예비동결 온도에 따른 숙지황 분말의 이화학적 특성 (Physicochemical Characteristics of Rehmanniae Radix Preparata Powder by Milling with Pre-freezing Temperatures)

  • Background: Rehmanniae radix preparata (RRP) has been used as a traditional medicine and is one of the most important oriental herbal medicines. However, the physical characteristics of RRP are not suitable for use in industry. The present study was under-taken to determined the preparation method of RRP powder and the physicochemical characteristics of RRP powder by milling under different pre-freezing temperatures. Methods and Results: Moisture content, powder yield, particle size, bulk density, compressive stress, extraction yield, and 5-HMF content of PRR powders by milling with pre-freezing temperatures (-20, -40, -60, and $-80^{\circ}C$) were analyzed, and correlation among these factors was determined. Powder yield increased and particle size decreased in a pre-freezing temperature-dependent manner from -20 to $-60^{\circ}C$. The Hausner ratio increased from 1.186 to 1.225 with decreasing temperature from -20 to $-80^{\circ}C$, whereas compressive stress showed the opposite trend. Extraction yield and 5-HMF content were not significantly different between RRP powder. Significant correlations were observed among pre-freezing temperature and physical characteristics (e.g., yield, particle size, Hausner ratio, and compressive stress). Conclusions: These results suggest that the pre-freezing temperature is an important factor affecting the physical characteristics of PRR powder and applicable to the industrial production of RRP powder.

숙지황 농축액 첨가 양갱의 품질특성 및 항산화성 (Quality Characteristics and Antioxidant Activities of Yanggeng with Added Rehmanniae radix Preparata Concentrate)

  • This study was performed to evaluate the quality characteristics of yanggeng prepared with different amounts of Rehmanniae radix Preparata (RRP) concentrate. Moisture, acidity, soluble solids, and reducing sugar contents of the control were the lowest, followed in order by the 3%, 6%, and 9% RRP addition. The pH of 9% RRP added yanggeng was the lowest, followed in order by the 6% and 3% additions. The lightness value of the Hunter color system decreased based on the amount of RRP concentrate added to yanggeng. In the texture analysis, the hardness and cohesiveness of the 9% RRP added yanggeng were the highest among the treatments. Total phenol content was the highest in 9% RRP added yanggeng. Antioxidant activities such as DPPH radical scavenging activity and hydroxyl radical scavenging activity, were the highest in the 9% RRP added yanggeng. In the sensory evaluation for yanggeng, the sensory scores for overall preference and texture were the highest in the 6% RRP added yanggeng The result was suggested that the optimum amount of RRP concentrate to add to yanggeng was 6%.

생지황(生地黃) 30% ethanol 추출물의 급성위염 및 위궤양 억제 효과 (Inhibitory Activities of Rehmanniae Radix 30% Ethanol Extract on Acute Gastritis and Peptic Ulcers)

  • Objective : This study was designed to evaluate the protective effect of Rehmanniae Radix Crudus (RC) in 150 mM HCl/ethanol induced acute gastritis mice. Methods : ICR mice were divided into 5 groups (normal group, control group, 10 mg/kg sucralfate treated group, 50 mg/kg RC treated group, 100 mg/kg RC treated group, n=8). Normal group was not take any treatment. Control group induced gastritis 1 hour after ingestion of distilled water. 10 mg/kg sucralfate induced group was induced gastritis 1 hour after ingestion of distilled of sucralfate 10 mg/kg. 50 mg/kg and 100 mg/kg RC treated groups were induced gastritis 1 hour after ingestion of distilled of RC 50 mg/kg and 100 mg/kg. After 1 hour of gastritis induction, removed the stomach tissue. We examined histological observations, oxidative stress biomarkers, antioxidant proteins, inflammatory mediators and cytokines. Results : In this study, the RC treatment group showed gastritis and gastric ulcer inhibition, and the area of injury decreased. The oxidative stress biomarkers such as reactive oxygen species (ROS) and peroxy nitrite ($ONOO^-$) in the serum were reduced in the RC treated group. Inaddition, antioxidant proteins (nuclear factor erythroid 2-related factor 2, Heme oxygenase 1) were increased in RC treated group, and the expression of inflammatory mediators and cytokines induced by nuclear factor-kappa B activation was inhibited. Conclusion : According to the results, RC may have an excellent inhibitory effect on acute gastritis and gastric ulcer.

숙지황(熟地黃)이 남성생식세포 GC-1의 항산화에 미치는 영향 (Antioxidant Effects of Rehmanniae Radix Preparat in GC-1 Cells)

  • Objectives : The purpose of this study is to examine the antioxidant activity in the germ cells of the extract of Rehmanniae Radix Preparat (RR). Methods : The extract was studied for diphenyl-picryl-hydrazyl (DPPH) radical scavenging activity, GC-1 cell viability by a modified MTT assay, the effects on H2O2-induced cytotoxicity by MTT assay and lipid peroixidation by malondialdehyde (MDA) formation, respectively. Results : The results showed that the extract scavenged DPPH radical in a dose-dependent manner by up to 43.1%. The extract at concentrations of 100 ${\mu}g/ml$ showed peak level of 136.5% in growth of GC-1 cell. The hydrogen peroxide-induced cytotoxicity was blocked by the extract at concentrations of 10, 50 and 100 ${\mu}g/ml$. The extract (50 and 100 ${\mu}g/ml$) also displayed a dose-dependent reduction of MDA formation on hydrogen peroxide-induced lipid peroxidation. Conclusions : In conclusion, the extract of Rehmanniae Radix Preparat has strong antioxidant activity.

생지황(Rehmanniae Radix)과 배의 Phenolic Compound가 Streptozotocin으로 유발된 고혈당 생쥐에 미치는 영향 (Effect of Rehmanniae Radix and Pear Phenolic Compound on the STZ-Treated Mice for Induction of Diabetes)

  • 선행연구에서 배의 과피에서 추출한 phenolic compound(PC)의 항당뇨 효능이 입증되었기에, 본 연구에서는 생지황 추출물과 배의 과피에서 추출한 PC가 streptozotocin(STZ)로 유발된 고혈당 생쥐에 미치는 영향을 규명하고자 하였다. 0.2 mL의 생지황 추출물을 사료에 혼합하여 공급한 실험군(S군), 0.2 mL 생지황과 PC(90 mg/kg/day)를 혼합하여 공급한 실험군(SPC군) 및 사료만 공급한 대조군으로 구분하여 6주 동안 실시하였다. 체중 계측은 매주 1회, 혈당검사는 꼬리정맥 혈을 취하여 매주 1회씩, 당내성 검사는 실험이 완료되기 24시간 전에 각각 실시하였다. 6주 후 BUN과 creatinine을 검사하기 위하여 심장채혈을 하였으며, 관류고정 후 췌장을 적출하여 $\beta$-세포에 대한 insulin-면역조직화학적 검사를 실시하였다. 체중은 S군이 가장 높았으며, 대조군에 비하여 약 2 g 정도 많았다. 혈당은 S군과 SPC군에서 대조군에 비하여 유의성있게 (p<0.05) 감소하였으며, BUN과 creatinine농도는 SPC군에서 대조군에 비하여 유의성있게 (p<0.05) 감소하였다. 당내성 검사결과 S군과 SPC군은 대조군에 비하여 개선된 결과를 보여주었다. 랑겔한스섬의 $\beta$-세포에 대한 insulin-면역조직 화학적 검사 결과 insulin 양성 반응을 보이는 랑겔한스섬들의 수가 S군(p< 0.01)과 SPC군(p<0.05)에서 대조군에 비하여 유의성있게 증가하였으며, 면역반응성은 SPC군에서 가장 높게 나타났다. 이상의 결과로 보아 생지황의 추출물과 배의 과피에서 추출한 PC가 STZ로 유발된 고혈당 생쥐에서 항당뇨 효능이 있다고 사료되며, SPC군이 S군에 비하여 신장의 기능이 보다 양호하게 나타나는 것으로 보아 배의 과피에서 추출한 PC가 당뇨병신증으로부터 보호하는 작용이 있는 것으로 사료되나 기전에 대해서 앞으로 연구가 필요하다고 생각된다.

식·약공용 농·임산물의 다환방향족탄화수소 오염도 조사 및 위해도 결정 (Contamination Investigation and Risk Characterization on the Polycyclic Aromatic Hydrocarbon of Agricultural Products Used for Food and Medicine)

  • Polycyclic aromatic hydrocarbons(PAHs) contents were analyzed by measuring benz(a)anthracene(BaA), chrysene(Chr), benzo(b)fluoranthene(BbF) and benzo(a)pyrene(BaP), and the related risk characterization was conducted for 113 samples out of 14 different agricultural products used for food and medicine. Detection rate of PAHs was 90.3% as a whole, and the highest one was 80.5% for BaP. The detection rate of BaP exceeding the maximum permitted concentration of Rehmanniae Radix Preparata and Rehmanniae Radix, 5.0 ㎍/kg was 1.8%, and the detection rates of BaA, Chr and BbF were within the range of 2.7~10.6%. The highest average concentration of BaA was 3.41 ㎍/kg detected from Lycii Fructus, while those of Chr, BbF, BaP and PAH4(sum of detected BaA, Chr, BbF and BaP) were 5.00, 1.79, 2.36, 12.36 ㎍/kg, respectively, detected from Rehmanniae Radix Preparata. As for the risk characterization on PAHs, the overall MOE(Margin of Exposure) values were measured within the range of 105~107, which is unlikely to cause direct health concerns, but the worring values of MOE were measured 6.57×104 for BaP and 6.10×104 for PAH4 from Rehmanniae Radix Preparata, which may require an improvement plan to reduce BaP contents.

한약재내 중금속 함량분석 및 물세척 효과 (Determination of Heavy Metal Contents in Oriental Medical Materials and the Effect of Washing)

  • 쌍화탕의 재료로 사용되는 9가지 한약재에 함유된 6가지 인체에 유해한 중금속 함유량은 생강과 당귀의 경우 식약청 기준치인 30ppm의 1.47배와 1.24배 이었고 기타 한약재는 기준치와 거의 동등하거나5% 이내에서 초과하였다. 가장 간단한 수치법인 물세 척 만으로 중금속이 20-38% 제거되었고 따라서 모든 실험 대상 약재 에 대하여 중금속 함량을 식약청 기준치보다 낮출 수 있었다.

사물탕(四物湯)과 사물탕(四物湯) 구성약물(構成藥物)이 혈구감소증(血球減少症)에 미치는 영향(影響)에 관(關)한 연구(硏究) (A study on Effects of Samultang and Samultang Composition on Lucopenia, Thrombocytopenia and Anemia)

  • In order to inverstigate interactive effects of Samultang(SMT) and SMT composition and the principle of prescription, I examined the effects of SMT and combination of three components of SMT, In C57BL/6 mouse with oral administration of SMT and cyclophosphamide(CTX) treatment, white blood cells, platelet were counted. Also, those were anti-anemia effect for rat which have hemolytic anemia induced by phenylhydrazine. The results were summerized as follows : 1. In vivo analysis of the effect of the SMT by orally administrated C57BL/6 mice with the SMT and was treated with cyclophosphamide (CTX). When the number of white blood cell. platelet was counted, there was ameliorative effects of leucopenia or thrombocytopenia as a protection to CTX. 2. It was acknowledged that SMT and its composition increased RBC count and hemlytic significantly in rat which have hemolytic anemia. 3. Among SMT composition Angelicae gigantis Radix and Cnidii Rhizoma solution and among the combination group Rehmanniae Radix and Cnidii Rhizoma combination and SMT solution showed significant effects for hemoglobin and weight of spleen in mouse which have hemolytic anemia. Thus it was acknowledged that Rehmanniae Radix and Cnidii Rhizoma combination had effect to improve hemoglobin and weight of spleen. From above these results it was acknowledged that SMT has effects that ameliorative effects of leucopenia or thrombocytopenia as a protection to CTX and anti-anemic effect for the rat which have hemolytic anemia induced by phenylhydrazine and these effects depend upon its combination of SMT composition. Especially it was acknowleged, though there are the slight difference according to combination, that Paeoniae Radix Alba combination had improving effect for physical change. So it is though that Paeoniae Radix Alba is main material for this kind of disease and continuous study is needed.

생맥산가미방 추출물이 멜라닌 생합성 저해 효과와 SOD 활성에 미치는 연구 (Study of ShengmaisanJiaweifang Extracts on the Inhibitory Effects of Melanin Synthesis and Superoxide Dismutase Activity)

  • This study aims to evaluate the effects of Shengmaisan (SMS) and three types of ShengmaisanJiaweifang on the inhibitory effect of melanin synthesis in B16F10 cells, the mechanism of action through tyrosinase, and the antioxidant effect through superoxide dismutase (SOD) activity. In this study, we used ShengmaisanJiaweifangs (SMS, SMSRR, SMSAD, SMSAR) to research the whitening effects in B16F10 cell lines. Shengmaisan (SMS) was a herbal medicine composed of Ginseng Radix, Liriopis Tuber, and Schisandrae Fructus. ShengmaisanJiaweifangs included SMSRR (SMS added with Rehmanniae Radix), SMSAD (SMS added with Asparagi Radix) and SMSAR (SMS added with Astragali Radix). We measured the cell viability, the inhibition rate of the melanin biosynthesis, and the activity of tyrosinase and SOD in malignant melanoma, B16F10 cells, to survey the whitening effect and the mechanism of the impact on the sample. As a result, SMSRR significantly suppressed the cell viability of B16F10 at more than $500{\mu}g/m{\ell}$ and significantly inhibited the generation of melanin induced by ${\alpha}$-MSH at more than $250{\mu}g/m{\ell}$. SMSRR ($500{\mu}g/m{\ell}$) decreased the activity of tyrosinase while increased the activity of SOD. Therefore, we considered that the SMSRR would be able to produce high value-added products more SMS if used as a commercial.

팔물탕이 복강 마크로파지의 탐식능에 미치는 영향 (Effect of Palmultang on the Phagocytosis of Murine Peritoneal Macrophage)

  • Palmultang(PMT) consists of Ginseng Radix Alba, Atractylodis Rhizoma Alba, Hoelen, Glycyrrhizae Radix, Rehmanniae Radix Preparata, Paeoniae Radix, Cnidii Rhizoma and Angelicae Gigantis Radix. PMT enhanced the lucigenin chemiluminescence and the engulfment of fluorescein-conjugated E. coli particles and inhibited the production of nitric oxide in murine peritoneal macrophage. PMT enhanced the production of ${\gamma}-interferon$, interleukin-2 and the cell viability in murine thymocyte, but did not affect the production of interleukin-4. These results indicate that PMT enhances the phagocytosis of macrophage via the stimulation of ${\gamma}-interferon$ production in $T_H1$ cells and the reduction of nitric oxide production in peritoneal macrophage.
