The purpose of this study is to investigate how elementary school student understands or students understand the illustrations on the phase change of the Moon in science textbook and to find out how textbook illustration helps students form the conception of the phase change of the Moon. To identify this purpose, we selected illustrations on the phase change of the Moon in the science textbook revised in 2007 and 2009 revised science textbook. For this study we selected and interviewed 20 students in the fifth grade. We integrated all data collected through interviews and created a transcription and a protocol and then, confirmed scientific conceptions related to the phase change of the Moon in students' illustration reading. The result are as followings: First, students read more scientific conceptions related to the phase change of the Moon in illustration of the 2009 revised science textbook which is presented with the universal observer's view point and the earth observer's view point. Second, students who find meaning in the various elements of the illustration and interpret with the integration of the various elements, get a lot of relevant information from illustration. All students have no differences recognizing the elements presented in illustration. But there are differences of contents of illustration reading depending on how students interpret the illustrations with integration of the various elements and if students cannot figure out the four scientific concepts needed to understand the phase change of the Moon, they ignore the information provided by illustration or analysis in their own way according to information provided by illustration. So misconception appears in this process.

The purpose of this study is to see and compare the contemporary Korean-Thai female poems from a feminine standpoint to newly clarify the textuality of their poems. The textuality defined in this manuscript is the text of Korean-Thai contemporary female poems. To newly clarify the textuality of their poems are to go against the existing discussion method and to newly read out the text as re-vision method. This discussion is to analyse deeply how the central exis composing a text which is the identity of woman in a body, appearance of uttrance, or action of abjection is exposed in gender space and to identify the poem's textuality. In other words, through in-depth analysis of the text of poems, which are very complicated as a skein of yarn, place a high value of Korean-Thai female poems. Transcending time, nations and races, if the text of female poem would not free from a biased male-dominated thinking or make a mystery of female poem textuality without critics or tend to be stereotype the text of poem as pathos of female, it would not get out from man-centered reading. To escape from the state of sexual discrimination, the new reading method was seriously analysed and found out that the female text poems not only implicate sexual discrimination but also link to expansive cultural and social structure. And for that reason, this study raise a question to male-dominant sexual discriminated norm. It is very significant that through this elaborate and in-depth text poem analysis, a creation process of female poem is traced. Eventually, the comparative study on Korean-Thai female poems is meaningful and worthy in regard to the contribution to promotion of cultural exchange between korea-Thai two nations and furthermore extend to East Asia to make a basement for the vitalization of Asia comparative literature.

본 연구는 대학도서관의 이용행태와 서비스의 혜택에 대한 인식이 이용자의 진술을 통해 가치가 도출되는 CVM을 적용해 측정한 도서관 서비스의 가치에 미치는 영향을 파악하고자 하였다. 이용행태의 변인은 도서관 방문 이용, 도서관 웹사이트 이용, 소속도서관 의존도이며, 혜택인식의 변인은 수업 및 강의, 논문 및 연구, 취업 및 외부활동에 대한 혜택의 만족도이다. 대학도서관 서비스는 도서대출, 해외 및 국내 논문, 참고서비스, 이용교육, 열람실 이용 서비스로 나누어 WTP를 측정하였다. 이용행태에 관한 가설 검증 결과 도서관의 방문 이용이 도서대출, 해외 논문 이용, 참고서비스, 이용교육에 대한 WTP에 영향을 미치며, 소속도서관의 의존도가 대출과 이용교육에 영향을 미치는 것으로 나타났다. 대학도서관이 제공하는 혜택에 대한 인식에 관한 가설 검증에서는 수업 및 강의의 혜택에 대한 만족도가 열람실 이용에 대한 WTP에 영향을 미치는 것으로 나타났다.

The word play on h(e)art-hunting has become a virtual commonplace in criticism of Chaucer′s Book of the Duchess. Less widely discussed is the third meaning of ME herte, "hurt." The "hart"/ "heart" pun is, however, only implicit in the poem, while the rhyme of "heart" and "hurt" in lines 883-84 makes clear the close association of the terms for Chaucer. Earlier commentators insisted that this was in fact an instance of rime riche or "identical rhyme," but if it is so it is striking that it is the unique instance of the rhyme in Chaucer, whose works are full of occasions for hurt hearts. The essay argues that this is, instead, an instance of near-rhyme and that the confusion in scribal spellings of ME hurten(with ′u,′ ′0,′ ′i,′ ′y,′ and ′e′ ) suggests uncertainties about its root vowel that modem linguistic study has not clarified completely. If the rhyme of herte ("hurt") with herte ("heart") is, however, established by these lines in BD, then it is probably reasonable to ask about all the occasions where characters in the poem are hurt by emotional or physical distress. In the cases of A1cyone and the Man in Blak, the hurt is revealed plainly as the death of a loved one, and Alcyone′s death and the Man in Blak′s return "homwarde" offer contrasting responses to the realization and acknowledgement of their loss. In the case of the Narrator, however, the exact nature of his "hurt" is nowhere made clear and the questions this Jack of clarity raises for the reader remain unanswered when the poem declares its "hert-huntyng" done. Further examination of the Narrator′s character and his role in the poem may reveal him to be a physician himself in need of healing, and this reading of his character may identify him as an ancestor as much of Chaucer′s Pardoner as of the Pilgrim Narrator of Canterbury Tales.

A novel gene coding for an endo-${\beta}$-1,4-mannanase (manA) from Bacillus subtilis strain G1 was cloned and overexpressed in P. pastoris GS115, and the enzyme was purified and characterized. The manA gene consisted of an open reading frame of 1,092 nucleotides, encoding a 364-aa protein, with a predicted molecular mass of 41 kDa. The ${\beta}$-mannanase showed an identity of 90.2-92.9% ${\leq}95%$) with the corresponding amino acid sequences from B. subtilis strains deposited in GenBank. The purified ${\beta}$-mannanase was a monomeric protein on SDS-PAGE with a specific activity of 2,718 U/mg and identified by MALDI-TOF mass spectrometry. The recombinant ${\beta}$-mannanase had an optimum temperature of $45^{\circ}C$ and optimum pH of 6.5. The enzyme was stable at temperatures up to $50^{\circ}C$ (for 8 h) and in the pH range of 5-9. EDTA and most tested metal ions showed a slightly to an obviously inhibitory effect on enzyme activity, whereas metal ions ($Hg^{2+}$, $Pb^{2+}$, and $Co^{2+}$) substantially inhibited the recombinant ${\beta}$-mannanase. The chemical additives including detergents (Triton X-100, Tween 20, and SDS) and organic solvents (methanol, ethanol, n-butanol, and acetone) decreased the enzyme activity, and especially no enzyme activity was observed by addition of SDS at the concentrations of 0.25-1.0% (w/v) or n-butanol at the concentrations of 20-30% (v/v). These results suggested that the ${\beta}$-mannanase expressed in P. pastoris could potentially be used as an additive in the feed for monogastric animals.

The $\alpha$1,6-mannosyltransferase encoded by Saccharomyces cerevisiae OCH1 plays a key role for the outer chain initiation of the N-linked oligosaccharides. A search for Hansenula polymorpha genes homologous to S. cerevisiae OCHI (ScOCH1) has revealed seven open reading frames (ORF100, ORF142, ORF168, ORF288, ORF379, ORF576, ORF580). All of the seven ORFs are predicted to be a type II integral membrane protein containing a transmembrane domain near the amino-terminal region and has a DXD motif, which has been found in the active site of many glycosyltransferases. Among this seven-membered OCH1 gene family of H. polymorpha, we have carried out a functional analysis of H. polymorpha ORF168 (HpOCH2) showing the highest identity to ScOCH1. Inactivation of this protein by disruption of corresponding gene resulted in several phenotypes suggestive of cell wall defects, including hypersensitivity to hygromycin B and sodium deoxycholate. The structural analysis of N-glycans synthesized in HpOCH2-disrupted strain (Hpoch2Δ) and the in vitro $\alpha$1,6-mannosyltransferase activity assay strongly indicate that HpOch2p is a key enzyme adding the first $\alpha$1,6-mannose residue on the core glycan Man$_{8}$GlcNAc$_2$. The Hpoch2Δ was further genetically engineered to synthesize a recombinant glycoprotein with the human compatible N-linked oligosaccharide, Man$_{5}$GlcNAc$_2$, by overexpression of the Aspergillus saitoi $\alpha$1,2-mannosidase with the 'HDEL” ER retention signal.gnal.

A Brevibacterium lactofermentum gene coding for a glucose-specific permease of the phosphoenolpyruvate-dependent phosphotransferase system (PTS) was cloned, by complementing an Escherichia coli mutation affecting a ptsG gene with the B. lactofermentum genomic library, and completely sequenced. The gene was identified as a ptsG, which enables an E. coli transformant to transport non-metabolizable glucose analogue 2-deoxyglucose (2DG). The ptsG gene of B. lactofermentum consists of an open reading frame of 2,025 nucleotides encoding a polypeptide of 674 amino acid residues and a TAA stop codon. The 3' flanking region contains two stem-loop structures which may be involved in transcriptional termination. The deduced amino acid sequence of the B. lactofermentum enzyme $II^{GIe}$ specific to glucose ($EII^{GIe}$) has a high homology with the Corynebacterium glutamicum enzyme $II^{Man}$ specific to glucose and mannose ($EII^{Man}$), and the Brevibacterium ammoniagenes enzyme $II^{GIc}$ specific to glucose ($EII^{GIc}$). The 171-amino-acid C-terminal sequence of the $EII^{Glc}$ is also similar to the Escherichia coli enzyme $IIA^{GIc}$ specific to glucose ($IIA^{GIc}$). It is interesting that the arrangement of the structural domains, IIBCA, of the B. lactofermentum $EII^{GIc}$ protein is identical to that of EIIs specific to sucrose or $\beta$-glucoside. Several in vivo complementation studies indicated that the B. lactofermentum $EII^{Glc}$ protein could replace both $EII^{ Glc}$ and $EIIA^{Glc}$ in an E. coli ptsG mutant or crr mutant, respectively.
