CRM1 inhibitor S109 suppresses cell proliferation and induces cell cycle arrest in renal cancer cells

  • Abnormal localization of tumor suppressor proteins is a common feature of renal cancer. Nuclear export of these tumor suppressor proteins is mediated by chromosome region maintenance-1 (CRM1). Here, we investigated the antitumor effects of a novel reversible inhibitor of CRM1 on renal cancer cells. We found that S109 inhibits the CRM1-mediated nuclear export of RanBP1 and reduces protein levels of CRM1. Furthermore, the inhibitory effects of S109 on CRM1 is reversible. Our data demonstrated that S109 significantly inhibits proliferation and colony formation of renal cancer cells. Cell cycle assay showed that S109 induced G1-phase arrest, followed by the reduction of Cyclin D1 and increased expression of p53 and p21. We also found that S109 induces nuclear accumulation of tumor suppressor proteins, Foxo1 and p27. Most importantly, mutation of CRM1 at Cys528 position abolished the effects of S109. Taken together, our results indicate that CRM1 is a therapeutic target in renal cancer and the novel reversible CRM1 inhibitor S109 can act as a promising candidate for renal cancer therapy.

N,N'-Dimethylethylenediamine-N,N'-di-α-butyric Acid Cobalt(III) Complexes Utilizing Oxidation of Sulfur of S-Methyl-L-cysteine

  • The Reaction of S-methyl-S-cysteine(L-Smc) with racemic $s-cis-[Co(demba)Cl_2]-1$ (Hydmedba = $NN'-dimethylethylenediamine-NN'-di-\alpha-butyric$, acid) yields ${\Delta}$-s-cis-[Co(dmedba)(L-Smc)] 2 with N, O-chelation. Oxidation of sulfur of 2 with $H_2O_2$ in a 1 : 1 mole ratio gives ${\Delta}$-s-cis[Co(dmedba)(L-S(O)mc)] 3 having an uncoordinated sulfenate group. Oxidation of sulfur of L-Sm with $H_2O_2in$ a 1: 1 mole ratio produces S-methyl-L-cysteinesulfenate (L-S(O)me) 5. Direct reaction of 1 with 5 in basic medium gives an N.O-chelated ${\Delta}$s-cis[Co(dmedba)(L-S(O)mc)-N.O], which turmed out be same as obtained by oxidation of 2, while an N, S-chelated ${\Delta}$-s-cis-[Co(dmedba)(S-S(O)mc)-N,O] complex 4 is obtained in acidic medium from the reaction of 1 with 5. This is one of the rare $[$Co^{III}$(N_2O_2-type$ ligand)(amino acid)] type complex preparations, where the reaction conditions determine which mode of N, O and N, S caelation modes is favored.

축산폐수에서 분리된 질소 산화균에 의한 축산 폐수의 질산화

  • 축산폐수에서 분리된 세균 S. sp1(S. capitis)와 S. sp2(3. xylosus)를 이용한 축산폐수 중 질소화합물 분해정도를 파악한 결과 1) 증식에 알맞은 조건은 LB broth, $27^{\circ}C$, pH7로 나타났다. 2) S. sp1은 배양 7일 초기 농도의 90.3%, 7일째 32.9%로 감소되어 14일까지 다소 증가하면서 유지되었다. 3) S. sp1과 S. sp2를 혼합배양한 결과 S. sp1과 S. sp2 단독배양보다 분해 효율이 높고 기간도 2일정도 단축되었다.

Volatile Flavor Components in the Mashes of Takju Prepared Using Different Yeasts (효모종류를 달리한 탁주 술덧의 휘발성 향기성분)

  • In this study, we examined the volatile flavor components in the mashes of takju prepared using different yeasts such as Saccharomyces coreanus, S. ellipsoideus, S. carlsbergensis, S. cerevisiae (Baker's yeast), and S, rouxii by GC and GC-MS. Fourteen alcohols, 13 esters, 5 acids, 3 aldehydes, 7 amines, and 2 other compounds were identified in the mash after 6 days of fermentation. On day 6, the takju fermented by S. coreanus had the greatest variety of volatile flavor components. Fifteen flavor components, including ethanol, isobutyl alcohol, isoamyl alcohol, methyl pentanol, 1,3-butanediol, 3-methylthio-1-propanol, benzeneethanol, ethyl lactate, acetic acid, acetaldehyde, and 1,3-cyclohexane diamine, were typically detected in all the treatments. The relative peak areas of the volatile components were as follows: alcohols (96.758-99.387%), esters (0.081-0.968%), acids (0.040-0.640%), aldehydes (0.266-0.959%), and amines (0.011-0.047%). In particular, 1-propanol, isobutyl alcohol, 3-methyl-1-butanol, 2,3-butanediol, trimethyl benzylalcohol, heptene-2,4-diol, ethyl lactate, diethyl succinate, ethyl nonanoate, methyl hexadecanoate, linoleic acid, hexadecanoic acid, and acetaldehyde were hish in the takju made by S. coreanus. Also, ethyl stearate was high in the takju made by S. carlsbergensis, and hexanoic acid was high in the takju made by S. cerevisiae. Finally, methyl pentanol, 1,3-butanediol, 3-methylthio-1-propanol, benzene ethanol, ethyl octadecanoate, acetic acid, pentanal, and 1,3-cyclohexane diamine were high in the takju made by S. rouxii.

Secretory Expression of Human $\alpha_{s1}$-Casein in Saccharomyces cerevisiae

  • A recombinant human $\alpha_{s1}$-casein was expressed as a secretory product in the yeast Saccharomyces cerevisiae. Three different leader sequences derived from the mating factor $\alpha$l (MF$\alpha$l), inulinase, and human $\alpha_{s1}$-casein were used to direct the secretion of human $\alpha_{s1}$-casein into the extracellular medium. Among the three leader sequences tested, the native leader sequence of human $\alpha_{s1}$-casein was found to be the most efficient in the secretory expression of human $\alpha_{s1}$-casein, which implies that the native leader sequence of human $\alpha_{s1}$-casein might be used very efficiently for the secretory production of other heterologous proteins in yeast. The recombinant human $\alpha_{s1}$-casein was proteolytically cleaved as the culture proceeded. Therefore, an attempt was made to produce human $\alpha_{s1}$-casein using a S. cerevisiae mutant in which the YAP3 gene encoding yeast aspartic protease 3 (YAP3) was disrupted. After 72 h of culture, most of the human $\alpha_{s1}$-casein secreted by the wild type was cleaved, whereas more than 70% of the human $\alpha_{s1}$-casein secreted by yap3-disruptant remained intact. The results suggest that YAP3 might be involved in the internal cleavage of human $\alpha_{s1}$-casein expressed in yeast

Effect of Nutrient Solution Strength on Growth of Phalaenopsis in an Ebb and Flow System (Ebb and Flow 시스템을 이용한 호접란 수경재배시 생장에 미치는 배양액 농도의 영향)

  • This experiment was conducted to investigate the effect of nutrient solution strength on the growth of miniature multiflora P. amabilis and P. Taisuco Red Jewel at two growth stages during four months cultivation in an ebb and flow system. Early stage plants gained the biggest leaf length and width when nutrient solution strength was EC 0.5 or $1.0dS{\cdot}m^{-1}$ in P. amabilis, and EC $1.5dS{\cdot}m^{-1}$ in P. Taisuco Red Jewel. Root length and weight were the greatest when nutrient solution strength was EC $0.5dS{\cdot}m^{-1}$ in both cultivars and this trend was also found in middle stage plants. Fresh and dry weights of leaves increased as nutrient solution strength was elevated and were the greatest when nutrient solution strength was EC $1.5dS{\cdot}m^{-1}$ in P. amabilis. On the contrary elevation of nutrient solution strength decreased fresh weight of roots, being the greatest when nutrient solution strength was EC $0.5dS{\cdot}m^{-1}$. This trend was also found in P. Taisuco Red Jewel and P. amabilis at middle growth stage. The T/R ratio and chlorophyll content increased with nutrient solution strength and were the greatest regardless of cultivar and growth stage when nutrient strength was EC $1.5dS{\cdot}m^{-1}$.

The Study on the Composting by Using Dam Suspended Particle Sawdust and Sewage Sludge (댐부유물 톱밥과 하수슬러지를 이용한 퇴비화 연구)

  • This study was carried out on the composting of the most part of the plant waste materials inflowed-drifting into the dam during the localized heavy rain and the rainy season, due to the abnormal climate change, and for the sewage sludge banned to dispose legally into the ocean from the year of 2012. It was analysed the distinctive physicochemical qualities of the compost with treatment S-1(dam suspended particle sawdust : oak tree bark : sewage sludge : chicken manure = 30 : 20 : 40 : 10) and treatment S-2(dam suspended particle sawdust : oak tree bark : sewage sludge : chicken manure = 30 : 30 : 30 : 10). Both S-1 and S-2 maintained for 10 days at above $65^{\circ}C$ of the compost pile temperature, and the most of its pathogen were destroyed. In case of pH, until the 90th day into composting, S-1 with the pH value of 7.78 was slightly higher than S-2. The C/N value of S-1 was 15.3 and that of S-2 was 16.9. The quality of its final product was satisfied to the manufacture-standards. The GI value of S-1 was 91 higher than that of S-2, which was 84. In conclusion, it is highly recommendable to manufacture S-1 for its frequent usage of dam suspended particle sawdust and sewage sludge, and for its excellent quality and safety.

  • In this paper, we will show that the multiplicative group G in a finite ring R with identity 1 has a (B, N)-pair satisfying the following conditions; (1) G=BNB where B and N are subgroups of G. (2) B.cap.N is a normal subgroup of N and W = N/(B.cap.N), is generated by a set S = { $s_{1}$, $s_{2}$, .., $s_{k}$} where $s_{i}$.mem.N/(B.cap.N), $s_{i}$$^{2}$.iden.1 and $s_{i}$.neq.1. (3) For any s.mem.S and w.mem.W, we have sBw.contnd.BwB.cup.BswB. (4) We have sBs not .subeq. B for any s.mem.S. When G, B, N and S satisfy the above conditions, we say that the quadruple (G, B, N, S) is a Tits system. The group W is called the Weyl gorup of the Tits system.ystem.m.
