A report on 38 unrecorded bacterial species in Korea in the class Gammaproteobacteria

  • During an investigation of indigenous prokaryotic species in the Republic of Korea, a total of 38 bacterial strains belonging to the class Gammaproteobacteria were isolated from diverse environments. Samples were collected from soil, seawater, sand, sedimentary soil, rabbit feces, rat intestines, marine wetland, and tidal flats. The strains were identified to the species level using the high 16S rRNA gene sequences and showed high similarity (>98.7%) with the closest bacterial species and formed a robust clade in the neighbor-joining phylogenetic tree; it was determined that each strain belonged to independent, predefined bacteria species within the class Gammaproteobacteria. The 38 strains of Gammaproteobacteria analyzed in this study have not been reported in the Republic of Korea. Therefore, this study describes 20 genera of 13 families in 8 orders: Aeromonadales, Alteromonadales, Cellvibrionales, Enterobacterales, Lysobacterales, Oceanospirillales, Pseudomonadales, and Vibrionales. For each species, we describe Gram reaction, strain ID, isolation source, colony and cell morphology, cultural, physiological, and basic biochemical characteristics.

다시마 첨가 배추김치의 체중 감량 및 지질 저하 효과 (Weight Reduction and Lipid Lowering Effects of Sea Tangle Added Korean Cabbage Kimchi)

  • 체중감량 효과가 있는 기능성 배추김치를 개발하기 위하여 기호도가 우수하고 섬유소가 풍부하여 지질저하 기능성 물질을 함유한 다시마를 첨가한 해조첨가 배추맛김치(이하 해조김치) 레시피를 개발하고, 잘 숙성된 해조김치를 고지방식이를 섭취한 쥐에게 공급하여 지질저하 및 비만억제 효과를 살펴보았다. 배추김치에 다시마를 20% 정도 첨가하였을 때(해조김치) 종합적인 관능평가가 가장 우수하였다. 해조김치의 숙성 특성은 일반 김치와 같이 전형적인 발효양상을 보였다. 해조김치의 비만억제 효과를 살펴보기 위하여 20% 고지방식이에 동결 건조한 김치 10%를 첨가하여 쥐를 8주간 사육하였다. 실험군으로는 고지방식이군, 대조김치군, 해조김치군 그리고 시판용 공장김치를 첨가한 J-김치군으로 하였다. 식이 섭취량, 식이효율은 각 군간에 유의적인 차이가 없었다. 김치첨가에 의한 효과를 고지방식이군과 비교하여 보았을 때 상대적인 내장 지방량은 해조김치군(-17%), J-김치군(-11%) 그리고 대조김치군(-9%) 순으로 감소하였다(p<0.05). 혈청 leptin 농도는 해조김치군(-59%)에서 감소하였는데(p<0.05), 이는 해조김치군의 지방세포의 수가 대조군에 비해 감소함을 의미한다. 혈청 빌리루빈 농도는 해조김치군(-24%)에서 감소하였다(p<0.05). 혈청 중성지방과 콜레스테롤 농도는 고지방군에 비해 각각 해조김치군(-51%와 -21%), J-김치군(-40%, -23%) 그리고 대조김치군(-39%, -13%) 유의적으로 낮았다(p<0.05). 분변의 중성지방과 콜레스테롤 농도는 각각 해조김치군(49%와 71%), 대조김치군(36%, 61%), 그리고 J-김치군(32%, 65%)에서 증가하였다. 김치 종류에 따른 비만억제 효과를 살펴보았을 때 해조김치의 상대적인 내장 지방량은 다른 김치군에 비해 유의적으로 낮았고(p<0.05), 렙틴, 혈청지질, 분변지질 및 빌리루빈의 농도는 김치군간 유의적인 차이는 없었으나, 해조김치군의 비만억제 효과가 가장 높았다. 이러한 결과를 살펴보면, 일반 김치에 비해 해조김치를 섭취하면 체중 감량의 효과가 있을 것으로 생각된다. 이러한 해조김치의 비만 억제 기전은 다시마 첨가에 의해 식이섬유소의 함량이 7% 증가함으로써 소장에서 식이지방 흡수를 방해하여 혈청 중성지방의 농도를 낮추고 이로 인하여 지방세포로 중성지질의 이동이 낮아져 복부지방 축적이 억제되었기 때문으로 생각된다.

Psychobiotic Effects of Multi-Strain Probiotics Originated from Thai Fermented Foods in a Rat Model

  • This work aimed to investigate the psychobiotic effects of six bacterial strains on the mind and behavior of male Wistar rats. The probiotic (PRO) group (n=7) were rats pre-treated with antibiotics for 7 days followed by 14-day probiotic administration, antibiotics (ANT) group (n=7) were rats treated with antibiotics for 21 days without probiotics. The control (CON) group (n=7) were rats that received sham treatment for 21 days. The six bacterial strains with probiotic properties were mostly isolated from Thai fermented foods; Pedicoccus pentosaceus WS11, Lactobacillus plantarum SK321, L. fermentum SK324, L. brevis TRBC 3003, Bifidobacterium adolescentis TBRC 7154 and Lactococcus lactis subsp. lactis TBRC 375. The probiotics were freeze-dried into powder (6×109 CFU/5 g) and administered to the PRO group via oral gavage. Behavioral tests were performed. The PRO group displayed significantly reduced anxiety level and increased locomotor function using a marble burying test and open field test, respectively and significantly improved short-term memory performance using a novel object recognition test. Antibiotics significantly reduced microbial counts in rat feces in the ANT group by 100 fold compared to the PRO group. Probiotics significantly enhanced antioxidant enzymatic and non-enzymatic defenses in rat brains as assessed using catalase activity and ferric reducing antioxidant power assay, respectively. Probiotics also showed neuroprotective effects with less pyknotic cells and lower frequency of vacuolization in cerebral cortex. This multi-strain probiotic formulation from Thai fermented foods may offer a potential to develop psychobiotic-rich functional foods to modulate human mind and behaviors.

Sprague Dawley 쥐에서 장내 유래 Lactobacillus sp. KP-3의 중금속 축적 저해 효과 (The Inhibitory Effects of Intestine-oriented Lactobacillus sp. KP-3 on the Accumulation of Heavy Metals in Sprague Dawley rats)

  • 유산 균주들을 신생아 및 성인의 분변으로부터 분리하여 그 중에서도 가장 중금속 흡착능이 크고 중금속에 내성이 있다고 사료되는 균주 KP-3를 선별하여 동정한 결과 Lactobacillus sp.으로 밝혀졌다. 동물 실험에서는 생후 4주령의 Sprague dawley male rat에게 선별한 균주 KP-3를 투여하여 혈액 및 장기 내의 중금속 축적 저해효과가 어느 정도인지 조사하였다. 실험군으로는 SD rat 한 마리당 Lactobacillus sp. KP-3와 중금속 10 ppm을 혼합하여 약 100 ml/day씩 7일간 투여하였다. 대조군으로 중금속과 멸균 생리 식염수를 혼합하여 투여한 군과 Lactobacillus sp. KP-3와 멸균 생리 식염수를 혼합하여 투여한 군, 멸균 생리 식염수 만을 투여한 군으로 분리하여 SD rat 한 마리당 약 100 ml/day씩 7일간 투여하였다. 혈액, 간장 및 신장의 시료를 채취하여 카드뮴(Cd), 크롬(Cr), 납(Pb) 등의 중금속 축적량을 측정하였다. 측정 결과 크롬(Cr)은 신장에 주로 축적되었고, 납(Pb)은 간장과 신장에 주로 많은 축적을 보였고, 실험군인 Lactobacillus sp. KP-3와 중금속을 혼합하여 투여한 군에서는 중금속만을 투여한 군보다 중금속 축적량이 낮은 것으로 나타났다. 카드뮴(Cd), 크롬(Cr), 납(Pb)의 평균 중금속 축적 저해율은 각각 41.8%, 33.4% 44.2%로 나타났다. 특히, 혈액에서의 중금속 축적 저해율은 약 70%로 매우 높게 나타났고 혈액에서의 카드뮴 축적 저해율은 100%로 나타났다. 본 실험에서 탐색하여 최적화된 유산균이라고 생각되는 Lactobacillus sp. KP-3균주는 실험 결과에 의하면 중금속 축적 저해에 효과가 있는 것으로 여겨지며, 또한 유산균이 함유된 식품을 지속적으로 섭취함으로써 체내의 중금속의 축적 억제에 도움이 크게 되리라 생각된다.

실험동물에서 기능성 요구르트(R&B Rhythm®)의 변비개선효과 (Effect of Functional Yogurt (R&B Rhythm®) on the Improvement of Constipation in Animal Models)

  • 본 연구는 mouse와 rat에 대한 발효유의 급여에 의한 장 기능 개선과 변비 예방 효과 및 변비 치료 효과를 알아보고자 수행하였다. 동물실험을 통하여 발효유의 투여 농도에 따라 유의적인 차이를 확인할 수 있었으며, 소화관 이 실험에서는 중농도와 고농도 투여에서 유의적인 수준으로 이동률의 증가가 관찰되었고, 변비 예방 및 치료 효과에 대한 실험에서는 변비 유발 물질인 loperamide 단독 투여군에 비하여 발효유를 함께 투여한 결과, 변의 개수는 고농도 투여에 의하여 유의적인 증가가 관찰되었고, 분변량과 변의 수분함량은 중농도와 고농도 투여군에서 유의적인 증가가 관찰되었다. 이상의 동물 실험 결과를 통하여, 기능성 발효유의 반복적인 투여는 변비 예방과 변비 치료에 효과가 있는 것으로 확인되었다.

한국인 분변으로부터 생균제용 Lactobacillus acidophilus 의 선발 및 그 특성 (Selection and Characteristics of Lactobacillus acidophilus Isolated from Korean Feces)

  • 유산균식품, 발효유제품, 의약품 및 동물용 생균제 등에 사용되는 유산균을 국내에서 자체 개발하기 위하여 한국인 41명($2{\sim}35$세, 남자 26명, 여자 15명)의 분변으로부터 내산성, 내담즙산성 및 병원성균 억제능력이 우수한 Lactobacillus 균주들을 선발하였다. 최종적으로 선발한 균주를 L. acidophilus KY 2104로 명명하였으며, 생리적인 특성을 조사하였다. 상기 균주는 pH 3.0 완충용액에서 2시간동안 거의 100% 생존하였으며, pH 2.0에서는 초기 접종농도 $10^7\;cfu/mL$에서 2시간 동안 $10^4\;cfu/mL$ 이상 생존하였다. 또한 선발균주는 0.5%의 담즙산에서도 정상적으로 성장하였으며, 장내에서 주로 설사를 유발하는 병원성균(E. coli, Shigella dysenteriae, Staphylococcus aureus, Salmonella)에 대하여 생육억제능력을 나타내었다. Rat에 대한 L. acidophilus KY 2104 균주의 급성독성시험 결과, 모든 시험군에서 사망동물은 없었으며, 임상증상, 체중변화 및 부검소견에서도 다른 이상소견이 발견되지 않았다.

고지방 식이내 식이섬유질이 흰쥐의 레티놀 결합 단백질 (RBP, cRBP I, cRBP II) 유전자 발현에 미치는 영향 (Effect of Dietary Fibers on Retinol Binding Protein (RBP, cRBP I, cRBP II) Gene Expression in Rats Fed High Fat Diet)

  • Recently it has been reported that vitamin A and retinol binding proteins (RBPs) in blood and urine were changed in the condition of diabetes mellitus or hyperlipidemia. Fruits and vegetables are recommended to consume for the people suffered from these chronic degenerative diseases. The main components of fruits and vegetables are dietary fibers, for example cellulose and pectin, of which function to affect the absorption and excretion of dietary fat and fat-soluble substances. This study was conducted to investigate the effect of dietary fibers on RBPs mRNA expression in liver, small intestine and serum of rat fed high fat diet during 4 weeks. Sprague-Dawley rats, weighing 121g on average, were divided into four groups; (Control; $17\%$ fat & cellulose supplement diet, HF0: $25\%$ fat & fiber free diet, B:.Uc: $25\%$ fat & cellulose supplement diet and HF0: $25\%$ fat & pectin supplement diet) . The rats fed high fat diet groups (HF0, HFC, HFP) tended to consume the food less than the control group, but FER of HF0 groups was significantly higher than the control (p < 0.05) . The weight of adrenal gland in high fat diet groups (HF0, HFC, HFP) was significantly less than the control. Total lipid in feces daily excreted and in liver did not show any significant differences among the groups. Total cholesterol in HFP group was significantly different from that of HFC group. Serum total cholesterol and triglyceride in other group tended to lower than other groups and HDL cholesterol higher. Consequently, AI (atherogenic index) was the lowest in HFP group. Vit A contents in feces daily excreted tended to lower in high fat diet groups (HF0, HFP) compared to the control group. That content in adrenal gland was the lowest in HF0 group, but not in liver. In HFP group were down-regulated cRBPI mRNA in liver and cRBPII mRNA in small intestine and up-regulated RBP and transthyretin expression in serum compared to the other groups. In conclusion, dietary fibers, especially pectin, in high fat diet might down-regulate the expression of CRBP I, CRBP II mRNA in liver and small intestine, but increase the secretion of RBP into serum and therefore inhance the bioavailability of Vit A through the body. 단백질(蛋白質) 최저요구량(最低要求量) 측정(測定)에 관한 연구(測定) (A Study on Estimation of Minimum Protein Requirement)

  • This study was carried out to estimate the minimum protein requirement for the Korean college girls, and to see the effect of carbohydrate intake on the minimum protein requirement in the rat. In the first experiment, three students attending Ewha Womans University were given protein free diet for 7 days while caloric intake were adjusted to maintain constant body weight. Endogenous nitrogen excretion was measured to estimate their minimum protein requirement. Second experiment was carried out in 3 parts to find out the effect of carbohydrate intake on the minimum protein requirement using 15 female, 50 days old rats weighing 120g. Inexperiment 2-1, the rats were fed protein free diet for 9 days and endogenous nitrogen excretion was measured to determine the minimum protein requirement of the rats. In experiment 2-2, the minimum casein requirement of the rats needed to maintain nitrogen equilibrium was estimated by the least squares regression method feeding 3 different levels of casein determined from the result of experiment 2-1. Finally, in experiment 2-3, the rats were given the minimum casein requirement for 7 days with 3 different levels of carbohydrate to find out the effect of carbohydrate intake on the nitrogen balance. The results of this study were as follows. Experiment I. Daily endogenous nitrogen losses of 3 college girls were 1.89g in urine, 1.10g in feces and 0.0108g from skin since the total daily endogenous nitrogen loss was 3.0g and the total daily nitrogen intake was 0.52g, the minimum nitrogen requirement of these subjects was about 2.48g/day (15.5g protein/day). Experiment II. Experiment 2-1: daily endogenous nitrogen losses of the rats fed protein free diet were $22.63{\pm}0.48\;mg$ through urine and $13.12{\pm}0.47\;mg$ in feces. The total daily endogenous nitrogen ouput was $35.71{\pm}1.27\;mg$ and the minimum protein requirement of the rats was 225mg/day in this study. Experiment 2-2: the minimum casein requirement needed to maintain the rats in nitrogen equilibrium was determined to be 340mg/day. Experiment 2-3: the rats were given 350mg/day casein with 3 different levels of carbohydrate (12,26 or 46 Cal by sugar-cornstarch mixture). The nitrogen balances were measured to be +0.0021, +0.0073 and +0.0143 respectively and there were significant differences among 3 groups.

표고버섯 헤미셀룰로즈의 식이가 쥐의 혈당과 콜레스테롤 함량에 미치는 영향 (Dietary Effect of Hemicellulose from Shiitake Mushroom(Lentinus edodes) on Blood Glucose and Cholesterol Content in Rats)

  • The effect of hemicellulose extracted from Shiitake mushroom(Lentinus edodes) on the level of blood sugar and cholesterol in the diabetes-induced rat by streptozotocin(STZ) was investigated. The yield of hemicellulose by extraction process of 5% salt extraction, preparation of alcohol insoluble substance, IN KOH extraction, acid precipitation(pH 3.0), and dialysis was 9.24%. The experimental plots divided to 1% cellulose group(control), 0.5% hemicellulose group(H-l) and 1% hemicellulose group(H-2). The groups were fed for 6 weeks, then continuously fed for 1 week after induction of diabetes by STZ. Feed intakes, weight gain and feed efficiency of the each groups were not significantly different, while water intakes and liver weight of H-2 group were lower than those of control and H-l group. Weight of liver in the H-2 group was significantly lower than those of control and H-l groups. The amounts of feces were 0.32 g/day in the control group, 0.43∼0.44 g/day in the H-l and H-2 groups, while the amounts of urine were 15.28 mL/day in the control group, 10.83∼11.20 mL/day in the H-l and H-2 groups. The content of blood glucose before diabetes induction(fed for 3∼5 weeks) was 111.2-132.6 mg/dL in the control group, not significantly different from others; After diabetes induction, however, the contents were 212.8 mg/dL in the control group, 140.0-144.0 mg/dL in the H-l and H-2 groups, which showed significant difference. Urine glucose contents of H-2 group before and after diabetes induction were lower than those of control and H-l groups. There was no significant difference in the content of neutral lipid between each groups. Total cholesterol contents were 101.6 mg/dL in the control group, 56.∼64.0 mg/dL in the hemicellulose groups. HDL-cholesterol content and atherogenic index of hemicellulose groups were lower than those of control group, respectively. In conclusion, the hemicellulose extracted from Shiitake mushroom represented improving and preventing effects for diabetes.

산마늘 기능성 활성물질 탐색 및 자원화 (Effect of Allium victorialis var.platyphyllum Leaves on Triton WR-1339-induced and Poloxamer-407-induced hyperlipidemic Rats and on Diet-Induced Obesity Rats)

  • The herb of Allium victorialis var. platyphyllum (Liliaceae) has been used as an edible wild herb and to treat heart failure and gastritis. We have already reported antihyperlipidemic anti-tumor effects of this plant. To enlarge the commercial availability of this food, it was investigated whether the extracts of A. victorialis var.platyphyllum reduce hyperlipidemia and obesity or not. The plants tested in this experiment were collected from two eco-types of IS. Ullung and Mt. Odae cultivated at Pyongchang. Extracts were prepared by extracting the fresh leaves and those dried at $36^{\circ}C$ and $90^{\circ}C$, respectively. Pretreatment with the ethanolic extracts for two weeks (p.o.) reduced serum triglyceride-, total cholesterol- and LDL-cholesterol contents in rats induced by Triton WR-1339, respectively. Furthermore, oral administration of the extracts also inhibited the hyperlipidemia induced with oral diet of 30% corn oil. In the other attempt to find to alleviate the obesity, the model rats with obesity were induced by the high fat-diet for six weeks. Post-treatment with the extracts for two weeks significantly reduced the hyperlipidemia. Retroperitoneal-, epidymal- and total abdominal fat pad weights were considerably reduced at 100 mg/kg oral administration of the extracts. Increased feces lipid contents were also found in the rat treated with the extracts. The extract of Mt. Odae eco-type showed more potent activity than that of Is. Ullung one. These results suggest that use of the fresh leaves may lead to the higher activity in treatment of hyperlipidemia and obesity than of the dried one.
