Efficient Labeling of Porcine Hematopoietic Cells by Fluorescence-Conjugated Nanoparticles

  • Nanotechnology is currently receiving considerable attention in various fields of biotechnology. The uptake of nanoparticles by cells for labeling and tracking is a critical process for many biomedical therapeutic applications. However, nanoparticle labeling of porcine hematopoietic cells has not been demonstrated so far. In the present study, silica-coated nanoparticles conjugated with rhodamine B isothiocyanate (SR-RITC) were used to investigate the uptake of nanoparticles by porcine hematopoietic cells. Flow cytometric and confocal microscopic analyses reveled that the cells were efficiently internalized by the silica-coated nanoparticles. Furthermore, biocompatibility tests demonstrated that the SR nanoparticles were not cytotoxic, and they had no impact on proliferation. Our study demonstrates that silica-coated nanoparticles are taken up very rapidly and with high efficiency into porcine hematopoietic cells, with no apparent deleterious effects. Therefore, silica-coated nanoparticles appear to be a promising tool for tracking porcine hematopoietic cells.

  • These days Women have repented their past when they were inactive in attaining their own rights and they've payed attentions in gaining their fundamental rights, especially health rights. The spread of contraception use is important to fulfill women's health right. There are a lot of meanings in the contraception use. It makes women can control their reproductive activities such as pregnancy, delivery, and raising children etc. It gives each woman has autonomy of her life. The review reveals high contraceptive rate and high experienced artificial abortion rate in Korea. It means Korean married couples use contraception inefficiently. From 1970's to 1980's, most of the researches in contraceptions were descriptive and survey studies. Since 1990's, the studies have researched in the feministic perspectives. In order to understand experiences of contraceptive behaviors, qualitative researches through indepth interview did. But the number of researches were limited. The relating factors to contraceptive behavior are knowledge, communication between couples, efficiency of contraceptives, self efficacy, attitudes toward contraception, national policy, mass media, public opinion, age, parity, abortion history, ever-used contraceptive method and educational level. Theses factors affect contraception directly and indirectly. Therefore. researches about contraceptives in the future must focus on development of contraceptive behavior assessment tools and further study should be comprehensive and multidementional.

  • Biotechnological researches in Central Asian Research Institute of Sericulture were developed since 1981 when laboratory of artificial diets for mulberry silk­worm was created. The researches were directed not only to creation of accessible artificial diets (AD) for off-season mass rearing of economical beneficial insect and for industrialization of sericulture but also to obtaining of easy reproducible source of animal proteins, carbohydrates, lipids, enzymes, enzymes inhibitors and other metabolites - bioresource for manufacture of oriental medicine preparations. Space experiments with silkworms became possible with use of AD. These experiments used mulberry silkworm as high effective board test organism with outstanding developmental, reproductive and transgenic properties having impor­tant significance in future of cosmonautics. Introduction to practice of bioprotective and ecologically pure method of drying and sterilizing of green cocoons and by-products of silk manufacture allows to increase efficiency of cocoons reeling technology as well as to preserve nativity of biological active substances in mulberry silkworm pupae that is unachievable by traditional methods of raw material processing. New spheres of application of two important silk proteins - fibroin and sericin in medicine, cosmetics, for bio- and nanotechnology are opening with use of biotechnological approaches. At present the achievements and experience of R & D in these fields are used in creation of modern pests biocontrol agents. Viability of Somatic Cell Nuclear Transfer Embryos following Embryo Transfer in Korean Native Striped Cattle (Bos namadicus Falconer, Chikso)

  • This study was conducted to examine the viability of Korean native striped cattle (Bos namadicus Falconer, Chikso) clone embryos after embryo transfer. Chikso somatic cell nuclear transfer (SCNT) embryos were produced by fusion of ear skin cells derived from a female Chikso with enucleated oocytes matured in vitro for 18-24 hr. After in vitro culture of SCNT embryos for 7 to 8 days, fresh or vitrified blastocysts derived from SCNT were transferred into a uterine horn of recipient cows. Fifteen of total 43 recipients were pregnant at Day 50 and 4 recipients were maintained to term. Three IVF-derived calves and 1 clone Chikso calf were born. Pregnancy rate was higher when fresh embryos were transferred to recipients compared to vitrified embryos, but development to term was not different between both groups. The clone Chikso calf died at 5 days after birth due to the fullness of amniotic fluid in rumen and the infection of umbilical cord. The result of the present study shows that clone Chikso calf can produced from the embryo transfer of SCNT embryos, however, solution of abortion problem is necessary to improve the cloning efficiency.

  • Effectiveness of transgene transfer into genome is crucially concerned in mass production of the bio-pharmaceuticals using genetically modified transgenic animals as a bioreactor. Recently, the mammary gland has been considered as a potential bioreactor for the mass production of the bio-pharmaceuticals, which appears to be capable of appropriate post-translational modifications of recombinant proteins. The mammary gland tissue specific vector system may be helpful in solving serious physiological disturbance problems which have been a major obstacle in successful production of transgenic animals. In this study, to minimize physiological disturbance caused by constitutive over-expression of the exogenous gene, we constructed new retrovirus vector system designed for mammary gland-specific expression of the hEPO gene. Using piggyBac vector system, we designed to express hEPO gene under the control of mammary gland tissue specific and lactogenic hormonal inducible goat ${\beta}$-casein or mouse Whey Acidic Protein (mWAP) promoter. Inducible expression of the hEPO gene was confirmed using RT-PCR and ELISA in the mouse mammary gland cells treated with lactogenic hormone. We expect the vector system may optimize production efficiency of transgenic animal and reduce the risk of global expression of transgene.

Lifetime Production of Kajli Ewes at Khushab and Khizerabad : Reproduction and Lamb Production as Affected by Ewe Longevity

  • Data from 5,311 ewes and 13,076 lambing from 1977 through 1994 were used to analyse both annual and cumulative outputs in terms on total number of lambs born, total lamb weight weaned and total wool produced per ewe for ewe longevity 1 to 8 depending on their productive life in the flock. Ewes at Khushab produced 0.08 more lambs per parturition than ewes at Khizerabad; however, 0.39 less lambs were weaned at Khushab than at Khizerabad. Similarly, cumulative number of lambs born was more at Khushab flock than Khizerabad flock (p < .01). However, total weight of lambs weaned was greater at Khizerabad than Khushab flock (p < .01) for each ewe longevity. Most ewes (35%) were sold/replaced just after their first parturition (i. e. ewe longevity 1). The overall mean for annual sale/replacement was 32 and 23% at Khushab and Khizerabad, respectively. Distribution of growth and reproductive traits from 1977-94 did not show upward or downward trend inspite of heavy sale/replacement except yearly variation. Lack of any genetic progress over the year suggested that random breeding was employed without any scientific selection programme. Annual means for lambs born, lambs weaned and weight of lambs weaned per ewe present in the flock were the highest for ewe longevity 2 compared with other ewe longevity groups. Relative efficiency in terms of net income was highest for ewe longevity 5 followed by ewe longevity 4 and 6 in both flocks.

Artificial Microparticle Diets for Culturing Rotifer, Brachionus plicatilis

  • Rotifer culture fed on five types of artificial microparticle diets were evaluated to substitute the natural diets such as Chlorella or w-yeast. These microparticle diets including solidified blood using squid oil (SBSO), solidified blood using soybean oil (SBSB), nylon protein walled particle (NPW) simple coacervation oil capsule (SCO), complex coacervation oil capsule(CCO), were tested for the evaluation of feeding efficiency. The prepared micro particle diets had diameters ranging from 3 to 30 Jim. Rotifer culturing experiments were carried out in 3-liter beakers for 13-16 days. The initial inoculum density of rotifers was 10 ind./ml. The rotifers fed on Chlorella or $\omega-yeast$ showed maximal densities of 2,000 ind./ml in 9 days or 500 ind./ml in 7 days, respectively. Those fed on SBSO, SBSB or NPW showed maximal densities of 1568 ind./ml, 586 ind./ml or 503 ind./ml, respectively and the reproductive rates for those diets were equivalent to or better than w-yeast. However, the coacervated oil capsule showed lower maximal densities of 400 ind./ml for SCO and less than 100 ind./ml for CCO due to the unbalanced diet formulation and indigestibility.

miR-153 Silencing Induces Apoptosis in the MDA-MB-231 Breast Cancer Cell Line

  • MicroRNAs (miRNAs) are small, non-coding RNAs (18-25 nucleotides) that post-transcriptionally modulate gene expression by negatively regulating the stability or translational efficiency of their target mRNAs. In this context, the present study aimed to evaluate the in vitro effects of miR-153 inhibition in the breast carcinoma cell line MDA-MB-231. Forty-eight hours after MDA-MB-231 cells were transfected with the miR-153 inhibitor, an MTT (3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide) assay was utilized to determine the effects of miR-153 on cell viability. Flow cytometry analysis and assessment of caspase 3/7 activity were adopted to determine whether miR-153 affects the proliferation rates and apoptosis levels of MDA-MB-231 cells. Our results showed that silencing of miR-153 significantly inhibited growth when compared to controls at 48 hours, reducing proliferation by 37.6%, and inducing apoptosis. Further studies are necessary to corroborate our findings and examine the potential use of this microRNA in future diagnostic and therapeutic interventions.

Lentivirus-mediated Gene Transfer to Bovine Embryos

  • Pronuclear DNA microinjection has been the most universal method in transgenic animal production but its success rate of transgenesis in mammals are extremely low. To address this long-standing problem, we used retrovirus- and lentivirus-based vectors carrying the enhanced green fluorescent protein (EGFP) gene under the control of ubiquitously active cytomegalovirus (CMV) promoter to deliver transgenes to bovine embryos. The rate of transgenesis was evaluated by counting EGFP positive blastocysts after injection of concentrated virus stock into the perivitelline space of the bovine oocytes in metaphase II. Among two different types of lentivirus vectors derived from FIV (feline immunodeficiency virus) and HIV (human immunodeficiency virus), the former scored the higher gene transfer efficiency; almost 100% of the blastocysts developed from the oocytes infected with FIV-based vector were EGFP positive. As for the vectors derived Com HIV lentivirus, the transgenesis rate of the blastocysts was reduced to 39%.

  • This study was undertaken to clarify the relationship between the cause of repeat-breeder and the luteal dysfunction in repeat-breeder dairy cows that failed to conceive to three or more artificial insemination(AI) at a regular interval. Progesterone concentrations were measured in milk fat for 20 to 22 days after AI. From the 15 repeat-breeder dairy cows, six cows had a normal progesterone profiles. Five cows showed a delayed rise of the progesterone concentrations until 7 to 10 days after AI, two cows had a comparatively low concentration of milk progesterone below 150 ng/ml through most of the luteal phase, and two cows had a combined pattern of a delayed rise and a low concentration of milk progesterone during luteal phase. It is suggest that luteal dysfunction as indicated by progesterone profiles is one of the causes of repeat-breeder in dairy cows.
