Isolation and Elucidation of Specific RNAs by Treatment of Rhus verniciflua Stokes Extract to U937 Cell (옻추출물 처리에 의한 U937 세포에서의 특정 RNA 발현 양상)

  • Differential display RT-PCR was used for screening the differentially expressed specific genes by Rhus verniciflua extract treatment to U937 cell, human leukemic monocyte. As a result, 19 clones differentially expressed were detected. Among the detected clones, one clone was confirmed to be over-expressed by R. verniciflua extract treatment in Northern blot analysis. Nucleotide sequence of the clone showed 100% homology with H2A histone family member Z gene. Therefore, it is concluded that the treatment of R. verniciflua extract to U937 cell specifically induces the expression of H2A.Z gene but its role should be elucidated by future works.

  • It was thought that the stiffness of a coated specimen became lower than that of the solid wood because the stiffness of the dried film of coating material is lower than that of the solid wood. The authors were trying to compare the effect of lacquer varnish and rhus lacquer on the resonant frequency of the solid woods for the violin, spruce and maple. Vibration modal shape of coated specimens were same to those of solid woods, but the frequency became lower at each mode as were expected regardless of coat. frequency decrement of coated specimen was getting larger at upper mode in both European violin woods, however, rhus lacquer coated spruce specimens were less affected than lacquer varnish coated specimens.

The Bending and Compression Strength Properties in Rhus verniciflua(I) (한국산 옻나무의 휨 및 종압축 강도적 성질(I))

  • The bending and compression strength properties of two types Rhus verniciflua specimens, which made of no heat-treated wood and heat-treated wood for urushiol extraction, were measured. The heat-treated specimens were finger-jointed with either resorcinol-phenol or polyurethane resin adhesives, and the vertical type bending strength property was also measured in these specimens. The results obtained are as follows ; 1. The correlation coefficient between the compression strength and specific gravity in the specimens of no heat-treated and heat-treated wood was high. However there was no difference in compression strength property as affected by heat treatment. 2 The correlation coefficient between the bending strength and specific gravity in the specimens of no heat-treated and heat-treated wood was also high. However, there was no difference in bending strength property as affected by heat treatment. 3 The bending test showed high correlation between modulus of elasticity and modulus of rupture for the specimens made of no heat-treated and heat-treated wood. However, there was no difference in bending strength property between the specimens made of heat-treated and no heat-treated wood. 4. The efficiencies of bending strength test on the finger-jointed specimens of heat-treated wood with resorcinol-phenol and polyurethane resin adhesives were 0.85, 0.81. respectively.

Effects of Climatic Factors and Tapping Date on Yield and Quality of Lactree (Rhus verniciflua) Sap (기상요인과 채취시기가 옻나무 칠액채취량 및 칠액의 質에 미치는 영향)

  • This study was conducted to understand the effects of weather and tapping date on yield and quality of lactree(Rhus verniciflua) sap yield showed a significant positive correlation with the minimum temperature of one day before sap collection at 1% level and with theminimum humidity of theday of sap collection at 5% level. However, the differences between the maximum and the minimum temperatures and humidities of the day of sap collection were negatively correlated with the sap yield at 5% level. Multiple regression analysis indicated that the minimum temperature of one day bofore sap collection and the minumum humidity of the day sap collection were important factors for increasing sap yield. The high sap yield of lactree by Japanese tapping method was recorded during mid-July and early August. Seasonal variation in lactree sap constituents was observed. The sap collected on 15th of August contained the highest urushiol content (68.3%) and the lowest water content resulting in high quality of lactree sap. By reversed-phase HPLC analysis, fove urushiol components were separated from each other depending on the number of doulbe bonds in the side-chain , and seaxonal variation of urushiol composition was noticed. The 3-C15 triene content fo the sap collected on 5th of July was the highest(77.56%) indicating the major component of urushiol which affects lactree sap quality.

Effects of Fermented Rhus Verniciflua Stokes Extract on Diet-induced Hyperlipidemia in Rats (발효옻 추출물이 흰쥐의 식이성 고지혈증에 미치는 영향)

  • This study was designed to investigate the antihyperlipidemic activity of Fermented Rhus Verniciflua Stokes Extract(FRVSE) on diet-induced hyperlipidemia in rats. The animals were divided into 4 groups of the normal group(fed with normal diet), control group(fed with high fat diet), sample A group(fed with high fat diet and medicated FRVSE 192 ㎎/㎏/day), sample B group(fed with high fat diet and medicated FRVSE 384 ㎎/㎏/day), and received oral administration of each prescription with diet for a period of six weeks. The changes in the body weight, the liver weight, the epididymal fat weight, and the plasma levels of total cholesterol, triglyceride(TG), high density lipoprotein(HDL) cholesterol and low density lipoprotein(LDL) cholesterol were measured. In sample A group, the weight of liver and plasma level of TG decreased significantly. There was a significant decrease in the plasma level of total cholesterol, TG and LDL cholesterol in the sample B group. These results suggest that the FRVSE have a good antihyperlipidemic activity and a potential to treat hyperlipidemia.

Rhus Verniciflua Stokes Extract Suppresses Expression of Metalloproteinases, iNOS and COX-2 in THP-1 Cells Via Inhibiting NF-𝜅B and MAPK Phosphorylation

  • Objectives: The aim of this study is to investigate the mechanisms involved in the anti-inflammatory and anti-tumor effects of Rhus verniciflua Stokes (RVS) on PMA-differentiated human monocytic leukemia THP-1 cells. Methods: Cells were treated with various concentrations of RVS decoction (0-300㎍/ml) for 24, 48, and 72h. Cell viability was evaluated by MTS/PMS assay. The expressions of MMP-2, MMP-9, TIMP-1, TIMP-2, iNOS and COX-2 mRNA and proteins were measured using RT-PCR and western blotting, respectively. Results: RVS suppressed expression of MMP-2 and MMP-9 mRNA. It also down-regulated iNOS and COX-2 mRNA and protein expression. RVS inhibited NF-𝜅B p65 activity and the phosphorylation of Akt and MAPK (ERK and p38 MAPK). Instead, the phosphorylation of JNK is increased at a very low concentration but decreased at higher concentrations. Conclusion: RVS is regarded to inhibit the expression of MMP and TIMP as well as iNOS and COX-2 gene expression via directly inhibiting the activation of NF-𝜅B and phosphorylation of MAPK pathway in THP-1 cells. This suggests RVS have potential to be used as a therapeutic agent for acute myeloid leukemia (AML).

Antimicrobial Activity of Extracts from Medicinal Herbs Against Escherichia coli (Escherichia coli에 대한 한약재의 항균활성)

  • The antimicrobial activity of 10 different traditional medicinal herbs extracts against E. coli was determined using paper disc method. Caesalpinia sappan and Rhus javanica, extracts in 5 mg/ml, and Paeonia suffruticosa and Seutellaria baicalensis extracts in 10 mg/ml showed significant antimicrobial activity against E. coli. Minimum inhibitory concentrations of medicinal herbs extracts were in the range of $1.4{\sim}8mg/ml\;and\;1.2{\sim}12mg/ml$, for MeOH extracts and EtOH extracts, respectively. In addition, the antimicrobial activity of each solvent fraction was most significant with the EtOAc layer. Optical density at 620nm after 24 hours incubation of E. coli in the presence of 100, 300 or 500 ppm of Caesalpinia sappan extract ranged from 0.02 to 0.1 compared to 0.6 in the absence of Caesalpinia sappan extract, indicating that growth of E. coli was significantly inhibited within 24 hours by the addition of at least 300 ppm of Caesalpinia sappan extract. Optical density at 620 nm after 24 hours incubation of E. coli in the presence of 300 ppm of Rhus javanica extract ranged from 0.02 to 0.2 compared to 0.5 in the absence of Rhus javanica extract, indicating that growth of E. coli was also significantly inhibited within 24 hours by the addition of at least 500 ppm of Rhus javanica extract. In conclusion, these findings suggest that extracts from medicinal herbs may play important roles in antimicrobial activities against E. coli.

Bark Production Analysis on Top Branch of Rhus verniciflua (옻나무 우죽의 수피생산량 분석)

  • This study was conducted to investigate the bark production from a top branch(Woojuk) of lacquer tree(Rhus verniciflua) that collected Rhus lacquer in summer, analyzing a relationship between several factors - diameter, tree weight, weight of Woojuk and bark. The weight of Woojuk averaged was made up $48\%$ of the tree on average. As the weight of the tree increased, however, the Woojuk showed the rising trend. It was turned out that the tree size for making 1kg of dry bark must be at least DBB 8cm. The result on Woojuk's bark was that $43.3\%$ of total trees produced a green bark of 1.1kg to 1.5kg, and most of them put out 1.5kg and less. Also the rate of trees to make 0.6kg to 1.0kg of dry bark was highest in $50.8\%$, but its production in most trees was 1.0kg and below. As the moisture content of a green bark averaged $39\%$, it is estimated that the production of 1kg in dry bark requires at least 6kg of top branch in lacquer tree.

Changes of Volatile Organic Compounds of Rhus verniciflua S. Bark by Fermentation (발효에 의한 옻나무 수피의 휘발성 유기성분 변화)

  • To investigate effects of fermentation on volatile components, we analyzed volatile organic compounds of raw and fermented Rhus verniciflua S. bark. A 50%(w/v) sugar solution was used for fermentation. Volatile organic compounds of raw and fermented Rhus verniciflua S. were extracted by the simultaneous steam distillation and extraction(SDE) method, with a mixture of n-pentane and diethylether(1:1, v/v) and analyzed by gas chromatograph-mass spectrometer. A total of 51 and 27 volatile organic compounds were detected in raw and fermented samples, respectively, and were mainly alcohols. Compounds such as ethyl acetate, 2-methyl-3-buten-2-ol, 3-methylbutanal, 1-octen-3-o1, 3-methyl-2-butanone, hexanal and pentanal were detected as the primary compounds in the raw sample. The fermented sample showed sour different volatile compounds, such as ethanol, ethyl acetate, ethyl lactate and 3-methylbutanol. Thus, a number of volatile organic compounds were synthesized after fermentation of Rhus verniciflua S. bark.

Isolation and Identification of Antioxidative Components from Bark of Rhus javanica Linne (붉나무 수피로부터 항산화 물질의 분리 및 동정)

  • The crude extracts from Rhus javanica Linne showed comparatively strong antioxidative activity in test oils. Antioxidative components were isolated and identified by column chromatography, thin layer chromatography, UV, and NMR. These antioxidative components were added to several oils to compare antioxidative activity with several commercial antioxidants, such as BHA, BHT, and tocopherol. After the sixth column chromatography, one fraction (R-18-9-3-2-4-2) was separated from chloroform layer of Rhus javanica Linne. The R-18-9-3-2-4-2 fraction was identified as methyl gallate by $^1H-NMR$ and $^{13}C-NMR$ and confirmed with methyl gallate standard as authentic. The R18-9-3-2-4-2 fraction from chloroform layer of Rhus javanica Linne showed stronger activity than that of the ${\alpha}-,\;{\delta}-tocopherol$, BHT, and BHA at the same concentration.
