Biological Activities from the Roots of Symplocarpus renifolius Schott (앉은부채(Symplocarpus renifolius Schott) 뿌리의 생리활성)

  • Symplocarpus renifolius, a folk medicinal herb has been used for treatment of hypertension, rheumatis, tetanus, ringworm, cough and epilepsy in north and middle part of Korea. The extracts from the root of S. renifolius were evaluated for antibacterial, antihypertensive and analgesic activities. The crude extract of the root of S. renifolius showed antibacterial activity against Gram(+) bacteria and dose dependantly decrease the blood pressure of SHR. The chloroform extract of the roots of S. renifolius was also exibits analgesic action in mice.

  • 비병원성 세균인 고acirlus subtilis(ATCC6633)를 등검은메뚜기(Euprepocnemis shr'rakii Bolivar)의 성충 복강에 주입한 후, 혈구의 반응양상을 투과전자현미경으로 관찰하였다. 세균에 대한 혈구의 면역반응은 plasmatocyte와 granulocyte에 의한 식 세포작용(phagocytosis)의 형태로 이루어졌으며, 결절형꽁(nodule formation)은 일어나지 않았다. 식 세포작용의 초기 반응은 혈구의 세포질 돌기가 돌출되 면서 이물질을 둘러싼 후, 원형질막과의 융합에 의해 식포가 형성되었고, 식포의 주변부에서는 일차 리소조옴의 집적이 관찰되었으며, 이런 과정은 세균 주입후, 10분 이내에 완료되었다. 일차 리소조옴은 세균 주입 후 1시간 이내에 식포의 한계막과 융합하여 이차 리소조옴을 형성하였으며, 소화된 이물질은 9시간 이내에 exocytosis에 의해 배출 되었다.

  • Normally, monetary penalties for breach of agreement will be stipulated in the contract. The contractor parties, who fail to fulfill the agreement, are required to pay the other parties a certain amount or proportion of money as a fine. However, it is worth our study - whether or not the scope of monetary penalty implementation and bases for determination of a fine will cover the losses of social and administrative costs incurred by the interruption of the contract. This research is about the best timing for government to cancel the freeway repair work contracts. Under the goal of the maximum social welfare, the limitation of government spending for the social and administrative costs invoked by interruption of contracts will have to be considered to attain the best timing of contracts' suspension or deferment. According to the factors of social and administrative costs, the best time point is calculated to reduce the loss of the aforesaid costs, which can also be used as theoretical basis for the future road-widening construction at home.

Antihypertensive Effects of Casein Hydrolysate in Spontaneously Hypertensive Rats (자연발증고혈압쥐에서 카제인 가수분해물의 혈압강하효과)

  • The aim of this study was to determine if a low-molecular weight casein hydrolysate has an anti- hypertensive effect in spontaneously hypertensive rats (SHR). Prior to the in vivo experiment, the casein hydrolysate was confirmed to be resistant to gastrointestinal digestion by confirming the retention of its potency as an inhibitor of angiotensin I-concerting enzyme after incubation with pepsin, trypsin, or chymotrypsin. The in vivo anti-hypertensive effect of the hydrolysate was determined by the tail cuff method. Following an oral administration of the hydrolysate solution, the systolic blood pressure (SBP) decreased by 12.9% (-28.9mmHg; P<0.05) at 3 h after the administration at a dose of 500mg/kg body weight. When the hydrolysate was administered as an emulsion with 30% egg yolk, its anti-hypertensive effect was even more greater at the same dose(-30.8mmHg or -15.9%; P<0.01). In a 50-day long-term trial where the casein hydrolysate was administered once a day, the SBP-lowering effect of the hydrolysate was apparent (P<0.05) from day 35 through the end. Moreover, organ weights and plasma glutamate oxaloacetate transaminase and glutamate pyruvate transaminase activities of the administered SHR were not significantly different from those of controls at the end of the long-term trial.

The Antihypertensive and Vasodilating Effects of Adventitious Root Extracts of Wild Ginseng (산삼 배양근 추출물의 혈압강화 및 혈관이완 효과)

  • Nitric oxide (NO) is a potent antihypertensive and vasodilator which plays an important role in regulating vascular tones. In this study, we investigated the effects of adventitious root extracts of wild ginseng on NO production and NO linked physiological activities. When human endothelial cell line (ECV304) was incubated with either water extracts of wild ginseng adventitious root (WE) or aqueous fraction of butanol extracts of wild ginseng adventitious root (ABE), considerable amounts of NO were released by the cells. The level of endothelial nitric oxide synthase (eNOS) expression was unchanged and about 6% of the angiotensin converting enzyme (ACE) was inhibited with treatment of ABE. The vasodilating activities of pulmonary artery rings in response to different doses of extracts were shown as 44.8% and 91.3% in 2.5 mg/ml WE and 0.1 mg/ml ABE, respectively. The blood pressure lowering effect was observed from the oral administered spontaneously hypertensive rat (SHR) with the lowest blood pressure (154.5${\pm}$8.6 mmHg) after 8 h. The blood pressure was recovered to the initial level after 24 h.

Antihypertensive Effects of Novel Isoflavone-Free Black Soy Peptide Mixture as HO-1 Inducer (Heme 산화효소 발현 유도체로서 Isoflavone-Free 검은콩 펩타이드의 항고혈압 활성)

  • We previously reported that the novel isoflavone-free peptide mixture (black soybean peptide, BSP) had several beneficial effects like antiobesity and hypotriglyceridemic effect. However, there are no reports for BSP on anti-hypertensive activity. BSP induced heme oxygenase-1 (HO-1) in HUVECs, thus investigated the HO-1-induced activity in HUVECs and the anti-hypertensive effects in SHR animal model. BSP significantly induced HO-1 expression both at transcriptional and protein levels in a time- and dose-dependent manner as measured by RT-PCR and Western blot analysis, respectively. These inductions were abolished by pretreatment of N-acetyl-cystein (NAC, 1~10 mM), but not by employing Tempol, a superoxide dismutase (SOD) mimetic (1~5 mM). As expected, enzymatic activity (~2 fold) determined by bilirubin formation assay and cGMP concentration (~6 fold) were significantly increased in BSP-treated cells. Based on the numerous evidences on the beneficial effects of HO-1 and our results, we investigated in vivo effects of BSP on the antihypertensive activity. The administration of BSP (1% in drinking water) significantly decreased mean blood pressure (BP) (from $218.6{\pm}6.99$ to $190.0{\pm}3.42$ mm Hg, p<0.01). This result indicates that BSP is strong inducer of HO-1 expression, which may be triggered by oxidative stress, and has anti-hypertensive activity.

Attenuating Development of Cardiovascular Hypertrophy with Hydrolysate of Chicken Leg Bone Protein in Spontaneously Hypertensive Rats

  • This study developed a natural ingredient as a functional food possessing properties of attenuation of hypertension and cardiovascular hypertrophy. In a previous study hydrolysates obtained from chicken leg bone protein using Alcalase strongly inhibited angiotensin I converting enzyme (ACE) in vitro. In particular, hydrolysate (A4H) from four hours of incubation exhibited the highest ACE inhibitory activity (IC50 = 0.545 mg/ml). A4H was selected as a potent ACE inhibitor and orally administrated to spontaneously hypertensive rats (SHR) for eight weeks to investigate attenuating effects on age-related development of hypertension and cardiovascular hypertrophy. Results showed that treatment with A4H of SHRs attenuated the development of hypertension as effectively as the clinical antihypertensive drug captopril. Moreover, a significantly lower heart to body weight ratio and thinness of coronary arterial wall was observed in SHRs that had been treated with A4H or captopril. The results suggest that A4H can be utilized in developing an ACE inhibitor as a potential ingredient of functional foods to alleviate hypertension and cardiovascular hypertrophy.

Effects of Euphorbiae lathyridis Semen on cell apoptosis in HT-29 human colon cancer cells (속수자가 HT-29 대장암세포의 활성 및 세포사멸에 미치는 영향)

  • Objectives : In this study, we investigate that Euphorbiae lathyridis Semen extract contributes to growth inhibitory effect and anti-cancer activity on the HT-29 human colon cancer cells. Methods : Euphorbiae lathyridis Semen was extracted from the Semen of the plant using 80% Methanol. The Euphorbiae lathyridis Semen extract was treated to different concentrations for 24 hr, 4Shr or 72hr. Growth inhibitory effect was analyzed by measuring FACS study and MTT assay. Cell apoptosis was confirmed by surveying caspases cascades activation using Westem blot. Results : Exposure to Euphorbiae lathyridis Semen extract (0.4mg/ml) results in an inhibitory effect on cell growth in HT-29 cells. Growth inhibition by Euphorbiae lathyridis Semen extract in HT-29 cells was related with the inhibition of proliferation and induction of apoptosis. The Euphorbiae lathyridis Semen extract induces DNA fragmentation in HT-29 cells. Furthermore, Euphorbiae lathyridis Semen extract induces cell apoptosis through the activation of caspases-3, caspase-9 and PARP cleavage. Conclusion : Euphorbiae lathyridis Semen extract induces apoptosis in human colon cancer cells, therefore, we suggest that Euphorbiae lathyridis Semen extract can be used as a novel class of anti-cancer drugs.

The Effect of Eicosanoids on Cerebral Energy Metabolism and the $Ca^{++}$Concentration in Ischemic Rats (Eicosanoids가 뇌허혈증 흰쥐의 에너지대사 및 $Ca^{++}$이동에 미치는 영향)

  • We studied the effect of eicosanoids on the content of energy metabolites and the intrasynaptosomal $Ca^{++}([Ca^{++}]_i)$ concentration in cerebral ischemic rats. An ischemic model was made by bilateral carotid artery ligation (BCAL) and by incubation of synaptosomes under aglycemic and $N_2$ gas bubbling condition. The content of ATP, creatine phosphate and glucose decreased at 15 minutes after BCAL while that of lactate increased in male Wistar rats. Oral administration of EPA(100 mg/ml/Kg/day) or DHA(16 mg/ml/Kg/day) for 6 weeks improved both the decreases and the increase of the cerebral energy metabolites. In addition, the increase of $[Ca^{++}]_i$, under BACL was suppressed by EPA or DHA treatment. When the both Wistar rats and SHR were administered orally with EPA or DHA for 6 weeks, the effect on the increase of $[Ca^{++}]_i$ under ischemia by $N_2$ gas bubbling were protected. From these results, it may be that EPA or DHA treatment were greatly contributed to preservation of ischemic cerebral energy metabolism and $Ca^{++}$ concentration.

Effect of Gamidojeok-san on Hypertension (가미도적산이 고혈압에 미치는 영향)

  • This study was Investigated to prove the effect of Gamidojeok-san (Jiaweidaochi-san) on the hypertension. After administer Gamidojeok-san (Jiaweidaochi-san) extract to SHR(Spontaneous Hypertensive Rat) for 5 weeks, we examined changes of blood pressure, pulse rate, aldosterone and catecholamine in serum. Results : Following results were obtained; Gamidojeok-san(Jiaweidaochi-san) showed significant antihypertensive effect. Gamidojeok-san(Jiaweidaochi-san) decreased pulse rate but insignificant. Gamidojeok-san(Jiaweidaochi-san) showed a significant decrease in the aldosterone. Gamidojeok-san(Jiaweidaochi-san) was ineffective on the dopamine. Gamidojeok-san(Jiaweidaochi-san) decreased the norepinephrine but insignificant. Gamidojeok-san(Jiaweidaochi-san) showed a significant decrease in the epinephrine. These results indicate that Gamidojeok-san(Jiaweidaochi-san) can be used in hypertension. Further study will be needed about the functional mechanism and etc..
