Optimal Conditions for the Expression of Glycoprotein E2 of Classical Swine Fever Virus using Baculovirus in Insect Cells

  • The structural proteins of classical swine fever virus (CSFV) consist of nucleocapsid protein C and envelope glycoprotein $E^{rns}$ (E0), E1 and E2. Among them, E2, the most immunogenic of the CSFV glycoproteins, induces a protective immune response in swine. In this study, to determine the optimal expression conditions of glycoprotein E2 using baculovirus system, we investigated the influence of insect cells and media to the expression of recombinant E2. Recombinant virus containing glycoprotein E2 coding gene was constructed with bApGOZA DNA. Expression of the glycoprotein E2 was analyzed by SDS-PAGE and Western blot analysis using anti-CSFV E2 monoclonal antibodies. Expression of glycoprotein E2 in Sf21 cells was first observed after 3 days and reached a maximum on the 5th day after infection. Furthermore, the highest levels of glycoprotein E2 expression were observed at multiplicity of infection (MOI) of 5. When three different insect cell lines (Sf21, High-Five and Se301) were tested, High-Five cells showed the highest production. In addition, four different serum-free and serum-supplemented media, respectively, were tested for the expression of glycoprotein E2 and the budded virus (BV) titers. As a result, serum-supplemented medium provided the best conditions for protein production and the BV yield.

  • To avoid imaging artifacts and interpretation mistakes, an improvement of the uniformity in gamma camera systems is a very important point. We can expect excellent uniformity using cadmium zinc telluride (CZT) photon counting detector (PCD) because of the direct conversion of the gamma rays energy into electrons. In addition, the uniformity performance such as integral uniformity (IU), differential uniformity (DU), scatter fraction (SF), and contrast-to-noise ratio (CNR) varies according to the energy window setting. In this study, we compared a PCD and conventional scintillation detector with respect to the energy windows (5%, 10%, 15%, and 20%) using a $^{99m}Tc$ gamma source with a Geant4 Application for Tomography Emission simulation tool. The gamma camera systems used in this work are a CZT PCD and NaI(Tl) conventional scintillation detector with a 1-mm thickness. According to the results, although the IU and DU results were improved with the energy window, the SF and CNR results deteriorated with the energy window. In particular, the uniformity for the PCD was higher than that of the conventional scintillation detector in all cases. In conclusion, our results demonstrated that the uniformity of the CZT PCD was higher than that of the conventional scintillation detector.

  • Superconducting equipment has been actively investigated for securing the environment and energy technology (ET) in various parts of the world. Despite these movements, a high voltage cryogenic bushing, which plays an important role of interconnection between the electric power systems and superconducting devices, has not been fully developed due to severe insulation requirements. A gas insulated cryogenic bushing has been investigated as one of our projects since 2010. As a basic step to obtain the design parameters for cryogenic bushing, we focused on the surface flashover characteristics of glass fiber reinforced plastic (FRP) in several insulation gases. For the surface flashover tests, several insulation gases including $SF_6$, $CF_4$ and $N_2$ gas were prepared. Various length of FRP specimens were fabricated in order to obtain the fundamental data for creepage distance of FRP. The first specimen group was from 2 mm to 10 mm with 2 mm intervals and the second specimen group was from 20 mm to 100 mm with 20 mm intervals. And the gas pressure was varied from 1 bar to 4 bar. An AC overvoltage test and a lightning impulse test were performed. Then the experimental results of surface flashover were obtained and analyzed. Based on these results, it would be possible to design the optimum creepage distance of FRP in a cryogenic bushing.

  • Power plant is that very high reliability when industrial and economic impact on the overall electric power system is required, it is essential to improve the reliability, especially the fault prediction diagnosis. Since an accident caused by the partial discharge in the power plant is above state has a faster response characteristic than the other indications in the case of any, the partial discharge generated in the power plant immediately detect the deterioration of insulation due to the accident of the power plant and the non-drawn It should prevent or reduce. Partial Discharge Measuring Systems for UHV SF6 Gas Insulated Switchgear and power transformer on site installed has some probability of abnormal recognition in case of non-flexible deal with on site noise. Many methode to eliminate these kinds of noises, UHF Detection System is chosen as purchase description in Korea, but this system having a bandwidth between 500MHz 1.5GHz wide band. Initial install periods(about 20 years ago), this band had no strong signal source, but in these days this wide band have strong signals, such as LTE. So, module described in this paper is designed as simultaneously use with wide and narrow band for solve this noise problem, and introduce this system.

Characterizations of the Mechanical Properties and Wear Behavior of Ni Plate Fabricated by the Electroforming Process (Electroforming을 이용하여 제조한 Ni 기판의 기계적 특성 및 내마모 거동 분석)

  • We fabricated the Ni plate by electroforming process and evaluated the microstructure, mechanical properties and wear behavior of the Ni plate. Specifically, the effects of addition of wetting agents, SF 1 and SF 2 solutions, on the microstructure and properties were investigated. The microstructure and surface morphology were characterized by transmission electron microscopy (TEM) and atomic force microscopy (AFM), respectively, and friction coefficient was measured by the ball-on-disk method. We found that the microstructure and mechanical properties of Ni plate were changed with kind and amount of wetting agents used. The hardness and tensile strength of Ni plate formed without wetting agents was 228 Hv and 660.7 MPa, respectively, whiled when wetting agent was added, those were improved to be 739 Hv and 1286.3 MPa. These improvements were probably due to the finer grain size and less crystallization of Ni. In addition, when both wetting agents were added, the friction coefficient was reduced from 0.73 to 0.67 which is partially caused by the improved hardness and smooth surface.

Study on Treatment Characteristics of Perfluorinated Compounds Using a High Temperature Plasma (고온 플라즈마를 이용한 과불화화합물의 처리 특성 연구)

  • In this study, the decomposition characteristics of perfluorinated compounds generated in semiconductor manufacturing process were investigated by using a high temperature plasma. The analysis results revealed that $CF_4$ and $SF_6$ showed the highest efficiency at 12.8 kW power, but no significant difference was observed at the power above. Experimental results showed that the maximum efficiency was obtained at the flow rate of about 14 mL/min and the treatment efficiency decreased as the flow rate increased or decreased with respect to the flow rate of 14 mL/min. As a result, the decomposition characteristics of perflurocompounds (PFCs) using a high temperature plasma could be grasped, and also the basis for the treatment of PFCs and greenhouse gases generated in the semiconductor manufacturing process could be obtained.

  • Digital Twin is a technology that creates a photocopy of real-world objects on a computer and analyzes the past and present operational status by fusing the structure, context, and operation of various physical systems with property information, and predicts the future society's countermeasures. In particular, 3D rendering technology (UAS, LiDAR, GNSS, etc.) is a core technology in digital twin. so, the research and application are actively performed in the industry in recent years. However, UAS (Unmanned Aerial System) and LiDAR (Light Detection And Ranging) have to be solved by compensating blind spot which is not reconstructed according to the object shape. In addition, the terrestrial LiDAR can acquire the point cloud of the object more precisely and quickly at a short distance, but a blind spot is generated at the upper part of the object, thereby imposing restrictions on the forward digital twin modeling. The UAS is capable of modeling a specific range of objects with high accuracy by using high resolution images at low altitudes, and has the advantage of generating a high density point group based on SfM (Structure-from-Motion) image analysis technology. However, It is relatively far from the target LiDAR than the terrestrial LiDAR, and it takes time to analyze the image. In particular, it is necessary to reduce the accuracy of the side part and compensate the blind spot. By re-optimizing it after fusion with UAS and Terrestrial LiDAR, the residual error of each modeling method was compensated and the mutual correction result was obtained. The accuracy of fusion-based 3D model is less than 1cm and it is expected to be useful for digital twin construction.

Predictive modeling of the compressive strength of bacteria-incorporated geopolymer concrete using a gene expression programming approach

  • The performance of gene expression programming (GEP) in predicting the compressive strength of bacteria-incorporated geopolymer concrete (GPC) was examined in this study. Ground-granulated blast-furnace slag (GGBS), new bacterial strains, fly ash (FA), silica fume (SF), metakaolin (MK), and manufactured sand were used as ingredients in the concrete mixture. For the geopolymer preparation, an 8 M sodium hydroxide (NaOH) solution was used, and the ambient curing temperature (28℃) was maintained for all mixtures. The ratio of sodium silicate (Na2SiO3) to NaOH was 2.33, and the ratio of alkaline liquid to binder was 0.35. Based on experimental data collected from the literature, an evolutionary-based algorithm (GEP) was proposed to develop new predictive models for estimating the compressive strength of GPC containing bacteria. Data were classified into training and testing sets to obtain a closed-form solution using GEP. Independent variables for the model were the constituent materials of GPC, such as FA, MK, SF, and Bacillus bacteria. A total of six GEP formulations were developed for predicting the compressive strength of bacteria-incorporated GPC obtained at 1, 3, 7, 28, 56, and 90 days of curing. 80% and 20% of the data were used for training and testing the models, respectively. R2 values in the range of 0.9747 and 0.9950 (including train and test dataset) were obtained for the concrete samples, which showed that GEP can be used to predict the compressive strength of GPC containing bacteria with minimal error. Moreover, the GEP models were in good agreement with the experimental datasets and were robust and reliable. The models developed could serve as a tool for concrete constructors using geopolymers within the framework of this research.

Is central pancreatectomy an effective alternative to distal pancreatectomy for low-grade pancreatic neck and body tumors: A 20-year single-center propensity score-matched case-control study

  • Backgrounds/Aims: Central pancreatectomy (CP) is associated with a higher rate of postoperative pancreatic fistula (POPF), and it is less preferred over distal pancreatectomy (DP). We compared the short- and long-term outcomes between CP and DP for low-grade pancreatic neck and body tumors. Methods: This was a propensity score-matched case-control study of patients who underwent either CP or DP for low-grade pancreatic neck and body tumors from 2003 to 2020 in a tertiary care unit in southern India. Patients with a tumor >10 cm or a distal residual stump length of <4 cm were excluded. Demographics, clinical profile, intraoperative and postoperative parameters, and the long-term postoperative outcomes for exocrine and endocrine insufficiency, weight gain, and the 36-Item Short Form Survey (SF-36) quality of life questionnaire were compared. Results: Eighty-eight patients (CP: n=37 [cases], DP: n=51 [control]) were included in the unmatched group after excluding 21 patients (meeting exclusion criteria). After matching, both groups had 37 patients. The clinical and demographic profiles were comparable between the two groups. Blood loss and POPF rates were significantly higher in the CP group. However, Clavien-Dindo grades of complications were similar between the two groups (p = 0.27). At a median follow-up of 38 months (range = 187 months), exocrine sufficiency was similar between the two groups. Endocrine sufficiency, weight gain, SF-36 pain control score, and general health score were significantly better in the CP group. Conclusions: Despite equivalent clinically significant morbidities, long-term outcomes are better after CP compared to DP in low-grade pancreatic body tumors.

Effect of Adding Ankle-Joint-Pumping Exercise to Gluteal-Muscle-Strengthening Exercise on Thigh Swelling, Gait ability and Pain level in Patients With Total Hip Arthroplasty (볼기근 근력강화 운동과 병행한 발목관절 펌핑 운동이 엉덩관절 전치환술 환자의 허벅지 둘레, 보행능력, 통증 수준에 미치는 영향 )

  • PURPOSE: This study was conducted to investigate the effect of gluteal muscle strengthening exercises (GMSE) with ankle joint pumping exercises (AJPE) on thigh swelling, gait ability, and pain level in patients who underwent total hip arthroplasty. METHODS: A total of 38 patients who had undergone total hip replacement surgery >1 week prior participated in this study. Participants were randomly assigned to a group that performed only GMSE (CG; n = 19) and a group that performed GMSE and AJPE (EG; n = 19). The CG group performed GMSE for 30 min, and the EG group performed GMSE for 30 min followed by AJPE for 15 min. Exercises were performed five times a week, for a total of 20 times over 4 weeks in both groups. Thigh swelling (thigh size), 10 m walking test (10MWT) and timed up and go test (TUG) results, pain level (visual analogue scale, VAS) scores, Short Form 36 health survey (SF-36), and hip outcome scale (HOS) scores were evaluated before and after the intervention. RESULTS: After 4 weeks of intervention, significant differences were observed in the thigh size, 10MWT, TUG, VAS, SF-36, HOS before and after intervention in both groups (p < . 05). However, only thigh size showed a significant interaction between group and measurement time (p < . 05). CONCLUSION: GMSE combined with AJPE might be effective in improving the gait ability and pain level in patients with total hip arthroplasty, and GMSE may be more effective in improving thigh swelling and gait ability than GMSE without AJPE.
