Optimization of Makgeolli Manufacture Using Several Sweet Potatoes (다양한 고구마를 이용하여 제조한 막걸리의 최적화)

  • The objective of this study was to manufacture three kinds of domestic sweet potato Makgeolli using a mixture design and an optimization technique. The effects of four different manufacture methods, such as simultaneous saccharification and fermentation (SSF) with or without malt and separate hydrolysis and fermentation (SHF) with or without malt were determined. The SSF methods of Makgeolli produced higher alcohol content than that of SHF methods. The sensory score was not influenced by different making methods. Fourteen experimental points were selected, and rice (10~50%), sweet potato (10~50%) and water (40~60%) were chosen as independent variables. The measured responses were sensory preference, total polyphenol content, and DPPH radical scavenging activities. The ratio of the optimum sweet potato Makgeolli mixture formulation was developed as 15.11 (rice): 44.89 (sweet potato): 40 (water) using the optimization technique. The desirability of the optimum mixture formulation was 0.839. Yellow sweet potato Makgeolli using the optimum mixture formulation produced higher soluble sugar content compared to others. Regular sweet potato Makgeolli produced higher pH. The purple sweet potato Makgeolli's total polyphenol content and DPPH radical scavenging activity were measured to be the highest at $771.91{\pm}1.42mg\;GAE/{\ell}$, $131.55{\pm}4.03%$.

Bioethanol Production from Wasted Corn Stalk from Gangwon Province : from Enzymatic Hydrolysis to Fermentation (강원지역 폐옥수수대로부터 바이오에탄올 생산 : 효소 당화부터 발효까지)

  • Among the samples prepared by various pre-treatment methods, the one pretreated by dilute sulfuric acid showed the highest glucose yield in the enzymatic hydrolysis. Statistical analysis of enzymatic hydrolysis revealed that the glucose yield was in proportion to the enzyme dosage, the ratio of the pre-treated sample to the buffer solution, and the reaction time and that the effect of enzyme dosage was predominant in the experiment range. In addition, the glucose yield was estimated to be 76.1% at an optimal enzymatic hydrolysis condition. In a separate hydrolysis and fermentation (SHF), Saccharomyces cerevisiae converted over 80% of glucose from the enzymatic hydrolysis of pre-treated wasted corn stalk by dilute sulfuric acid to bioethanol with 37% of ethanol yield and 0.42 $g/L{\cdot}hr$ of productivity. In the simultaneous saccharification and fermentation (SSF), 59.5% of conversion from glucan to ethanol and 0.20 $g/L{\cdot}hr$ of productivity were achieved. In both SHF and SSF, approximately 88 g of bioethanol could be obtained from 1 kg of wasted corn stalk. The possible amount of bioethanol in Gangwon province were estimated to be 1.9 kiloton with the assumption of the 50% of collection ratio.

Effects of photoperiod on nutrient digestibility, hair follicle activity and cashmere quality in Inner Mongolia white cashmere goats

  • Objective: This study investigated the effects of photoperiod on nutrient digestibility, hair follicle (HF) activity and cashmere quality in Inner Mongolia white cashmere goats. Methods: Twenty-four female (non-pregnant) Inner Mongolia white cashmere goats aged 1 to 1.5 years old with similar live weights (mean, $20.36{\pm}2.63kg$) were randomly allocated into two groups: a natural daily photoperiod group (NDPP group:10 to 16 h light, n = 12) and a short daily photoperiod group (SDPP group: 7 h light:17 h dark, n = 12). All the goats were housed in individual pens and fed the same diets from May 15 to October 15, 2015. The digestibility of crude protein (CP), dry matter (DM), and neutral detergent fiber (NDF) were measured in different months, along with secondary hair follicle (SHF) activity, concentration of melatonin (MEL), and cashmere quality. Results: Although there was no significant difference in the live weights of goats between the SDPP and NDPP groups (p>0.05), the CP digestibility of goats in the SDPP group was significantly increased compared to the NDPP group in July, September, and October (p<0.05). For the DM and NDF digestibility of goats, a significant increase (p<0.05) was found during in September in the SDPP group. Furthermore, compared to the NDPP group, the SHF activity in July, the MEL concentration in July, and the cashmere fiber length and fiber weight in October were significantly increased in the SDPP group (p<0.05). Conclusion: The cashmere production of Inner Mongolia white cashmere goats was increased without obvious deleterious effects on the cashmere fibers in the SDPP group (metabolizable energy, 8.34 MJ/kg; CP, 11.16%; short daily photoperiod, 7 h light:17 h dark).

  • This study introduces about the analysis, verification and implementation of Digital Communication Satellite transponder. Through the pre-performance analysis of each component by the circuit level simulation, each component's performance was checked whether satisfying the specifications. When each of the components was connected in order, System's performances were evaluated through system harmonic balanced simulation whether satisfying the specifications. Through pre-analysis of the system performance, specification of each component was defined. On the basis of that specification, Components which comprise the Digital Communication Satellite repeater were manufactured, and finally, the overall system performance check was made by integrating the components of Digital Communication Satellite transponder.

  • In the defence wireless communications, conventional Anti-Jamming techniques(Frequency Hopping/Spread Spectrum or Direct Sequence/Spread Spectrum) are used to overcome a intentional interfering signals which are single/multitone or partial band jammer etc. DS/SS techniques is very strong on tone jamming signal but not to be on a partial band jammer. So FH/SS AJ performances are expected method of an substitution of DS/SS, however FH/SS could not have good performance on some BMTJ(Band Multi-tone Jammer). So this paper proposes FH-CSS (Frequency Hopped - Chirp Spread Spectrum) to get more robustness against jammers(BMTJ, PBNJ) and analyze the AJ performances.

Evaluation of Galactose Adapted Yeasts for Bioethanol Fermentation from Kappaphycus alvarezii Hydrolyzates

  • Bioethanol was produced from Kappaphycus alvarezii seaweed biomass using separate hydrolysis and fermentation (SHF). Pretreatment was evaluated for 60 min at 121℃ using 12% (w/v) biomass slurry with 364 mM H2SO4. Enzymatic saccharification was then carried out at 45℃ for 48 h using Celluclast 1.5 L. Ethanol fermentation with 12% (w/v) K. alvarezii hydrolyzate was performed using the yeasts Saccharomyces cerevisiae KCTC1126, Kluyveromyces marxianus KCTC7150, and Candida lusitaniae ATCC42720 with or without prior adaptation to high concentrations of galactose. When non-adapted S. cerevisiae, K. marxianus, and C. lusitaniae were used, 11.5 g/l, 6.7 g/l, and 6.0 g/l of ethanol were produced, respectively. When adapted S. cerevisiae, K. marxianus, and C. lusitaniae were used, 15.8 g/l, 11.6 g/l, and 13.4 g/l of ethanol were obtained, respectively. The highest ethanol concentration was 15.8 g/l, with YEtOH = 0.43 and YT% = 84.3%, which was obtained using adapted S. cerevisiae.

  • Ethanol fermentations were performed using separate hydrolysis and fermentation (SHF) processes with monosaccharides from pretreated seaweed, Eucheuma spinosum as the biomass. The pretreatment was carried out with 11% (w/v) seaweed slurry and 150 mM $H_2SO_4$ at $121^{\circ}C$ for 40 min. Enzyme hydrolysis after $H_2SO_4$ pretreatment was performed with Celluclast 1.5 L at $45^{\circ}C$ for 24 h. Five % active charcoal were added to hydrolysate to removed 5-hydroxy methylfurfural. Ethanol fermentation with 11% (w/v) seaweed hydrolysate was performed for 72~96 h using Kluyvermyces marxianus, Pichia stipits, Saccharomyces cervisiae and Candida tropicalis. Ethanol concentration was reached to 18 g/L by K. marxianus, 16 g/L by P. stipitis, 15 g/L by S. cerevisiae and 10 g/L by C. tropicalis, respectively. The ethanol yield from total monosugar was obtained 0.50 and ethanol productivity was obtained 0.38 g/L/h by K. marxianus.

PCR-Based Sensitive Detection of Wood-Decaying Fungus Phellinus linteus by Specific Primer from rDNA ITS Regions

  • Based on the rDNA ITS sequences data, specific primer set for PCR detection of wood-decaying fungus Phellinus linteus was designed. The length of PCR products using designed primer set(SHF and SHR) was about 540 bp. Among 11 species, 17 isolates of Phellinus spp. including Phellinus linteus, P. pomaceus, P. spiculosus, P. baumi, P. pini, P. igniarius, P. gilvus, P. biscuspidatus, P. weirii, P. johnsonianus, P. robutus, and P. igniarius, seven isolates of Phellinus linteus showed about 540 bp-sized single band. This molecular technique could offer a useful tool for detecting and identifying Phellinus linteus.

Performance Investigation of Visible Light Communication Using Super Bright White LED and Fresnel Lens (조명용 고출력 백색 LED와 프레넬 렌즈를 이용한 가시광 통신 성능연구)

  • White light-emitting diode (WLED) is growing interest in using both illumination and communications. This paper reports visible light communication (VLC) composed of a super bright white light-emitting diode, low cost commercial photo-diode and a Fresnel lens. LED driver is consisted of the power MOSFET and MOSFET driver that switches the LED on and off. The modulation bandwidth of the LED used was determined to be 8 MHz. However, it was possible to communicate up to 1 Mbps under illumination of 500 lx because of the weak signal power and a low spectral sensitivity of the SHF213 as a PIN photodiode. In order to enhance the system bandwidth, the LED light was focused on the PIN photodiode by use of the Fresnel lens. As a result of that, visible light link was operated up to modulation bandwidth of the LED. The signal to noise ratio can be improved by 40 dB using an optical concentration at the receiver.

  • The Simultaneous Saccharification and Fermentation(SSF) of ethanol production from potato starch studied with respect to growth pH, temperature, substrate concentration. The glucoamylase and Saccharomyceses cerevisiae have a capacity to carry out a single stage SSF process for ethanol production. The characteristics, termed as starch hydrolysis, accumulation of glucose, ethanol production and biomass formation, were affected with variation in pH, temperature and starch concentration. The maximum ethanol concentration of 12.9g/l was obtained using a starch concentration 30g/l, which represent an ethanol yield of 86%. The optimum conditions for the maximum ethanol yield were found to be a temperature of 38, pH of 4.0 and fermentation time of 18hr. Thus by using the control composite design, it is possible to determine the accurate values of the fermentation parameters where maximum production of ethanol occurs.
