We report a highly sensitive surface-enhanced Raman scattering (SERS) platform for the selective trace analysis of mercury (II) ions in drinkable water using aptamer-conjugated silver nanoparticles. Here, an aptamer designed to specifically bind to $Hg^{2+}$ ions in aqueous solution was labelled with a TAMRA moiety at the 5' end and used as a Raman reporter. Polyamine spermine tetrahydrochloride (spermine) was used to promote surface adsorption of the aptamer probes onto the silver nanoparticles. When $Hg^{2+}$ ions are added to the system, binding of $Hg^{2+}$ with T-T pairs results in a conformational rearrangement of the aptamer to form a hairpin structure. As a result of the reduced of electrostatic repulsion between silver nanoparticles, aggregation of silver nanoparticles occurs, and the SERS signal is significantly increased upon the addition of $Hg^{2+}$ ions. Under optimized assay conditions, the concentration limit of detection was estimated to be 5 nM, and this satisfies a limit of detection below the EPA defined limit of 10 nM in drinkable water.
Surface-Enhanced Raman Scattering (SERS) spectroscopy is a multifaceted surface sensitive methodology which exploits spectroscopy-based analysis for various applications. This technique is based on the massive amplification of Raman signals which were feeble previously in order to use them for appropriate identification at qualitative and quantitative in chemical as well as biological systems. This novel powerful technique can be utilized to identify pathogens such as bacteria and viruses. As far as SERS is concerned, one of the most studied problems has been functionalization of SERS active substrate. Metal colloids and nanostructures or microstructures synthesized using noble metals such as Au, Ag and Cu are considered to be SERS active. Silver and gold are extensively used as SERS active substrates due to chemical inertness and stability in air compare to copper. However, use of Cu as a suitable alternative has been taken into account as it is cheap. Herein, we have synthesized air-stable copper microstructures/nanostructures by chemical, electrochemical and microwave-assisted methods. In this paper, we have also discussed the use of as synthesized copper micro/nanostructures as inexpensive yet effective SERS active substrates for the fast identification of micro-organisms like Staphylococcus aureus and Escherichia coli.

To investigate only the effects of the stratified mixture distribution on initial flame propagation and combustion characteristics, the instantaneous equivalence ratio in the spark plug gap and combustion pressure were measured simultaneously In a constant volume chamber, To induce the stratified propane-air mixture distribution near the spark plug, counter-flow typed mixture injection system was used under the constant mean equivalence ratio $\Phi$$\_$mean/= 1.0 The instantaneous equivalence ratio was measured by a single-shot Raman scattering with narrow-band KrF excimer laser. The measuring error was within the limit of $\pm$ 3.5% provided that the proposed method was applied to the measured Raman signals. Judging from mass fraction burned derived from the measured pressure, the optimum combustion characteristics were shown under the condition that the local equivalence ratio in the spark plug was near 1.28$\pm$0.04, and these characteristics were more remarkable at the initial stage of combustion.

Purpose: This study examined the performance of two spectroscopy methods and multivariate classification methods to discriminate viable pepper seeds from their non-viable counterparts. Methods: A classification model for viable seeds was developed using partial least square discrimination analysis (PLS-DA) with Fourier transform near-infrared (FT-NIR) and Raman spectroscopic data in the range of $9080-4150cm^{-1}$ (1400-2400 nm) and $1800-970cm^{-1}$, respectively. The datasets were divided into 70% to calibration and 30% to validation. To reduce noise from the spectra and compare the classification results, preprocessing methods, such as mean, maximum, and range normalization, multivariate scattering correction, standard normal variate, and $1^{st}$ and $2^{nd}$ derivatives with the Savitzky-Golay algorithm were used. Results: The classification accuracies for calibration using FT-NIR and Raman spectroscopy were both 99% with first derivative, whereas the validation accuracies were 90.5% with both multivariate scattering correction and standard normal variate, and 96.4% with the raw data (non-preprocessed data). Conclusions: These results indicate that FT-NIR and Raman spectroscopy are valuable tools for a feasible classification and evaluation of viable pepper seeds by providing useful information based on PLS-DA and the threshold value.

In this work, we describe a very simple electroless deposition method to prepare moderate-SERS-active nanostructured Pd films deposited on the glass substrates. To the best of our knowledge, this is the first report on the one-pot electroless method to deposit Pd nanostructures on the glass substrates. This method only requires the incubation of negatively charged glass substrates in ethanol-water mixture solutions of $Pd(NO_3)_2$ and butylamine at elevated temperatures. Pd films are then formed exclusively and evenly on glass substrates. Due to the aggregated structures of Pd, the SERS spectra of benzenethiol and organic isonitrile could be clearly identified using the Pd-coated glass as a SERS substrate. This one-step fabrication method of Pd thin film on glass is cost-effective and suitable for the mass production.
We studied the detection of the Hg(II) concentration in an aqueous solution using rhodamine dyes on citrate-reduced Au nanoparticles (NPs). The quenching effect from Au NPs was found to decrease as the Hg(II) concentration increased under our experimental conditions. As the fluorescence signals intensified, the surface-enhanced Raman scattering (SERS) intensities reduced on the contrary due to less rhodamine dyes on Au NPs as the Hg(II) concentration increased. The rhodamine 6G (Rh6G) and rhodamine 123 (Rh123) dyes were examined via fluorescence and SERS measurements depending on Hg(II) concentrations. Fast and easy fluorescence detection of an Hg (II) concentration as low as a few ppm could be achieved by naked eye using citrate-reduced Au NPs.
In coherent anti-Stokes Raman scattering (CARS) microscopy reported until now, conventional microscope objectives are used, so that they are limited for introduction into a living body. Gradient-index (GRIN) rod lenses might be a solution for miniaturized microscope objectives for in-vivo CARS microscopy. However, due to the inherent large amount of chromatic aberration, GRIN rod lenses cannot be utilized for this purpose. CARS imaging catheter, composed of miniaturized microscope objective and fiber bundle, can be introduced into a living body for minimally invasive diagnosis. In order to design the catheter, we have to first investigate design requirements. And then, the optical design is processed with design strategies and intensive computing power to achieve the design requirements. We report the miniaturized objective lens system with diffraction-limited performance and completely corrected chromatic aberrations for an in-vivo CARS imaging catheter.
Single crystal 3C-SiC(cubic silicon carbide) thin-films were deposited on Si(100) wafers up to a thickness of 4.3 ${\mu}m$ by APCVD method using HMDS(hexamethyildisilane) at $1350^{\circ}C$. The HMDS flow rate was 0.5 sccm and the carrier gas flow rate was 2.5 slm. The HMDS flow rate was important to get a mirror-like crystal surface. The growth rate of the 3C-SiC films was 4.3 ${\mu}m/hr$. The 3C-SiC epitaxial films grown on Si(100) were characterized by XRD, AFM, RHEED, XPS and raman scattering, respectively. The 3C-SiC distinct phonons of TO(transverse optical) near 796 $cm^{-1}$ and LO(longitudinal optical) near $974{\pm}1cm^{-1}$ were recorded by raman scattering measurement. The hetero-epitaxially grown films were identified as the single crystal 3C-SiC phase by XRD spectra($2{\theta}=41.5^{\circ}$).
The adsorption structure and binding of maleimide (MI) and succinimide (SI) on silver nanocolloidal surfaces have been comparatively investigated by means of pH-varied surface-enhanced Raman scattering (SERS). The two molecules appeared not to adsorb onto Ag surfaces at pH values below 5. The appearance of a ring ν (CH) band at ~3100 $cm^{-1}$ denoted the standing geometry of MI’s aromatic ring on Ag. The absence or weakness of in-plane vibrational modes of MI and SI also supported a perpendicular orientation of MI and SI on Ag from the electromagnetic selection rule. Density functional theory (DFT) calculations were employed to examine the vibrational frequencies of MI’s and SI’s neutral and anionic states.

[ $Gd_{1+x}Ba_{2-x}Cu_3O_7$ ] (GdBCO) coated conductors on IBAD-MgO templates have grown by pulsed laser deposition. Critical current of the films were measured as about 90 A/cm by a four-probe method. The optical response of the films was investigated by Raman scattering spectroscopy. According to the Raman scattering spectra, the peaks at $328\;cm^{-1}$, $451\;cm^{-1}$, $504\;cm^{-1}$ were found and assigned to one $B_{1g}$ mode and two $A_{1g}$ modes, respectively. The high critical-current carrying behaviors of the GdBCO coated conductors are ascribed to their 123-structure without exchange of cation and incorporation of oxygen into the cuprates.
