Effect of Ripe Fruits of Rubus coreanus on Anaphylactic Allergic Reaction and Production of Cytokines

  • The anaphylactic allergic reaction is involved in many allergic diseases such as asthma and allergic rhinitis. In this report, I investigated the effect of ripe fruits of Rubus coreanus Miq. (Rosaceae) (RFRC) on the anaphylactic allergic reaction and studied its possible mechanisms of action. RFRC inhibited compound 48/80-induced systemic anaphylactic reaction in mice. RFRC dose-dependently decreased the IgE-mediated passive cutaneous anaphylaxis. In addition, RFRC decreased the gene expression and production of tumor necrosis factor-$\alpha$ (TNF-$\alpha$) and interleukin (IL)-6 in phorbol 12-myristate 13-acetate (PMA) plus calcium ionophore A23187 (A23187)-stimulated human mast cells. These findings provide evidence that RFRC could be a candidate as an anti-allergic agent.

Rubus속 식물에 존재하는 $19{\alpha}-Hydroxyursane-type$ Triterpenoid의 특성과 이용 (Chemical Characterization and Utilization of $19{\alpha}-Hydroxyursane-type$ Triterpenoids in Rubus species)

  • The plant Rubus species (Rosaceae) mainly contains $19{\alpha}-hydroxyurane-type$ triterpenoids $(19{\alpha}-HUT)$ as bioactive components. Available functional food includes blackberry (the fruit of thornless Rubus sp.), red raspberry (R. idaeus) and black raspberry (R. occidentalis). However, the fruit of R. coreanus, which is used in Korea as a functional food, substitutes black raspberry. Rubi Fructus, which has been traditionally used as an oriental medicinal drug, designates only unripe fruit of R. coreanus but not its ripe fruit which indicates that it needs high content of $19{\alpha}-HUT$ as a crude drug. Throughout our experiment, we found that ripe fruits contain very little amount of $19{\alpha}-HUT$ when compared to unripe fruits. In addition, various and rich $19{\alpha}-HUT$ has been reported from Rubus species. The most common structure of $19{\alpha}-HUT$ of Rubus species, euscaphic acid or tormentic acid with $3{\alpha}-OH$ or $3{\beta}-OH$, respectively, can be glycosylated or oxidized to produce a number of $19{\alpha}-HUTs $as euscaphic acid and tormentic acid derivatives and even esterified to form dimeric triterpenoids. In this review, the bioactivity and biosynthetic pathway and chemical characterization of $19{\alpha}-HUTs$ found in Rubus species are discussed.

Quantitative Determination of the Triterpenoids and Total Tannin in Korean Rubus species by HPLC

  • The triterpenoids contained in four Rubus species (Rosaceae) were quantitatively analyzed using HPLC to select plants with large quantities of niga-ichigoside $F_1$. Unripe fruits, ripe fruits, and leaves were extracted to estimate the quantity of niga-ichigoside $F_1$, together with Rubus-specific $19{\alpha}$-hydroxyursane-type triterpenoids, euscaphic acid, tormentic acid, and kaji-ichigoside $F_1$, and a dimeric triterpenoid coreanoside $F_1$. Niga-ichigoside $F_1$ was most abundant in the leaves of R. crataegifolius (23.4 mg/g dry weight). The amount of triterpenoid in the black, ripe fruits of R. coreanus was lower than the unripe fruits of the same plant. On the other hand, the ripe fruits of three plants, R. crataegifolius, R. parvifolius and R. pungens var. oldhami, which are reddish, contained higher or similar level of triterpenoids than their unripe fruits. In addition, the concentration of niga-ichigoside $F_1$ in the ripe fruit of R. crataegifolius was 20.5 mg/g, suggesting that the fruits could be used as a functional food. Methyl gallate and ellagic acid were used as quantitative indices of total tannin. Methyl gallate levels were higher in ripe fruits than unripe fruits in R. crataegifolius, R. pungens var. oldhami, and R. parvifolius. In R. crataegifolius, the quantity of methyl gallate was 30.5 mg/g in ripe fruit, but 1.19 mg/g in unripe fruit.

복분자 딸기 추출물의 효능에 관한 연구 (The Study on the Effect of Rubus coreanus Miquel Extract as a Comsetic Ingredient)

  • 통상적으로 식물의 열매, 잎, 뿌리 등의 추출물이 가지는 다양한 효능이 밝혀짐에 따라 점차 천연소재에 대한 관심이 증가하고 있는 추세이다. 본 연구에서는 복분자 딸기로부터 얻은 4종(미숙과, 완숙과, 잎/줄기, 뿌리의 에탄을 추출물)의 각기 다른 추출물에 대한 항산화, 미백, 항주름 효과를 비교하였다. 복분자 딸기의 항산화 및 항염 효과에 대한 몇몇의 보고가 있지만 효능에 대한 연구는 충분하지 않다. 따라서 복분자 추출물의 화장품적 효능 효과를 비교한 결과, 미백효과 및 주름개선효과에 있어, 미숙과부위의 추출물이 다른 부위의 추출물에 비하여 효과가 높은 것으로 관찰되었다. 이 결과를 바탕으로 복분자 딸기의 미숙과는 완숙과나 잎/줄기, 뿌리보다 화장품 원료로 더 높은 응용 가능성을 가지고 있음을 알 수 있었다.

대립 다수성 복분자딸기 "두륜" (Selection of New Variety of Rubus coreanus Miq., "Doo-Ryun", with Large Fruit and High Yield)

  • 전국 15개 지역에서 수집된 227 클론으로 조성된 클론보존원에서 선발을 통해 육성된 새로운 대립 다수확성 복분자 딸기 품종 '두륜'은 평균 개체와 비교하여 줄기의 가시가 적고, 잎의 상면 광택이 강하며 크기가 큰 검은색의 과실 특성을 갖는다. 1998년부터 개화 결실 특성 조사를 실시하여 2001년 17개 클론이 2차 선발되었으며 이들 17개 클론에 대한 지역적응성 및 안정성 검정을 통해 최종 육성된 신품종 '두륜'은 결실지당 개화 수에서 17.2개, 결실지당 과실 수에서 16.9개, 과실 입중 1.73 g, 본당 수확량에서 4.50 kg을 나타내 대조클론과 비교하여 각각 103.6%, 107.6%, 140.6%, 155.7% 의 우수한 선발효과를 나타내는 대립 다수확성 복분자 딸기 품종이다.

다공성 합성흡착제를 이용한 복분자 딸기(Rubus coreanus) 농축액의 항산화 효과 (Antioxidant Activity of Ripe Fruit Components of Rubus coreanus: Extraction Using Porous Polymer Resins)

  • 4종류의 다공성 합성흡착제(Sepabeads 207, 700, 850, Diaion HP 20)를 사용하여 Rubus coreanus 과실의 물 추출물로부터 과당 및 비polyphenol성분을 효과적으로 제거하여 DPPH라디칼 소거능 및 화학발광법을 이용한 superoxide 라디칼 소거능이 증가된 농축물을 효율적으로 얻을 수 있었다. 또한, 실시간 HPLC $ABTS^{{\cdot}+}$ 활성분석 시스템을 활용하여 6개의 항산화 능력을 갖는 활성피크를 확인할 수 있었으며 이들 대상으로 LC-MS/MS분석을 실시하여 5개의 anthocyanin인 cyanidin-3-sambubioside, cyanidin-3-glucoside, cyanidin-3-xylosylrutinoside, cyanidin-3-rutinoside, pelargonidin-3-rutinoside를 mass pattern 및 분자량 확인 과정을 통해 잠정적 화학구조 동정을 하였다. 수율과 항산화 효능을 고려해 볼 때, 4종의 합성흡착제 중에서 Diaion HP 20이 가장 우수한 결과를 나타내었다. 이처럼 적절한 합성흡착제의 선택으로 표적물질에 대한 polyphenol성 물질의 농축뿐만 아니라 생리학적 활성증대효과를 가져올 수 있어 적절한 합성흡착제의 선택은 식품 및 의약품의 정제 및 분리에 유용하게 활용될 수 있음이 확인되었다.

복분자 술이 가토의 testosterone에 미치는 영향 연구 (Effects of Black Raspberry Wine on Testosterone Level of New Zealand White Rabbits)

  • This study was conducted to investigate the effects of black raspberry wine, administered per os on testosterone levels in rabbits. Black raspberry wine containing 13% alcohol(v/v) was prepared from ripe fruits of Rubus coreanus fermented with Saccharomyces cervisiae. Experimental rats were divided into 2 groups(A and B) with 2 animals group: Group A received black raspberry wine. Group B received 13% concentrated alcohol('Soju') over a period of 6 weeks. all animals were monitored weekly for testosterone levels by radioimmunoassay using $^{125}I$ labeled anti-testosterone monospecific antibody. There was a significant(1.7-2.33 fold) increase in testosterone levels in Group A with a peak at 1 week post administration of black raspberry wine. There was no significant change in testosterone levels of rabbits in Group B. These data point to yet uncharacterized phytotestronegenic mechanisms of black raspberry wine that appear to be independent of ethyl alcohol. Further investigation of the mechanism of action of this fruit is warranted. These observations have potential implications for human reproductive health.
