Protective Effect of Sachungwhan against CCl4-induced Hepatotoxicity

  • Sachungwhan reduced hepatotoxicity induced by carbon tetrachloride($CCl_4$). Improved liver function was observed by measuring the activities of aspartate aminotransferase(AST), alanine aminotransferase(ALT), alkaline phosphatase(ALP), blood urea nitrogen(BUN), creatinine(CRE), total cholesterol(TCHO), triglyceride(TG), low density lipoprotein cholesterol(LDL-CHO), high density lipoprotein cholesterol(HDL-CHO), total protein(TP), albumin(ALB) and total bilirubin(BIL) in serum. Hepatic parameters monitored were levels of cholesterol(CHO), triglyceride(TG), malondialdehyde(MDA), content of cytochrome P450(CYP), level of glutathione(GSH), and activities of NADPH-CYP reductase, superoxide dismutase(SOD), catalase(CAT), glutathione S-transferase(GST), glutathione reductase(GR), glutathione peroxidase(GPx). The histopathological examination showed that the treatment of Sachungwhan relieved the ballooning degeneration of hepatocytes which had been generated by $CCl_4$. The results suggested that hepatoprotective effects of Sachungwhan possibly are due to their promising antioxidative activity.

Exposure to Sublethal Concentrations of Copper Pyrithione Reduces Cholinergic Activity and Induces Oxidative Stress in a Marine Polychaete

  • Despite concerns about the significant toxicity of copper pyrithione (CuPT) at environmental concentrations, effects of CuPT on benthic organisms have received little attention. Here, we analyzed the detrimental effects of CuPT at sublethal concentrations (1/50, 1/20, and 1/10 of the 96 h-LC50 value) for 14 days in the marine polychaete Perinereis aibuhitensis. Reduced burrowing activity and significantly decreased the acetylcholinesterase activity in response to relatively high concentrations of CuPT were identified as CuPT-triggered cholinergic inhibition. The lipid peroxidation marker, malondialdehyde levels were dose-dependently increased, whereas intracellular glutathione was depleted by relatively high concentrations. In the CuPT-treated polychaete, significant fluctuations in the enzymatic activities of the antioxidant defense system (catalase, superoxide dismutase, glutathione reductase, and glutathione peroxidase) were observed with significantly modulated glutathione 𝘚-transferase activity. These results indicate that even sublethal levels of CuPT would have detrimental effects on the health status of the marine polychaete.

β-Carotene의 섭취가 당뇨 유도 흰쥐의 간조직 항산화효소 활성과 Glutathione 함량에 미치는 영향 (Effect of Dietary Supplementation of β-Carotene on Hepatic Antioxidant Enzyme Activities and Glutathione Concentration in Diabetic Rats)

  • 본 연구에서는 당뇨병의 치료에 중요한 저해요인으로 제기되고 있는 당뇨합병증을 예방하고자 녹황색 채소 등을 통해 식사 시 한국인들이 쉽게 섭취할 수 있는 ${\beta}$-carotene의 혈관계 합병증 예방효과를 분석하고자 하였다. 간 미토콘드리아에서의 catalase 활성은 ${\beta}$-carotene을 급여한 비당뇨군에서 유의적으로 높은 활성을 나타내었으나 당뇨군에서는 ${\beta}$-carotene 급여에 의한 영향을 나타내지 않았다. 간 사이토졸 내의 superoxide dismutase 활성은 ${\beta}$-carotene을 급여하지 않은 당뇨군에서 그 활성이 가장 저하되었으나 ${\beta}$-carotene 공급에 의해 유의적으로 증가되었다. 간 사이토졸 내의 glutathione-S-transferase 활성은 ${\beta}$-carotene의 급여에 의한 차이가 없었다. 간 마이크로솜에서의 glucose-6-phosphatase 활성은 대조군에 비하여 ${\beta}$-carotene을 급여하지 않은 당뇨군에서 유의적으로 높게 나타났으나 ${\beta}$-carotene을 급여한 당뇨군에서는 활성이 유의적으로 저하되었다. 간조직의 환원형 glutathione 함량은 당뇨군에서 유의적으로 감소되었으며, 당뇨군에서 ${\beta}$-carotene의 급여로 증가하였다. 이상의 결과에서 볼 때 ${\beta}$-carotene의 섭취는 당뇨 유도에 의한 산화스트레스가 증가된 생리적 상태에서 glutathione의 소모를 감소시키고, 항산화 효소계 중 특히 superoxide dismutase 활성을 유도하여 과산화 라디칼을 환원시킴으로써 활성산소에 의해 유발되는 산화적 손상의 일차적 방어에 관여한 것으로 여겨진다. 따라서 인체시험 등을 통한 후속연구와 연계하여 ${\beta}$-carotene 수준을 적절하게 보충 섭취한다면 당뇨로 인한 항산화계 활성의 변화를 조절함으로써 산화스트레스에 대한 보호효과를 나타내어 당뇨병 합병증의 예방에 기여할 수 있을 것으로 여겨진다.

Effects of Hydroxybrazilin on Glutathione Depletion Induced by $\textrm{BrCCl}_3$ and Menadione in Cultured Rat Hepatocytes

  • In this study we investigated the effect of hydroxybrazilin on glutathione depletion induced by BrCCl$_3$ and menadione in cultured hepatocytes to understand the cellular mechanisms of hepatoprotective effect of hydroxybrazilin. Hydroxybrazilin alone had no effect on total glutathione level and the ratio of reduced glutathione/total glutathione (GSH/(GSSG+GSH)). BrCCl$_3$ dramatically decreased total glutathione level and hydroxybrazilin significantly prevented glutathion depletion by BrCCl$_3$. The ratio of GSH/(GSSG+ GSH) was also decreased by BrCCl$_3$ and recovered by hydroxybrazilin treatment. Menadione decreased total glutathione level and the ratio of GSH/(GSSG+GSH) but hydroxybrazilin showed no significant effects on menadione-induced glutathione depletion. These data suggest that hydroxybrazilin might prevent the hepatotoxicity induced by chemicalderived radicals but not the toxicity linked with oxidative stress.

Suppression of oxidative stress by grape seed supplementation in rats

  • Polyphenol-rich grape seeds have a beneficial effect on human health. The present study was performed to investigate the effects of grape seeds on antioxidant activities in rats. Male Sprague-Dawley rats were randomly divided into a control diet group (C), a high-fat diet group (HF), a 5% grape seed-supplemented control diet group (G), and a 5% grape seed-supplemented high-fat diet group (HG). Dietary supplementation with grape seeds reduced serum concentrations of lipid peroxides compared with those in the C and HF groups. The hepatic level of lipid peroxides decreased significantly in the grape seed groups compared with that in the C and HF groups. Superoxide dismutase activity in the G group increased significantly compared with that in the C group. Catalase activity tended to be higher by feeding grape seeds. The grape seed diet increased glutathione peroxidase activity in the C group. Glutathione-S-transferase activity increased significantly in the G group compared with that in the C group. Hepatic content of total glutathione increased significantly in the HG group but decreased significantly in the HF group. The ratio of reduced glutathione and oxidized glutathione increased by feeding the grape seed diet. Total vitamin A concentration was significantly higher in HG group than in other groups. Liver tocopherol content of the G and HG groups was significantly higher than that of the control groups. These results suggest that dietary supplementation with grape seeds is beneficial for suppressing lipid peroxidation in high fat-fed rats.

흰쥐에 있어서 사염화탄소에 의한 간손상에 allopurinol의 영향 (Effect of Allopurinol Pretreatment on the Liver Damage in $CCl_4$-treated Rat)

  • To evaluate the effect of xanthine oxidase on liver injury by $CCl_4$, liver damage was induced both in allopurinol pretreated rats (500 mg/kg. ip) and control group by twice intraperitoneal injection of $CCl_4$ (0.1 ml/100 g body wt. 50% in olive oil) at interval of one day. Increases in the levels of serum alanine aminotransferase and liver weight/body weight (%) by $CCl_4$ were significantly smaller inallopurinol pretreated rats than in control whereas the hepatic microsomal glucose-6-pholphatase activities were significantly higher in allopurinol pretreated rats than control group by $CCl_4$ treatment. These results indicates that allopurinol pretreatment may reduce the liver damage in $CCl_4$ intoxicated rats. In rats either with $CCl_4$or not, hepatic type O xanthine oxidase activities were significantly reduced by allopurinol pretreatment and the increasing rate of these enzymes to each control was remarkably lower in allopurinol pretreated rats than control. Liver cytosolic protein contents and aniline hydroxylase, aminopyrine demethylase activities were higher in allopurinol pretreated rats than coirol rats when animals were treated with $CCl_4$. On the other hand, neither allopurinol pretreated nor $CCl_4$ treatment caused any significant changes in hepatic superoxide dismutase and catalase activities. Hepatic glutathione contents were higher in $CCl_4$-treated rats than control, but no significant changes were found in both between the allopurinol treated rats and $CCl_4$-treated rats pretreated with allopurinol, and glutathione and glutathione S-transferase activities were significantly reduced in $CCl_4$-treated rats than control whereas these enzyme activities showed on significant change in both between allopurinel treated and $CCl_4$-treated rats pretreated with allopurinol. It is concluded that xanthine oxidase reaction system augment $CCl_4$ induced liver injury via even oxygen free radical system.

Chemopreventive Effect of Amorphophallus campanulatus (Roxb.) blume tuber against aberrant crypt foci and cell proliferation in 1, 2-dimethylhydrazine induced colon carcinogenesis

  • Colorectal cancer is one of the leading causes of cancer death, both in men and women. This study investigated the effects of Amorphophallus campanulatus tuber methanolic extract (ACME) on aberrant crypt foci (ACF) formation, colonic cell proliferation, lipid peroxidative damage and the antioxidant status in a long term preclinical model of 1, 2-dimethylhydrazine (DMH) induced colon carcinogenesis in rats. Male Wistar rats were divided into six groups, viz., group I rats served as controls; group II rats treated as drug controls receiving 250 mg/kg body weight of ACME orally; group III rats received DMH (20 mg/kg body weight) subcutaneously once a week for the first 15 weeks; groups IV, V and VI rats received ACME along with DMH during the initiation, post-initiation stages and the entire period of the study, respectively. All the rats were sacrificed at the end of 30 weeks and the intestinal and colonic tissues from different groups were subjected to biochemical and histological studies. Administration of DMH resulted in significant ($p{\leq}0.05$) intestinal and colonic lipid peroxidation (MDA) and reduction of antioxidants such as catalase, glutathione peroxidase, glutathione reductase, glutathione-Stransferase and reduced glutathione. Whereas the supplementation of ACME significantly ($p{\leq}0.05$) improved the intestinal and colonic MDA and reduced glutathione levels and the activities of antioxidant enzymes in DMH intoxicated rats. ACME administration also significantly suppressed the formation and multiplicity of ACF. In addition, the DMH administered rats showed amplified expression of PCNA in the colon and decreased expression of this proliferative marker was clearly noted with initiation, post-initiation and entire period of ACME treatment regimens. These results indicate that ACME could exert a significant chemopreventive effect on colon carcinogenesis induced by DMH.

백화사설초 메탄올 추출물이 acetaminophen으로 유도된 생쥐의 급성 간손상에 대한 효능 연구 (Effects of Oldenlandiae Diffusae Herba Methanol Extract on Acetaminophen Induced Acute Liver Injury in Mice)

  • Objectives : Oldenlandiae Diffusae herba has been used as a natural drug for tumor, inflammation and liver disease in traditional medicine. This study was performed in order to investigate the antioxidative effects of Oldenlandiae Diffusae herba methanol extract(ODHM) on acetaminophen induced acute liver injury in mice. Methods : In order to investigate the protective effect of ODHM on acute hepatic injury in vivo, ICR mice were pretreated with ODHM, and then treated with acetaminophen(500mg/kg). And the levels of LPO and glutathione(GSH), antioxidative enzyme activities were measured. The levels of LPO were measured by TBA method. And catalase activity was measured as the decrease in hydrogen peroxide absorbance at 240nm on spectrophotometer using 30mM hydrogen peroxide. Superoxide dismutase(SOD) was assayed by recording the inhibition of nitro blue tetrazolium reduction with xanthine and xanthine oxidase. Glutathione peroxidase(GPX) activity was determined by the modified coupled assay developed by Paglia and Lawrence. The reaction was started by addition of 2.2mM hydrogen peroxide as substrate. The change in absorbance at 340nm was measured for 1min on spectrophotometer. Glutathione-S-transferase(GST) activity was assayed with CDNB as substrate and enzyme activity of GST towards the glutathione conjugation of CDNB. And Total SH and GSH levels were measured. Results : In vivo study, LPO levels of acetaminophen treatment group were significantly higher than other groups. This increased level was significantly reduced by ODHM pretreatment. The acetaminophen treatment resulted in a decrease of catalase, GPX, SOD and GST activities. By contrast, ODHM pretreatment markedly increased compare to those of untreated groups. Total SH and GSH levels were reduced by of acetaminophen treatment, and ODHM pretreatment significantly increased GSH levels.

Multimerization of Bovine Thyroglobulin, Partially Unfolded or Partially Unfolded/Reduced; Involvement of Protein Disulfide Isomerase and Glutathionylated Disulfide Linkage

  • Fate of the nascent thyrolglobulin (Tg) molecule is characterized by multimerization. To establish the formation of Tg multimers, the partially unfolded/reduced Tg or deoxycholate-treated/ reduced Tg was subjected to protein disulfide isomerase (PDI)-mediated multimerization. Oxidized glutathione/PDI-mediated formation of multimeric Tg forms, requiring at least an equivalent molar ratio of PDI/Tg monomer, decreased with increasing concentration of reduced glutathione (GSH), suggesting the oxidizing role of PDI. Additional support was obtained when PDI alone, at a PDI/Tg molar ratio of 0.3, expressed a rapid multimerization. Independently, the exposure of partially unfolded Tg to GSH resulted in Tg multimerization, enhanced by PDI, according to thiol-disulfide exchange. Though to a lower extent, a similar result was observed with the dimerization of deoxycholate-pretreated Tg monomer. Consequently, it is implied that intermolecular disulfide linkage may be facilitated at a limited region of unfolded Tg. In an attempt to examine the multimerization site, the cysteine residue-rich fragments of the Tg were subjected to GSH-induced multimerization; a 50 kDa fragment, containing three vicinal dithiols, was multimerized, while an N-terminal domain was not. Present results suggest that the oxidase as well as isomerase function of PDI may be involved in the multimerization of partially unfolded Tg or deoxycholate-treated Tg.

양파 추출물에서의 글루타티온 전달효소 활성 저해제에 관한 연구 (A Study of Glutathione S-transferase Inhibitors obtained from Allium cepa var. cepa Extract)

  • In this study, an ethanol extract being obtained from Allium cepa var. cepa examins the inhibitory effects on the glutathione S-transferase and the separation had been done by silica-gel column chromatography using various eluents, such as ethyl acetate, methanol, and 50% methanol. A volume of column fraction was 50ml and evaporation has been performde by the rotary evaporator under reduced pressure. Each fraction is being examined by thin layer chromatography and the UV spectrum at 365 nm was used to investigate separation patterns of spots on thin layer chromatography. When the eluent was changed, the spot patterns showed another different pattern on thin layer chromatography, so on. Fractions showing similar pattern are combined and eventually, three fractions are obtained. Each fraction is testified to examine the inhibition effects on glutathione S-transferase. All of these showed inhibition effects on glutathione S-transferase. The GC-MS shows that each fraction contains more than 2 compounds.
