc extracts and purified extracts of Orostachys japonicus were measured by assay of DPPH, xanthine/xanthine oxidase and mammalian cells(2-12). Scavenging of DPPH radical and inhibition of xanthine/xanthine oxidase of EtOAc extracts were showed the highest activity in the arable land and in September. The S-4 fraction showed the highest activity among the silica-gel column chromatography fractions. LH-4 fraction showed higher activity than the other fractionsins in assay of DPPH and xanthine/xanthine oxidase. Fatty acids and phenolic compounds were identified by GC/MS and main compounds were 1,2,3-benzenetriol, alpha-androsta-7,14-diene in LH-4 fraction. The activities of POD and SOD in samples havested on different habitats were high such as arable land> intermountain> seashore. That of POD and SOD in crude extracts of late stage were higher than early stage. Isozyme bands of crude extracts samplinged in all habitats and all growing stages showed two bands and the signal intensity showed strongly according to passage of growing stage. The purified extracts of LH-4 fraction showed excellent inhibition effect in proliferation of HL-60 cells and markedly suppressed colony formation in mouse fibroblast cells. Dose response between partially purified extracts(400ppm) and negative control did not produced statistically significant reduction in colony formation.

This study was conducted to investigate the effects of pine (Pinus densiflora) needle extracts on hepatic antioxidative system in rats fed high-cholesterol diet. Eight-week old Sprague Dawley rats were divided into four groups; normal diet group (CON), 0.5% cholesterol diet group (0.5% CHOL), 0.5% cholesterol diet and pine needle extracts prepared by using hot water group (HOT water), and 0.5% cholesterol diet and pine needle extracts prepared by using sub-supercritical $CO_2$ group (Sub-supercritical). The serum ALP, AST, and ALT activities were increased by 0.5% cholesterol diet supplementation, but treatment with pine needle extracts showed decrease compared with 0.5% CHOL group. The SOD activity in liver at 0.5% cholesterol diet group was more decreased than control group while Sub-supercritical group had significantly higher SOD activity than 0.5% CHOL group. Moreover, groups with pine needle extracts supplementation had higher level of GPx in liver than 0.5% cholesterol diet group. These results suggest that pine needle extracts increased antioxidative activities.
Successful germination and establishment of seedlings in flooded paddy are critical in direct seeding cultivation of rice. In this study, we examined the relationship between antioxidant enzymes and alcohol dehydrogenase (ADH) activities and coleoptile elongation under submergence of deep water with two rice cultivars, Iksan429 and Woodrose, which show characteristic coleoptile elongation under hypoxic condition. The growth of shoot under submerged in water was faster than the root. The survival duration was longer in Iksan429 than in Woodrose under submerged in water. The alcohol dehydrogenase (ADH) activities were significantly increased under hypoxia compared to in aerated condition. The ADH activity was increased in Iksan429 more than in Woodrose under hypoxia. The superoxide dismutase (SOD) activity in Iksan429 was gradually increased up to 5 days after treatment (DAT) then decreased slowly till 14 DAT under water, whereas in Woodrose it was dramatically decreased after 5 DAT. The peroxidase (POX) activity in Iksan429 was significantly increased until 7 DAT under hypoxia, whereas it was not significantly different in Woodrose during hypoxic treatment. However, in non-treated condition, POX activity in Woodrose was increased more than Iksan429. The changes of catalase (CAT) activities showed no differences in both cultivars. We suggest that the overexpression of ADH, SOD and POX activities is responsible for the hypoxic tolerance and plays an important role in the surviving of rice seedling.
