Effect of Monensin and Fish Oil Supplementation on Biohydrogenation and CLA Production by Rumen Bacteria In vitro When Incubated with Safflower Oil

  • An in vitro study was conducted to examine the effect of monensin or fish oil addition on bio-hydrogenation of $C_{18^-} unsaturated fatty acids and CLA production by mixed ruminal bacteria when incubated with safflower oil. Commercially manufactured concentrate (1%, w/v) with safflower oil (0.2%, w/v) were added to mixed solution (600 ml) of strained rumen fluid and McDougalls artificial saliva (control). Monensin $Rumensin^{(R)}$, 10 ppm, w/v, MO), mixed fish oil (0.02%, w/v, absorbed to 0.2 g alfalfa hay, FO) or similar amounts of monensin and fish oil (MO+FO) to MO and FO was also added into the control solution. All the culture solutions prepared were incubated in the culture jar anaerobically at $39^{\circ}C$ up to 12 h. Higher pH (p<0.047) and ammonia concentration (p<0.042) were observed from the culture solution containing MO at 12 h incubation than those from the culture solutions of control or FO. The MO supplementation increased (p<0.0001-0.007) propionate proportion of culture solution but reduced butyrate proportion at 6 h (p<0.018) and 12 h (p<0.001) of incubations. Supplementation of MO or MO+FO increased (p<0.001) the proportions of $C_{18:2}$. The MO alone reduced (p<0.022-0.025) the proportion of c9,t11-CLA compared to FO in all incubation times. The FO supplementation increased the proportion of c9,t11-CLA. An additive effect of MO to FO in the production of c9,t11-CLA was observed at 6 h incubation. In vitro supplementation of monensin reduced hydrogenation of $C_{18^-}$UFAs while fish oil supplementation increased the production of CLA.

Cottonseed Extract Improves the Function of Osteoblastic MC3T3-E1 Cells

  • We have investigated the effects of cottonseed extract on the proliferation, differentiation and lipopolysaccharide (LPS)-induced production of local factors in murine clonal osteoblastic MC3T3-E1 cells. Ethanol extract of cotton seed ($4{\sim}63{\mu}g/mL$) significantly increased the proliferatin of MC3T3-E1 cells (p<0.05). Moreover, cottonseed extract ($10{\sim}50{\mu}g/mL$) caused a significant elevation of alkaline phosphatase (ALP) activity and collagen content in the cells. Lipopolysaccharide (LPS) is a potent stimulator of bone resorption in inflammatory diseases. We examined the effect of cottonseed extract on the LPS-induced production of tumor necrosis factor a (TNF-$\alpha$) and nitric oxide (NO) in MC3T3-E1 cells. Treatment with cottonseed extract ($10{\sim}50{\mu}g/mL$) decreased the $5{\mu}g/mL$ LPS-induced production of TNF-$\alpha$ and NO in osteoblasts, suggesting that the antiresorptive action of cottonseed extract may be mediated by decrease in these local factors. This study suggests that cottenseed may contribute to antiresorptive action against osteoblastic cells, resulting in a beneficial effect in promoting the function of osteoblastic cells.

The Effect Nursing Organizational Culture and Happiness Index on Turnover Intention among Nurses (간호사가 지각한 간호조직문화와 행복지수가 이직의도에 미치는 영향)

  • The purpose of this study was to identify the effect nursing organizational culture and happiness index on turnover intention among nurses. The subjects of this study were 377 nurses who were working at 3 general hospitals in B city. The data were collected by structured questionnaire from July 1 to August 28 of 2013. The data collected were analyzed with SPSS Win 20.0 using descriptive methods, t-test, ANOVA, Pearson correlation coefficient and Stepwise multiple regression. Turnover intention were significant negative correlation for affiliative oriented organizational culture(r=-.137, p=.008), happiness index(r=-.290, p<.001). There were significant positive correlation for innovative oriented organizational culture(r=.123, p=.017), rank oriented organizational culture(r=.126, p=.015), task oriented organizational culture(r=.218, p<.001). Factors affecting for turnover intention were happiness index(${\beta}$=-.297, p<.001), rank oriented organizational culture(${\beta}$=.266, p<.001), nursing experience(${\beta}$=.199, p=.009), affiliative oriented organizational culture(${\beta}$=-.142, p=.034). The explained variances for were turnover intention among nurses 17.2%. Based on the study consider to development and education program of happiness index and affiliative oriented organizational culture for nurses in the hospital setting.

The Effects of Nursing Organizational Culture, Work Performance and Workplace Bullying Type on Workplace Bullying Consequence of Nurses (간호조직문화, 업무수행능력, 직장 내 괴롭힘 유형이 간호사의 직장 내 괴롭힘 결과에 미치는 영향)

  • Purpose: The purpose of this study was to investigate the relationship among nursing organizational culture, work performance, and workplace bullying of nurses, and to identify the factors that affect workplace bullying. Methods: The design of this study was descriptive research, and the data were collected from October 26 to November 5, 2018. The research participants were 210 nurses in six general hospitals in G province. The data were analyzed by descriptive statistics, a t-test, one-way ANOVA, Scheff? post-hoc test, Pearson's correlation coefficient, and a stepwise multiple regression using SPSS 22.0. Results: The workplace bullying type showed negative correlations with innovation-oriented culture, relation-oriented culture, and work performance. The workplace bullying consequence showed negative correlations with the innovation-oriented culture, relation-oriented culture, and work performance, and showed positive correlations with the workplace bullying type. The factors influencing the results of the workplace bullying consequence were workplace bullying type followed by relation-oriented culture, and unmarried status. Conclusion: Workplace bullying among nurses was related to the nursing organizational culture and work performance. Therefore, in order to reduce the turnover rate of nurses and to help them adapt well to the nursing organizational culture, a bullying prevention and intervention program is necessary.

Teenager's Hiphop Fashion is subcultures, the fashion culture of teenager reflects their unique identity. 2. The formative characters of Rap Music and Hiphop Fashion are the incompatibility ignored their body size and the popularity caused street fashion imitation, the unbalance ignored a principle of fashion design. 3. Hiphop Fashion is consist of jump-style jacket, hood -shirt, stripe T-shirt and drum pants, baggy pants, snowboard pants and so on. 4. The characters of Hiphop Fashion is the breaking of rule and the destruction. 5. They select something which they like, even though the price is high, so they is tendency to editing coordinate 6. Fashion business for teenager, it needs an ideal conversion not for 'company' but for 'individual' As this results, fashion culture of teenager came out to the surface of water, because of Hiphop Fashion.

Addition Effect of Seed-associated or Free Linseed Oil on the Formation of cis-9, trans-11 Conjugated Linoleic Acid and Octadecenoic Acid by Ruminal Bacteria In Vitro

  • The effects of seed-associated or free linseed oil on fermentation characteristics and long-chain unsaturated fatty acids composition, especially the formation of conjugated linoleic acid (CLA) and octadecenoic acid (trans-11 $C_{18:1}$, $t-C_{18:1}$) by mixed ruminal bacteria were examined in vitro. Concentrate (1% of culture solution, w/v, as-fed basis) with ground linseed (0.6% of culture solution, w/v, DM basis) or linseed oil as absorbed onto ground alfalfa hay was added to 600 ml mixed solution consisting of strained rumen fluid and artificial saliva at the ratio of 1:1 in a glass culture jar. The culture jar was covered with a glass lid with stirrer, and placed into a water-bath ($39^{\circ}C$) and incubated anaerobically up to 24 h. Seed-associated or free linseed oil did not significantly affect the pH and ammonia concentration in the culture solution. Molar percent of acetate tended to increase while that of propionate decreased with the addition of free oil treatment throughout the incubation. Differences in bacterial number were relatively small, regardless of the form of supplements. Decreasing trends in the compositions of linoleic acid ($C_{18:2}$) and linolenic acid ($C_{18:3}$) but increasing trends of stearic acid ($C_{18:0}$), $t-C_{18:1}$ and CLA compositions were found from culture contents up to 12h incubation when incubated with both ground linseed and linseed oil. The compositions of $C_{18:0}$, $C_{18:2}$ and $C_{18:3}$ were greater but those of oleic acid ($C_{18:1}$), $t-C_{18:1}$ and CLA were smaller in a culture solution containing ground linseed than those containing linseed oil. The ratio of $t-C_{18:1}$ to CLA was lower in the culture solutions containing linseed oil up to 12h incubations as compared to those containing ground linseed.

  • With suffering financial crisis in 1997, Korea's fashion market has grown in the direction that satisfies the reasonable price and diverse consumer's sensitivity and needs. The aim of this study is to examine whether the cutting portion in the waist line is surely necessary for designing the jacket pattern in men's wear, by closely examining the effects of the cutting portion in the waist line on the external appearance and functionality given designing the jacket pattern in men's wear for the standard body type in 20s. The results of the study are as follows. As a result of evaluating the external appearance, when having evaluated by dividing into 14 items such as the whole surface, the side, and the back side, the experimental clothing, which wasn't cut the waist line, obtained the highest mark in 13 items and, the significant difference was indicated in 10 items except 4 items. As a result of evaluating performance, the experimental clothing, which wasn't cut the waist line, obtained the highest mark in 13 items among 25 items in the total. Next, the cutting portion in the waist line obtained the highest mark in 10 items for the experimental clothing with opening in 1.0cm. The next was indicated to be in order of the experimental clothing with 1.5cm and the experimental clothing with 0.5cm. Given seeing the above result, the cutting portion in the waist line given designing the jacket pattern for the standard body type in 20s could be known that the experimental clothing with the most excellent mark in the evaluation of performance was all the experimental clothing with 0.0cm whose waist line wasn't cut.

Lipid A of Salmonella typhimurium Suppressed T-cell Mitogen-Induced Proliferation of Murine spleen Cells in the Presence of Macrophage (Salmonella typhimurium lipid A를 처리한 식세포 존재 조건에서 mitogen에 유도되는 이자 세포의 증식억제)

  • Infection with virulent or attenuated Salmonella typhimuriumhas known to induce reduction in proliferative responses of spleen cells. We investigated a role of lipid A from S. typhimurium, a B cell mitogen, on proliferation of spleen cells by T cell mitogens such as concanavaline A and phytohemagglutinin under in vitro and ex vivo conditions. Lipid A alone induced proliferation of spleen cells in vitroin a dose-dependent manner. However, subsequent treatment of concanavaline A or phytohemagglutin in after lipid A treatment induced proliferation suppression of murine spleen cells in vitro and ex vivo. Removal of macrophages from spleen cells, which were obtained from a lipid A-injected mouse, restored proliferation by concanavaline A and phytohemagglutinin, indicating that macrophages appeared to play a role in lipid A-induced suppression. Secreted molecules from macrophages did not accounted for the suppression because suppressive effect was not achieved when the supernatant from macrophage-containing spleen cell culture was conditoned to macrophage-depleted spleen cell culture. Co-culture of spleen cells from lipid A-treated and - untreated mice showed proliferation suppression as increasing cell numbers of lipid A-treated mouse. These data suggested that the cell-to-cell contact of macrophage with splenic lymphocyte cells is responsible for immune responses against lipid A, which is applicable to the case of human S. typhi infection.

THE INHIBITORY EFFECT OF FRUCTAN-PRODUCING S. SALIVARIUS ON THE FORMATION OF ARTIFICIAL PLAQUE (Fructan 생성 S. salivarius의 인공치태 억제효과)

  • S. mutans is the most important causative bacteria of dental caries among the oral bacteria. S. salivarius is a normal inhabitant in the human oral cavity. Nine strains of S. salivarius in this study were isolated from the oral cavities of children and identified, and their effect on S. mutans and S. oralis was studied. 1. The mean weight of produced artificial plaque on the wires in the beaker was 204.9 mg in the culture of S. mutans only, whereas being reduced to 1.9 mg through 20.6mg in the combined culture of S. mutans and each S. salivarius isolate (p<0.05). The viable cell didn't show the difference between them after culturing. 2. When S. mutans was cultured in the media containing culture supernatant of each S. salivarius isolate in M17 broth, the mean weight of produced artificial plaque was 117.1 mg on the wires, whereas being 47.7 mg in the media containing culture supernatant of each S. salivarius isolate in M17 broth containing 5% sucrose. 3. The polymer produced by S. salivarius isolates was on the thin layer chromatography. 4. Inulin and levan didn't inhibit the formation of artificial plaque by S. mutans in the beaker test. These results suggested that fructan-producing S. salivarius isolates inhibited the formation of artificial plaque by S. mutans.

Effects of Combined Micronutrient(Fe, Mn, Cu, Zn, Mo and B) Application on Forage Traits in Pure and Mixed Swards of Orchardgrass and White Clover IV.   • This pot experiment was conducted to investigate the effects of combined micronutrient application($T_1$;control, $T_2$; Fe, $T_3$; Fe+Mn, $T_4$: Fe+Mn+Cu, $T_5$ ; Fe+Mn+Cu+Zn, $T_6$ ; Fe+Mn+Cu+Zn+Mo, $T_7$ ;Fe+Mn+Cu+Zn+Mo+B) on forage performance of pure and mixed cultures of orchardgrass and white clover. This 4th part was related to the changes in the contents of micronutrients(Fe, Mn, Cu, Zn, Mo, and B) in forages. The results obtained are summarized as follows: 1. General differences have been showed in the contents of micronutrients based on the treatments, forage species, pure/mixed culture, cutting order, and additional fertilization, especially N. Compared to pure culture, orchardgrass showed relatively high contents of Mn and Zn, and low contents of B and Fe in mixed culture. White clover, however, tended to be exactly opposed to the above trends. The contents of Cu and Mo did not show any differences between pure and mixed cultures. 2. In relative comparison, the $T_7$ influenced negatively on the contents of Cu, Zn, and Mo in orchardgrass. The $T_7$ also influenced negatively on the contents of Mo in white clover. However, the $T_7$ influenced positively on the contents of Mn in orchardgrass, and also influenced positively on the contents of Fe, Mn, and Cu in white clover. Because of the antagonism between Fe and Mn, the Fe contents in both forages were significantly decreased by the $T_3$. Under the various conditions, the differences among Fe contents tended to be more significant in white clover than in orchardgrass. 3. At the $T_6$ and $T_7$, the Mo contents in both forages tended to be relatively high. The Mo contents, however, were somewhat decreased by the $T_7$ 7/. The Mo-toxicity, which was caused by the high Mo-contents, tended to be diminished, and was likely to be prevented by the optimum B/Mo ratio and B application($T_7$ ).
