Superoxide dismutases (SOD) are metalloenzymes that convert $O_2^-\;to\;H_2O_2$. Rehmannia glutinosa is highly tolerant to paraquat-induced oxidative stress. The primary objective of this study was to characterize regulation of SOD gene expression in R. glutinosa in response to oxidative stresses and hormones. A full-length putative SOD clone (RgCu-ZnSOD1) was isolated from the leaf cDNA library of R. glutinosa using an expressed sequence tag clone as a probe. RgCu-ZnSOD1 cDNA is 777 bp in length and contains an open reading frame for a polypeptide consisted of 152 amino acid residues. The deduced amino acid sequence of the clone shows highest sequence similarity to the cytosolic Cu-ZnSODs. The two to three major bands with several minor ones on the Southern blots indicate that RgCu-ZnSOD1 is a member of a small multi-gene family. RgCuZnSOD1 mRNA was constitutively expressed in the leaf, flower and root. The expression of RgCu-ZnSOD1 mRNA was increased about 20% by wounding and paraquat, but decreased over 50% by ethylene and $GA_3$. This result indicates that the RgCu-ZnSOD1 expression is regulated differentially by different stresses and phytohormones at the transcription level. The RgCu-ZnSOD1 sequence and information on its regulation will be useful in investigating the role of SOD in the paraquat tolerance of R. glutinosa.
Background: Rehmannia glutinosa is a perennial herb belonging to the family Scrophulariaceae. Its root has been utilized as a traditional medicine but the aerial parts (flower, flower stalk, leaf) are not used. We aimed to determine the content of three compounds [aucubin, catalpol, and ${\gamma}$-aminobutyric acid (GABA)] in the different organs of R. glutinosa cultivars (Dakang, Tokang, and Suwon 9) Methods and Results: The flower, flower stalk, leaf, and root of R. glutinosa were harvested at the end of August. The aucubin and catalpol contents were analyzed by LC/MS, whereas the GABA content was analyzed by GC/MS. The aucubin content was the highest in the leaf, while catalpol and GABA were the highest in the flower. The aucubin contents of leaf in Dakang, Tokang, and Suwon 9 were 1.43, 0.81, and 1.07 mg/g, respectively. The catalpol contents of flower in Dakang, Tokang, and Suwon 9 were 41.06, 28.78, and 37.48 mg/g, respectively, the GABA contents were 0.79, 0.76, and 0.65 mg/g, respectively. Conclusions: The aucubin, catalpol, and GABA contents were higher in the leaf and flower than that in the root. This study show that R. glutinosa leaf and flower can be used as a potential supplement.
Background: Rehmannia glutinosa is a perennial herb belonging to the family Scrophulariaceae. Its roots have been utilized as a traditional medicine. The aim of this study was to elucidate the basic information of the roots of the R. glutinosa cultivars and their utilization. Methods and Results: The roots of R. glutinosa cultivars were harvested in the end of March. The two iridoid glycosides, aucubin and catalpol, were analyzed by liquid chromatography/mass spectrometry (LC/MS), whereas ${\gamma}-aminobutyric$ acid (GABA) was analyzed by gas chromatography/mass spectrometry (GC/MS). The aucubin content was the highest in the Dakang cultivar, whereas no aucubin was detected in the five cultivars. All cultivars had more than 12 mg/g catalpol content, and the maximum catalpol content was found in Jihwang 1. The GABA content was the highest in Suwon 1, and it was 40 times more than that in the Yeongang cultivar. Conclusions: The highest aucubin, catapol and GABA contents were detected in the Dakang, Jihwang 1, and Suwon 1, cultivars respectively. This study provides the crucial information regarding the versatile utilization and pedigree selection of R. glutinosa cultivars.
Background: The use of Rehmannia glutinosa of the family Scrophulariaceae, in traditional medicine is well known. This study was conducted to elucidate the effect of air curing of the seed rhizome of R. glutinosa on its storability and yield. Methods and Results: The root of the R. glutinosa cultivar (Dagang) was harvested in at the end of November. The seed rhizomes were air cured for one to seven days. They were subsequently wrapped with newspaper and further stored in a plastic container at $1^{\circ}C$. It was observed that the weight loss and decay rates were significantly lower in the air cured seeds than in the untreated ones. Moreover, the decay rate of the control was approximately 50%, 120 days after storage. However. the decay rate of all the air cured treatment groups was less than 1%. Additionally, air curing led to an increase in the germination rate of the seeds and the root yield when compared with the untreated groups. Taken together, the ideal treatment period for air curing was found to be 3 days. Under these conditions, the germination rate and yield were 88.7% and 2,185 ㎏/10a, respectively. Conclusions: This study successfully demonstrated that the air curing of seed rhizomes can lead to a considerable increase in the storability and yield of R. glutinosa.
This experiment was carried out in order to collect the basic data on the standardization of the manufacturing process of Rehmannia glutinosa Libosch. var. purpurea Makino drying. By the drying methods of R. glutinosa, the content of water, inorganic components, reducing sugar, catalpol and benzo[${\alpha}$]pyrene were investigated. The water content was 15.6~17.2% when R. glutinosa was dried by cold-warm air moisture absorption drying method (CAMAD) at $60^{\circ}C$ during 6 days. Among of the inorganic components of R. glutinosa the K content was the most followed by P, Na, Ca and Mg. The reducing sugar content of R. glutinosa by the hot air drying method (HAD) was much more than that by the CAMAD. The catalpol content of R. glutinosa was not different by the drying temperature when it was dried by the CAMAD. The catalpol content of the large size tuber (about 50.0 g/unit) showed a tendency to increase from $60^{\circ}C$ until $70^{\circ}C$ drying temperature, but that of the small size tuber(about 4.0 g/unit) was decreased as being a trend as the drying temperature high when R. glutinosa was dried by the HAD, But the catalpol content R. glutinosa had a tendency to drop significantly at drying temperature above $80^{\circ}C$. The benzo[${\alpha}$]pyrene content was little detected when R. glutinosa was dried by both the SLD and the CAMAD, and the sampling by the HAD indicated within the scope of 5 ${\mu}g/kg$ which was the scope to regulate by Korean food and drug administration. In conclusion, it seemed that an appropriate drying temperature of R. glutinosa by the CAMAD and the HAD was about $60^{\circ}C$ and about $70^{\circ}C$, respectively, when we consider the catalpol content and benzo[${\alpha}$]pyrene detection in the manufacturing process of drying R. glutinosa.
Objectives : The aim of the present study is to evaluate the effects Rehmannia glutinosa, Radix puerariae, Semen coicis, Fructus hordei, Cortex acanthopanacis, Fructus chaenomelis, or Radix glycyrrhizae extracton 3T3-L1 preadipocytes differentiation. Methods : We adopted Oil Red O staining methods to observe the formed lipid droplets. And the amount of lipids in adipocytes was measured using absorptiometric analysis. Results : The extracts of Rehmannia glutinosa, Radix puerariae, Semen coicis, Fructus hordei, Cortex acanthopanacis, Fructus chaenomelis, or Radix glycyrrhizae stimulated the preadipocytes differentiation and lipid droplet formation. And the complex extract of the traditional herbal medicines stimulated there actions more than single extracts. Conclusions : The extract of Rehmannia glutinosa, Radix puerariae, Semen coicis, Fructus hordei, Cortex acanthopanacis, Fructus chaenomelis, or Radix glycyrrhizae has stimulatory effects. on adipogenesis. Moreover, the complex extract of the traditional herbal medicines has more effect than single extracts.
Background: Type I allergy is involved in allergic asthma, allergic rhinitis, and atopic dermatitis which are accompanied by an acute and chronic allergic inflammatory responses. Rehmannia glutinosa is a traditional medicine in the East Asian region. This study examined whether a Rehmannia Glutinosa pharmacopuncture solution (RGPS) had anti-allergic or anti-inflammatory effects in antigen-stimulated-RBL-2H3 cells. Methods: We determined the effect of RGPS on cell viability using the 3-[4,5-dimethylthiazolyl]-2,5-diphenyltetrazolium bromide (MTT) assay. We also examined the effect of RGPS on the release of ${\beta}$-hexosaminidase and the secretion of IL-4 and TNF-${\alpha}$ using ELISA. In addition, we evaluated the effect of RGPS on the mRNA expression of various cytokines; IL-2, IL-3, IL-4, IL-5, IL-13 and TNF-${\alpha}$ using RT-PCR. Furthermore, we assessed the activation of mitogen-activated protein kinases (MAPKs) and nuclear factor (NF)-${\kappa}$B using Western blotting after RGPS treatment. Results: We found that RGPS ($10^{-4}$ to $10^{-1}$ dilution) did not cause any cytotoxicity. We observed significant inhibition of ${\beta}$-hexosaminidase release and suppression of the protein secretion of IL-4 and TNF-${\alpha}$ and mRNA expression of multiple cytokines in antigen-stimulated-RBL-2H3 cells after RGPS treatment. Additionally, RGPS suppressed not only the phosphorylation of MAPKs, but also the transcriptional activation of NF-${\kappa}$B in antigen-stimulated-RBL-2H3 cells. Conclusions: These results suggest that RGPS inhibits degranulation and expression of cytokines including IL-4 and TNF-${\alpha}$ via down-regulation of MAPKs and NF-${\kappa}$B activation in antigen-stimulated-RBL-2H3 cells. In conclusion, RGPS may have beneficial effects in the exerting anti-allergic or anti-inflammatory activities.
1998년 $6{\sim}9$월의 강수량이 1,042mm, 강수일수가 60일이나 되어 배수조건이 나쁜 포장의 밭작물에 습해가 심하게 발생되었던 안동지역에서 습해정도에 따른 식방풍과 지황의 생육특성 및 수량을 농가포장에서 조사하였다. 식방풍은 습해가 심할수륵 지상부보다 지하부의 생육장해가 더 심하였으며, 주근의 신장생장이 비대생장보다 현저하게 억제되었다. 습해 정도별로 ha당 생근수량은 정상생육에 비하여 $21{\sim}80%$ 감소되었다. 지황은 습해 정도별로 고사주율이 52%였으며, 습해에 의하여 근경의 형성 및 발육이 장해를 받아 습해가 심할수록 주당 근경수와 근경의 직경이 크게 감소되었다. 따라서 습해가 심할수록 상품성이 높은 근경의 수량이 현저하게 감소되었으며, 습해 정도별로 ha당 근경의 생체수량은 정상생육에 비하여 $19{\sim}83%$감소되었다.
지황 (Rehmannia glutinosa) 우량종묘 생산을 위하여 조직배양에 의해 생산된 종묘의 포장수량성을 평가하고 지표성분인 Catalpol과 유리당 함량 특성을 비교한 결과는 다음과 같다. 조직배양주의 종근저장성은 재래종에 비하여 약간 양호한 경향이었다. 또한 근장, 근경 등의 뿌리생육이 왕성하여 조직배양주의 수량성은 서천재래종 384kg/10 a에 비하여 1년차 묘는 112%, 2년차 묘는 246%까지 증수되는 결과를 보였다. 성분함량 특성비교에서는 총당함량이 서천재래 1.86%에 비하여 조직배양 1, 2년차 묘는 각각 1.21, 1.10%를 보였고, 지표성분인 Catalpol도 서천재래 0.46%에 비하여 조직배양 1, 2년차묘는 0.4%로 약간 감소되는 경향이었다. 이상의 결과에서 지황 조직배양주의 수량성은 재래종에 비하여 증수되는 특성을 보였다. 그러나 유리당과 Catalpol 함량은 약간 감소되는 경향이었는데 모본식물과 큰 차이를 보이지는 않았다.
지황의 안정적인 저장으로 종근 비용을 절감하고자 재래종과 생육특성이 상이한 지황 1호를 공시하여 저장 후 120일 동안의 저장방법에 따른 종근 특성을 분석한 결과는 다음과 같다. 지황의 저장방법별 온도변화는 움저장에서 7.2∼13.8$^{\circ}C$로 가장 적었고 스치로폼상자내 흙을 충진한 저장에서 높았으며, 습도는 움저장에서 98.2%로 가장 높았다. 당도와 중량 감모율 그리고 부패율은 저장기간이 경과할수록 증가하는 경향을 보였으며, 왕겨를 충진한 망사와 스치로폼상자내 저장에서 당도와 중량 감모율이 높았으나, 부패율은 움저장에서 27.5%로 가장 높았다. 저장중 경도의 변화는 저장기간이 경과할수록 표피의 경도는 증가하는 경향이었으나, 내부 경도는 감소하는 경향을 보였다.
