개 세르토리세포종 케이스에서 면역조직화학적 마커를 통한 진단 (Diagnosis of immunohistochemical marker expressed by a canine Sertoli cell tumor case)

  • Sertoli cell tumor (SCT) of the testicle arises from the supporting cells within the seminiferous tubules. SCT is common in dogs, especially in cryptorchid testicles, but also has been reported in the stallion, ram, cat, and bull. Sertoli cell tumor sample was collected from 7-years male german shepherd. In this study, SCT arose from one testicle. Sample size is approximately 1.7 cm in diameter and it has a round form. In the microscopic, cells within the tumor variably resemble Sertoli cells (SCs) that normally populate the seminiferous tubules and interstitial area. There is abundant stroma of dense, mature fibrous connective tissue in SCT. In the immunohistochemical staining, cytokeratin AE1/AE3 was not expressed in the control and SCT. S-100 protein was expressed by SCs, germ cells and fibrous connective tissue of SCT. Melan A was expressed by leydig cells (LCs) of SCT. A study by using S-100 and melan A in canine SCT was almost never carried out. S-100 and melans A is considered to suggest for diagnosis and pathogenesis of canine SCTs. Inhibin-alpha and Vimentin were well known as the marekers of SCTs. Also, they were expressed by Sertoli cells and LSs of SCT in this study.

SCT 세라믹 박막의 제조 및 구조적 특성 (Fabrication and Structural Properties of SCT Ceramic Thin Film)

  • The (Sr$\sub$0.85/Ca$\sub$0.15/)TiO$_3$(SCT) thin films are deposited on Pt-coated electrode(Pt/TiN/SiO$_2$/Si) using RF sputtering method. The crystallinity of SCT thin films is increased with increase of substrate temperature in the temperature range of 100[$^{\circ}C$]∼500[$^{\circ}C$]. Also, the crystallinity of SCT thin films are obtained at the substrate temperature above 400[$^{\circ}C$]. SCT thin films had (111) preferred orientation. The dielectric constant changes almost linearly in temperature ranges of-80∼+90[$^{\circ}C$]. The temperature properties of the dielectric loss have a stable value within 0.1. SCT thin films used in this study show the phenomena of dielectric relaxation with the increase of frequency, and the relaxation frequency is observed above 200[kHz].

  • The (S $r_{0.85}$C $a_{0.15}$) Ti $O_3$(SCT) thin films are deposited on Pt-coated electrode (Pt/TiN/ $SiO_2$/Si) using RF sputtering method at various deposition temperature. The crystallinity of SCT thin films were increased with increase of deposition temperature in the temperature range of 200~500[$^{\circ}C$]. Also, the composition of SCT thin films were closed to stoichiometry (1.080~1.111 in A/B ratio). V-I characteristics of SCT thin films show the increasing leakage current with the increases of deposition temperature. The conduction mechanism of the SCT thin films observed in the temperature range of 25~100[$^{\circ}C$] can be divided into four regions with different mechanism by the increasing current. The region I below 0.8[MV/cm]shows the ohmic conduction. The region II between 0.9~2[MV/cm] is in proportion to J∝ $E^{1.5}$ , the region III between 2~4[MV/cm] can be explained by the Child’s law, and the region IV above 4[MV/cm]is dominated by the tunneling effect.ect.

미더덕껍질의 유수추출물이 과산화수소에 의해 유발된 HepG2간암세포의 세포사멸에 미치는 보호 효과 (Protective Effect of Aqueous Extracts of Styela Clava Tunic Against Apoptosis of HepG2 Cells Induced by Hydrogen Peroxide)

  • 미더덕껍질(Styela clava tunic, SCT)은 항염증 복합체, 창상필름, 골재생 유도 등을 포함한 다양한 의학적인 치료영역에 이용 되고 있다. 본 연구에서는 미더덕껍질 열수추출물(aqueous extract of Styela clava tunic, AE-SCT)의 $H_2O_2$에 의해 유발된 세포사멸의 보호 효과를 알아보기 위하여 세포 활성도의 변화에 관련된 요인을 측정하였다. 그 결과, AE-SCT는 3.3 mg/g의 플라보노이드와 32.3 mg/g의 페놀화합물을 포함하고 있었으며, HepG2 세포주에 독성을 유발하지 않았다. 또한, $H_2O_2$ 처리 후 AE-SCT를 처리하는 실험에서 AE-SCT는 $H_2O_2$에 의해 유발된 세포사멸을 개선하는 효과를 나타내지 않았다. 그러나, $H_2O_2$ 처리전에 AE-SCT를 사전 처리하는 예방효과 실험에서, 세포생존율은 $H_2O_2$만 처리한 그룹에 비하여 AE-SCT를 처리한 그룹에서 유의적으로 증가하였으며, 특히 AE-SCT를 $50{\mu}g/ml$ 처리한 농도에서 가장 높았다. 또한, FACS분석과 DAPI 염색에서도 사멸 세포의 수는 $H_2O_2$만 처리한 그룹에 비하여 AE-SCT를 처리한 그룹에서 유의적으로 감소하였다. 더불어, $H_2O_2$의 처리에 의해 유도된 Bax/Bcl-2 발현비율은 AE-SCT처리에 의해 농도의존적으로 감소되었다. 이러한 연구 결과는 AE-SCT가 $H_2O_2$에 의해 유발된 세포사멸을 예방하는 우수한 효과를 가지고 있음을 제시하고 있어 향후 다양한 항산화 제품 개발을 위한 기초자료로 사용될 수 있을 것으로 사료된다.

알칼리처리에 따른 미더덕 껍질의 이화학적 특성 (Characterization of Styela clava Tunic after Alkaline Treatment)

  • 본 연구에서는 미더덕 껍질(SCT)의 전처리과정에 사용되는 수산화나트륨을 농도별로 처리하여 알칼리처리에 따른 SCT 셀룰로오스의 물리화학적 특성 및 효소학적 특성에 대한 연구를 수행하였다. 알칼리처리 한 SCT를 주사전자현미경으로 분석한 결과 알칼리처리에 의해 표면에 농도 의존적으로 상당히 많은 손상을 초래하는 것을 확인할 수 있었으며, X-선 회절 분석을 통한 결정화도의 분석으로 알칼리처리에 따라서 SCT 셀룰로오스의 결정화도가 증가함을 알 수 있었다. Celluclast를 이용한 효소의 초기반응에서는 알칼리처리에 따른 SCT 표면 손상과 효소반응이 밀접한 관계가 있었으나 장시간 동안 완전분해를 하는 경우에는 알칼리처리로 인한 결정화도의 증가로 인해 분해되지 않고 남는 SCT가 많음을 확인할 수 있었다. 따라서 SCT의 전처리 시에 알칼리 사용을 최대한 적게 사용하거나 결정화도를 감소시킬 수 있는 대체 기술에 대한 연구가 필요할 것으로 판단된다.

The Effect of Sopungchungyoung-tang on Activity of CD4 T cell

  • Sopungchungyoung-tang (SCT) has been widely used in Korea as a treatment of atopic dermatitis. SCT consists of Talcum, Rehmannia glutinosa, Angelica sinensis, Paeonia lactiflora, Cnidium officinale, Ledebouriella divaricata, Schizonepeta tenuifolia, Scutellaria baicalensis, Glycyrrhiza uralensis, Mentha arvensis, Cordyceps cicadae. We examined the immunological effect of SCT in vitro. We studied about the effect of SCT on Th cells' differentiation. In the case of CD4 T cells under neutral condition where there was only rIL-2 stimulus, SCT inhibited IFN-γ secretion by 70-80 %. Likewise, SCT also inhibited the IL-4 secretion of neutral Th cells by 85-90 %. We also experimented with the polarized Th1 cells/ Th2 cells and their production of IFN-γ and IL-4, respectively. There also were inhibitory effects on the polarized cells like there was on neutral cells, they were not as strong on the polarized cells. Under Th1 polarized condition, SCT acted dose-dependently, while in Th2 cells, the IL-4 production was inversely proportional to the doses of SCT. From the current study, it can be concluded that SCT exerts inhibitory effects on cytokine production without interfering with immune cells' activity. The result that SCT inhibits IFN-γ and IL-4 confirms that it does have a effect on immunomodulation.

The optimum conditions of RF power and Ar/O₂ ratio were 140[W] and 80/20, respectively. Deposition rate of SCT thin film was about 18.75[Å/min]. The crystallinity of SCT thin films were increased with increase of substrate temperature in the temperature range of 100~500[℃]. The dielectric constant of SCT thin films were increased with the increase of substrate temperature, and changed almost linearly in temperature ranges of -80~+90[℃]. The current-voltage characteristics of SCT thin films showed the increasing leakage current as the substrate temperature increases.
