난소절제 동물모델을 이용한 폐경 후의 비만에 대한 건강 추출물의 효능에 대한 실험적 고찰 (The Experimental Study of the Beneficial Effect of Zingiberis Rhizoma on Post-menopausal Obesity Using Ovariectomized Rats)

  • Objectives: This study was performed to investigate anti-obesity effects of Zingiberis Rhizoma on ovariectomized rats in order to determine the possibility of the clinical use in preventing and treating post-menopausal obesity. Methods: To investigate how menopause affects obesity in woman, rats were treated with Zingiberis Rhizoma extracts. We measured various biomarkers including GOT,GPT, leptin, ghrelin, adiponectin, $PPAR-{\gamma}$ mRNA, total cholesterol, triglyceride, HDL-cholesterol, liver weight, estradiol, uterine weight, and calcitonin, which are linked with obesity and menopause. Results: There was a significant decrease in group which was given Zingiberis Rhizoma extracts 100 mg/kg and lipid level found in blood(total cholesterol, triglyceride). Fat accumulation of liver cells was repressed, liver function was improved and leptin and adipomectin levels were significantly normalized. In addition, expression of $PPAR-{\gamma}$ was significantly increased. Conclusions: The results indicated that Zingiberis Rhizoma extracts have anti-obesity effects on ovariectomized rats through improving liver function and lipid metabolic function.

대황(大黃)이 뇌허혈 손상에 의한 뇌부종에 미치는 영향 (Effect of Rhei Rhizoma on Brain Edema Induced by MCAO in Rats)

  • Brain edema is a major importance in the pathophysiology of CNS injuries including stroke. Ischemic brain edema results from both cytotoxic edema, which is severe in astrocytes at early stage, and vasogenic edema caused by excessive blood-brain barrier (BBB) permeability. The present study was performed to determine the effect of Rhei Rhizoma on brain edema induced by middle cerebral artery occlusion (MCAO) in the rats. The neurological symptom, total infarct volume and edema index caused by MCAO were measured. The changes of Matrix Metalloproteinase-9 (MMP-9) and inducible nitric oxide synthase (iNOS) immunoreactivities were also observed. We found that Rhei Rhizoma extract improved the neurological symptom and attenuated the total infarct volume and brain edema caused by ischemic insult. Rhei Rhizoma extract also attenuated the expression of MMP-9 and iNOS. This results suggest that Rhei Rhizoma has a protective effect on the brain edema caused by ischemic insult.

흡광도측정법에 의한 제제 중 황련 알칼로이드의 정량 (Determination of Coptidis Rhizoma Alkaloids in Preparations by Spectrophotometric Method)

  • The Coptidis rhizoma is known for containing protoberberine alkaloids. Berberine, coptisine and palmatine are the major constituents of protoberberine alkaloids. The alkaloids were isolated and determined by forming complex compounds from Coptidis rhizoma in preparation I(Sam-Hwang-Sa-Sim-Tang) and II(Hwang-Ryen-Tang). For the determination of these alkaloids, a new spectrophotometric method was developed with a simple and selective sample clean-up using thiocyanatocobaltate[II] complex compound ion. The absorbance of alkaloidal complex compounds in l.2-dichloroethane solution was measured at 625 nm. Calibration curve for the alkaloids isolated from Coptidis rhizoma was linear over the concentration range of 0.2-0.3 mg/ml. The method was proved to be rapid, simple and reliable for the isolation and the determination of the alkaloids in Coptidis rhizoma preparation I and II.

Effect of Rhizoma gastrodiae on glucose oxydase induced neurotoxicity in cultured mouse spinal dorsal root ganglion neurons

  • Effects of Rhizoma gastrodiae on glucose oxidase-induced neurotoxicity was investigated in cultured newborn mouse spinal dorsal root ganglion(DRG) neurons that were treated in the media with or without glucose oxidase. In addition, the protective effect of Rhizoma gastrodiae extract against glucose oxidase-induced neurotoxicity was examined. Cytotoxic values were expressed as a percentage of number of living cells by 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide (MTT) assay. In this paper, exposure of neurons to glucose oxidase resulted in a significant call death in a dose- and time-dependent manners in DRG neuron cultures. The decrease in cell viability induced by the glucose oxidase was blocked by Rhizoma gastrodiae extract. These results indicate that the neuroprotective effect of Rhizoma gastrodiae extract against glucose oxidase-induced neurotoxicity may result from a prevention or attenuation of oxidative damage induced by glucose oxidase.

수종의 전통약제가 일차 배양 간세포에서 $CCl_4$ 유발 세포독성에 미치는 영향 (Effects of Traditional Drugs on $CCl_4-induced$ Cytotoxicity in Primary Cultured Rat Hepatocytes)

  • 80% Methanol extracts of 44 traditional drugs used for the treatment of liver diseases or tonic effects were screened for anti-hepatotoxic activity by in vitro assay using $CCl_4-induced$ cytotoxicity in primary cultured rat hepatocytes. $CCl_4-induced$ cytotoxicity was evaluated by determination of LDH, GOT or GPT activity in the medium. Rehmaniae Radix Preparata and Gelantina nigra inhibited the release of LDH, GOT or GPT from $CCl_4-treated$ hepatocytes. Gibotii Rhizoma and Eucommiae Cortex showed inhibitory effect on release of LDH from normal hepatocytes as well as $CCl_4-treated$ hepatocytes. Eucommiae Cortex and Lili Bulbus decreased release of GOT and LDH from normal hepatocytes, respectively. Astragali Radix inhibited release of GPT in $CCl_4-treated$ hepatocytes. Phlomidis Radix, Imperatae Rhizoma, Cistanchis Herba, Broussonetiae Fructus, Asparagi Tuber, Trigonellae Semen and Polgonati Rhizoma inhibited release of LDH from $CCl_4-treated$ hepatocytes. Among 44 traditional drugs, most of them released LDH, GOT or GPT at the dose of 1 mg/ml in normal hepatocytes, and Drynariae Rhizoma, Acanthopanacis Cortex, Longanae Arillus, Atratylodis Rhizoma and Ecliptae Herba increased $CCl_4-induced$ cytotoxicity.

인진(茵蔯), 울금(鬱金), 상기생(桑寄生), 와송(瓦松)이 HepG2 cell의 혈관생성인자 발현에 미치는 영향 (The Effects of Artermisiae Capillaris Herba, Curcumae Rhizoma, Loranthi Ramulus, and Orostachys Herba on Expression of Angiogenic Factors in HepG2 Cells)

  • Objectives : This study was designed to investigate the effects of Artermisiae Capillaris Herba, Curcumae Rhizoma, Loranthi Ramulus, and Orostachys Herba on expression of angiogenic factors in HepG2 cells. Materials and Methods : The mRNA expression level and protein secretion level of angiogenic factors were measured using quantitative RT-PCR, western blot and ELISA assay respectively in Artermisiae Capillaris Herba, Curcumae Rhizoma, Loranthi Ramulus, Orostachys Herba -treated and untreated HepG2 cells. Results : Curcumae Rhizoma, Loranthi Ramulus, and Orostachys Herba reduced mRNA expression level and protein secretion level of angiogenic factors, but Artermisiae Capillaris Herba increased it, especially VEGF and bFGF in HepG2 cells. Conclusions : The results indicate that Artermisiae Capillaris Herbapromotes expression of angiogenic factors but Curcumae Rhizoma, Loranthi Ramulus, and Orostachys Herba inhibit expression of angiogenic factors in HepG2 cells. The result is expected that Curcumae Rhizoma, Loranthi Ramulus, Orostachys Herba have an inhibitive effect of angiogenesis in HCC.

골쇄보(骨碎補) 부탄올 추출물의 U937세포주 세포자멸사에 미치는 효과 (Effects on the Apoptosis of U937 cell line by n-butanol Extracts of Drynariae Rhizoma)

  • To investigate the possible mechanism of Drynariae Rhizoma extracts as a candidate of anti-cancer drug, I examined the effects of Drynariae Rhizoma extracts on the apoptosis of U937 cell line. MTT assay, flow cytometric analysis, SDS-polyacrylamide gel electrophoresis, Western blot analysis, and RT-PCR were performed. Drynariae Rhizoma extracts treatment reduced the cell viablilty of U937 cells in a dose-dependent manner, which was associated with induction of apoptotic cell death. Drynariae Rhizoma extracts treatment also reduced the levels of Bcl-xL anti-apoptotic protein expression and increased the levels of caspase-3, p53, pro-apoptotic protein, in U937 cells. RT-PCR data revealed that the level of bcl-2, bcl-xL mRNA expressions decreased in a dose-dependent manner. These findings suggest that Drynariae Rhizoma extracts may have induction of apoptotic cell death via regulation of several growth regulatory gene products. 감초 함유 처방의 글리치리진 대사와 몇가지 효소저해효과 (Metabolism of Glycyrrhizin in Polyprescriptions Containing Glycyrrhizae Radix by Human Intestinal Bacteria and Their Inhibitory Effects on Some Enzymes)

  • To analyze scientifically the prescription principle of polyprescriptions (Gamchotang, Daewhanggamchotang, Jakyakgamchotang, Gamchogungangtang and Gilkyungtang) containing Glycyrrhizae Radix, the transforming rate of glycyrrhizin in these polyprescriptions to 18 ${\beta}-glycyrrhetinic$ acid and their inhibitory effect on ${\beta}-glucuronidase$, hyaluronidase, phosphodiesterase and trypsin were investigated. When Glycyrrhizae Radix containing polyprescriptions were extracted with water, the contents of glycyrrhizin in water extract of Glycyrrhizae Radix with Rhei Rhizoma or with Zingiberis Rhizoma were higher than that of Glycyrrhizae Radix only, but that in water extract of Glycyrrhizae Radix with Platicodi Radix was lower than that of Glycyrrhizae Radix only. By human intestinal bacteria, glycyrrhizin was metabolized to 18 ${\beta}-glycyrrhetinic$ acid. These metabolism of glycyrrhizin in polyprescriptions containing Glycyrrhizae Radix was inhibited by Rhei Rhizoma, Paeoniae Radix and Platicodi Radix, but was not affected by Zingiberis Rhizoma. The inhibitory activity of Glycyrrhizae Radix on hyaluronidase and ${\beta}-glucuronidase$, was synergistic with Rhei Rhizoma or Zingiberis Rhizoma, but was antagonistic by Platicodi Radix.

  • Objectives : The purpose of this study was to compare the anticancer effects of Smilacis Chinae Rhizoma (SCR) and Smilacis Glabrae Rhizoma (SGR) on stomach cancer SNU-1 cells. Methods : The cell proliferation, apoptosis and cytokine level from the extracts were examined in order to compare the anticancer effects of SCR and SGR on SNU-1 cells. Results : SCR had greater anticancer effects than SGR in terms of the inhibition of cell proliferation and apoptosis in the SNU-1 cells. SCR decreased TGF-$\beta$ and SGR increased IL-2 in SNU-1 cells. SCR decreased more TGF-$\beta$ and increased more TNF-$\alpha$ compared to SGR. Conclusions : There were few significant differences according to the concentration and fraction, but a greater anticancer effect of SCR was shown as compared with SGR.

Quantitative and Pattern Recognition Analyses for the Quality Evaluation of Cimicifugae Rhizoma by HPLC

  • In this study, quantitative and pattern recognition analysis for the quality evaluation of Cimicifugae Rhizoma using HPLC/UV was developed. For quantitative analysis, three major bioactive phenolic compounds were determined. The separation conditions employed for HPLC/UV were optimized using ODS $C_{18}$ column ($250{\times}4.6mm$, $5{\mu}M$) with isocratic elution of acetonitrile and water with 0.1% phosphoric acid as the mobile phase at a flow rate of 1.0 mL/min and a detection wavelength of 323 nm. These methods were fully validated with respect to the linearity, accuracy, precision, recovery, and robustness. The HPLC/UV method was applied successfully to the quantification of three major compounds in the extract of Cimicifugae Rhizoma. The HPLC analytical method for pattern recognition analysis was validated by repeated analysis of twelve reference samples corresponding to five different species of Cimicifugae Rhizoma and seventeen samples purchased from markets. The results indicate that the established HPLC/UV method is suitable for the quantitative analysis and quality control of multi-components in Cimicifugae Rhizoma.
