Exploring mixotrophy of dinoflagellate species is critical to understanding red-tide dynamics and dinoflagellate evolution. Some species in the dinoflagellate genus Karenia have caused harmful algal blooms. Among 10 Karenia species, the mixotrophic ability of only two species, Karenia mikimotoi and Karenia brevis, has been investigated. These species have been revealed to be mixotrophic; however, the mixotrophy of the other species should be explored. Moreover, although K. mikimotoi was previously known to be mixotrophic, only a few potential prey species have been tested. We explored the mixotrophic ability of Karenia bicuneiformis, Karenia papilionacea, and Karenia selliformis and the prey spectrum of K. mikimotoi by incubating them with 16 potential prey species, including a cyanobacterium, diatom, prymnesiophyte, prasinophyte, raphidophyte, cryptophytes, and dinoflagellates. Cells of K. bicuneiformis, K. papilionacea, and K. selliformis did not feed on any tested potential prey species, indicating a lack of mixotrophy. The present study newly discovered that K. mikimotoi was able to feed on the common cryptophyte Teleaulax amphioxeia. The phylogenetic tree based on the large subunit ribosomal DNA showed that the mixotrophic species K. mikimotoi and K. brevis belonged to the same clade, but K. bicuneiformis, K. papilionacea, and K. selliformis were divided into different clades. Therefore, the presence or lack of a mixotrophic ability in this genus may be partially related to genetic characterizations. The results of this study suggest that Karenia species are not all mixotrophic, varying from the results of previous studies.
Yellow clay dispersion has been applied to minimize fisheries impact by the red tide Cochlodinium polykrikoides blooms in Korean coasts since 1995. The present preliminary study documents the effect of yellow clay on Korean rockfish, Sebastes schlegelii, in terms of oxygen consumption rate (OCR). The OCR in the low clay suspension (0.05 and 0.23 %, w/w) showed normal level compared to the control. In contrast, the OCR for each one of three replicates in the high clay suspension (1.16 and 5.58 %, w/w) was not returned to the previous level that clay was not treated, indicating that high clay suspension (${\geq}1.16%$, w/w) might give negative effect on Korean rockfish. Overall, this result suggests that field application of clay to control Harmful Algal Blooms (HABs) may not give impact on Korean rockfish once the clay is dispersed in a low concentration (${\leq}0.23%$). In order to understand the changes of OCR in the repeated exposure to clay, it is required to do further studies on the changes of OCR when the fish is exposed to clay repeatedly after recovery in the normal seawater.

Heterotrophic dinoflagellate Pfiesteria piscicida (Dinophyceae) has been claimed to produce potent ichthyotoxins that cause disorientation and eventually death of fish and other marine animals. A real-time PCR probe targeting for SSU rRNA gene was used for detection of P. piscicida in Chinhae Bay, Korea. PCR inhibitors were successfully removed by dilution of template DNA. Positive detections were shown from surface water samples indicating the presence of P. piscicida in Chinhae Bay.
Periodically, harmful algal blooms (HABs) have occurred, with impacts on various areas including public health, tourism, and aquatic ecosystems, especially aquacultured and caged fisheries. To prevent or manage invasions of HABs into fish farms on an emergency basis, many methods have been proposed. Frequently over the past 30 years in coastal countries, treatments of clay and clay mixed with polyaluminum chloride (PAC) and chitosan have been tested for HAB-removal effectiveness in both the laboratory and the field. In Korea, yellow loess clay (hwangto) has been dispersed using electrolytic clay dispensers, both to decrease the amount of yellow loess clay's usage in containers and enhance HAB-removal efficiency. However, this emergency method has limitations, among which is the requirement for more effective controlling agents for field applications. Thus, in this paper, we review technologies for clay-based red tides prevention and control and their limitations, and, further, introduce next-generation algicidal technologies for the emergency protection of fish farms.
Relative DNA contents in some harmful algae were measured using DAPI staining and image analysis system. This method was useful to identify some morphologically similar species and isolates from harmful algal blooms (HABs). In exponential phase, Prorocentrum micans had higher relative DNA content (RD) of $1.83\pm0.52$ than any other isolates, followed by Cochlodinium polykrikoides $(1.10\pm0.46)$ Alexandrium tamarense $(0.93\pm0.32)$ Gyrodinium impudicum $(0.56\pm0.17)$, Scrippsiella trochoidea $(0.41\pm0.26)$ and P. minimum$(0.05\pm0.01)$. When they were fixed with Lugol's solution, it was difficult to d,iscern C. polykrikoides from G. impudicum under the light microscope, but the DNA contents were quite different in two species. C. polykrikoides contained about twice as much RD as G. impudicum under the same culture conditions and exponential phase. DAPIstained DNA feature in C. polykrikodes showed concentrated in the peripheral part of the cell, but in G. impudicum showed a compact structure in the central part. Although A. tamarense and S. trochoidea were morphologically similar under the light microscope, nuclear DNA content of A. tamarense was twice as much as that of S. trochoidea.
Algicidal bacterium was isolated from sea water during the declining period of Cochlodinium polykrikoides blooms and this bacterium had a significant algicidal activity against C. polykrikoides. In this study, algicidal bacterium was identified on the basis of biochemical and chemotaxonomic characteristics, and analysis of 16S rDNA sequences. The algicidal bacterium showed 98.6% homology with Micrococcus luteus ATCC $381^T$. Therefore, this bacterium was designated Micrococcus luteus SY-13. The optimal culture conditions of the algicidal bacterium was $25^{\circ}C$, initial pH 8.0, and 3.0% NaCl concentration. M. luteus SY-13 is assumed to produce secondary metabolites which have algicidal activity. When 10% culture filtrate of this strain was applied to C. polykrikoides ($1.0\;{\times}\;10^4\;cells/ml$) cultures, over 98% of C, polykrikoides cells were destroyed within 6 hours. The culture filtrate of M. luteus SY-13 exhibited similar algicidal activity after heat-treatment at $121^{\circ}C$ for 15 min. While algicidal activity remained in filtrates with pH adjusted to 8.0, loss of algicidal activity occurred when the pHs of filtrates were adjusted to over 9.0 or heat-treated at $121{\times}180^{\circ}C$ for 1 hour. M. luteus SY-13 showed significant algicidal activities against C. polykrikoides (98.9%) and a wide algicidal range against various harmful algal bloom (HAB) species. However, there was no algicidal effect on diatom and marine livefood organisms except Isocrysis galbana. These results suggest that M. luteus SY-13 could be a candidate for use in the control of HABs.

Harmful algal blooms (HAB) caused by the dominant species Cochlodinium polykrikoides (C. polykrikoides) appear in the South Sea of Korea and are particularly present in summer and fall seasons. Environmental factors such as water temperature, weather conditions (air temperature, cloud cover, sunshine, precipitation and wind) influence on the initiation and subsequent development of HAB. The purpose of this research was to study spatial and temporal variations of HAB in the Yeosu area using environmental (oceanic and meteorological) and satellite data. Chlorophyll-a concentrations were calculated using Sea-viewing Wide Field-of-view Sensor (SeaWiFS) images by an Ocean Chlorophyll 4 (OC4) algorithm, and HAB were estimated using the Red tide index Chlorophyll Algorithm (RCA). We also used the surface velocity of sequential satellite images applying the Maximum Cross Correlation method to detect chlorophyll-a movement. The results showed that the water temperature during HAB occurrences in August 2002-2008 was $19.4-30.2^{\circ}C$. In terms of the frequency of the mean of cell density of C. polykrikoides, the cell density of the HAB found at low (<300 cells/ml), medium (300-1000 cells/ml), and high (>1000 cells/ml) levels were 27.01%, 37.44%, and 35.55%, respectively. Meteorological data for 2002-2008 showed that the mean air temperature, precipitation, wind speed and direction, and sunshine duration were $22.39^{\circ}C$, 6.54 mm/day, 3.98 m/s (southwesterly), and 1-11.7 h, respectively. Our results suggest that HAB events in the Yeosu area can be triggered and extended by heavy precipitation and massive movement of HAB from the East China Sea. Satellite images data from July to October 2002-2006 showed that the OC4 algorithm generally estimated high chlorophyll-a concentration ($2-20\;mg/m^3$) throughout the coastal area, whereas the RCA estimated concentrations at $2-10\;mg/m^3$. The surface velocity of chlorophyll-a movement from sequential satellite images revealed the same patterns in the direction of the Tsushima Warm Current.

The dinoflagellate genus Alexandrium is known to often form harmful algal blooms causing human illness and large-scale mortality of marine organisms. Therefore, the population dynamics of Alexandrium species are of primary concern to scientists and aquaculture farmers. The growth rate of the Alexandrium species is the most important parameter in prediction models and nutrient conditions are critical parameters affecting the growth of phototrophic species. In Korean coastal waters, Alexandrium affine and Alexandrium fraterculus, of similar sizes, often form red-tide patches together. Thus, to understand bloom dynamics of A. affine and A. fraterculus, growth rates and nitrate uptake of each species as a function of nitrate ($NO_3$) concentration at $100{\mu}mol\;photons\;m^{-2}s^{-1}$ under 14-h light : 10-h dark and continuous light conditions were determined using a nutrient repletion method. With increasing $NO_3$ concentration, growth rates and $NO_3$ uptake of A. affine or A. fraterculus increased, but became saturated. Under light : dark conditions, the maximum growth rates of A. affine and A. fraterculus were 0.45 and $0.42d^{-1}$, respectively. However, under continuous light conditions, the maximum growth rate of A. affine slightly increased to $0.46d^{-1}$, but that of A. fraterculus largely decreased. Furthermore, the maximum nitrate uptake of A. affine and A. fraterculus under light : dark conditions were 12.9 and $30.1pM\;cell^{-1}d^{-1}$, respectively. The maximum nitrate uptake of A. affine under continuous light conditions was $16.4pM\;cell^{-1}d^{-1}$. Thus, A. affine and A. fraterculus have similar maximum growth rates at the given $NO_3$ concentration ranges, but they have different maximum nitrate uptake rates. A. affine may have a higher conversion rate of $NO_3$ to body nitrogen than A. fraterculus. Moreover, a longer exposure time to the light may confer an advantage to A. affine over A. fraterculus.

The ichthyotoxic Cochlodinium polykrikoides red tides have caused great economic losses in the aquaculture industry in the waters of Korea and other countries. Predicting outbreak of C. polykrikoides red tides 1-2 weeks in advance is a critical step in minimizing losses. In the South Sea of Korea, large C. polykrikoides red tide patches have often been recorded offshore and transported to nearshore waters. To explore the processes of offshore C. polykrikoides red tides, temporal variations in 3-dimensional (3-D) distributions of red tide organisms and environmental parameters were investigated by analyzing 4,432 water samples collected from 2-5 depths of 60 stations in the South Sea, Korea 16 times from May to Nov, 2014. In the study area, the vegetative cells of C. polykrikoides were found as early as May 7, but C. polykrikoides red tide patches were observed from Aug 21 until Oct 9. Cochlodinium red tides occurred in both inner and outer stations. Prior to the occurrence of large C. polykrikoides red tides, the phototrophic dinoflagellates Prorocentrum donghaiense (Jun 12 to Jul 11), Ceratium furca (Jul 11 to Aug 21), and Alexandrium fraterculus (Aug 21) formed red tides in sequence, and diatom red tides formed 2-3 times without a certain distinct pattern. The temperature for the optimal growth of these four red tide dinoflagellates is known to be similar. Thus, the sequence of the maximum growth rates of P. donghaiense > C. furca > A. fraterculus > C. polykrikoides may be partially responsible for this sequence of red tides in the inner stations following high nutrients input in the surface waters because of heavy rains. Furthermore, Cochlodinium red tides formed and persisted at the outer stations when $NO_3$ concentrations of the surface waters were < $2{\mu}M$ and thermocline depths were >20 m with the retreat of deep cold waters, and the abundance of the competing red-tide species was relatively low. The sequence of the maximum swimming speeds and thus potential reachable depths of C. polykrikoides > A. fraterculus > C. furca > P. donghaiense may be responsible for the large C. polykrikoides red tides after the small blooms of the other dinoflagellates. Thus, C. polykrikoides is likely to outgrow over the competitors at the outer stations by descending to depths >20 m and taking nutrients up from deep cold waters. Thus, to predict the process of Cochlodinium red tides in the study area, temporal variations in 3-D distributions of red tide organisms and environmental parameters showing major nutrient sources, formation and depth of thermoclines, intrusion and retreat of deep cold waters, and the abundance of competing red tide species should be well understood.

Species belonging to the dinoflagellate genus Prorocentrum are known to cause red tides or harmful algal blooms. To understand the dynamics of a Prorocentrum sp., its growth and mortality due to predation need to be assessed. However, there are only a few Prorocentrum spp. for which heterotrophic protist predators have been reported. We explored feeding by the common heterotrophic dinoflagellates Gyrodinium dominans, Oxyrrhis marina, Pfiesteria piscicida, Oblea rotunda, and Polykrikos kofoidii and the naked ciliate Strombidinopsis sp. (approx. 90 ㎛ cell length) on the planktonic species Prorocentrum triestinum, P. cordatum, P. donghaiense, P. rhathymum, and P. micans as well as the benthic species P. lima and P. hoffmannianum. All heterotrophic protists tested were able to feed on the planktonic prey species. However, O. marina and O. rotunda did not feed on P. lima and P. hoffmannianum, while G. dominans, P. kofoidii, and Strombidinopsis sp. did. The growth and ingestion rates of G. dominans and P. kofoidii on one of the seven Prorocentrum spp. were significantly different from those on other prey species. G. dominans showed the top three highest growth rates when it fed on P. triestinum, P. cordatum, and P. donghaiense, however, P. kofoidii had negative growth rates when fed on these three prey species. In contrast, P. kofoidii had a positive growth rate only when fed on P. hoffmannianum. This differential feeding on Prorocentrum spp. between G. dominans and P. kofoidii may provide different ecological niches and reduce competition between these two common heterotrophic protist predators.
