Seasonal changes of tide signal(s), temperature, salinity and current were studied during the years 2004-2005 in the northernmost Gulf of Aqaba, which is under developmental activities, to obtain scientific bases for best management and sustainability. Spectrum analysis revealed permanent signals of tide measurements during all seasons, which represented semidiurnal and diurnal barotropic tides. The other signal periods of 8.13, 6.10-6.32, 4.16 and 1.02-1.05 h were not detected in all seasons, which were related to shallow water compound and overtides of principle solar and lunar constituent and to seiches generated in the Red Sea and the Gulf of Aqaba. Spatial and temporal distribution of temperature, salinity and density showed significant differences between months in the coastal and offshore region and no significant differences among the coastal sites, between the surface and bottom waters and between coastal and offshore waters. Therefore, the temporal and spatial variation of water properties in the northernmost Gulf of Aqaba behave similarly compared to other parts. The coastal current below 12 m depth was weak $(3-6\;cms^{-1})$ and fluctuated from east-northeastward to west-southwestward (parallel to the shoreline), which may be related to the effect of bottom topography and/or current density due to differential cooling between eastern and western parts in the study area, and wind-induced upwelling and downwelling in the eastern and western side, respectively. The prevailing northerly winds and stratification conditions during summer were the main causes of the southward current at 6 and 12 m depths with average speed of 28 and $12cms^{-1}$ respectively.

Species belonging to the dinoflagellate genus Prorocentrum are known to cause red tides or harmful algal blooms. To understand the dynamics of a Prorocentrum sp., its growth and mortality due to predation need to be assessed. However, there are only a few Prorocentrum spp. for which heterotrophic protist predators have been reported. We explored feeding by the common heterotrophic dinoflagellates Gyrodinium dominans, Oxyrrhis marina, Pfiesteria piscicida, Oblea rotunda, and Polykrikos kofoidii and the naked ciliate Strombidinopsis sp. (approx. 90 ㎛ cell length) on the planktonic species Prorocentrum triestinum, P. cordatum, P. donghaiense, P. rhathymum, and P. micans as well as the benthic species P. lima and P. hoffmannianum. All heterotrophic protists tested were able to feed on the planktonic prey species. However, O. marina and O. rotunda did not feed on P. lima and P. hoffmannianum, while G. dominans, P. kofoidii, and Strombidinopsis sp. did. The growth and ingestion rates of G. dominans and P. kofoidii on one of the seven Prorocentrum spp. were significantly different from those on other prey species. G. dominans showed the top three highest growth rates when it fed on P. triestinum, P. cordatum, and P. donghaiense, however, P. kofoidii had negative growth rates when fed on these three prey species. In contrast, P. kofoidii had a positive growth rate only when fed on P. hoffmannianum. This differential feeding on Prorocentrum spp. between G. dominans and P. kofoidii may provide different ecological niches and reduce competition between these two common heterotrophic protist predators.

Mixotrophic dinoflagellates act as primary producers, prey, and predators in marine planktonic food webs, whereas exclusively autotrophic dinoflagellates are primary producers and prey. Species of the dinoflagellate genus Scrippsiella are commonly found in marine ecosystems and sometimes cause harmful red tides. Among the 28 formally described Scrippsiella species, S. acuminata has been found to be mixotrophic and two unidentified species have been found to be mixotrophic. To determine whether the other species in this genus are similarly mixotrophic, the mixotrophic ability of S. donghaiensis SDGJ1703, S. lachrymosa SLBS1703, S. masanensis SSMS0908, S. plana SSSH1009A, and S. ramonii VGO1053 was explored using 15 potential prey items, including 2-㎛ fluorescently labeled microspheres (FLM) and heterotrophic bacteria (FLB), the cyanobacterium Synechococcus sp., and various microalgal prey species. The ability of S. acuminata to feed on FLM and FLB was also investigated. We found that S. donghaiensis, S. lachrymosa, S. masanensis, S. plana, and S. ramonii did not feed on any potential prey tested in this study, indicating a lack of mixotrophy. However, S. acuminata fed on both FLM and FLB, confirming its mixotrophic ability. These results lowered the proportion of mixotrophic species relative to the total number of tested Scrippsiella species for mixotrophy from 100% to 29-38%. Owing to its mixotrophic ability, S. acuminata occupies an ecological niche that is distinct from that of S. donghaiensis, S. lachrymosa, S. masanensis, S. plana, and S. ramonii.
Sodium hypochlorite (NaOCl) has been demonstrated to be effective against algae that cause red tides. To secure the environmental safety of this chemical for practical use, toxic effects of NaOCl were evaluated in two important aquaculture fishes, flounder and rockfish. At concentrations above 2.5 and 5 ppm for 1 hr, mortality occurred in flounder and rockfish, respectively. Flounder, however, did not die at 2 ppm up to an 8 hr exposure, nor did lipid peroxide level change at the same concentration in 1 hr. Plasma glucose level significantly increased in 1 hr with more than 2 ppm exposure. The results indicate that NaOCl may not induce persistent severe toxicity in flounder and rockfish at the algicidal concentration, 0.5 ppm.

It has been mown that the global warming has an effectet on marine ecosystem and marine environments. Then, fisherman's activity and fishing production were decreased by changing of marine plankton composition and increasing of harmful marine organisms such as jellyfish, starfish and green laver bloom. Harmful red tides algae bloom and the deserted sea bottom often occurred due to increasing of sea water temperature and sea level rising in Korea. Phytoplankton forms the base of sea ecosystems. Various environmental factors and anthropogenic pollution, primarily, affect the concentration and photosynthetic activity algal cells, and the changes in the phytoplankton photosynthesis influence other elements of aquatic ecosystems. The increase in anthropogenic pollution markedly damages natural aquatic ecosystems, particularly, in the coastal zones, where an intense blooming of microalgae occurs, including the release of highly dangerous ecotoxic substances of various chemical natures (red tides). In this study, we tried to apply as a parameter for the algal blooming prediction in the ocean from fluorescence values in the taken samples around Busan coastal area. F0 value was almost constant but Fv/Fm value showed the irregular pattern. We presume that these results are due to the changes of the ocean environment and climate. To predict or give early warning the algal blooming, we need to investigate the specific area or fixed area through real-time monitoring. Especially, algal blooming prediction or warning can be achieved via continuously monitoring and interpretation of fluorescence changes.

Quantitative real-time polymerase chain reaction (qPCR) was applied for the early detection of red tides in the coastal areas of South Gyeongsang in 2019. Cochlodinium polykrikoides (Dinophyceae) was detected at very low cell densities (0.0015~0.0058 cells mL-1) in early June, but its cell density increased by up to 0.163 cells mL-1 in mid-August. Higher cell densities were detected mainly in Namhaedo using both qPCR and microscopy (maximum 24 cells mL-1) in late-August. Accordingly, a red tide alert was issued on September 2 (maximum 200 cells mL-1) on this island. C. polykrikoides cell density in Namhaedo peaked on September 11 (12,000 cells mL-1). Our results indicate that C. polykrikoides was detected at very low cell density in Namhaedo prior to bloom, which occurred in the same area. Therefore, qPCR is a useful tool to detect even at very low cell densities of C. polykrikoides for early warning of blooms.
Myrionecta rubra, a mixotrophic ciliate, is a cosmopolitan red tide species which is commonly found in neritic and estuarine waters. M. rubra had long been listed as an “nculturable protist”until 2 different laboratory strains were finally established in 2 research groups at the beginning of this century, enabling us to perform initiative investigation into various aspect of the live M. rubra strains (Gustafson et al. 2000; Yih et al. 2004b; Johnson and Stoecker 2005). Field sampling was carried out on high tide at 2 fixed stations around Kunsan Inner Harbor (St.1 near the Estuarine Weir and St.2 off Kunsan Ferry Station) every other day for 4 months from mid-February 2004 to understand detailed figure of the recurrent spring blooms of M. rubra following the onset of the water gates operation of the Keum River Estuarine Weir on August 1994. With its maximum abundance of 272 cells mL$^{-1}$ in St.1, fluctuation pattern of the M. rubra population at the 2 stations was strikingly similar. Notable growth of M. rubra population started on late April, to cause M. rubra red tides during one month from mid-May in which “xceptionally low salinity days”without its red tide were intermittently inserted. High abundance of M. rubra over 50 cells mL$^{-1}$ was recorded at samples with their water temperature and salinity higher than 15${^{\circ}C}$ and 4.0 psu, respectively. During pre-bloom period when salinity fluctuation is moderate and the water temperature is cooler than 15°C, Skeletonema costatum, a chain-forming centric diatom, was most dominant. Cyanobacterial species such as Aphanizomenon flos-aquae and Phormidium sp. replaced other dominant phytoplankters on the days with “xceptionally low salinity”even during the main blooming period of M. rubra. To summarize, M. rubra could form spring blooms in Keum River Estuary when the level of salinity fluctuation was more severe than that for the dominant diatom Skeletonema costatum and milder than that for the predominance by freshwater cyanobacteria. Therefore, optimal control of the scale and frequency of freshwater discharges might lead us to partially modify the fluctuation pattern of M. rubra populations as well as the period of spring blooms by M. rubra in Keum River Estuary. Sampling time interval of 2 days for the present study or daily sampling was concluded to be minimally required for the detailed exploration into the spring blooms by M. rubra populations in estuaries with weirs like Keum River Estuary.

To determine the composition of benthic species on the southwest coast, a macrobenthos study was conducted at 18 sites on the islands of Wando and Jodo for seven summers (2014-2021) from June to September. Two hundred and twenty species(645 ind./m2) were collected on the rocky shores of Wando and Jodo. A similar number of species was observed in Wando (165 species) and Jodo (167 species). The mean density was higher in Wando (726 ind./m2) than in Jodo (564 ind./m2). The number of species was the highest at Site 15 (116 species), and the density was highest at Site 7 (1,664 ind/m2). The dominant species were Nodilittorina radiata with 175 ind./m2 (27.1%), Littorina brevicula with 97 ind./m2 (15.1%), and Heminerita japonica with 41 ind./m2 (6.3%). The climate change indicator species, Tetraclita japonca, appeared in 15 sites, and the endemic species, Ptilohyale bisaeta, appeared in some sites. Fusinus spectrum, which appears on the Red List as a Near Threatened (NT) species, the lowest level of extinction risk, appeared in Jodo. The macrobenthos of Wando and Jodo demonstrated the general characteristics of rocky shores in Korea. Based on the tides and substrate, the rocky shores revealed a vertical distribution trend where Nodilittorina radiata, Littorina brevicula, Chthamalus challengeri, and Reishia clavigera were dominant. Wando and Jodo exhibited similar habitat distribution patterns.

Noctiluca scintillans is a heterotrophic dinoflagellate that causes red-colored oceans during the day (red tides) and glowing oceans at night (bioluminescence). This species feeds on diverse prey, including phytoplankton, heterotrophic protists, and eggs of metazoans. Thus, many scientists have conducted studies on the ecophysiology of this species. It is easy to cultivate N. scintillans at a scale of <1 L, but it is difficult to cultivate them at a scale of >100 L because N. scintillans cells usually stay near the surface, while prey cells stay below the surface in large water tanks. To obtain mass-cultured N. scintillans cells, we developed an automatic system for cultivating N. scintillans on a scale of 100 L. The system consisted of four tanks containing fresh nutrients, the chlorophyte Dunaliella salina as prey, N. scintillans for growth, and N. scintillans for storage, respectively. The light intensities supporting the high growth rates of D. salina and N. scintillans were 300 and 20 µmol photons m-2 s-1, respectively. Twenty liters of D. salina culture from the prey culture tank were transferred to the predator culture tank, and subsequently 20 L of nutrients from the nutrient tank were transferred to the prey culture tank every 2 d. When the volume of N. scintillans in the predator culture tank reached 90 L 6 d later, 70 L of the culture were transferred to the predator storage tank. To prevent N. scintillans cells from being separated from D. salina cells in the predator culture tank, the culture was mixed using an air pump, a sparger, and a stirrer. The highest abundance of N. scintillans in the predator culture tank was 45 cells mL-1, which was more than twice the highest abundance when this dinoflagellate was cultivated manually. This automatic system supplies 100 L of N. scintillans pure culture with a high density every 10 d for diverse experiments on N. scintillans.
