신한약복합물 HT005의 성장기 흰쥐 장골길이 성장에 미치는 효과 (New Multi-herb Mixture, HT005, Induces Longitudinal Bone Growth in Male Rats)

  • Objectives : It was investigated new herbal prescription HT005 supported by traditional Korean medicine has a activity on the longitudinal bone growth of rats. HT005 were consisted of the root of Angelica sinensis, the fruit of Alpinia oxyphylla, the root of Astragalus membranaceus, the stem of Eleutherococcus senticosus, and the root of Dioscorea japonica, and Poria cocos. Method : To investigate the effect on longitudinal bone growth in adolescent male Sprague-Dawley rats, the longitudinal length of growth plate was measured directly by the tetracycline fluorescent labeling method and chondrocyte staining method. HT005 administered orally for four days, and the tetracycline was injected twice on the growth plate of the animals for fluorescent dying. The rate of longitudinal bone growth was measured the length between the tetracycline bands which fixed on the 3rd day and 5th day after injection. Cresyl violet was also used to stain the chondrocytes in the growth plate. The length of growth plate after administration was compared. Expression of IGF-1 and BMP-2 in the growth plate was investigated by immunohistochemistry. Results : HT005 showed a significant longitudinal bone growth which was $301.0{\pm}6.1\;{\mu}m/day$ at the dose of 100 mg/kg and $283.8{\pm}1.25\;{\mu}m/day$ (p < 0.001)at the dose of 10 mg/kg of HT005, compared to control group by the tetracycline fluorescent labeling method. HT005 showed a significant chondrocyte length on growth plate which was measured $797.19{\pm}3.31\;{\mu}m$ (p < 0.001) at the dose of 100 mg/kg and $720.14{\pm}2.19$ (p < 0.001) ${\mu}m$ at the dose of 10 mg/kg compared to the control group by cresyl-violet staining method. Both the number and intensity of BMP-2 and IGF-1 positive cells were increased in the hypertrophic zone of growth plate. There was a significant correlation between BMP-2 and IGF-1 expression and heights of chondrocytes in growth plate. Conclusions : Treatment of HT005 on Sprague-Dawley rats markedly increased the longitudinal bone growth. Therefore, HT005 may be an alternative herbal source to growth hormone as it promotes bone growth in children afflicted by growth retardation.

Growth retardation and differential regulation of expansin genes in chilling-stressed sweetpotato

  • We report here a first evaluation of chilling-responsive gene regulation in the sweetpotato. The growth of sweetpotato plants was severely retarded at $12^{\circ}C$; the lengths of the leaf, petiole, and root were markedly reduced and microscopic observation revealed that the elongation growth of the epidermal cells in each of these organs was significantly reduced. We examined the transcriptional regulation of three sweetpotato expansin genes (IbEXP1, IbEXP2 and IbEXPL1) in response to various chilling temperatures (12, 16, 22, and $28^{\circ}C$). In the leaf and petiole, the highest transcript levels were those of IbEXP1 at $28^{\circ}C$, whereas IbEXPL1 transcript levels were highest in the root. IbEXP1 mRNA levels in the $12^{\circ}C-treated$ petiole showed a fluctuating pattern (transient decrease-recovery-stable decrease) for 48 h. In the leaf and petiole, IbEXP1 and IbEXPL1 exhibited a similar response to chilling in that their mRNA levels decreased at $22^{\circ}C$, increased at $16^{\circ}C$, and decreased dramatically at $12^{\circ}C$. In contrast, mRNA levels of IbEXP2 in the leaf fell gradually as the temperature fell from 28 to $12^{\circ}C$, while they remained unaltered in the petiole. In the root, mRNA levels of IbEXPL1 and IbEXP1 reached maximum levels at $16^{\circ}C$, and decreased significantly at $12^{\circ}C$. These data demonstrated that expression of these three expansin genes was ultimately down-regulated at $12^{\circ}C$; however, transcriptional regulation of each expansin gene exhibited its own distinctive pattern in response to various chilling temperatures.

저산소 조건하에서 맥류 유묘의 근생장 및 혐기발효 효소의 반응 (Effects of Hypoxia on Root Growth and Anaerobic Fermentative Enzymes in Winter Cereal Seedlings)

  • 본 연구에서는 맥류 습해의 원인인 과습에 의한 근권의 산소부족이 맥류 뿌리의 생장 및 혐기 발효대사 효소(alcohol dehydrogenase, ADH; lactate dehydrogenase, LDH)에 미치는 영향을 조사하였다. 양액재배를 이용하여 1.5엽기 맥류 유묘에 용존산소 1.5-2 ppm 정도의 혐기처리를 1, 3, 5, 7일간 실시하였다. 혐기조건에서 뿌리의 생장은 모든 맥종에서 감소하였으며, 감소정도는 내습성이 낮은 보리가 내습성이 높은 밀과 호밀보다 낮았다. 정상조건에서 ADH 활성은 보리가 LDH 활성은 호밀이 가장 높았으나 두 효소 모두 맥종 간의 차이는 크지 않았다. 혐기처리에 의해 ADH와 LDH 활성은 모든 맥종에서 공통적으로 증가하였으며 증가 정도는 호밀과 밀에서 가장 높았고 보리에서 가장 낮았다. 혐기조건에서 이들 효소의 활성증가는 모든 맥종에서 유사하게 항시발현 동위효소의 증가 및 새로운 동위효소의 발현유도에 의해 나타났다. 혐기조건에서 ADH와 LDH 활성 증가 정도는 맥종의 재해저항성과 정의 상관관계가 인정되었다. 이러한 결과는 맥류의 내습성에 혐기 발효계 효소가 관여하고 있음을 의미하는 것으로 판단된다.

부갑상선 기능항진증 진단에서의 교정의사의 역할 : 치근흡수를 동반한 희귀증례 (The role of the orthodontist in diagnosis of hyperparathyroidism : rare case with general root resorption)

  • 부갑상선 호르몬이 과도하게 분비되는 경우 전반적인 골의 탈석회화에 의해 골막성 골흡수 및 골의 동통 등의 증상이 발현될 수 있다. 본고에서 소개된 환자의 경우 광범위한 치근 흡수를 동반한 부정 교합 환자로, 임상 및 두부 방사선 계측사진 분석에서 상악 열성장과 하악 과성장이 동반된 골격성 III급 부정 교합으로 진단되 었으며, 병리검사결과 부갑상선 호르몬 및 성장 호르몬의 수치가 증가된 것으로 보아 부갑상선 기능항진증으로 판단되었다. 방사선 사진 분석 결과 광범위한 치근의 흡수를 관찰할 수 있었으며 골다공증과 유사하게 골소주 형태가 매우 성기면서 ground glass 양상을 보였다. 이와 같이 부갑상선 기능항진증에 의해 골질뿐만 아니라 치근이 영향을 받은 것은 매우 드문 경우로 생각된다. 또한 부갑상선 기능항진증에 의해 내장두개의 골침착이 감소될 수 있다는 동물실험 결과를 볼 때 III급 부정교합의 원인이 상악 열성장인 점과 연관하여 매우 흥미로운 점으로 지적된다. 따라서 부갑상선 기능항진증의 다양한 증상 및 양태에 대한 조기진단 및 이에 대한 지식은 교정진단 및 치료에 중요한 역할을 할 수 있다고 생각된다.

  • 저자들은 저출생 체중과 호흡 곤란을 주소로 출생 직후 이송 된 여아에서, 진찰상 안면과 골격계 기형 소견 보여 말초 혈액에서 시행한 염색체 핵형 검사에서 trisomy 9, low level mosaic type으로 진단되었던 1례를 경험하였기에 보고하는 바이다.

Ultraviolet-activated peracetic acid treatment-enhanced Arabidopsis defense against Pseudomonas syringae pv. tomato DC3000

  • Disinfecting water containing pathogenic microbes is crucial to the food safety of fresh green agricultural products. The UV-activated peracetic acid (UV/PAA) treatment process is an efficient advanced oxidation process (AOP) and a versatile approach to disinfecting waterborne pathogens. However, its effects on plant growth remain largely unknown. This study found that low-dose UV/PAA treatment induced moderate oxidative stress but enhanced the innate immunity of Arabidopsis against Pseudomonas syringae pv. (Pst) DC3000. When applied as water sources, 5- and 10-ppm UV/PAA treatments slightly reduced biomass and root elongation in Arabidopsis seedlings grown under hydroponic conditions. Meanwhile, treatments of the same doses enhanced defense against Pst DC3000 infection in leaves. Accumulation of hydrogen peroxide and callose increased in UV/PAA-treated Arabidopsis samples, and during the post-infection period, UV/PAA-treated seedlings maintained vegetative growth, whereas untreated seedlings showed severe growth retardation. Regarding molecular aspects, priming-related defense marker genes were rapidly and markedly upregulated in UV/PAA-treated Arabidopsis samples. Conclusively, UV/PAA treatment is an efficient AOP for disinfecting water and protecting plants against secondary pathogenic attacks.

Inhibitory Activity of Sedum middendorffianum-Derived 4-Hydroxybenzoic Acid and Vanillic Acid on the Type III Secretion System of Pseudomonas syringae pv. tomato DC3000

  • The type III secretion system (T3SS) is a key virulence determinant in the infection process of Pseudomonas syringae pv. tomato DC3000 (Pst DC3000). Pathogen constructs a type III apparatus to translocate effector proteins into host cells, which have various roles in pathogenesis. 4-Hydroxybenozic acid and vanillic acid were identified from root extract of Sedum middendorffianum to have inhibitory effect on promoter activity of hrpA gene encoding the structural protein of the T3SS apparatus. The phenolic acids at 2.5 mM significantly suppressed the expression of hopP1, hrpA, and hrpL in the hrp/hrc gene cluster without growth retardation of Pst DC3000. Auto-agglutination of Pst DC3000 cells, which is induced by T3SS, was impaired by the treatment of 4-hydroxybenzoic acid and vanillic acid. Additionally, 2.5 mM of each two phenolic acids attenuated disease symptoms including chlorosis surrounding bacterial specks on tomato leaves. Our results suggest that 4-hydroxybenzoic acid and vanillic acid are potential anti-virulence agents suppressing T3SS of Pst DC3000 for the control of bacterial diseases.

재배토양의 수분 및 토성이 시호의 생육상황 및 항산화효소 활성에 미치는 영향 (Effect of Soil Moisture and Texture on Saikosaponins Content and Antioxidative Enzyme Activities in Bupleurum falcatum L.)

  • 토성 및 토양수분에 따른 시호근의 생육특성과 saikosaponin함량 및 항산화효소 활성 변화를 조사해 본 결과, 수분과다 토양에서 는 삼도시 호는 장수시호보다 생육 억제정도가 매우 높았고, 건물중은 과습, 적습, 과건 재배 순으로 높았고, 수분조건에 따른 토성별로는 과건은 식양토, 사양토, 사토 순으로, 적습과 과습시는 사토, 사양토, 식양토 순으로 높았다. 토양수분조건별 saikosapon a, d, c함량은 과건, 적습, 과습 순으로 높았고, 토성간에는 사토, 사양토, 식양토 순으로 높았다. POD 활성은 지상부는 장수시호가 삼도에 비하여 매우 높았고, 토양 수분함량간에는 장수는 과건, 과습, 적습 조건 순으로 높았으나, 삼도시 호는 과건, 적습, 과습 순으로 높았다. SOD활성은 지상부와 지하부 공히 장수시호가 높은 경향이나, 수분조건별로는 과건, 과습, 적습순으로 높았으며, 건조상태의 경우 두 품종 모두 사토에서 높았다. 시호에서 뿌리의 Saikosaponin함량과 POD 및 SOD 함량과는 정의상관을 나타내는 경향이었고 줄기 및 뿌리길이와 POD함량과는 부의 상관을 나타내었다.

  • Objectives : The process through which mesenchymal cells condense and differentiate into chondrocytes to form new bone is known as endochondral bone formation. Chondrogenic differentiation and hypertrophy are essential steps in bone formation and are influenced by various factors. The stem bark and root bark of Eleutherococcus sessiliflorus (ES) have been widely used to treat growth retardation and arthritis in traditional Korean Medicine. In this study, we aimed to investigate the possible role of the stem bark of ES in the stimulation of chondrogenic differentiation in clonal murine chondrogenic ATDC5 cells. Methods : In ATDC5 cells treated with ES extract, cell viability and extracellular matrix production were determined using CCK-8 assay and Alcian blue staining, respectively, and alkaline phosphatase activity was measured. We also examined mRNA and protein expression levels of genes related to chondrogenic expression in ATDC5 cells using reverse transcription-polymerase chain reaction and western blot analyses. Results : ES extract increased the accumulation of Alcian blue-stained cartilage nodules and alkaline phosphatase activity in ATDC5 cells. It increased the mRNA expressions of chondrogenic markers including bone sialoprotein (BSP), cartilage collagens, Runt-related transcription factor-2 (RUNX-2), osteocalcin (OCN), β-catenin, and bone morphogenetic protein-2 (BMP-2), as well as the protein expressions of β-catenin, RUNX-2, BMP-2, and alkaline phosphatase (ALP). Conclusion : Taken together, these results suggest that ES extract exhibits a chondromodulating activity and therefore may be a possible agent for the treatment of bone growth disorders.

목탄분말 시용이 고추의 생육 및 토양미생물상 변화에 미치는 영향 (Effects of Charcoal Powder on the Growth and Development of Red Pepper and the Changes of Soil Microflora)

  • 유기$.$자연농업 등 환경친화적 농업을 수행하는 농가에서 많이 사용하고 있는 목탄분말에 대한 특성파악과 작물생육, 토양화학성 및 토양미생물에 대한 영향을 검토하여 목탄분말의 효율적인 이용방안에 대한 기초자료를 얻고자 본 연구를 수행하였다. 목탄분말을 시용하고 고추를 재배한 결과 토양의 pH와 EC는 큰 변화가 없었으며, 대립목탄분말 처리시 뿌리생육이 저하되어 목탄분말 입경과 뿌리 발육은 밀접한 상관이 있는 것으로 나타났으며, 목탄분말의 입도가 작을수록(5mm 〈1mm 〈0.5mm) 고추의 수량이 증가되었다. 0.5∼1mm정도의 소립 목탄분말은 식물체의 양분흡수와 이용율을 향상시키나, 과다한 목탄분말 시용은 양분집적을 초래할 수 있기 때문에 고추에 대한 목탄분말의 적정시용량은 300kg 10a$^{-1}$ 정도인 것으로 판단되었다. 목탄분말 처리로 토양 중의 Rhizobium 속 질소고정균과 사상균 특히, 인 가용화 사상균수가 고추 생육시기에 따라 변화를 나타냈으며, 소립 목탄분말을 처리하였을 때 그 효과가 현저하였다.
