고성능 액체 크로마토그래피/방사흐름 전기화학전지를 이용한 안트라사이클린계 항생제의 동시 정량 (Simultaneous determinations of anthracycline antibiotics by high performance liquid chromatography coupled with radial-flow electrochemical cell)

  • 방사흐름 전지를 연결한 고성능 액체 크로마토그래프를 사용하여 독소루비신, 에피루비신, 노갈라마이신, 다우노루비신 및 아이다루비신을 동시에 정량 할 수 있는 역상 크로마토그래피법을 개발하였다. 안트라사이클린계 항생제들은 이동상 용매 중에서 은/염화은 (0.01 M NaCl) 기준전극에 대하여 확산 전류를 나타내는 -0.74 V에서 검출되었다. 부피 흐름속도 ($V_f$)를 1.0 mL/min로 고정하였을 때 독소루비신, 에피루비신, 다우노루비신 및 아이다루비신은 각각 6.4 분, 7.4분, 12.7분 및 18.4분의 머무름 시간 ($t_r$)에서 나타났으며, $V_f$ 0.6 mL/min 에서는 독소루비신, 에피루비신, 노갈라마이신, 다우노루비신 및 아이다루비신의 $t_r$은 각각 9.9분, 11.5분, 13.5분, 19.6분 및 28.7분에서 나타났다. 주입된 각각의 안트라사이클린계 항생제의 농도 ($2.40{\times}10^{-7}M{\sim}1.42{\times}10^{-5}M$)에 대하여 봉우리 면적 (전하)를 도시하였을 때 상관계수의 제곱 ($R^2$)은 0.999 이상으로 직선 성이 우수하였다. 다섯 가지 안트라사이클린계 항생제의 검출한계는 $1.0{\times}10^{-8}M{\sim}1.5{\times}10^{-7}M$이었으며 정밀도는 $0.7{\mu}M$ 이하의 농도를 제외하고는 상대 표준편차3%($1.00{\times}10^{-6}M{\sim}1.42{\times}10^{-5}M$) 미만이었다. 사람의 혈청 중에 포함된 $1.00{\times}10^{-5}M$ 에피루비신, $0.48{\times}10^{-5}M$ 노갈라마이신 및 $1.52{\times}10^{-5}M$ 다우노루비신을 $C_{18}$ 카트리지로 고상 추출하였을 때 회수율은 각각 97%, 100% 및 90% 이었다.

흰쥐에서 에탄올과 phenobarbital이 벤지딘의 헤모글로빈 부가체 형성에 미치는 영향 (Effects of ethanol and phenobarbital on hemoglobin adducts formation in rats exposed to benzidine)

  • Recently, biochemical analysis using hemoglobin adduct is frequently performed to evaluate the exposure to chemical carcinogens. However, data on the effect of co-exposure with other chemicals on hemoglobin adduct formation are seldom provided. The objective of this study is to evaluate the effects of pretreatment of ethanol(EtOH) and phenobarbital(PB), which are known to affect metabolism of xenobiotics, on the formation of hemoglobin adducts in the rats(Sprague-Dawley) administered benzidine(BZ). The experimental rats were divided into control, EtOH, and P8 groups. Rats were pretreated with EtOH or PB 24 hours before the oral administration of BZ. Blood sampling was taken before the administration of the chemicals and 0.5, 3, 6, 9, 12, 24, 48, 72, 96, and 144 hours after the administration of the BZ in 5 rats each. The blood was separated into hemoglobin and plasma immediately after taking the blood samples, and the adducts were undergone basic hydrolysis to convert them into aromatic amines. Hydrolyzed BZ, monoacetylbenzidine (MABZ), and 4-aminobiphenyl(4ABP) were separated by reversed-phase liquid chromatography without derivatization, and quantitative analyses of them were performed by a highperformance liquid chromatograph equipped with electrochemical detector. The quantitative amount of the metabolites was expressed by hemoglobin binding index(HBI), BZ-, MABZ-, and 4ABP-HBI of EtOH and PB groups were increased more than those of control group. These results are attributable to the fact that EtOH and PB induced N-hydroxylation related to the hemoglobin adduct formation. The ratio of N-acetylation (viz, MABZ-HBI/BZ-HBI) showed no significant difference between EtOH group and control group. It means that EtOH increased N-hydroxylation and N-acetylation in a similar degree. The N-acetylation ratio of PB group was relatively lower than control group because the PB increased N-hydroxylation induction. The N-acetylation ratios of all groups were higher than 1 during the entire experimental period. This result suggests that the effects of EtOH or PB need to be considered in the biochemical monitoring for the assessment of intermittent exposure of benzidine.

국내에서 소비되는 상용 식품의 비타민 K1 함량 분석 (Analysis of Vitamin K1 in Commonly Consumed Foods in Korea)

  • 본 연구에서는 국내에서 소비되는 일부 상용식품의 비타민 $K_1$ 함량을 측정하고자 하였으며, 이 분석방법을 검증하며 결과의 신뢰도를 확보하고자 하였다. 각 식품군의 비타민 K는 용매추출법을 이용하여 추출하였으며, 포스트칼럼이 장착된 역상 HPLC를 통하여 분리, 정량하였다. 채소류에서는 고구마 잎이 $1,467.3{\mu}g/100g$으로 가장 높았으며, 과일류는 녹색 키위가 $42.7{\mu}g/100g$으로 가장 높게 분석되었다. 두류에서는 녹두가 $63.4{\mu}g/100g$으로 가장 높은 값을 나타내었다. 분석방법을 검증하기 위하여 직선성, 재현성, 반복성, 회수율 및 peak purity를 구하였으며 분석관리 차트를 작성하여 분석 결과 값을 관리하였다. 그 결과 본 시험법에서 표준물질의 피크 머무름시간과 모시잎 시료의 피크 머무름시간이 일치하여 특이성을 확인하였다. 검량선의 상관 계수는 0.9999 이상으로 높은 유의 수준을 보여 분석에 적합함을 알 수 있었으며, 반복성과 재현성의 변동계수는 각각 2.33, 3.93%로 나타났고 회수율은 103.67%로 우수하게 나타났다. 본 연구에서 분석된 자료는 표준화된 식품성분표의 기초 자료로 이용될 수 있을 것으로 판단된다.

국내 콩(Glycine max L.)자원의 lutein 함량 (Lutein Contents of Soybeans (Glycine max L.) Cultivated in Korea)

  • 본 연구에서는 국내 콩 자원의 lutein 함량 변이를 분석함으로써 우수 품종을 선발하고 신품종 육성연구와 기능성 식품 개발을 위한 우수 품종을 선발하고자 하였다. 본 연구에서는 장려품종 23종과 육성품종 140종(장콩품종 56종, 나물콩품종 44종, 지역재래품종 40종)을 lutein 분석에 사용하였다. 검화방법을 이용하여 lutein을 추출하였고 역상 HPLC를 이용하여 분석하였다. 연구 결과 장려품 23종의 평균 lutein 함량은 $240.27{\pm}90.74{\mu}g/100g$으로 IT No. 24634가 $402.48{\mu}g/100g$으로 가장 높은 함량을 나타내었다. 장콩은 $777.73{\mu}g/100g$으로 IT No. 153398이, 나물콩은 $588.88{\mu}g/100g$으로 IT No. 25490이, 지역재래종은 $861.07{\mu}g/100g$으로 IT No. 22564가 높은 lutein 함량을 나타내었다. 장려품종과 육성품종간의 그 평균치는 통계학적으로 유의적인 차이를 나타내지 않았으나 장려품종 보다는 육성품종에서 상대적으로 높은 lutein 함량을 나타내는 품종이 많은 것으로 나타났다. 본 연구결과는 국내 콩 유전자원의 품종개량 및 고 부가가치 기능성 식품의 개발에 대한 기초 자료가 될 것으로 생각된다.

간편한 DNPH 유도체화 HPLC 분석법을 이용한 화장품 중 포름알데하이드 분석 (Determination of Formaldehyde in Cosmetics Using a Convenient DNPH Derivatization Procedure Followed by HPLC Analysis)

  • 대한민국 식품의약품안전처(식약처)는 포름알데하이드 분석법으로 2,4-dinitrophenylhydrazine(DNPH) 유도체화-고성능액체크로마토그래프법(HPLC)을 고시하고 있다. 본 연구는 고시법의 복잡한 시료 전처리 과정을 개선하여 화장품 분석에 편리하게 사용할 수 있는 유도체화법을 개발하고자 수행되었다. 전처리법을 간단하게 하기 위하여 pH, 시간 및 온도 등 반응조건을 최적화하였다. 이 전처리법은 초산염 완충액(pH 5.0)을 사용한 검액의 pH 조정, 디클로로메탄을 사용한 액-액 분획 그리고 감압농축기를 사용한 증발건조와 같이 식약처 고시법의 복잡한 과정이 필요 없다. 유도체화 과정을 통하여 생성된 formaldehyde dinitrophenylhydrazone(formaldehyde-DNP)는 식약처의 시험방법을 약간 변형한 역상 HPLC법으로 분리하고 정량하였다. 2 ~ 40 ppm 농도 범위의 표준액들을 가지고 수행한 검량선 작성 결과, 본 시험법은 상관계수 값이 0.9999로 좋은 직선성을 보여주었다. 본 실험의 최소검출한계(LOD)와 최소정량한계(LOQ)는 각각 0.2 ppm과 0.5 ppm이었다. 또한 회수율 실험결과는 실험방법이 매우 정확하고 재현성이 높음을 보여주었다. 따라서 본 연구에서 제안된 시험법은 화장품 중 포름알데하이드를 신속하게 분석하는데 적용될 수 있을 것이다.

잎새버섯 균사체를 이용한 감초추출발효물의 플라보노이드 생성과 항염 활성 연구 (Production of Flavonoid Compounds and Anti-inflammatory Property of Fermented Licorice Extract with the Basidiomycete Grifola frondosa HB0071)

  • 리퀴리티게닌과 이소리퀴리티게닌은 감초의 주요 플라보노이드 성분이다. 이들 플라보노이드는 수용성 감초 추출물과 ${\beta}$-glucosidase를 생성하는 잎새버섯 HB0071 균사체 발효배양을 통하여 생산하였다. 감초추출물 내 리퀴리티게닌과 이소리퀴리티게닌은 잎새버섯 발효배양 동안 현저히 증가하였다. 이 균주의 ${\beta}$-glucosidase의 활성은 배양 96시간을 기준으로 최고 91.5 mU/mL로 확인되었으며, 감초추출발효물로부터 생성된 리퀴리티게닌과 이소리퀴리티게닌의 함량은 HPLC 분석을 통하여 최대 $568.5{\mu}g/mL$$89.6{\mu}g/mL$로 확인되었다. 본 연구에서는 감초추출물의 잎새버섯 발효 전 후의 시료가 처리된 각질형성세포를 이용하여 자외선 UVB에 조사로 발현된 염증유발인자(COX-2)와 사이토카인(IL-$1{\beta}$, IL-6) 모두 감초추출발효물(FLEx)에서 농도의존적으로 발현이 억제되는 것을 확인하였다. 결론적으로 리퀴리티게닌과 이소리퀴리티게닌의 함량이 증가된 감초추출발효물은 자외선으로부터 손상된 피부 염증반응을 완화시켜줄 것으로 사료된다.

Vitamin D3 and Beta-carotene Deficiency is Associated with Risk of Esophageal Squamous Cell Carcinoma - Results of a Case-control Study in China

  • Objective: The aim was to evaluate roles of vitamin D3 (VD3) and beta-carotene (BC) in the development of esophageal squamous cell carcinoma (ESCC) in a high-risk area, Huai'an District, Huai'an City, China. Methods: 100 new ESCC diagnosed cases from 2007 to 2008 and 200 residency- age-, and sex-matched healthy controls were recruited. Data were collected from questionnaires, including a food frequency questionnaire (FFQ) to calculate the BC intake, and reversed phase high-performance liquid chromatography (RP-HPLC) was used to measure the serum concentrations of BC and VD3. Odds ratios (OR) and 95% confidence intervals (CI) were calculated in conditional logistic regression models. Results: The average dietary intake of BC was $3322.9{\mu}g$ (2032.4-5734.3) in the case group and $3626.8{\mu}g$ (1961.9-5827.9) in control group per capita per day with no significant difference by Wilcoxon test (p>0.05). However, the levels of VD3 and BC in the case group were significantly lower than in the control group (p<0.05). The OR values of the highest quartile and the lowest quartile of VD3 and BC in serum samples were both 0.13. Conclusion: Our results add to the evidence that high circulating levels of VD3 and BC are associated with a reduced risk of ESCC in this Chinese population.

소시호탕에서 baicalin과 glycyrrhizin 동시 분석 및 항염 활성 (Simultaneous determination of baicalin and glycyrrhizin in Sosiho-tang by HPLC and anti-inflammatory activity)

  • Objectives : To quantitate the main compounds and investigate the biological activity of Sosiho-tang (Xiao-Chai-Hu-Tang, SST), simultaneous determination of baicalin and glycyrrhizin, and anti-inflammatory activity were estimated. Methods : A quantitative analysis was performed using a high performance liquid chromatography (HPLC). Reference compounds were separated on a reversed-phase column using gradient elution with water and acetonitrile each containing acetic acid at a flow rate of 1 mL/min. And the productions of nitric oxide (NO) and prostaglandin $(PE)E_2$ were examined by lipopolysaccharide (LPS)-treated RAW 264.7 cells in the presence of the SST. The anti-inflammatory activity of SST was investigated by carrageenin-induced paw edema in rats. The paw volume was measured at 2 and 4 hr following carrageenin-induced paw edema in rats. Results : The correlation coefficients of the compounds showed good linearity ($r^2$ > 0.9992) over the linear range. The precisions of intra- and inter-day were less than 7.0% of relative standard deviation (RSD) values for baicalin and less than 3.5% of RDS valuse for glycyrrhizin. Recovery rates were within the range of 95.41-101.5%. The contents of baicalin and glycyrrhizin in SST were average 70.52, 6.18 mg/g, respectively. And SST exhibited inhibitory effect on NO production in LPS-treated RAW 264.7 cells but not on $PGE_2$ production. Oral administration of SST (1 g/kg) showed a reduction in carrageenin-induced paw edema on rats. Conclusions : The analytical method was applied successfully to measure the contents of baicalin and glycyrrhizin in SST which exhibited anti-inflammatory activities.

High-Level Expression of T4 Endonuclease V in Insect Cells as Biologically Active Form

  • T4 endonuclease V (T4 endo V) [EC 3. 1. 25. 1], found in bacteriophage T4, is responsible for excision repair of damaged DNA. The enzyme possesses two activities: a cyclobutane pyrimidine dimer DNA glycosylase (CPD glycosylase) and an apyrimidic/apurinic endonuclease (AP lyase). T4 denV (414 bp cDNA) encoding T4 en do V (138 amino acid) was synthesized and expressed using either an expression vector, pTriEx-4, in E. coli or a baculovirus AcNPV vector, pBacPAK8, in insect cells. The recombinant His-Tag/T4 endo V (rHis-Tag/T4 endo V) protein expressed from bacteria was purified using one-step affinity chromatography with a HiTrap Chelating HP column and used to make rabbit anti-His-Tag/T4 endo V polyclonal antibody for detection of recombinant T4 endo V (rT4 endo V) expressed in insect cells. In the meantime, the recombinant baculovirus was obtained by cotransfection of BacPAK6 viral DNA and pBP/T4 endo V in Spodoptera frugiperda (Sf21) insect cells, and used to infect Sf21 cells to overexpress T4 endo V protein. The level of rT4 endo V protein expressed in Sf21 cells was optimized by varying the virus titers and time course of infection. The optimal expression condition was set as follows; infection of the cells at a MOI of 10 and harvest at 96 h post-infection. Under these conditions, we estimated the amount of rT4 endo V produced in the baculovirus expression vector system to be 125 mg/l. The rT4 endo V was purified to homogeneity by a rapid procedure, consisting of ion-exchange, affinity, and reversed phase chromatographies, based on FPLC. The rT4 endo V positively reacted to an antiserum made against rHis-Tag/T4 endo V and showed a residual nicking activity against CPD-containing DNA caused by UV. This is the first report to have T4 endo V expressed in an insect system to exclude the toxic effect of a bacterial expression system, retaining enzymatic activity.

소변 중 4-hydroxyproline 분석에 관한 연구 (Determination of Free 4-hydroxyproline with Dansylchloride by HPLC in Human Urine)

  • Objectives : The level of 4-hydroxyproline (4-Hyp) in human urine was measured using high performance liquid chromatography (HPLC) with a fluorescence detector. This method is useful for medical examinations and investigating the radicals induced by physical, chemical, mental stresses. This method is superior to many published several methods in terms of its low cost and ability to analyze many samples. Methods : The urine from workers in a tire manufacturing company (22 male pre- and post-shift workers) and 18 office-workers as controls were analyzed. Data concerning age, the cumulative drinking amount and the cumulative smoking amount was collected with a questionnaire. The optimum applied amount of dansyl-Cl, the optimum reaction temperature and time, the recoveries and the optimum pH of the eluent and buffer were determined.4-Hyp from human urine was derivatized with dansyl-Cl (dimethylamino-naphthalene-1-sulfonyl chloride) after removing the a-amino acid by a treatment with phthalic dicarboxaldehyde (OPA) and cleaned with Bond Elut C18 column. The 4-Hyp derivatives were separated on a reversed phase column by gradient elution with a phosphate buffer (5 mmol, pH 8.0) and acetonitrile, and detected by fluorescence measurements at 340 nm (excitation) and 538 nm (emission). Results : The detection limit for the urinary free 4-Hyp was $0.364{\mu}mol/l$. The recovery rate of 4-Hyp was 99.7%, and the effective pH of the phosphate buffer and borate buffer were 3.0 and 8.0, respectively. From statistical analysis, age, drinking and smoking did not affect the urinary free 4-Hyp in both the controls and workers. The range of urinary 4-Hyp in the controls, pre-shift, and post-shift workers were 0.33-16.44, N.D-49.06, and $0.32-56.27{\mu}mol/l$. From the pared-sample t-test, the urinary 4-Hyp levels in post-shift workers ($11.82{\pm}6.73\;nmmol/mg\;Cre$) were 2-fold higher than in pre-shift workers ($5.36{\pm}5.53\;nmol;/mg\;Cre$) and controls ($4.91{\pm}4.89\;nmol;/mg\;Cre$). Conclusions : This method was developed with high sensitivity, accuracy, and precision. The present method was effectively applied to analyze the urinary free 4-Hyp in both controls and workers.
