Screening for Herbs Having Inhibitory Effect on Egg Hatching of Strongyloides venezuelensis

  • In vitro bioassays of aqueous extracts of 25 herbal species were performed by ovicidal activity against Strongvloides venuelensis. 500 mg of feces from Mongorian gerbil infected with S. venuelensis were placed into polyvinyl culture with extracts of herb and anthelmintic drugs in triplicates. Larval activity inside the polyvinyl bag was recorded microscopically at 12 hours, and every 24 hours thereafter for up to 72 hrs incubation. Eggs were not hatched within 72 hrs and degeneration was evident in the presence of extracts. The ovicidal activity characterized by the absence of L$_3$ in fecal cultures inside the polyvinyl bag. 1. Among 25 herbs, 12 herbs had ovicidal effects and caused degeneration of larvae in egg. S. angustifolia, I. helenium, A. korenum, P. tenuifolia, F. virridissima, A. asiatica, S. chinensis, A. tataricus, S. baicalensis, M. rimosa, S. glabra and Z. piperitum were the most ovicidal effect. However, P. thunbergiana, T. chinensis, C. flos, G. uralensis, C. obutusifolia, C. fortunei, A. koreana, P. grandiflorum, P. tenuifolia (Radix), P. mume and A. asphodelodes showed no ovicidal effects in vinyl bag culture. 2. Of 25 hrebs, 4 herbs had ovicidal and wormicidal effects, P. thunbergiana, C. flos, B. Striata and T. chinensis were characterized by degeneration of larvae.

A Study of Pectinesterase, Polygalacturonase, Lipoxygenase and Peroxidase in Hot Pepper (고추내 함유 Pectinesterase, Polygalacturonase, Lipoxygenase와 Peroxidase의 특성에 관한 연구)

  • This study was carried out 19 investigate the characteristics of pectinesterase (PE), polygalacturonase (PG), lipoxygenase(LOX) and peroxidase (POD) in hot pepper to know the effect of hot pepper on food quality during food processing and storage. The results were as follows : 1. The optimum pH of PE was pH 7.5 and the activity of PE below pH 5.5 was revealed scarcely, The concentration of NaCl and $CaCl_2$ that showed the highest activity of PE were 0.2M and 0.05M, respectively. 2. The optimum pH of PG was pH 6.0 and the activity of PG in acidity was higher than that in alkalinity. The activity of PG was maximum at 0.3M NaCl and 0.2mM $CaCl_2$. Above the concentration of 0.5M NaCl and 0.5M $CaCl_2$, the activity of PG was lower than that of PG not adding these salts 3. The optimum pH of LOX was pH 7.0 and pH 8.5. 4. The optimum pH of POD was pH 6.0 and the activity of POD was higher in weak acidity and neutrality than in alkalinity. POD activity was slightly decreased by the increase of NaCl and $CaCl_2$ concentration.

Protective Effects of Cinnamic Acid Derivatives on Gastric Lesion

  • P-methoxycinnamic acid and 3,4,5-trimethoxycinnamic acid are the compounds found in Polygalae Radix, the root of Polygala tenuifolia Willdenow, and have been reported to have hepatoprotective and anti-neurodegenerative effects. On the other hand, there are no reports of their effects on gastric lesions. This study examined the inhibitory effects of cinnamic acids, including p-methoxycinnamic acid, 3,4,5-trimethoxycinnamic acid, and 8 compounds (cinnamic acid, 2-(trifluoromethyl) cinnamic acid, 3-(trifluoromethyl) cinnamic acid, trans-4-(trifluoromethyl) cinnamic acid, 4-(dimethylamino) cinnamic acid, 3,4-(methylenedioxy) cinnamic acid and 3,4-dihydroxycinnamic acid), which were selected based on their presence in medicinal herbs and molecular weight, against gastric lesions. Animal models were used to confirm the protective effects on acute gastritis caused by the administration of HCl/EtOH. Gastric acid inhibition was examined by an acid-neutralizing test and the proton pump ($H^+/K^+$-ATPase) inhibiting activity. In addition, antioxidant tests were performed and the gastric emptying rate was determined. The results showed that cinnamic acid, p-methoxycinnamic acid, and 3,4,5-trimethoxycinnamic acid had an inhibitory effect on gastric lesions.

Effect of Achyrantis Radixs and Ultrasound in Osteoarthritis Rats Articular Cartilage (우슬과 초음파가 퇴행성관절염 백서의 관절 연골에 미치는 영향)

  • Osteoarthritis(OA) diseases are characterized by joint pain, tenderness, limitation of movement, crepitus, occasional effusion, and variable degrees of inflammation without systemic effects. We investigated the effects of Achyrantis radixs cream treatment and low intensity ultrasound in monosodium iodoacetate(MIA) induced experimental osteoarthritis rat. Sprague-Dawley 40 rats of 7-8 weeks, weight $250\;{\pm}\;50$ g were divided into four groups including the control group and ostoarthritis group(30 rats). Histopathological examination, Mankin's score, and immunohistochemical were performed. Histological findings in control group that are similar to those observed in human osteoarthritis, such as disorganization of chondrocytes, erosion and fibrillation of cartilage surface, and subchondral bone exposure. Safranin O-fast green staining revealed that marked diffuse reduction of proteoglycans and chondrocyte treated with MIA. The Mankin's score were closely correlated to the grade of histological findings. The level of Bax and caspase-3 expression decreased experimental groups. This study shows that a Acyranthes Radix cream treatment and low intensity ultrasound exerts a beneficial influence on the severity of chondral lesion in osteoarthritis rats. This treatments could related to a reduced level of chondrocyte apoptosis through anti-apoptotoc capacities of MIA-induced apoptotic protein overexpression.

  • The antidiabetic activity-guided fractionation and isolation of the 80% EtOH extracts from Peucedani Radix (Peucedanum japonicum, Umbelliferae) led to the isolation and characterization of a coumarin and a cyclitol as active principles, that is, peucedanol 7-O-${\beta}$ -D-glucopyranoside (1) and myo-inositol (2). Their structures were identified by spectroscopic methods. Compound 1 showed 39% inhibition of postprandial hyperglycemia at 5.8 mg/kg dose, and compound 2 also significantly inhibited postprandial hyperglycemia by 34% (P<0.05).

  • A Literature study on the external treatment of nose diseases, the results are as follows; 1. Nose was called "Cheun-bin(天牝)", was outer orifice of the lung, was the gate of vital energe(氣), The main function of nose is divided three pacts. One is the respiration, another is the sense of smell, the other is the sounding consonance. 2. Nose is concerned with The Lung(肺), The Spleen(脾), The Heart(心), The Stomach(胃), The Large intestine(大腸), The Bladder(膀胱) in the organs, is concerned with Lung Meridean, Large intestine Meridean, Stomach Meridean, Heart Meridean, Spleen Meridean, Governor Vessel Meridean etc. 3. The method on the external treatment of nose diseases is classified with obstructive method, pouring method, blowing method, attaching method, plaster method, rubbing method, cleansing method, poulticing method. In frequency of practical use, Obstructive method is used 276 cases, Blowing method is used 123 cases, Attaching method is used 103 cases, Plaster method is used 76 cases. 4. In frequency of the medicinal works on the external treatment of nose diseases, Herba Asari(細辛) is used 134 cases, Pedicellus Melo(瓜 ) is used 85 cases, Radix Aconiti Praeparata(附子) is used 52 cases, Herba Agastachis(麝香) is used 51 cases, Semen Armeniacae Amarurn(杏仁) is used 44 cases etc. In frequency of a prescription on the external treatment of nose diseases, Tongcho-san(通草散) is used 9 cases, $Sesin-go{\cdot}Sinyi-go{\cdot}Changpo-san$ are used 8 cases, $Kwache-san{\cdot}Wungwhang-san$ are used 7 cases etc.

Analysis of Sulfur Dioxide Residue in Commercial Medicine Prepared from Crude Drugs (생약제제 중 이산화황 잔류실태 시중 유통 생약제제 중 이산화황의 잔류실태 조사)

  • This study was carried out to investigate the content of sulfur dioxide residues in the medicine prepared from crue drugs. Total 29 pills that contains salvia miltiorrhiza root, anemarrhena rhizome, fritillaria bulb, glehnia root, asparagus tuber, lycium fruit, moutan root bark, cimicifuga rhizome, pinellia tuber, dipsaci radix, codonopsis pilosula root, liriope tuber, achyranthes root, prepared aconite, aurantii fructus immaturus, corydalis tuber, dioscorea rhizome were examined by the modified Monier_Willams method. Sulfur dioxide were detectd in all the tested samples, and were found over MRLs (30 mg/ kg to medicinal herbs) in 8 samples (27.6%). Especilly, "i2" pills contains 132.46 mg/kg sulfur dioxide that is 4 times over than MRLs.

Chemopreventive Effect of Saponins Derived from Roots of Platycodon grandiflorum on 4-(Methylnitrosamino)-1-(3-Pyridyl)-1-Butanone-Induced Lung Tumorigenesis in A/J Mice

  • This study examined the chemopreventive effect of saponins that were isolated from the roots of Platycodon grandiflorum A. DC (Campanulaceae), Changkil saponins (CKS), against the tobacco-specific carcinogen, 4-(methylnitrosamino)-1-(3-pyridyl)-1-butanone (NNK), -on lung tumorigenesis in A/J mice. The mice were treated with a single NNK dose (100 mg/kg b.w., i.p.). CKS (0.5, 1, 4 mg/kg body wt.) was administered orally daily for 3 days/week beginning 1 day after the NNK treatment and was maintained throughout the experiment. The administration of CKS suppressed the NNK-induced increase in the level of proliferating cell nuclear antigen, which are a marker of cell proliferation, in the lungs of the mice 4 weeks after the NNK injection. Twenty-five weeks after the NNK treatment, the mice were sacrificed and the number of surface lung tumors was measured. CKS significantly reduced the number of lung tumors induced by NNK in a dose dependent manner. These results suggest that CKS suppresses the development of lung tumors and has a chemopreventive effect against NNK-induced mouse lung tumorigenesis.

Effects of Naetakcheonkeumsan and It’s Gamypang on the Lymphocytes and Cancer cells (內托千金散 및 그 加味方이 마우스의 免疫細胞 및 癌細胞에 미치는 效果)

  • Naetakcheonkeumsan(NCS) was a drug that treated carbuncle and cellulitis. So, the purpose of this Study was to investigate effect of NCS on the anti-cancer and proliferation of lymphocytes in normal mouse group, L1210 cells-transplanted mouse group and anti-cancer drug (vincristine) 0.005mg/kg were injected mouse(Ll210 cells-transplanted) group. We used NCS extract with freeze-dried, 8wks-old male mice, and Ll210 cell lines for this Study, The proliferation of cells was tested using a colorimetric tetrazoliun assay(MTT assay). The results of this Study were obtained as follow ; Group C(NCS plus Rehmanniae Radix Preparat administered group) inhibited proliferaion of lymphocytes in normal mouse group and Ll210 cells transplanted mouse group. Group A(NCS administered group) and Group B(NCS plus Cervi pantotrichum Cornu administered group) inhibited proliferation of Ll210 cells in Ll210 cells-transplanted mouse group and anti-cancer drug were injected mouse(Ll210 cells-transplanted) group. Group C incresed proliferation of L1210 cells in L1210 cells-transplanted mouse group, but inhibited in anti-cancer drug(vincristine) 0.005mg/kg were injected mouse(L1210 cells-transplanted) group.

Effects of Rosa rwgosa Radix on Lipid Metabolism in Rats (흰쥐의 지질대사에 미치는 해당화 뿌리 추출물의 효과)

  • The feeding effects of Rosa rugosa roots powder extracted with two different extracting solvents on lipid metabolism in rats were evaluated. Rats were fed with purified diets containing methanol or 70% aqueous acetone extracts at a 1% level for 4 weeks. In addition, to examine the effects of age, rats with different ages (4weeks old vs. 3 months old) were fed with purified diets containing methanol extracts at a 1% level for 4weeks. Feeding of methanol extracts decreased the concentration of triacylglycerol in serum and liver, whereas the aqueous acetone extracts lowered the concentration of liver triacylglycerol with concomitant elevation of the concentration of serum triacylglycerol. The methanol extracts ameliorated the degree of increase in the serum triacylglycerol level in relation to age and decreased especially the concentration of free fatty acid in serum of adult rats. The results suggested possibility that the regulatory mechanism responsible for triacylglycerol-lowering effects of Rosa rugosa roots extracts in the liver could be modulated by differences in the components of extracts due to the variation in extracting solvents, and that the methanol extracts might be effective in lowering an age-dependent increase in the concentration of serum and liver triacylglycerol in rats.
