Transmission and Histochemical Detection of Mulberry Dwarf Mycoplasma in Several Herbaceous Plants (뽕나무 오갈병 마이코플라스마의 몇가지 초본식물에의 전염과 조직화학적 검정)

  • Transmission of mulberry dwarf mycoplasma (MDM) from diseased mulberry to 5 herbaceous plants (periwinkle, white clover, Ladino clover, red clover, and Chinese milk vetch) through insect vector, Hishimonus sellatus, was confirmed by symptom expression and microscopic evidences. The earliest symptom appearance was noticed on periwinkle in which incubation period was 25-30 days, while it ranged 35-40 days in the other plant species. The common symptoms of MDM infected plants were characterized by poor plant growth with accompanying discolorations of leaves (chlorosis with vein clearing in periwinkle, reddish in red clover, brownish in white and Ladino clovers, and yellowish in Chinese milk vetch). Mycoplasmal infections were diagnosed light microscopically by Dienes' and toluidine blue staining of hand-cut and Epon-embedded sections, respectively. In Dienes' stain, all the plants infected with MDM showed specific staining reaction in phloems. In toluidine blue stain, mycoplasmal existence was noted by granular appearance in sieve tubes which were confirmed to be mycoplasma-like organisms under an electron microscope.

Effect of N Levels on the Herbage Yield and Quality of Orchargrass-Red Clover Mixtures (Orchargrass-Red Clover 초지의 N 시비수준이 목초의 수량과 품질에 미치는 영향)

  • The objective of this experiment was to suggest the optimum nitrogen fertilization level for orchardgrass(0G)- red clover(RC) mixtures. The field trials were conducted from 1993 to 1994 in order to evaluate the dry matter yield, botanical composition, chemical composition, dry matter digestibility(DMD), crude protein dry matter (CPDM) and digestible dry matter(DDM) yields on the nitrogen levels( 0, 50, 100, 150 and 300kgha). The results obtained were summarized as follows : 1. With increasing nitrogen levels, the DM yield of OG-RC mixttures significantly increased(P<0.05). The maximum total DM yield (7,964kgha) was obtained at N 300kgha level, However, there was no significant difference between N 150 and 300kgha. 2. With increasing the nitrogen levels, the botanical composition of OG in OG-RC mixttures was significantly increased(P< 0.05), whereas that of RC was remarkably decreased(P< 0.05). 3.   • This experiment was conducted to select acid tolerant cells from red clover suspension culture and obtained following results. The most effective condition of plant growth regulators for suspension culture of red clover cells was 2 mg/l 2,4-D as auxin source plus 0.5 mg/l BAP as cytokinin source. Among serveral basal media, PC medium brought the best result. The most suitable EMS concentration for the mutated cells tolerant to acid was 1 % with fours treatment and average 27 colonies/plate were selected. The frequency of the occurence of mutants tolerant to acid in 1 % EMS with four hours treatment was 8.9 $\times$ 10$^{-5}$ which was 100 times larger than that of control. The total colonies selected initially were 176 but the survived colonies after two subcultures with 4 weeks interval in a selection medium were 44.

Effect of Cover Crops on the Soil Properties and Fruit Quality in a Persimmon Orchard (녹비작물이 단감과원의 토양 특성과 과실품질에 미치는 영향)

  • This study was conducted into the effects of cover crops among the hairy vetch, red clover, rye, and hairy vetch+rye on the soil chemical and physical properties, and fruit yield in mature 'Fuyu' persimmon (Diospyros${\times}$kaki Thunb.) trees. The shallow-rooted red clover had poor dry matter production, resulting in the lowest coverage (66%) on the orchard floor. In contrast, the highest dry matter production observed in rye and hairy vetch+rye. Estimated N, P and K production from the cover crops were the highest on the hairy vetch+rye plots, increasing soil chemicals at a depth of 0-30 cm soil. Rye or hairy vetch+rye treatments decreased the soil bulk density and solid phase. As the hairy vetch+rye treatment increased fruit yield and sugar contents, it could be proposed as an suitable cover crop for improving productivity of persimmon trees.

A Comparative Study of Dry Matter Yield and Nutritive Value of Tall type and Tail + Short type Mixtures (상번초 및 상ㆍ하번초형 혼파초지의 건물수량 및 사료가치 비교 연구)

  • This study was conducted to investigate the effect of tall type mixtures and tall+short type mixtures on dry matter yield, botanical composition and quality. The experimental design includes two mixture types: Tall type mixtures(TM); orchardgrass(Potomac) 40+tall fescue(Fawn) 20+Festulolium braunii (Paulita) 10+perennial ryegrass(Reveille) 10+timothy(Climax) 10+red clover(Kenland) 5+alfalfa(Vernal) 5%) and tall+short type mixtures(TSM); orchardgrass(Potomac) 40+tall fescue(Fawn) 20+Kentucky bluegrass(Newport, turf type) 10+redtop(Barricuda, turf type) 10+perennial ryegrass(Palmer III, turf type) 10+red fescue(Flyer II, turf type) 5+white clover(Regal) 5%. The dry matter(DM) yield was higher obtained in TSM than that of TM, but there was no significant difference between TM and TSM. In the chemical composition, there was no significant difference between TM and TSM, but the content of cellulose, lignin were higher in TM than those in TSM(p<0.05). Also, the content of acid detergent fiber which affected dry matter digestibility was higher in TM than that of TSM. The crude protein dry matter yield was not observed significant difference between TM and TSM, but digestible dry matter(DDM) yield was higher in TSM than that of TM. In botanical composition, tall fescue, alfalfa and orchardgrass were highly maintained in TM but white clover, tall fescue and orchardgrass were highly, and redtop, red fescue were continuously maintained in TSM. In this experiment, DM yields and quality of two mixture types were no difference. However, the tall+short type mixtures(TSM) were more effective in enhancing the dry matter digestibility and DDM yield, and in maintaining the various botanical composition.

A Comparative Study on the Herbage Utilization from Tall type and Tall+short type Mixtures by Korean Native Goats (상번초 및 상ㆍ하번초형 혼파초지의 산양에 의한 초류이용성 비교 연구)

  • This study was conducted to investigate the effect of tall type mixtures(TM) and tall + short type mixtures(TSM) on nutritive utilization. DM intake, digestibility and utilization of nitrogen and energy of herbages from pastures by Korean native goats were determined. This experiment was conducted by total collection method in laboratory, 2000. The experimental herbage included two treatments: Tall type mixtures(orchardgrass, Potomac 40+tall fescue, Fawn 20+Festulolium braunii, Paulita 10+perennial ryegrass. Reveille 10+timothy, Climax 10+red clover, Kenblue 5+alfalfa, Vernal 5%), Tall+short type mixtures {orchardgrass, Potomac 40+tall fescue, Fawn 20+Kentucky bluegrass, New port(turf type) 10+redtop, Barricuda(turf type) 10+perennial ryegrass, Palmer III(turf type) 10+red fescue, Flyer II(turf type) 5+white clover, Regal 5%}. The voluntary dry matter(DM) intake of Korean native goats fed with herbages from tall+short type mixtures(TSM) was slightly higher than that of tall type mixtures(TM), but no difference was observed between TSM and TM(p > 0.05). The digestibility of DM, neutral detergent fiber, cellular contents and acid detergent fiber of herbage from TSM was slightly higher than that of TM, but no difference was detected between TSM and TM(p > 0.05). The utilization of nitrogen and energy by Korean native goats did not showed any difference in herbages from TSM and TM(p > 0.05). In conclusion, the herbage utilization by Korean native goats did not showed any significant difference. Thus, there is a possibility that turf grasses in mixtures can be used as a forage sources.

Effects of Rotatory Speed of Coating Pan on the Physical Characteristics of Coated Seeds (종자피복기의 피복통 회전속도가 피복종자의 물리적 특성에 미치는 영향)

  • Using a small pilot coating pan, red clover and tall fescue seeds were coated under different rotating speed of pan(20, 30, 40, 45, 55, 60 rpm) for standardization of seed coating. Vermiculite was used as particulate matter and polyvinyl alcohol as adhesive for the coating of seeds. Coating index was calculated based on the percent singles out, percent agglomerates out, weight of particulate matter fine, percent friability, and average weight per 100 seeds to evaluate the physical characteristics of coated seeds. The coated seed of red clover under 45 rpm rotating speed of pan, was best in terms of percent singles out, percent agglomerates out, weight of fines, and average 100 seed weight. The coated seed of tall fescue under 40 rpm rotating speed of pan, showed highest percent singles out, lowest percent agglomerates out, lowest weight of fines, and heaviest average 100 seed weight. Excellent coating results were obtained with the pan speed of 45rpm for red clover and 40 rpm for tall fescue seeds. As rotatory speed of coating pan affected much to the physical characteristics of coated seeds, standardization of the speed is required before seed coating.

Effect of mixed hay supplementation during fattening on carcass traits and meat quality of Hanwoo steers

  • Background: This study was aim to observe the effects of feeding mixed local hay (MH) consisted of 55% orchard grass (Dactylis glomerata L.), 35% tall fescue (Festuca arundinacea) and 10% red clover (Trifolium pratense) to Hanwoo steers on performance, carcass characteristics and meat quality (longissimus thoracis) compared with feeding imported timothy hay (TH) and local rice straw (RS). Results: Although no significant effects were found on animal performance and carcass yield grade, the carcasses of MH group had higher marbling score and quality grade than those of RS and TH group (P < 0.05). Therefore, higher fat content (P < 0.001), lower shear force and hardness value in the beef of MH group than that of other groups were observed. Furthermore, the beef of MH group had higher CIE $a^*$ value (redness) than that of other groups and feeding MH to Hanwoo steers lowered n-6 to n-3 fatty acids ratio in beef. Conclusions: Mixed hay provided benefits on meat quality and could be used for Hanwoo fattening program.

Effect of Some Factors Affecting Callus Formation of Red Clover (레드 클로버의 캘러스 유도에 미치는 몇가지 요인의 영향)

  • This experiment was conducted to determine factors affecting callus formation of some varieties of red clover. Growth regulators. basal medium, medium pH, agar concentration, and plant age for explant were investigated. The result obtained were as follows. As auxin source, 0.1 mg/ l picloram was found to be effective on callus formation. Callus formation was the highest on PC medium among several growth media. The callus produced on PC medium was more friable than those produced on the other media. The medium pH 5.8 gave the best response for callus formation. The effective seedling age for callus formatiom was around 5-7 days old. In agar concentration, 0.5-0.8%(WN) was suitable for callus formation.
