옻나무 추출액의 염색성 (A Study on the Chemical and Dyeing Properties of Rhus Verniciflua Extract)

  • This study was carried out to investigate K/S values, surface color, the fastness to washing, bacteria reduction rate of the silk and cotton fabrics dyed with Rhus verniciflua extract under the various dyeing conditions. The optimum dyeing temperature, dyeing time, dyeing pH and repetition of the silk fabrics dyed with Rhus verniciflua extract were $l00^{\circ}C, 30min., pH 5, five times repetition respectively, but in the cotton fabrics, it were $60^{\circ}C . 30min., pH 7, one times repetition. It were colored orangish Yellow in the silk fabrics and colored bright yellow in the cotton fabrics dyed with Rhus verniciflua extract. Surface color(munsell value) was not changed by the mordanting agents but those of the silk showed high tone when mordanting with stannous chloride, and it was decolored and darked when mordanting with ferric sulfate. The fastness to washing in the silk fabrics dyed with mordanting agents improved in 4~5 grade, but the cotton fabrics were 3~4 grade, so washing fastness of the silk and cotton fabrics were significantly improved when washing with the neutral detergent. The bacteria reduction rate of the silk fabrics increased drastically by dyeing of Rhus verniciflua extract.

  • In Korea, Rhus verniciflua Stokes is used to purge hardness, alleviate blood stasis, and treat cancer. However, the mechanisms of related anti-cancer activity are not fully understood in human cancer cells. This study investigated the anti-cancer effects and mechanisms of Rhus verniciflua Stokes on MDA-MB-231 human breast cancer cells and found that treatment with a Rhus verniciflua Stokes extract resulted in time- and concentration-responses that indicated growth inhibition of breast cancer cells by induced apoptosis. This was followed by a decrease in mitochondrial membrane potential; the activation of caspase-3, -8, and -9; and the up-regulation of tBid. Caspase-dependent apoptosis was induced through the inhibition of phosphatidylinositol 3-kinase (PI3K) and the Akt signaling pathway. This study provides evidence that Rhus verniciflua Stokes might be useful for the treatment of breast cancer.

ITS 및 rbcL 염기서열에 근거한 한국 자생 옻나무속의 계통분류 (Phylogeny of Korean Rhus spp. Based on ITS and rbcL Sequences)

  • 한국산 옻나무속 6종에 대하여 분자식물학적 방법으로 계통유연관계를 확인하기 위하여, nrDNA의 ITS 구간과 cpDNA rbcL 염기서열을 사용하여 계통분석한 결과 ITS 1의 길이는 $246{\sim}253\;bp$이었고, ITS 2는 $234{\sim}244\;bp$이었다. ITS 1의 길이는 Rhus sylvestris와 R. succedanea에서 246 bp로 가장 작았으며, R. verniciflua에서 253 bp로 가장 긴 것으로 나타났다. ITS 2의 길이는 R. verniciflua가 234 bp로 가장 짧았으며, R. trichocarpr가 244 bp로 가장 길게 나타났다. 이들 분류군의 G+C Content는 ITS 1에서는 $58.0{\sim}68.13%$의 범위를 나타냈고, ITS 2에서는 $59.75{\sim}68.46%$로 나타나 두 구간이 비슷한 비율을 보이고 있었다. ITS 1에서의 G+C content는 R. sylvestris가 58.0%로 가장 낮았으며, 가장 높은 값은 R. ambigua가 68.13%로 확인되었다. ITS 2에서는 외군인 Cotinus coggygria가 59.75%로 가장 낮았으며, R. ambigua가 68.46%로 가장 높게 나타났다. 한국산 옻나무 속에서 ITS 염기서열은 일반적으로 피자식물이 갖는 G+C content 범위 안에 포함되는 것으로 확인되었다. 한편, rbcL의 길이는 1,428 bp로 모든 종에서 동일하였다. 또한 rbcL의 G+C content는 $43.56%{\sim}43.77%$로 나타나 종간에 거의 차이가 없음을 확인하였다. 연구결과 rbcL gene은 옻나무속의 종간 계통유연관계를 해석하는데 유용하지 않았으며, ITS 1 구간의 염기서열 변이는 향후 옻나무속을 분류할 때 신속하게 분류할 수 있는 분류 marker로 이용할 수 있다고 판단되었다.

동유를 함유하는 목조주택 외장용 옻칠도료의 적용 (Application of Korean Rhus Lacquer Containing Tung Oil For Exterior Coatings)

  • 본 연구에서는 식물성 오일인 동유에 국내산 생칠 또는 정제칠을 10, 20, 30 및 40 wt.% 혼합하여 목조주택 외장용 도료를 제조하고 기존의 외장용 도료인 오일스테인과 내후성을 비교하여 외장용 도료로서의 가능성을 제시하려고 하였다. 생칠, 정제칠 및 동유를 혼합하여 제조된 도료의 성질에 대한 실험결과를 보면 생칠이 정제칠에 비해 화학구조 내에 존재하는 -OH group이 많으며 경화특성이 좋은 것으로 나타났다. 동적점도는 정제칠이 생칠에 비해 Frequency 150 rad/s 이하에서는 높은 것으로 나타났지만 150 rad/s 이상에서는 거의 비슷한 수준의 동적 점도를 보여 주었다. 또한 동유에 생칠을 혼합할수록 화학구조내의 -OH group이 증가하고 경화온도가 증가함을 보여 주었다. 내후성 실험결과는 제조된 도막의 색차변화가 기존에 목조주택 외장용 도료로 사용하는 오일스테인에 비해 높게 나타났지만 동유에 생칠을 40 wt.% 혼합한 경우와 정제칠을 30 wt.% 또는 40 wt.% 혼합한 경우 거의 비슷한 색차 변화를 보여 주었다. 색차를 제외한 광택도 변화, 접촉각 변화의 측정 결과는 오일 스테인의 변화정도와 유사한 결과를 보여 주어 목조주택 외장용 도료로써의 가능성을 보여 주었다.

식용유지에 대한 붉나무 추출물의 항산화 효과 (Antioxidant Activity of Ethanol Extract from Rhus javanica $Linn\'{e}$ on Edible Oil)

  • 천연 항산화 물질을 분리하고자 식용 또는 한약재로 이용되는 붉나무를 75% ethyl alcohol로 항산화 성분을 추출하여 chloroform, ethyl acetate 등의 용매를 사용하여 separatory funnel, column chromatography로 분획하여 얻은 분획물의 항산화 효과를 검토하였다. 붉나무 추출물중 항산화 효과를 나타내는 유효성분은 ethyl acetate와 chloroform에 잘 용해되는 물질로, ethyl acetate로 분리한 분획물을 팜유에 600 ppm 첨가시 AI는 1.60, 돈지에서는 AI가 3.90으로 이들 용매로 분리한 분획물의 항산화 효과는 붉나무 추출 원액보다 높은 현상을 보였다. 붉나무 추출물을 팜유, 돈지 및 대두유에 농도별로 첨가하여 $60^{\circ}C$에 저장하면서 POV, AV 및 TBA가를 측정하여 이들 유지에 대한 산화안정성을 시험한 결과 Rancimat test 결과와 비슷하여 팜유에는 항산화 효과가 뚜렷하였고, 돈지에는 탁월한 결과를 보였다.

화피, 석류피, 염부수백피 추출물의 항산화 및 항균 효과 (Antioxidative and Antimicrobial Activities of Water- and Ethanol-Extracts from Betula platyphylla var. japonica, Punica granatum and Rhus javanica)

  • Objectives : This study was conducted to measure the antioxidative and antimicrobial activities of water- and ethanol-extracts from Betula platyphylla var. japonica, Punica granatum and Rhus javanica against various species of anaerobic bacteria. Methods : In order to evaluate the antioxidative and antimicrobial activities of water- and ethanol-extracts, DPPH radical scavenging and superoxide dismutase (SOD)-like activities were measured in various species such as Staphylococcus epidermidis, Staphylococcus aureus, Escherichia coli, Pseudomonas aeruginosa, Candida albicans and Propionibacterium acnes that induce skin inflammation. Also the total amount of phenol in each water- and ethanol-extract was measured to identify its role in the antioxidative and antimicrobial activities of water- and ethanol-extracts from Betula platyphylla var. japonica, Punica granatum and Rhus javanica. The antibacterial activity of the extracts was measured by confirming the lowest concentrations in disk diffusion and minimum inhibitory concentration (MIC) tests. Results : It was observed that RJE (Rhus javanica-EtOH extracts) show the highest content of total phenol. In addition, for RJE the total phenol content was higher from samples taken from domestic sources than from samples taken from foreign sources. DPPH radical scavenging activity was increased by treatment with PGE (Punica granatum-EtOH extracts), RJE and RJW (Rhus javanica-water extracts). It was observed that SOD-like activity was highest in the treatment with PGE. All of the extracts showed antimicrobial activity on S. epidermidis, S. aureus, P. acnes and E. coli, including those from Punica granatum, and it was noted that the activity was higher with RJE than with RJW. Conclusions : These results provide evidence that ethanol extracts of Punica granatum and Rhus javanica may have a beneficial role as antioxidants and antibiotics. Extracts from domestic samples of Betula platyphylla var japonica appeared to have a greater efficacy than extracts from foreign samples.

옻나무 추출액의 음수투여가 육계의 생산성에 미치는 영향 (Effects of Drinking Rhus Tree-Extract on Performance of Broiler)

  • 본 연구는 육계 생산과정에서의 천연항생제 개발을 위해서 옻나무 추출액의 급여가 육계의 생산성에 미치는 영향에 대해서 조사하였다. 총320수의 1일령 육계에 옻나무 추출액 0 ppm (대조구), 500 ppm (T1), 1,000 ppm (T2) 및 2,000 ppm (T3)을 음용수와 함께 7주간 급여하였다. 증체중과 사료요구율은 35일령 까지 차이는 인정되지 않았지만, 그 이후는 옻나무추출액의 급여구가 개선되어지는 경향이 인정되었고 그 개선정도는 T3구에서 더 크게 나타났다. 맹장 내용물 및 분중의 대장균과 살모넬라균은 옻나무 추출액의 급이에 의해서 감소되었다. 사료중 건물, 조지방 및 조회분 소화율은 옻나무 추출액 급이에 의해서 개선되어지는 경향이 인정되었으며, 사료중 조단백질 소화율은 3개의 처리구 공히 대조구에 비해서 유의하게 개선되었다 (P<0.05). 이상의 결과를 종합해 보면, 증체량, 사료요구율, 맹장내용물과 분중의 대장균과 살모넬라균총의 감소 및 영양소 소화율은 옻나무 추출액의 음수투여가 효과적인 것으로 사료된다.

옻나무 전탕추출물 한방치료와 항암화학요법을 병용한 대장암 4기 환자 2례 (Two Cases of Stage IV Colorectal Cancer Patients by Combined Treatment of Rhus Vernifciflua Stokes Decoction and Chemotherapy)

  • Background & Objectives : Colorectal Cancer is 10% of all cancer incidence and the motality from colorectal cancer is nearly 450,000 a year. Since chemopreventive agents from Herbal medicine is hot issue recently, to prove antitumor effecicacy of Rhus vernifciflua STOKES decoction(Chijong-tang), clinical study was carried out. Here we report two patients with colorectal cancer of stage IV. One is colorectal cancer patient with liver, lung metastases underwent operation, chemotherapy. The other patient with liver metastasis underwent chemotherapy. All of two patients acquired tolerance. Methods : One patient visited Hana oriental medicine Clinic in 2010 and was treated using Chijong-tang for 17 months at Hana Oriental Clinic. The other patient visited Hana oriental medicine Clinic in 2011 and was treated using Chijong-tang for 5 months at Hana Oriental Clinic. Results : Chijong-tang showed no side effect, no more tolerance during its treatment with chemotherapy and tumor size was retarded based on CT scanning. Overall, this case report suggests that Rhus vernifciflua STOKES can be a potent cancer preventive agents for colorectal cancer but it is still required to verify the scientific and clinical evidences for Rhus vernifciflua STOKES decoction (Chijong-tang).

옻나무 추출 염액을 이용한 양모혼방직물의 염색성과 항균효과 (Dyeing Properties and Antibacterial Activity of Wool Blend Fabrics dyed with Rhus Verniciflua Extract)

  • The purpose of this study is to define optimal conditions for wool blend fabric dyeing using Rhus verniciflua extract. K/S value, surface color change, the fastness to light, fastness to dry-cleaning, metal mordanting effects and antimicrobial activities were investigated. The results were summarized as follows; As the storage time of Rhus verniciflua stokes dyeing solution was longer, K/S was improved. K/S value showed the highest one when dyeing temperature and time were at 100oC and 60 minutes respectively. As for the changes of pH values of the dyeing solution, fabric was dyed well with the pH value of acid or neutral. K/S value recorded the highest at pH6 and the lowest at pH10. Fastness to light showed the unmordanted fabric is 2.84, and Fe is 1.69 on color difference. Antibacterial activities of Rhus verniciflua dyeing solution recorded high figures in both dyed fabric with unmordanted and dyed fabric with mordanting.

  • Based on 11 Classics of Materia Medica designated by Ministry of health and welfare bulletin 1995-15 as legal basis in scope and preparation of herbal prescription, we reviewed indication, property and processing of Rhus Verniciflua Stokes which appear extensively in literature since earlier Joseon Dynasty. Following is the conclusion of this review after comparing with 'Korean Pharmacopoeia: commentary on herbal product standard'. 1. The properties of Rhus Verniciflua Stokes in 'Korean Pharmacopoeia :commentary on herbal product standard' should be changed as 'Tonifying the middle, restoring the muscle, fulfilling marrow, breaking old static blood, tonifying and activating after removing mass, unblocking the meridian, killing worms' and indication should be changed as 'Old static blood, deep rooted mass, wind-cold-dampness arthralgia, cough, 9 types of chest pain, abdominal or flank accumulation of stagnated qi[氣], amenorrhea, hernia mass, small bowel or bladder colic pain, abdominal pain due to worm accumulation'. 2. The processing of Rhus Verniciflua Stokes in 'Korean Pharmacopoeia: commentary on herbal product standard' should include 'natural drying or steaming drying followed by grinding and stir-baking until ripened or smoking appears'.
