Solution deposition planarization for IBAD-MgO texture template

  • In this work, the optimized process condition of chemical solution deposition which is used to planarize the surface of the metal tape (which is used to grow IBAD-MgO texture template) was investigated. $Y_2O_3$ films were dip-coated on the surface of the unpolished metal tape as the seed and barrier layer. The effects of $Y_2O_3$ concentration of the solution (0.5wt.%, 1.3wt.%, 2.8wt.%, 5.6wt.%) and the number of coatings on the surface morphology and barrier capability against the diffusion from the metal tape were examined. The surface morphology and the thickness of the film were observed using the scanning electron microscope and the atomic force microscope. The presence of elements in metal tape on the film surface was analyzed using the auger electron spectroscopy. The $Y_2O_3$ film thickness increases with increasing the $Y_2O_3$ concentration in the solution, and the surface became smoother with increasing the number of coating cycles. The best result was obtained from the $Y_2O_3$ film coated 4 cycles using 2.8wt.% solution.

  • The effect of process parameters such as the growth pressure and temperature on the AlN crystal growth has been investigated. AlN crystal was grown onto 6H-SiC seed crystal using PVT (Physical Vapor Transport) method. Crystal properties and morphology of AlN crystal was changed with growth pressure and temperature. Raman analysis confirmed that AlN crystals with different orientation were successfully grown on SiC seed crystal.

  • Anatomical characters such as stem, leaf, ovary, calyx lobe, ultrastructure of stigma, epidermis of leaf blade and midvein, corolla lobe, nectary, seed coat and pollen were examined on 6 taxa of Korean Ophelia, including 5 taxa distributed in south Korea and one taxon considered to be the variation type of Ophelia wilfordi, in order to clarify the limits of intersection and interspecies and to establish the taxonomic position. One taxon distributed in north Korea was also included in the description of species by observation of herbarium specimen of the University of Tokyo in Japan. The two sections were successfully distinguished by internal structure of ovary, morphology of nectary, surface sculpturing of corolla lobe and pollen, ultrastructure of seed and seed coat, which were useful characters to distinguish taxa higher than species. The variation type of Ophelia wilfordi was not distinguished with other species except for absent or present of purple spot in corolla lobe.

Photosynthesis and Leaf Anatomical Morphology on Different Leaf Shape of Soybean (엽형에 따른 콩 품종의 광합성 능력과 잎의 해부형태 비교)

  • To find ideal leaf types for soybean breeding program, we examined the relationships among leaf anatomical and morphological characteristics and $\textrm{CO}_2$ assimilation on the different leaflet shape of soybean (Glycine max). In anatomical characters of leaf, palisade and spongy cells were thicker in both small seed cultivars with narrow leaflet and large seed cultivars with wide leaflet than others. $\textrm{CO}_2$ uptake per plant and leaf thickness were significantly associated with seed yield per plant, showing difference among the soybean cultivars. Although the leaf area was lower for narrow leaflet cultivars, which had a significantly higher photosynthetic rate per plant comparable to the wide leaflet cultivars.

Significance of Semame Seedborne Fungi, with special Reference to Corynespora cassiicola (참깨의 종자전염성 진균과 그 병원성 : Corynespora cussiicola를 중심으로)

  • Alternaria sesami, A. sesamicola, A. tenuis, A. longissima, Cercospora sesami, Cephalosporium sp., Corynespora cassiicola, Fusarium equiseti, F. moniliforme, F. oxysporum, F. semitectum, Macrophomina phaseolina and Myrothecium roridum were detected from 40 seed samples of sesame. A sesami, A. sesamicola, A. tenuis and C. cassiicola were the predominant fungi. Except C. cassiicola, all fungi were almost completly reduced and wiped out the infection by pretreatment with chlorine. Plating components also indicate that C. cassiicola was well-established infections. Seedborne infection of C. cussiicola caused heavy seed rot and seedling mortality. Detailed description has been given on the habit character of C. cassiicola under stereoscopic microscope and the variation in colony character and spore morphology have been taken into account. In inoculation experiments, C. cassiicola produced severe leaf and stem spots and blights on sesame plants resulted in ultimate death of the plants. A. sesami, A. sesamicola A. longissima and C. sesami also produced mild to severe leaf spotting and leaf blight when suspension of their conidia were sprayed on to plants. In soil inoculation experiments, F. oxysporum and M. phaseolina were the most pathogenic causing seed rot and seedling blight.

Regeneration of plants from alginate-encapsulated axenic nodal segments of Paederia foetida L. - A medicinally important and vulnerable plant species

  • Paederia foetida L. is an important medicinal plant that has been used for the treatment of various gastrointestinal related ailments by different tribal communities in India. This plant is also known for its use as a food. Due to overexploitation, P. foetida has been classified as a vulnerable plant in some states of India. The propagation of P. foetida by conventional methods is easy but very slow. Synthetic seed technology offers incredible potential for in vitro propagation of threatened and commercially valuable plants, and can also facilitate the storage and exchange of axenic plant material between laboratories. However, synthetic seed production for P. foetida has not yet been reported. Thus, to the best of our knowledge, the present study is the first attempt to produce synthetic seeds of P. foetida by calcium alginate encapsulation of in vitro regenerated axenic nodal segments. Sodium alginate (3%) and CaCl2 (100 mM) were found to be the optimal materials for the preparation of ideal synthetic seeds, both in terms of morphology and germination ability. The synthetic seeds showed the best germination (formation of both shoot as well as root; 83.3%) on ½ MS medium augmented with 0.5 mg/L indole-3-acetic acid. The plantlets obtained from these synthetic seeds could be successfully acclimatized under field conditions. We also studied the storage of these synthetic seeds at low temperature and their subsequent sprouting/germination. The seeds showed a germination rate of 63.3% even after 21 days of storage at 4 ℃; thus, they could be useful for transfer and exchange of P. foetida germplasm.

Epitaxial growth of buffer layers for superconducting coated conductors (초전도 선재용 완충층의 결정성장 연구)

  • All three buffer layers of $Y_2O_3$, YSZ, and $CeO_2$ have been deposited on the biaxially textured metal substrates using rf-sputtering method, The first 50-70nm thick $Y_2O_3$ films were grown epitaxially on biaxially textured metal substrates as a seed layer and followed by the diffusion barrier ${\sim}100nm$ thick YSZ and subsequent capping layer ${\sim}200nm$ thick $CeO_2$ deposited epitaxially on top of $Y_2O_3$ seed layer. The epitaxial orientation of all three layers were all (100) grown with rocking curve Full Width at Half Maximum(FWHM) of $4-5^{\circ}$ and in plane phi-scan FWHM of $6-8^{\circ}$ using X -ray diffraction analysis. The NiO phases formed during the $Y_2O_3$ seed layer deposition seem to degrade the crystallinity and roughen the surface morphology of the following layer observed by AFM(Atomic Force Microscopy). The buffered tapes were used as substrates for long length YBCO coated conductors with high critical current density $J_c$. The five multi-turn of metal tapes was employed to increase the thickness of films and production rate to compensate the low growth rate of rf-sputtering method.

Morphological and Morphophysiological Dormancy in Seeds of Two Genera (Heloniopsis and Thalictrum) Native to Korea (자생 관상용 Heloniopsis와 Thalictrum속 종자의 형태적 및 형태생리적 휴면)

  • This study was conducted to find basic information for the research of morphological dormancy in seeds of two genera (Heloniopsis and Thalictrum) native to Korea. Seeds of Heloniopsis koreana, Heloniopsis tubiflora, Thalictrum rochenbrunianum, Thalictrum uchiyamai, and Thalictrum coreanum were collected, and embryo morphology and seed germination were investigated. All seeds of five species had underdeveloped embryos which occupied about 10-20% of the full seed length at maturity. The seeds of H. koreana and H. tubiflora had rudimentary embryos. The seeds of T. rochenbrunianum, T. uchiyamai, and T. coreanum had intermediate type between rudimentary and linear embryo. The small embryos of all seeds of five species grew to a critical length before radicles emerge from the seeds. At 30 days after sowing, the seeds of H. koreana and H. tubiflora germinated to 22.7% and 40.7%, respectively. On the other hand, the seeds of T. rochenbrunianum, T. uchiyamai, and T. coreanum germinated to 12.5%, 3.3%, and 0%, respectively. If embryo growth and germination in seeds are completed in approximately 30 days, the seed are said to have morphological dormancy (MD). However, if the seeds require > 30 days for germination, they are considered to have morphophysiological dormancy (MPD). Thus, the seeds of H. koreana and H. tubiflora had about 78% and 60% MPD, respectively. On the other hand, the seeds of T. rochenbrunianum, T. uchiyamai, amd T. coreanum had about 87%, 96%, and 100% MPD, respectively. There was a different level of dormancy (MD and MPD) within the same seed population examined. Therefore, germination of the seeds was delayed due to the underdeveloped embryos. This result could contribute to determine morphological seed dormancy and germination mechanism in seeds of the Korean Peninsula.

An updated taxonomy of the family Linderniaceae in Korea

  • In the present study, according to morphological observations followed by recent circumscriptions, we have classified the Korean taxa of the family Linderniaceae into Scrophulariaceae sensu lato has been considered in several works, though the taxa have remained undefined because identification work was mostly done according to vegetative morphological features, such as the leaf shape, leaf margins, and leaf venation. The taxa of Linderniaceae are mostly considered to be weeds and, for correct identification, it is necessary to clarify their taxonomic characteristics. Morphological studies were carried out using samples collected in the field. Micro-morphological observations of the vegetative and floral parts were also performed using light microscopy (LM) and scanning electron microscopy (SEM). We concluded that important characteristics are reproductive morphologies viz. calyx, stamen structure, capsule shape, calyx ratio with capsule, inflorescence morphology, and seed morphology. As a result, we formulated taxa descriptions and provided a key of the genera of Linderniaceae in Korea. Lindernia crustacea (L.) F. Muell. is transferred to Torenia crustacea (L.) Cham. & Schltdl. Lindernia micrantha D. Don and L. angustifolia (Benth.) Wettstein are a synonym of Vandellia micrantha (D. Don) Eb. Fisch., Schäferh. & Kai Müll. Lindernia attenuata Muhl. and L. dubia var. major (Pursh) Pennell are a synonym of Lindernia dubia (L.). Lindernia verbenifolia (Colsm.) Pennell is a synonym of Bonnaya antipoda Druce. Our study reports the presence of four genera: Bonnaya, Lindernia, Torenia, and Vandellia, comprising six taxa under the family Linderniaceae in Korea.

Morphology and Characteristics of Floral Organ in Highbush Blueberry (Vaccinium corymbosum) Cultivars (하이부쉬 블루베리의 품종별 화기 특성 및 형태적 관찰)

  • Morphology and characteristics of floral organ in highbush blueberry cultivars were studied to select suitable cultivars of highbush blueberry for domestic cultivation. The stamen consists an anther and a tape-like hairy filament with well-developed trichomes. When the anther was opened, the wall of anther was not dehiscent, and pollen grains were discharged into two tubes. Pollen was mature tetrad type without being separated after meiosis (Late March). The number of pollen granules per anther was 400~1,300, the germination rate was higher in the cultivars having many pollen grains. Pistil was composed of five carpels and a shipper without separate part. The number of ovules per ovary was 39~67, therefore, the coefficient of ranged from 11.6 to 31.0%. The seed pod formation by combination of ‘Bluejay’ and ‘Sharpblue’ was higher in the cross-pollination than in the self-pollination.
