Background: To refine facial transplantation techniques and achieve sound results, it is essential to develop a suitable animal model. Rat is a small animal and has many advantages over other animals that have been used as transplantation models. The purpose of this study was to describe a rat hemifacial transplantation model and to verify its convenience and reproducibility. Methods: Animals used in this study were Lewis rats (recipients) and Lewis-Brown Norway rats (donors). Nine transplantations were performed, requiring 18 animals. The hemifacial flap that included the ipsilateral ear was harvested based on the unilateral common carotid artery and external jugular vein and was transferred as a single unit. Cyclosporine A therapy was initiated 24 hours after transplantation and lasted for 2 weeks. Signs of rejection responses were evaluated daily. Results: The mean transplantation time was 1 hour 20 minutes. The anatomy of common carotid artery and external jugular vein was consistent, and the vessel size was appropriate for anastomosis. Six of nine allografts remained good viable without vascular problems at the conclusion of study (postoperative 2 weeks). Conclusion: The rat hemifacial transplantation model is suitable as a standard transplantation training model.
The insulin-like growth factor-I(IGF-I) is an important metabolic factor involved in cell growth and metabolism. Although secretion of IGF-I in rat liver is regulated by growth hormone, the effects of forskolin, adenylate cyclase activator, on secretion of IGF-I have not been reported. Therefore, a modified perfused rat liver model was used to investigate the regulatory effects of forskolin on IGF-I secretion in this experiment. The results were summerized as follows : 1. Modified perfused rat liver model was not changed to aspartate aminotransferase(AST), alanine aminotransferase(ALT) and lactic dehydrogenase(LDH) secretion in time. 2. The IGF-I secretion in hepatic cell was increased by forskolin($10^{-5}$, $10^{-6}$ and $10^{-7}M$) in a dose-dependent manner as compared with those of the controls, and significantly increased by $10^{-5}$ and $10^{-6}$ forskolin(p < 0.05). 3. Secretion of glucose in hepatic cell significantly was decreased by $10^{-5}$ forskolin as compared with those of controls(p < 0.05). These results suggest that forskolin may be involved in the regulation of IGF-I secretion in the perfused rat liver.
Spaceflight induces a number of cardiovascular physiological alterations. To study adaptations to microgravity on Earth, the tail-suspended, hindlimb-unloaded rat model has been used to simulate the effects of microgravity. Despite the extensive use of this model to infer physiological adaptations of many organs to microgravity, little information has been obtained on the effect of tail suspension(TS) on cardiac adaptations in the rat. This study was aimed to investigate the effects of simulated microgravity on the rat myocardium using the TS model. Twenty-four male Sprague-Dawley rats were randomly assigned to 3 experimental groups(1, 7 and 14 days of TS) and a control group. A microscopic examination was performed to assess histopathological changes in the myocardial morphology. The hearts from the control group, the 1 day-TS rats and the 7 day-TS rats revealed no evident abnormalities in cardiomyocyte size and morphology. At day 14 of TS, in contrast, the ventricular cardiomyocytes appeared more separated from each other and were slightly smaller in size compared with those of the control group. Also seen were scattered areas exhibiting focal disorganization of muscle fibers and some degenerating cardiomyocytes, of which the nuclei had become pyknotic or disappeared. In this study, we demonstrated that the ventricular cardiomyocytes underwent degeneration and atrophy at the microscopic level during exposure to simulated microgravity in TS rats.
The neuroprotective effects of herbal ethanol extracts from Gynostemma pentaphyllum (GP-EX) in 6-hydroxydopamine (6-OHDA)-lesioned rat model of Parkinson's disease treated with L-DOPA were investigated. Rats were prepared for the Parkinson's disease model by 6-OHDA-lesioning for 14 days. The rats were then treated with L-DOPA (10 and 20 mg/kg) with or without the oral administration of GP-EX (30 mg/kg, daily) for 28 days. L-DOPA (20 mg/kg) treatment for 28 days enhanced dopaminergic neuronal cell death in 6-OHDA-lesioned rat groups, but L-DOPA (10 mg/kg) did not. However, the oral administration of GP-EX (30 mg/kg) for 28 days ameliorated the enhanced neurotoxic effects induced by chronic L-DOPA treatment in 6-OHDA-lesioned rat groups by increasing tyrosine hydroxylase (TH)-immunohistochemical staining and the number of TH-immunopositive cells surviving in the substantia nigra. In addition, GP-EX administration (30 mg/kg) for 28 days recovered the levels of dopamine and norepinephrine of the striatum in 6-OHDA-lesioned rat groups, which were markedly reduced by L-DOPA treatment (20 mg/kg). GP-EX (30 mg/kg) did not produce any signs of toxicity, such as weight loss, diarrhea, or vomiting in rats during the 28-day treatment period. These results suggest that GP-EX has protective functions against chronic L-DOPA-induced neurotoxic reactions in dopaminergic neurons in the 6-OHDA-lesioned rat model of Parkinson's disease. Therefore, GP-EX may be beneficial in the prevention of adverse symptoms in parkisonian patients.
Objectives : Allergic rhinitis is highly increased in these days. We aimed to investigate therapeutic effect ofShinchulsanby observing changes in blood cells and the protein expression level of IL-4 in spleen of Sprague-Dawley(SD) rats with allergic rhinitis.Methods : Fifteen SD rats were divided into three groups: normal, control, and sample group. Allergic rhinitis was induced in the control and sample group by intraperitional and intranasal sensitization with 0.1% and 0.4% Ovalbumin solution. ThenShinchulsanwas orally administered only to the sample group for 28 days, while the rats in the control group was given normal saline.Results : 1.Shinchulsanshowed significantly decreased eosinophil level on the serum of the rat model.2.Shinchulsanshowed significantly decreased IgE level on the serum of the rat model.3.Shinchulsanshowed significantly decreased the protein expression levels of IL-4 in spleen of the rat model.4.Shinchulsanhad no hepatoxicity, according to AST and ALT on the serum.Conclusion : According to the above results, it is considered thatShinchulsanis helpful in treatment of allergic rhinitis.
Sodium polyacrylate based hydrogel patch was prepared and its wound healing efficacy in comparison with control groups was evaluated in thermal burn wound ($200^{\circ}C$, 10 sec burn) and excision wound rat model. Cytotoxicity of gamma irradiated (25 kGy) hydrogel patch was investigated in human fibroblasts and showed no significant cytotoxicity. Wound closure rates (H50%) in hydrogel patch group and BAND-$AID^{(R)}$ treated group were faster than that of control group (uncovered open wound). Hydrogel patch showed an enhancement in wound healing process.
Chronic liver diseases and hepatic cancer have been reported as 10% of cause of death in Koreans. Regardless of various causes, chronic liver disease accompanies commonly hepatic fibrosis. But still the mechanism of hepatic fibrosis remains poorly understood. Using the dimethylnitrosamine(DMN)-induced hepatic fibrosis rat model, We performed to evaluate the possible therapeutic effect of RIP(extracts of Phellodendron amurense and Patrinia scabiosaefolia) and to investigate the changes in referential connective tissue proteins($TGF-{\beta}_1$, ${\alpha}$-smooth muscle actin, and vimentin) as a marker of fibrogenesis. For these purposes, liver tissues were stained with H & E, and Azan staining for estimation of developing fibrosis. In the DMN-treated rat liver tissue, fibrosis were developed forming incomplete septal fibrosis. Whereas, in the RIP-treated rat liver tissues, the fibrosis were decreased recovering to normal morphology. The expressions of $TGF-{\beta}_1$, ${\alpha}$-smooth muscle actin($\alpha-SMA$), and vimetin were increased in the DMN-treated rat liver tissues, but decreased in the various areas of RIP-treated rat liver tissues. According to these results, RIP could be a possible therapeutic agent to reduce hepatic fibrosis, and the $TGF-{\beta}_1$, ${\alpha}$-SMA, and vimentin could be possible indicative markers of hepatic fibrosis development and recovery.
Objective : The aim of this study was to investigate voluntary wheel running behavior in the unilateral 6-hydroxydopamine (6-OHDA) rat model. Methods : Male Sprague-Dawley rats were assigned to 2 groups : 6-OHDA group (n=17) and control group (n=8). The unilateral 6-OHDA rat model was induced by injection of 6-OHDA into unilateral medial forebrain bundle using a stereotaxic instrument. Voluntary wheel running activity was assessed per day in successfully lesioned rats (n=10) and control rats. Each behavioral test lasted an hour. The following parameters were investigated during behavioral tests : the number of running bouts, the distance moved in the wheel, average peak speed in running bouts and average duration from the running start to the peak speed. Results : The number of running bouts and the distance moved in the wheel were significantly decreased in successfully lesioned rats compared with control rats. In addition, average peak speed in running bouts was decreased, and average duration from the running start to the peak speed was increased in lesioned animals, which might indicate motor deficits in these rats. These behavioral changes were still observed 42 days after lesion. Conclusion : Voluntary wheel running behavior is impaired in the unilateral 6-OHDA rat model and may represent a useful tool to quantify motor deficits in this model.
Introduction : In this study, the analgesic effect and its mechanism of bee venom aqua-acupuncture on complete Freund's adjuvant-induced arthritis in rats was investigated. It has been reported from a neurochemical standpoint that bee venom exerts antinociceptive effects on inflammation and that the opioid system and adrenergic system play important roles in acupuncture analgesia. however, it is not known whether central opioid and ${\alpha}2$-adrenergic components of the intrinsic descending analgesic system are activated after bee venom aqua-acupuncture. Methods : Bee venom(1mg/kg) was subcutaneously aqua-acupunctured into Joksamni($ST_{36}$) of rats with complete Freund's adjuvant(CFA)- induced arthritis and was checked of increase in TFL. Opioid and ${\alpha}_2$-adrenergic neurotransmitter system were examined by naloxone as an opioid receptor antagonist, and yohimbine as ${\alpha}_2$-adrenoceptor antagonist prior to bee venom aqua-acupuncture. Results : The following results have been obtained. 1. The tail flick latency in the rat model with adjuvant-induced arthritis was significantly decreased in 2 weeks. 2. The tail flick latency in the rat model with adjuvant-induced arthritis was increased in bee venom aqua-acupuncture group compared to the normal saline aqua-acupuncture group. 3. Analgesic effect of bee venom was antagonized by yohimbine not by naloxone pretreatment in the rat model adjuvant-induced arthritis. Conclusions : Bee venom aqua-acupuncture has an analgesic effect on the rat model of adjuvant-induced of adjuvant-induced arthritis and has antinociception mediated by ${\alpha}_2$-adrenergic system.
유전자 치료등 폐암에 대한 새로운 치료법의 개발및 그 효능의 검증에 있어 적절한 동물 모델이 없음은 큰 제 한점중의 하나이다. 특히 종양의 생물학적 특성이나 치료에의 효과등이 장기자체의 환경에 크게 영향 을 받는다는 사실은, 인체에서의 폐암의 특성을 가지며 폐에 정 위적으로 발생하는 폐암의 동물모델의 개발 을 시급하게 한다. 저자등은 Nude rat을 대상 동물로 하여, 개흉하에 종양세포 부유액을 원하는 폐말단 부위에 직접 주입함으로 폐에 정 위적으로 폐암의 발달을 유도하였으며 이를 이용하여 발생된 비소세포 폐암의 병태를 연구하였다. 종양은 실험 대상 등물에서 모두 발생하였으며 이용한 두 가지 종류의 세포주(NCI-H46O과 NCI-H1299)에서 모두 효과적으로 발생하였다. 발생된 폐종양은 시간 경과에 따라 주위 조직으로의 침윤과 종격동 전이의 양상를 보였다. 종양 숙주 동물의 평균 수명은 약 5주 정도였다. 저자등이 개발한 비소세포폐암의 동물 모델은 기관지를 통한 종양 세포 주입법에 의한 폐암 모델에 비해 국소적으로 진행된 폐암을 원하는 부위에 정확히 만들 수 있음은 물론 외과적 처치를 비롯한 국소적 치료 방법의 개발이나 ?과의 검증에 두루 이용되기에 적절하다고 사료된다.
