배추과 작물의 자가불화합성 유전자의 발현 및 조절 (Expression and regulation of self-incompatible genes in Brassica)

  • In most self-incompatible plant species, recognition of self-pollen is controlled by a single locus, termed the S-locus. The self-incompatibility (SI) system in Brassica is controlled sporophytically by multiple alleles at a single locus, designated as S, and involves cell-cell communication between male and female. Two highly polymorphic S locus genes, SLG (S locus glycoprotein) and SRK (S receptor kinase), have been identified, both of which are expressed predominantly in the stigmatic papillar cell. Gain-of-function experiments have demonstrated that SRK solely determines S haplotype-specificity of the stigma, while SLG enhances the recognition reaction of SI. The sequence analysis of the S locus genomic region of B. campestris (syn. rapa) has led to the identification of an anther-specific gene, designated as SP11/SCR, which is the male S determinant. Molecular analysis has demonstrated that the dominance relationships between S alleles in the stigma were determined by SRK itself, but not by the relative expression level. In contrast, the expression of SP11/SCR from the recessive S allele was specifically suppressed in the S heterozygote, suggesting that the dominance relationships in pollen were determined by the expression level of SP11/SCR. Furthermore, recent studies on recessive allele-specific DNA methylation of Brassica self-incompatibility alleles demonstrate that DNA methylation patterns in plants can vary temporally and spatially in each generation. In this review, we firstly present overview of self incompatibility system in Brassica and then describe dominance relationships in Brassica self- incompatibility regulated by allele-specific DNA methylation.

식품의 안전성 및 식품첨가물에 대한 소비자 인식에 관한 연구 (A Study on the Consumer Recognition of Food Safety and Food Additives)

  • This study was to investigate consumer recognition of food safety and food additives. The results of this study are as follows: 1. When shopping for food, the food safety was rated as foremost concern for the respondents and the details checked most often were manufacture date, packing condition and axpiration date, respectively. When asked their opinion about harmful factors in relation to food safety, the most important consideration was food additives. The highest rating for credibility regarding the sources of information on food was given to scientists in university and institute, lowest one was given to food manufacturer. 2. Government regulation on the use of food additives was known to exist, but control of the uses of food additive was considered inadequate by 60% of the respondents. These results showed significant differences for age (p<0.05), for education level (p<.01) and for income (p<.05) respectively. 3. 47.9% of the respondents indicated that they were willing to purchase the additive free foods, although it was expensive. More than 70% made an effort to eat food with less food additive and were concerned about possible effects of food additives on health, showing significant differences for age (p<0.05), for education level (p<0.05) and for income (p<0.05). 4. Labeling for food additive found on food package was shown to be unsatisfactory, showing significant difference only for income (p<0.05) and more information about food was needed by over 91.3% of the respondents, showing significant difference for education level (p<0.05). 5. Although approximately 80% of the respondents were concerned about artificial preservative, artificial flavor, artificial color and artificial sweetener, the concern about artificial preservative was the highest. Primary causes that respondents felt fear toward food additives were the unknown harms and cancer, and primary sources that respondents got information on food were televison and radio, respectively. From these results, it was shown that consumer considered food safety important and they felt a great deal of concern about food additives.

비례 문제 해결에 영향을 주는 인지적 변인 분석 (Analysis on cognitive variables affecting proportion problem solving ability with different level of structuredness)

  • 이 연구는 비례문제 해결에 영향을 주는 인지적 변인이 무엇인지 확인하는 것을 궁극적인 목적으로 한다. 이를 위해 비례 문제를 구조화 정도에 따라 잘-구조화된 문제, 구조화된 문제, 비-구조화된 문제로 분류하고, 이론적 고찰을 통해 비례문제 해결에 영향을 주는 인지적 변인으로 사실 알고리즘 지식, 개념적 지식, 문제유형 지식, 양의 변화 인식, 메타인지를 추출하였다. 중다회귀분석 방법을 사용해 구조화 정도가 다른 문제를 해결하는데 유의하게 영향을 주는 인지적 변인이 무엇인지를 분석하였다. 분석 결과 구조화 정도가 다른 문제를 해결하는데 서로 다른 인지적 변인이 영향을 주었다. 즉 잘-구조화된 문제 해결에는 사실 알고리즘 지식과 문제유형 지식, 그리고 구조화된 문제 해결에는 개념적 지식, 문제유형지식, 양의 변화 인식, 마지막으로 비-구조화된 문제해결에는 메타조절, 개념적 지식, 양의 변화 인식, 문제유형지식이 영향을 주었다. 이처럼 문제 유형에 따라 다른 인지적 변인이 영향을 미치기 때문에, 수학수업에서는 문제 유형에 따라 다른 교수학습 방법과 다른 평가 틀을 적용할 필요가 있으며, 더불어 학생들의 비례 문제 해결 능력을 계발하기 위해서는 수학 수업에서 구조화된 문제와 비-구조화된 문제를 적극 활용할 필요가 있다는 결론을 도출할 수 있었다.

DNA microarray를 이용한 항진균 활성세균 Bacillus lentimorbus WJ5의 유전자 발현 분석 (DNA Microarray Analysis of Gene Expression in Antifungal Bacterium of Bacillus lentimorbus WJ5)

  • 여러 항진균 활성 관련 유전자들의 발현 수준을 동시에 연구하기 위하여 DNA microarray를 이용하여 유전자들의 발현 패턴을 비교 분석하였다. 본 연구에서는 항진균활성을 가지는Bacillus lentimorbus WJ5의 genomic DNA를 무작위 하게 제한효소로 절단하여 2,000개의 DNA단편을 microarray하였으며, 감마선($^{60}Co$)조사로 유도된 7종의 항진균 활성 결핍 돌연변이체와 발현양상을 정량적으로 비교하였다. Gene Cluster (Michael Risen, Stanford Uniy.)를 이용한 DNA microarray의 분석 결과, 총 408개의 DNA 단편이 발현되는 것을 확인할 수 있었으며, 이들 중 20개의 DNA단편이 항진균 활성 결핍 돌연변이체에서 발현이 억제되는 것으로 나타났다. 특히,pbuX (xanthine permease, K222), ywbA (phosphotransferase system enzyme II, K393), ptsG (PTS glucose specific enzyme II ABC component, K877), yufO (ABC transporter(ATP-binding protein), K1301), 그리고 ftsY (signal recognition particle (docking protein), K868)는 모든 돌연변이체에서 동시에 발현되는 down-regulation된 유전자들로서 물질 이동과 관련된 것으로 보고되어 있으며, 항진균 활성 관련 신호 및 물질의 이동에 관여할 것으로 사료되어진다.

학교급식 원산지 표시제 시행에 따른 일부 서울지역 초등학생의 인식도 (Recognition of Elementary School Students for The Country-of-Origin Labeling at School Foodservice in Seoul)

  • This study was conducted to offer basic data that give effective ways to inform the country-of-origin labeling, where the ingredients they are serving are from, at school foodservice and to reconsider the importance of the labeling origin based on the survey by the elementary school students. 96.0% of the elementary school students agreed to the regulation about the country-of-origin labeling and the older students were influenced more by media and also supported the labeling. About the tendency of ingesting food from the country the students didn't like, 69% of them disagreed to eat. In the ways to label the country-of-origin labeling at school foodservice, elementary school students recognized easily the indication of origin designed by menu items, letter type. 76% of elementary school students checked the country-of-origin labeling posted at restaurants. When the students eat out, 68% of them were unwilling to have the food using the ingredients from the country they don't like. The country-of-origin for main ingredients such as beef, pork, chicken and other meat products, rice, kimchi had high importance scores. We found that the students think about the country-of-origin for main ingredients is important. Consequently, education and public relations of the country-of-origin labeling for elementary school students would be required.

노인음성신호처리: 젠더혁신 분석에 대한 체계적 문헌고찰 (Elderly Speech Signal Processing: A Systematic Review for Analysis of Gender Innovation)

  • 본 연구의 목적은 국내의 젠더혁신 기반 노인음성신호처리 연구에 대하여 체계적으로 문헌을 고찰하고, 젠더분석 방법의 효용성과 혁신성을 소개하는 것이다. 2000년부터 현재까지 국내 학회지에 게재된 37편의 연구 논문 중 중복되는 자료와 포함 및 배제 기준에 따라 적합한 25편의 논문이 선정되었다. 그리고 젠더분석 방법을 적용하여 남녀 연구대상과 젠더연구 설계로 구분하여 살펴보았다. 연구 결과 젠더혁신 측면 공학연구 개발에서 연구 분야의 다양성과 연구 개발팀의 높은 젠더 인식이 필요함을 보였다. 또한 노인음성신호처리 연구 및 논문에서 젠더혁신 연구과정 및 방법이 체계적으로 적용될 수 있도록 정부 차원의 규정 확립과 대학 차원의 다양한 젠더혁신 프로젝트 출범을 제언하는 바이다. 추후 노인음성 신호처리 연구에서의 젠더혁신은 남녀 모두의 요구를 반영한 음성인식 시스템과 서비스 개발로 우리 모두를 위한 새로운 시장을 창출할 것이다.

공업화주택 인정제도 개선 방향 설정에 관한 연구 (A Study on the Improvement Directions for the Industrial Housing Accreditation System in Korea)

  • 최근 우리나라 건설업계는 건설기술 인력 부족, 건설노동인력 고령화, 주 52시간 근무 등 다양한 건설환경 대응 방안으로 공업화공법 활성화에 대한 요구가 증가하고 있다. 공업화주택 인정제도는 공업화주택의 품질인정과 품질 향상 유도를 위하여 1992년에 도입되었으나 현재 6개 업체만 인정을 받았고, 인정 받은 공업화주택이 건설된 실적도 전무한 상황이다. 현재 우리나라 공업화주택 인정제도는 크게 인정 및 인정 취소기준, 공업화주택 건설촉진 기준으로 구성되어 있다. 이에 본 연구에서는 공업화주택인정제도의 관련 규정을 심층분석하여, 각 규정에 따른 문제점을 도출하고, 이를 기반으로 공업화주택 인정제도 활성화를 위한 제도 개선방향을 도출하였다. 구체적인 공업화인정제도 방안으로는 현행 공업화 주택 인정 범위를 준주택까지 확대하는 방안, 공업화 주택의 성능 및 생산기준 개정, 공업화주택 활성화 지원을 위한 건축기준 완화 및 금융지원 등을 제안하였다. 향후 공업화주택 인정제도와 별도로 국내 공업화 건축시장이 활성화되지 못하는 주요 원인 중 하나인 경제성 문제를 해결하기 위해 공업화공법의 사회·경제·환경·기술적 파급효과에 대한 체계적 분석과 가시화를 통해 건설시장 패러다임을 전환을 적극적으로 유도할 필요가 있다.

NLRC4 Inflammasome-Mediated Regulation of Eosinophilic Functions

  • Eosinophils play critical roles in the maintenance of homeostasis in innate and adaptive immunity. Although primarily known for their roles in parasitic infections and the development of Th2 cell responses, eosinophils also play complex roles in other immune responses ranging from anti-inflammation to defense against viral and bacterial infections. However, the contributions of pattern recognition receptors in general, and NOD-like receptors (NLRs) in particular, to eosinophil involvement in these immune responses remain relatively underappreciated. Our in vivo studies demonstrated that NLRC4 deficient mice had a decreased number of eosinophils and impaired Th2 responses after induction of an allergic airway disease model. Our in vitro data, utilizing human eosinophilic EoL-1 cells, suggested that TLR2 induction markedly induced pro-inflammatory responses and inflammasome forming NLRC4 and NLRP3. Moreover, activation by their specific ligands resulted in caspase-1 cleavage and mature IL-1β secretion. Interestingly, Th2 responses such as secretion of IL-5 and IL-13 decreased after transfection of EoL-1 cells with short interfering RNAs targeting human NLRC4. Specific induction of NLRC4 with PAM3CSK4 and flagellin upregulated the expression of IL-5 receptor and expression of Fc epsilon receptors (FcεR1α, FcεR2). Strikingly, activation of the NLRC4 inflammasome also promoted expression of the costimulatory receptor CD80 as well as expression of immunoregulatory receptors PD-L1 and Siglec-8. Concomitant with NLRC4 upregulation, we found an increase in expression and activation of matrix metalloproteinase (MMP)-9, but not MMP-2. Collectively, our results present new potential roles of NLRC4 in mediating a variety of eosinopilic functions.

Regulation of Inflammation by Bidirectional Signaling through CD137 and Its Ligand

  • Although the majority of research on CD137 has been directed to T cells, it is becoming clear that this molecule has distinct functions in other lineages of cells, including non-hematopoietic cells. In particular, emerging evidence suggests that the CD137-its ligand (CD137L) network involving immune cells and non-immune cells, directly or indirectly regulates inflammation in both positive and negative manners. Bidirectional signaling through both CD137 and CD137L is critical in the evolution of inflammation: 1) CD137L signaling plays an indispensible role in the activation and recruitment of neutrophils by inducing the production of proinflammatory cytokines and chemokines in hematopoietic and non-hematopoietic cells such as macrophages, endothelial cells and epithelial cells; 2) CD137 signaling in NK cells and T cells is required for their activation and can influence other cells participating in inflammation via either their production of proinflammatory cytokines or engagement of CD137L by their cell surface CD137: 3) CD137 signaling can suppress inflammation by controlling regulatory activities of dendritic cells and regulatory T cells. As recognition grows of the role of dysregulated CD137 or CD137L stimulation in inflammatory diseases, significant efforts will be needed to develop antagonists to CD137 or CD137L.

여성 소비자의 식생활 라이프스타일에 따른 체중 조절 행위에 관한 연구 (A Study on Weight Control Behavior by Female Consumers Eating Life Styles)

  • The study investigated the eating lifestyles of female consumers and whether they influenced weight control behavior. The findings of the study can be summarized as follows. First, the eating lifestyles of the subjects were presented four types: 'active eating life pursuit type', 'safe economical type', 'gourmet type' and 'passive negligence type'. In general, most cases were of the 'passive negligence type'. Second, weight control behavior was analyzed according to eating lifestyle. The results of the analysis indicated that the female consumers of the 'active eating life pursuit type' and 'safe economy type' had high behavior levels in each aspect of weight control attitude, subjective regulation, perceived behavior control, and behavior intention. Third, the 'gourmet type', with a strong preference for taste, and the 'passive negligence type', with fewer eating considerations in general, showed weaker attitudes towards weight control and the recognition of pressure from people around them. As such, the level of consideration for food or eating was correlated with weight control. These findings impliedy that the more active, safer, and economical women are in their general eating style, the stronger they perceive weight control issues.
