Quantitative Determination of the Thirteen Marker Components in Banhasasim-Tang Decoction Using an Ultra-Performance Liquid Chromatography Coupled to Electrospray Ionization Tandem Mass Spectrometry (LC-MS/MS를 이용한 반하사심탕 물 추출물 중 13종 성분의 함량분석)

  • Banhasasim-tang is a well-known traditional Korean herbal formula and has been used clinically for the treatment of gastric disease, including acute and chronic gastritis, diarrhea and gastric ulcers in Korea. In this study, an ultra-performance liquid chromatography-electrospray ionization-mass spectrometer method was developed for the quantitative determination of the 13 marker constituents, homogentisic acid (1), 3,4-dihydroxybenzaldehyde (2), spinosin (3), liquiritin (4), baicalin (5), ginsenoside Rg1 (6), liquiritigenin (7), wogonoside (8), ginsenoside Rb1 (9), baicalein (10), glycyrrhizin (11), wogonin (12), and 6-gingerol (13) in Banhasasim-tang decoction. Separation of the compounds 1-13 was using an UPLC BEH $C_{18}$ ($100{\times}2.1mm$, $1.7{\mu}m$) column and column oven temperature was maintained at $45^{\circ}C$. The mobile phase consisted of 0.1% (v/v) formic acid in water (A) and acetonitrile (B) by gradient elution. The injection volume and flow rate were $2.0{\mu}L$ and 0.3 mL/min, respectively. Calibration curves of the compounds 1-13 were showed with $r^2$ values ${\geq}0.9908$. The limit of detection and limit of quantification values of the compounds 1-13 were 0.04-1.11 ng/mL and 0.13-3.33 ng/mL, respectively. Among the these compounds, the compounds 1-3 were not detected, while the compounds 4-13 were detected in the ranges of $3.20-107,062.98{\mu}g/g$ in Banhasasim-tang sample.

High $f_T$ 30nm Triple-Gate $In_{0.7}GaAs$ HEMTs with Damage-Free $SiO_2/SiN_x$ Sidewall Process and BCB Planarization

  • A 30 nm $In_{0.7}GaAs$ High Electron Mobility Transistor (HEMT) with triple-gate has been successfully fabricated using the $SiO_2/SiN_x$ sidewall process and BCB planarization. The sidewall gate process was used to obtain finer lines, and the width of the initial line could be lessened to half by this process. To fill the Schottky metal effectively to a narrow gate line after applying the developed sidewall process, the sputtered tungsten (W) metal was utilized instead of conventional e-beam evaporated metal. To reduce the parasitic capacitance through dielectric layers and the gate metal resistance ($R_g$), the etchedback BCB with a low dielectric constant was used as the supporting layer of a wide gate head, which also offered extremely low Rg of 1.7 Ohm for a total gate width ($W_g$) of 2x100m. The fabricated 30nm $In_{0.7}GaAs$ HEMTs showed $V_{th}$of -0.4V, $G_{m,max}$ of 1.7S/mm, and $f_T$ of 421GHz. These results indicate that InGaAs nano-HEMT with excellent device performance could be successfully fabricated through a reproducible and damage-free sidewall process without the aid of state-of-the-art lithography equipment. We also believe that the developed process will be directly applicable to the fabrication of deep sub-50nm InGaAs HEMTs if the initial line length can be reduced to below 50nm order.

Analysis of Upper Limb Muscles Properties In Elite Swimmers Before and After Training (엘리트 수영선수들의 수중 훈련 전후의 상지 근육 특성 변화 분석)

  • Objective: The aim of this study is to verify the differences in muscle characteristics of elite level swimmers before and after a 2-hour practice session. Method: The study was conducted on 15 elite swimmers. Preliminary measurements for each muscle (Anterior Deltoid, Triceps Brachii, Biceps Brachii, Flexor Carpi Ulnaris) were taken using the MyotonPRO device before training. After approximately 2 hours of training, the same muscle areas were measured again. The collected data was analyzed through descriptive statistics and two-way 2×2 RG·RM ANOVA, and all statistical significance levels were set at α=.05. Results: After analyzing the characteristics of the Flexor Carpi Ulnaris (FCU) before and after training in both proficiency level swimmers (excellent, non-excellent), it was found that the interaction effect of group X repetition in muscle tension (F), muscle stiffness (S), and body recovery time (R) was statistically significant. Secondly, in the analysis of the Biceps Brachii (BB), the main effect of repetition in muscle tension (F), muscle stiffness (S), and body recovery time (R) was statistically significant. Furthermore, the interaction effect of group X repetition in muscle stiffness (S) and body recovery time (R) was statistically significant. Conclusion: The efficient use of FCU and BB suggests that it is an important factor distinguishing the performance of excellent and non-excellent swimmers in swimming. Therefore, if we develop and apply measures to efficiently utilize FCU and BB during training, it can help improve the performance of the athletes.

  • In this study, the ginseng sprout has produced through smart farm was classified according to its size and divided into above-ground (AG) and below-ground (BG) parts to compare ginsenoside contents and antioxidant activity. In the case of the AG part, the total phenolic contents were the highest at 5.16 mg/g in medium (M) size and the lowest at 2.23 mg/g in largest (L) size. The BG part also showed the highest content in the M size, but there was no significant difference. Also, the total flavonoid contents were also high in the M size in both the AG (5.16 mg/g) and BG (1.28 mg/g) parts. The major ginsenosides in the AG part were Re (20.33-24.15 mg/g) > Rd (11.36-27.42 mg/g) > Rg1 (4.48-5.54 mg/g) and the main ginsenosides in the BG part were Rb1 (5.09-8.61 mg/g) > Re (4.48-5.54 mg/g) > Rc (3.11-4.11 mg/g) in orders. In the case of M size, Re and Rd were approximately 4- and 19-folds higher at 24.15 mg/g and at 27.42 mg/g in the AG part and 5.20 mg/g and 1.43 mg in the BG part, respectively. In addition, F3 and Rh1 were detected in the AG part, but not in the BG part. 2,2-diphenyl-1-picrylhydrazyl (74.95%), 2,4,6-azino-bis (3-ethylbenzothiazoline-6-sulphnoic acid) diammonium salt (94.47%), and hydroxyl (70.39%) radical scavenging activities and FRAP (2.169) assay were the highest in M size than other sizes.

Ginsenoside Content of North American Ginseng (Panax quinquefolius L. Araliaceae) in Relation to Plant Development and Growing Locations

  • North American ginseng (Panax quinquefolius L.) was analysed for total ginsenosides and ten major ginsenosides (R$_{0}$ , Rb$_1$, Rb$_2$, Rc, Rd, Re, Rf, Rg$_1$, pseudoginsenoside F$_{11}$ and gypenoside XVII), and variations in ginsenoside content with age of plant (over a four-year-period) and geographic location (Ontario versus British Columbia) were investigated. In the roots the total ginsenoside content increased with age up to 58-100 mgㆍg$^{-1}$ dry weights in the fourth year, but in leaves it remained constant over time. Roots and leaves, moreover, had different proportions of individual ginsenosides. The most abundant ginsenosides were Rb$_1$ (56mgㆍg$^{-1}$ for Ontario; 37mgㆍg$^{-1}$ for British Columbia) and Re (21mgㆍg$^{-1}$ for Ontario; 15 mgㆍg$^{-1}$ for British Columbia) in roots, and Rd (28-38 mgㆍg$^{-1}$ ), Re (20-25 mgㆍg$^{-1}$ ), and Rb$_2$ (13-19 mgㆍg$^{-1}$ ) in leaves. Measurable quantities of Rf were found in leaves (0.4-1.8 mgㆍg$^{-1}$ ) but not in roots or stems. Our results show that ginsenoside profiles in general, and Rf in particular, could be used for chemical fingerprinting to distinguish the different parts of the ginseng plant, and that ginseng leaves could be valuable sources of the ginsenosides Rd, Re, and Rb$_2$.

Unraveling dynamic metabolomes underlying different maturation stages of berries harvested from Panax ginseng

  • Background: Ginseng berries (GBs) show temporal metabolic variations among different maturation stages, determining their organoleptic and functional properties. Methods: We analyzed metabolic variations concomitant to five different maturation stages of GBs including immature green (IG), mature green (MG), partially red (PR), fully red (FR), and overmature red (OR) using mass spectrometry (MS)-based metabolomic profiling and multivariate analyses. Results: The partial least squares discriminant analysis score plot based on gas chromatography-MS datasets highlighted metabolic disparity between preharvest (IG and MG) and harvest/postharvest (PR, FR, and OR) GB extracts along PLS1 (34.9%) with MG distinctly segregated across PLS2 (18.2%). Forty-three significantly discriminant primary metabolites were identified encompassing five developmental stages (variable importance in projection > 1.0, p < 0.05). Among them, most amino acids, organic acids, 5-C sugars, ethanolamines, purines, and palmitic acid were detected in preharvest GB extracts, whereas 6-C sugars, phenolic acid, and oleamide levels were distinctly higher during later maturation stages. Similarly, the partial least squares discriminant analysis based on liquid chromatography-MS datasets displayed preharvest and harvest/postharvest stages clustered across PLS1 (11.1 %); however, MG and PR were separated from IG, FR, and OR along PLS2 (5.6 %). Overall, 24 secondary metabolites were observed significantly discriminant (variable importance in projection > 1.0, p < 0.05), with most displaying higher relative abundance during preharvest stages excluding ginsenosides Rg1 and Re. Furthermore, we observed strong positive correlations between total flavonoid and phenolic metabolite contents in GB extracts and antioxidant activity. Conclusion: Comprehending the dynamic metabolic variations associated with GB maturation stages rationalize their optimal harvest time per se the related agroeconomic traits.

Glass Forming Ability and Characteristic Evaluation in Ca-Mg-Zn Alloy System (Ca-Ma-Zn 합금계에서 비정질 형성능 및 특성 평가)

  • The effect of alloy composition on the glass forming ability (GFA) of the Ca-rich Ca-Mg-Zn alloys has been investigated in $Ca_{65}Mg_{5+x}Zn_{30-x}$ and $Ca_{55+x}Mg_{15}Zn_{30-x}$ (x=0, 5, 10, 15, 20) alloys. In a wide composition range of 15-25% Zn and 10-20% Mg bulk metallic glass (BMG) samples with the diameter larger than 6 mm are fabricated by conventional copper mold casting method in air atmosphere. Among the alloys investigated, the $Ca_{65}Mg_{15}Zn_{20}$ alloy exhibits the highest GFA enabling to form BMG sample with the diameter of at least 15 mm. The crystalline phase formed during solidification of $Ca_{65}Mg_{15}Zn_{20}$ ($D_{max}=15\;mm$) could be identified as a mixture of $Ca_3Zn$ and $CaMg_2$ cause by the redistribution of the constituent elements on long-range scale. The compressive fracture strength and fracture elongation of the $Ca_{65}Mg_{15}Zn_{20}$ BMG are 602 MPa and 2.08% respectively. The ${\sigma}$ parameter which has been recently proposed for evaluating GFA exhibits better correlation with GFA of Ca-Mg-Zn alloys than other parameters suggested so far such as ${\Delta}T_x$, $T_{rg}$, K, ${\gamma}$, and ${\Delta}T^*$ parameters.

  • The Purpose of this study are to investigate the characteristics of the flame behavior of gasoline-methanol blended fuels in spark ignition engine. Ionization probe were installed at the cylinder head and piston in order to measure flame speed. Other parameter such as engine performance, fuel consumption rate and exhaust gas were measured. The results were as follows. 1. In the case of increase methanol contents in blend fuel, flame propagation speed were increased, and thermal efficiency of the engine were increased due to decrease of energy consumption rate. 2. In the case of fixed equivalance ratio, NO sub(X) in exhaust gas were increased in accordance with increase of spark advance, and mean effective pressure were decreased in accordance with increase of methanol contents. 3. CO and HC concentration were decreased in accordance with increase of methanol contents.

적외선 광대역 고분산분광기 IGRINS 개발 현황

  • 한국천문연구원은 미국 텍사스대학교와 공동으로 근적외선 광대역 고분산 분광기 IGRINS (Immersion Grating Infrared Spectrograph)를 개발하고 있다. IGRINS는 R=40,000의 높은 분광분해능으로 H-밴드 ($1.49\sim1.80{\mu}m$)와 K-밴드 ($1.96\sim2.46{\mu}m$) 전체 분광스펙트럼을 한 번에 관측할 수 있다. IGRINS를 구성하는 핵심 부품은 실리콘 담금격자 (Silicon Immersion Grating)와 VPH (Volume Phased Holographic) 격자, 그리고 HAWAII2RG 적외선 센서이다. 실리콘 담금격자를 사용함으로써 적외선 분광기의 크기를 일반적인 격자를 사용한 것보다 2-3배정도 줄일 수 있게 되었다. IGRINS는 개발 후 미국 맥도날드 천문대에 3년간 장착하여 관측연구에 활용될 예정이다. 이후 IGRINS는 4미터급 이상의 망원경에 장착될 예정이다. 한국천문연구원은 IGRINS 국제공동 개발을 위하여 2009년 8월 텍사스 대학과 양해각서(MOU)를 교환하였으며 현재 개념 설계를 끝내고 기본 설계를 진행하고 있다. 기본설계검토 회의 (Preliminary Design Review)은 12월에 실시할 예정이다. 또한 과학연구 활용 극대화를 위하여 사용자 그룹을 조직하였으며, 여름학교를 열어 적외선 분광 연구에 대한 교육을 실시하였다. 이 발표에서는 IGRINS의 개발 현황을 보고하고, IGRINS의 설계와 활용 분야에 대하여 소개한다.

  • IGRINS, the Immersion GRating INfrared Spectrometer, is a near-infrared wide-band high-resolution spectrograph jointly developed by the Korea Astronomy and Space Science Institute and the University of Texas at Austin. IGRINS employs three HAWAII-2RG focal plane array (FPA) detectors. The mechanical mounts for these detectors serves a critical function in the overall instrument design: Optically, they permit the only positional compensation in the otherwise "build to print" design. Thermally, they permit setting and control of the detector operating temperature independently of the cryostat bench. We present the design and fabrication of the mechanical mount as a single module. The detector mount includes the array housing, a housing for the SIDECAR ASIC, a field flattener lens holder, and a support base. The detector and ASIC housing will be kept at 65 K and the support base at 130 K. G10 supports thermally isolate the detector and ASIC housing from the support base. The field flattening lens holder attaches directly to the FPA array housing and holds the lens with a six-point kinematic mount. Fine adjustment features permit changes in axial position and in yaw and pitch angles. We optimized the structural stability and thermal characteristics of the mount design using computer-aided 3D modeling and finite element analysis. Based on the computer simulation, the designed detector mount meets the optical and thermal requirements very well.
