Effect of renal ischemia on amino acid transport in rabbit renal cortical slices (신장 허혈이 토끼 신피질 절편에서 아미노산 이동에 미치는 영향)

  • This study was carried out determine the effect of renal ischemia on amino acid transport in rabbit renal cortical slices. The animal models of renal ischemia induced experimentally by clamping the renal artery. These results were summarized as follows: 1. The uptake of amino acids lysine and ${\alpha}$-aminobutyrate(AIB), dicarboxylate succinate and organic anion PAH in cortical slices was normal or increased after 30 or 60 min of ischemia in vivo. 2. In a 30 min ischemic kidney, the slice uptake of amino acids was returned to the control level by 30 min of reflow. In a 60 or 90 min ischemic kidney, the lysine uptake was returned to the control level after of reflow, but the uptake of AIB and succinate was significantly reduced during reflow period of 30-120 min. 3. Oxygen consumption in cortical slices was increased after 30 min of ischemia but was not altered by 60 min of ischemia. This results indicat that transient ischemia caused increasing of amino acid uptake in renal cortical slices without metabolic disorder of renal proximal tubule.

Beneficial Effect of Scutellaria Balicalensis Georgi Extract ont-Buthylhydroperoxide-Induced Inhibition of Organic Cation in Rabbit Renal Cortical Slices (황금약침액(黃芩藥鍼液)이 토끼의 신장절편에서 t-BHP로 유발된 유기양이온의 이동장애에 미치는 영향(影響))

  • Objective : This study was undertaken to determine whether Scutellaria balicalensis Georgi (SbG) extract exerts the protective effect against oxidant-induced alterations in organic cation transport in the renal proximal tubule. Methods : Organic cation transport was estimated by examining alterations in tetraethylammon - ium(TEA) uptake in rabbit renal cortical slices. The slices were treated with 0.2 mM tBHP for 60 min at $37^{\circ}C$. tBHP caused an inhibition in TEA uptake by renal cortical slices. Such an effect was accompanied by depressed Na+-K+-ATPase activity and ATP depletion. tBHP also induced a significant increase in LDH release. Results : SbG prevented tBHP-induced inhibition of TEA uptake in a dose-dependent manner at the concentration ranges of 0.05-0.1%. tBHP-induced inhibition of Na+-K+-ATPase activity and ATP depletion were significantly prevented by 0.05% SbG. tBHP-induced LDH release also was blocked by SbG. tBHP caused a significant increase in lipid peroxidation and its effect was prevented by SbG. Conclusion : These results suggest that SbG prevents oxidant-induced alterations in organic cation transport in rabbit renal cortical slices. Such protective effects of SbG may be attributed to inhibition of peroxidation of membrane lipid.

  • This study was undertaken to determine whether Scutellaria baicalensis Georgi (SbG) extract exerts the protective effect against $HgCl_2$-induced alterations in membrane transport function in rabbit renal cortical slices. The slices were treated with 0.1 mM $HgCl_2$ for 60 min at $37^{\circ}C$. $HgCl_2$ caused an inhibition in PAH uptake by renal cortical slices. Such an effect was accompanied by depressed $Na^+-K^+$-ATPase activity and ATP depletion. SbG prevented $HgCl_2$-induced inhibition of PAH uptake in a dose-dependent manner at the concentration ranges of 0.01-0.1%. $HgCl_2$-induced inhibition of $Na^+-K^+$-ATPase activity and ATP depletion were significantly prevented by 0.05% SbG. These results suggest that SbG prevents $HgCl_2$-induced alterations in membrane transport function in rabbit renal cortical slices. Such protective effects of SbG may be attributed to inhibition of peroxidation of membrane lipid.

Role of Lipid Peroxidation on $H_2O$$_2$-Induced Renal Cell Death in Cultured Cells and Freshly Isolated Cells

  • This study was undertaken to determine the underlying mechanisms of reactive oxygen species-induced cell injury in renal epithelial cells and whether there is a difference in the role of lipid peroxidation between freshly isolated renal cells and cultured renal cells. Rabbit renal cortical slices were used as a model of freshly isolated cells and opossum kidney (OK) cells as a model of cultured cells. Cell injury was estimated by measuring lactate dehydrogenase (LDH) release in renal cortical slices and frypan blue exclusion in OK cells. $H_2O$$_2$ was used as a drug model of reactive oxygen species. $H_2O$$_2$ induced cell injury in a dose-dependent manner in both cell types. However, renal cortical slices were resistant to $H_2O$$_2$ approximately 50-fold than OK cells. $H_2O$$_2$-induced cell injury was prevented by thiols (glutathione and dithiothreitol) and iron chelators (deferoxamine and phenanthroline) in both cell types. $H_2O$$_2$-induced cell injury in renal cortical slices was completely prevented by antioxidants N,N-diphenyl-p -phenylenediamine and Trolox, but the cell injury was not affected by these antioxidants in OK cells. $H_2O$$_2$ increased lipid peroxidation in both cell types, which was completely inhibited by the antioxidants. These results suggest that $H_2O$$_2$ induces cell injury through a lipid peroxidation-dependent mechanism in freshly isolated renal cells, but via a mechanism independent of lipid peronidation in cultured cells.

Beneficial Effect of Pentoxifylline on Hypoxia-Induced Cell Injury in Renal Proximal Tubular Cells

  • Tumor necrosis factor-α (TNF-α) or its mRNA expression are increased in acute nephrosis of various types including ischemia/reperfusion injury. This study was undertaken to determine whether pentoxifylline (PTX), an inhibitor of TNF-α production, provides a protective effect against hypoxia-induced cell injury in rabbit renal cortical slices. To induce hypoxia-induced cell injury, renal cortical slices were exposed to 100% N₂ atmosphere. Control slices were exposed to 100% O₂ atmosphere. The cell injury was estimated by measuring lactate dehydrogenase (LDH) release and p-aminohippurate (PAH) uptake. Exposure of slices to hypoxia increased the LDH release in a time-dependent manner. However, when slices were exposed to hypoxia in the presence of PTX, the LDH release was decreased. The protective effect of PTX was dose-dependent over the concentrations of 0.05∼1 mM. Hypoxia did not increase lipid peroxidation, whereas an organic hydroperoxide t-butylhydroperoxide (tBHP) resulted in a significant increase in lipid peroxidation. PTX did not affect tBHP-induced lipid peroxidation. Hypoxia decreased PAH uptake, which was significantly attenuated by PTX and glycine. tBHP-induced inhibition of PAH uptake was not altered by PTX, although it was prevented by antioxidant deferoxarnine. The PAH uptake by slices in rabbits with ischemic acute renal failure was prevented by PTX pretreatment. These results suggest that PTX may exert a protective effect against hypoxia-induced cell injury and its effect may due to inhibition of the TNF-α production, but not by its antioxidant action.

  • This study was carried out to investigate whether water extract of Sagunja-Tang and its composing herbs have the inhibitory effects on cytotoxicity and lipid peroxidation induced by oxidant in rat renal cortical slices. Cytotoxicity was estimated by measuring lactate dehydrogenase (LDH) activity and lipid peroxidation was examined by measuring malondialdehyde (MDA). a product of lipid peroxidation. When rat renal cortical slices were treated with tert-butylhydroperoxicle (t-BHP) of 1 mM and water decocted herbs. LDH release from the slices was inhibited in dose dependent manner at low concentrations of herbs. It shows that herbs can reduce cytotoxicity, but overdose of herbs can be toxic to the slices. And MDA measurements show each herb has its own activities of preventing cytotoxicity from oxidants. So further studies should be followed to make clear the mechanisms of anti-oxidative effects of herbs.

  • Objective : This study was undertaken to determine whether Cordyceps sinensis Sacc. (CSS) extract exerts the protective effect against oxidant-induced alterations in membrane transport function in renal tubules. Methods : Membrane transport fucntion was estimated by examining alterations in p-aminohippurate (PAH) uptake in rabbit renal cortical slices. For induction oxidative stress, slices were treated with an organic peroxide cumene hydroperoxide for 60 min at $37^{\circ}C$. Cumene hydroperoxide inhibited PAH uptake in a time dependent manner. Results : CSS at 0.5-5% concentrations prevented cumene hydroperoxide-induced inhibition of PAH uptake. CSS at 1% also attenuated LDH release and lipid peroxidation induced by cumene hydroperoxide. When slices were treated with 0.2 mM mercury chloride, PAH uptake was inhibited and lipid peroxidation was increased. These changes by mercury were significantly prevented by CSS. Conclusion : These results suggest that CSS prevents oxidant-induced alterations in membrane transport function in rabbit renal cortical slices. Such protective effect of CSS may be attributed to inhibition of peroxidation of membrane lipid.

Effect of Renal Ischemia in Tetraethylammonium Transport in Rabbit Renal Coritcal Slices

  • This study was carried out to determine effect of acute renal ischemia on transport function of organic cation, tetraethylammonium (TEA), in rabbit kidney proximal tubule. Clamping of the renal artery for 30 and 60 min produced a polyuria which was accompanied by an increase in $Na^+$ excretion. The capacity of kidney cortical slices to accumulate TEA was increased after 30 and 60 min of ischemia. When blood flow was restored for 30 min after 30 and 60 min of ischemia, the augmented TEA uptake was recovered to the control values. Oxygen consumption of cortical slices was stimulated after 30 min of ischemia, whereas it was not altered by 60 min of ischemia. A 90-min ischemia produced a significant inhibition of TEA uptake and tissue oxygen consumption. These results suggest that the basolateral transport system for organic cation persists after ischemic periods of 60 min despite evidence that tubular reabsorptive mechanism of $Na^+$ and water is markedly impaired. This may indicate that the active secretory systems of proximal tubule are more resistant to ischemic injury than the reabsorptive systems.

Effect of Probenecid on Tetraethylammonium (TEA) Transport Across Basolateral Membrane of Rabbit Proximal Tubule

  • The effect of probenecid on the transport of tetraethylammonium (TEA) was studied in renal cortical slices and isolated membrane vesicles to investigate the interaction of organic anion with the organic cation transport system in proximal tubule. Probenecid reversibly inhibited TEA uptake by renal cortical slices in a dose-dependent manner over the concentration range of 1 and 5 mM. The efflux of TEA was not affected by the presence of 3 mM probenecid. Kinetic analysis indicated that probenecid decreased Vmax without significant change in Km. Probenecid inhibited significantly tissue oxygen consumption at concentrations of 3 and 5 mM. However, probenecid did not significantly reduce TEA uptake in brush border and basolateral membrane vesicles prepared from renal cortex even at a concentration as high as 10 mM. These results indicate that probenecid reduces TEA uptake in cortical slices by inhibiting tissue metabolism rather than by an interaction with the organic ration transporter.

Effect of Amino Acids on Anoxia-induced Cell Injury

  • This study was undertaken to examine the effect of amino acids on anoxia-induced cell injury in rabbit renal cortical slices. In order to induce anoxic cell injury, slices were exposed to a 100% $N_2$ atmosphere and control slices were exposed to 100% $O^2$. Irreversible cell injury was estimated by measuring lactate dehydrogenase (LDH) release and alterations in renal cell function were examined by measuring p-aminohippurate (PAH) uptake. Anoxia caused the increase in LDH release in a time-dependent manner. Glycine and glutathione almost completely prevented anoxia-induced LDH release. Of amino acids tested, glycine and alanine exerted the protective effect against anoxia-induced cell injury. However, asparagine with amide side chain, leucine and valine with hydrocarbon side chain, and basic amino acids (lysine, histidine, and arginine) were not effective. Anoxia-induced inhibition of PAM uptake was prevented by glycine. ATP content was decreased by anoxia, which was not affected by glycine. Anoxia-induced depletion of glutathione was significantly prevented by glycine. These results suggest that neutral amino acids with simple structure exert the Protective effect against anoxia-induced cell injury the involvement of specific interaction of amino acids and cell structure.
